Melatonin as a hub signal integrating redox and hormonal crosstalk with multi-omics regulation in plant heavy metal stress tolerance.

Heavy metal contamination is a major environmental constraint that negatively affects plant growth, metabolism, and agricultural productivity. Excess metals such as cadmium, lead, and copper disturb cellular functions mainly by inducing oxidative stress, disrupting nutrient balance, and causing toxicity at multiple levels of organization. To cope with these stresses, plants activate complex defense systems, among which melatonin (MT) (N-acetyl-5-methoxytryptamine) has recently emerged as a key regulatory molecule. This review highlights MT's central role in coordinating plant responses to heav

Integrated Metabolomic and Transcriptomic Profiling Reveals a Distinct Pathological Aging State in Diminished Ovarian Reserve of Advanced Reproductive-Age Women.

Women of advanced age exhibit significant heterogeneity in ovarian reserve, categorized as normal (NOR) or diminished (DOR). This study aims to distinguish physiological age-related decline from pathological accelerated aging in DOR, and to explore underlying molecular mechanisms for optimizing assisted reproductive strategies. In vitro fertilization-embryo transfer (IVF-ET) outcomes were retrospectively compared in advanced-age women (≥ 40 years) with NOR or DOR treated at our center (January 2022 - December 2024). Simultaneously, follicular fluid (FF) was collected from both groups (n = 20).

Lysolecithin reprogramming via LPCAT1 modulation restores endothelial function and prevents diabetes-associated dysmetabolism.

BACKGROUND: Dysregulation of lysophosphatidylcholines (LPCs) and phosphatidylcholines (PCs) is linked to endothelial dysfunction and impaired tissue repair. Nevertheless, the organ-specific modulation of lysolecithin remodeling in T2DM remains unexplored. Here, we investigate the LPC/PC remodeling dynamics in a T2DM model and propose a novel therapeutic approach using an orally bioavailable peptide (SP6) derived from Spirulina platensis. METHODS: LPC/PC levels were analyzed by UHPLC-HRMS. Membrane fluidity, VEGF/API5, LPCAT1, VE-cadherin, and GLUT1 were evaluated by merocyanine assay, qPCR, i

Mitochondrial Dysfunction in Renal Aging: A Vicious Cycle Driving Kidney Disease Progression.

SIGNIFICANCE: The aging of the global population is linked to an increase in age-related diseases. The kidneys undergo both structural and functional declines with age, and aging is a significant risk factor for kidney diseases. Mitochondrial dysfunction is recognized as a crucial factor affecting kidney aging. Although the importance of disrupted mitochondrial homeostasis in renal aging has gained increasing attention, the associations and causal mechanisms have not been systematically summarized. RECENT ADVANCES: Mitochondria are highly dynamic organelles that operate in various functions,

TNF-NF-κB signaling mediates immune-biomineralization crosstalk during shell repair under ocean acidification in Mytilus edulis.

Ocean acidification (OA) impairs biomineralization in bivalves, but its effects on immune-biomineralization crosstalk during shell repair remain unknown. Here, we exposed adult Mytilus edulis bearing standardized shell perforations to three pH levels (8.1, 7.9, and 7.7) for up to 40 days. OA slowed early repair and caused microstructural disorganization and an approximately 87% reduction of compressive strength at pH 7.7, yet the damaged area appeared largely closed by day 15, suggesting a decoupling between morphological closure and functional recovery. In addition, transcriptomic profiling o

Single-nucleus RNA sequencing uncovers cell type-specific alterations in OSA-related liver injury.

Chronic intermittent hypoxia (CIH), a hallmark feature of obstructive sleep apnea (OSA), is strongly implicated in the development of hepatic injury. However, the cellular mechanisms underlying CIH-induced liver dysfunction remain poorly understood. This study aimed to systematically characterize hepatic cellular responses to CIH at single-nucleus resolution. A rodent model of CIH was established and subjected to histological and transcriptomic analyses. Single-nucleus RNA sequencing (snRNA-seq) was performed to delineate transcriptional alterations across liver cell populations. Differentiall

Coordinated Cell-Wall and Starch Maturation Is Associated with Winter-Harvest Quality in Sparganium stoloniferum Tubers.

Sparganium stoloniferum tubers (SL), known medicinally as Sparganii Rhizoma, are commonly considered superior at the winter-harvest stage, when they show the traditional quality traits of heavy weight and firm texture. However, the developmental basis of this quality phenotype remains insufficiently understood. This study aimed to determine how tissue organization, cell-wall architecture, starch deposition, and related transcriptional patterns are associated with winter-harvest quality in SL. By comparing SL at different developmental stages, we found that maturation was accompanied by reduced

Nitric oxide and phytohormones molecular crosstalk regulating fruit quality and postharvest management.

Nitric oxide (NO) has emerged as a pivotal signaling molecule regulating postharvest fruit physiology, functioning far beyond its early recognition as a simple reactive nitrogen species. Increasing evidence highlights NO as a central signaling hub that integrates redox homeostasis, hormone signaling, transcriptional control, and post-translational regulation to modulate fruit ripening, senescence, and stress responses, and enhance disease resistance during postharvest storage. At the molecular level, NO participates in complex crosstalk with reactive oxygen species, reactive sulfur species, an

The endometriosis-adenomyosis spectrum: shared pathophysiology and microenvironment-driven disease divergence.

