Human Norovirus: Experimental Models of Infection.

Human noroviruses (HuNoVs) are a leading cause of acute gastroenteritis worldwide. HuNoV infections lead to substantial societal and economic burdens. There are currently no licensed vaccines or therapeutics for the prevention or treatment of HuNoVs. A lack of well-characterized in vitro and in vivo infection models has limited the development of HuNoV countermeasures. Experimental infection of human volunteers and the use of related viruses such as murine NoV have provided helpful insights into HuNoV biology and vaccine and therapeutic development. There remains a need for robust animal model

New approach methodologies in nanomedicine: bridging the gap between in silico, in vitro, and in vivo systems.

The rise of nanomedicine has led to the regulatory approval of more than 80 nanoparticle (NP)-based therapeutics over the past two decades. Further advancement in the field requires a deeper understanding of the nano-bio interface of NPs, including their uptake, delivery, efficacy, toxicity, and adverse effects. Despite the heavy reliance on conventional cell cultures for NP screening, these models do not recapitulate the in vivo microenvironment, complexity, and physiological responses; hence, follow-up animal studies are required. Nonetheless, because of species-specific differences and ethi

Genetic and environmental mouse models of autism reproduce the spectrum of the disease.

Genetic and environmental factors increase autism spectrum disorder (ASD) incidence, and this has led to the generation of corresponding animal models, with some showing strong construct and face validity. This short review focuses on results we have recently obtained with environmental and genetic mouse models of ASD and that are the valproic acid, the poly I:C and the Shank 3 models. This has allowed us to provide a comparative description of these widely used animal models providing an interesting perspective as to the pros and cons of each one of them, in our experimental settings. The antipsychotic drug, fluphenazine, effectively reverses mechanical allodynia in rat models of neuropathic pain.

RATIONALE: Fluphenazine is a potent antipsychotic drug used to treat schizophrenia and other psychotic symptoms. Its clinical benefit is mainly mediated by the antagonism of dopamine D2 receptors. We have recently discovered, however, that fluphenazine is also a potent sodium channel blocker, a property that may offer additional therapeutical indications, including analgesia. OBJECTIVES: The present study sought to determine the analgesic effect of fluphenazine on neuropathic pain in animal models. METHODS: The effect of fluphenazine on mechanical allodynia was assessed in three animal neuro

Large-scale gene expression changes in APP/PSEN1 and GFAP mutation models exhibit high congruence with Alzheimer's disease.

Alzheimer's disease (AD) is a complex neurodegenerative disorder with both genetic and non-genetic causes. Animal research models are available for a multitude of diseases and conditions affecting the central nervous system (CNS), and large-scale CNS gene expression data exist for many of these. Although there are several models specifically for AD, each recapitulates different aspects of the human disease. In this study we evaluate over 500 animal models to identify those with CNS gene expression patterns matching human AD datasets. Approaches included a hypergeometric based scoring system th

Computational approaches for skin sensitization prediction.

Drugs, cosmetics, preservatives, fragrances, pesticides, metals, and other chemicals can cause skin sensitization. The ability to predict the skin sensitization potential and potency of substances is therefore of enormous importance to a host of different industries, to customers' and workers' safety. Animal experiments have been the preferred testing method for most risk assessment and regulatory purposes but considerable efforts to replace them with non-animal models and in silico models are ongoing. This review provides a comprehensive overview of the computational approaches and models tha

Efficacy of phage therapy in preclinical models of bacterial infection: a systematic review and meta-analysis.

BACKGROUND: Antimicrobial resistance of bacterial pathogens is an increasing clinical problem and alternative approaches to antibiotic chemotherapy are needed. One of these approaches is the use of lytic bacterial viruses known as phage therapy. We aimed to assess the efficacy of phage therapy in preclinical animal models of bacterial infection. METHODS: In this systematic review and meta-analysis, MEDLINE/Ovid, Embase/Ovid, CINAHL/EbscoHOST, Web of Science/Wiley, Cochrane Central Register of Controlled Trials, Cochrane Database of Systematic Reviews, and Google Scholar were searched from inc

Fetal Cleft Lip/Palate Surgery: End of a Dream?

Recognition that a fetus can scarlessly heal in intrauterine life led to various animal studies in the mid 1980s exploring the possibility of fetal cleft lip/palate surgery. The idea of scarless cleft repair seemed like a possible dream after the promising results from the early animal studies. In this review, we analyze the progress made in the 30 years since our first experience with animal models.

Evaluation of the clinical relevance and limitations of current pre-clinical models of peripheral artery disease.

Peripheral arterial disease (PAD) usually results from atherosclerosis and associated thrombosis and limits blood supply to the lower limbs. Common presenting symptoms include intermittent claudication (IC), rest pain and tissue loss. When limb viability is threatened, known as critical limb ischaemia (CLI), surgical and endovascular interventions are frequently undertaken; however, these are not always successful and ultimately major amputation may be required. There is significant interest in developing