Endometriosis and adenomyosis are common gynecologic disorders associated with dysmenorrhea, chronic pelvic pain, and infertility. Although they share several molecular features, the mechanisms by which endometrium-derived tissues develop distinct pathological phenotypes in different tissue environments remain incompletely understood. This review summarizes shared and divergent pathogenic mechanisms, focusing on lesion-specific microenvironments. This narrative review was based on a PubMed literature search from the year of the first publication through December 2025 using terms related to end

A Redox-Lipid Transcriptional State Is Associated with High Aflatoxin Biosynthetic Activity in Aspergillus flavus.

Background/Objectives: Aflatoxin B1 (AFB1) contamination of maize and peanut is influenced by fungal responses to environmental and food-matrix cues, but the transcriptional coordination of redox adaptation, lipid metabolism, and aflatoxin biosynthesis remains unresolved. This study examined whether an expression-defined redox-lipid state is associated with high relative aflatoxin biosynthetic activity in Aspergillus flavus. Methods: A cross-dataset secondary analysis integrated 18 analytical datasets (247 samples) derived from eight public source records spanning defined-medium, maize, and pe

Comparison of Liquid Chromatography- and Nano-Electrospray Ionization-Mass Spectrometry Approaches for Single-Cell Metabolomics.

Live single-cell metabolomics is a rapidly growing area of research, which offers the potential to provide unique insights into cellular function and heterogeneity. Single-cell isolation approaches based on capillary sampling are in principle compatible with either nano-electrospray ionization-mass spectrometry (nano-ESI-MS), where the cell is lysed and sprayed directly into a mass spectrometer, or liquid chromatography-mass spectrometry (LC-MS) for metabolomics analysis. However, there are no data indicating which approach can provide the best performance (metabolite coverage, reproducibility

Integrated transcriptome-metabolome analysis reveals a GmMYB093-GmCYP90A1-BR module supporting soybean adaptation to iron deficiency.

Iron deficiency is a major abiotic constraint that limits soybean growth, nodulation, symbiotic nitrogen fixation, and yield, yet objective criteria for evaluating low-Fe tolerance and the regulatory mechanisms linking root-nodule responses with shoot adaptation remain insufficiently defined. Here, we established an entropy-weight-based evaluation system using 62 soybean accessions and identified Wanhuang506 (Wh506) as a highly tolerant cultivar and Flyer as a highly sensitive cultivar. Physiological validation showed that Wh506 maintained higher Fe accumulation, chlorophyll retention, antioxi

The root-knot nematode effector Mi1D08B targets a NINJA-family co-repressor to suppress jasmonate production and promote infection in soybean.

Plant-parasitic nematodes deploy secreted effector proteins that reprogram host cellular processes to establish parasitism. Here, we characterized Mi1D08B, a putative dorsal-gland effector from the root-knot nematode Meloidogyne incognita and defined its role in subverting soybean jasmonate-mediated immunity. Mi1D08B is conserved across several Meloidogyne species and localizes to the plant nucleus and cytoplasm. Overexpression of Mi1D08B significantly increased galling and egg production, demonstrating a strong virulence role. A gall-specific yeast two-hybrid screen identified the NINJA-famil

Molecular ontogeny of Bemisia tabaci life stages reveals miRNA-governed development and egg-stalk support for early embryogenesis.

BACKGROUND: Deciphering how developmental gene-regulatory programs interface with lineage-specific reproductive novelties in Bemisia tabaci demands a temporally resolved, integrative molecular framework. Using deep miRNA sequencing, high-throughput transcriptomics, quantitative proteomics, two-dimensional proteo-mapping, and spatially multiplexed FISH, we delineate the circuitry governing whitefly ontogeny. RESULTS: This atlas uncovers a previously unrecognized morphological innovation: a transporter-enriched egg stalk operating as an active, metabolically competent nutrient-harvesting append

Neuroimmune Cross-Talk and Multilevel Cascades in Fentanyl Toxicity: Interplay of Hypoxic Stress, Glial Activation, and Synaptic Dysregulation in Systems-Level Neurodegeneration.

Fentanyl, an ultra-potent synthetic opioid, has traditionally been characterized by its acute toxic effects, particularly respiratory depression. However, accumulating research indicates that its neurobiological influence extends far beyond its short pharmacological window, intersecting with several core mechanisms implicated in major neurodegenerative disorders. This review integrates multiscale evidence to propose a unified conceptual framework in which fentanyl may function not only as an acute neurotoxin but also as a putative accelerator of long-term neurodegenerative vulnerability. Differential peripheral immune dynamics underlie therapeutic response to chemotherapy and chemoimmunotherapy in triple-negative breast cancer.

Triple-negative breast cancer (TNBC) is an aggressive breast cancer subtype with limited treatment options and response to immune checkpoint inhibitors. Tumor-infiltrating lymphocytes have been extensively studied; however, the integration of peripheral immune dynamics with mechanistic regulation underlying therapeutic response remain poorly defined. Here, we integrate immune-state modeling with pathway-level mechanistic inference to analyze single-cell RNA sequencing of PBMCs from patients with advanced TNBC treated with paclitaxel alone or in combination with the anti-PD-L1 Ab atezolizumab.

Immune suppression in MTAP-deficient cancers via glutamate metabolism and CXCL10 downregulation.

BACKGROUND: Immune checkpoint inhibitors (ICIs) have transformed cancer therapy; however, their efficacy remains limited in certain tumor subtypes, including those deficient in methylthioadenosine phosphorylase (MTAP). MTAP-deficient cancers are characterized by immunosuppressive tumor microenvironments (TMEs) and poor T cell infiltration, as suggested by large-scale transcriptomic analyses. Yet, the underlying mechanisms and therapeutic vulnerabilities remain poorly defined. METHODS: We employed murine tumor models and transcriptomic profiling to investigate the immunosuppressive features of
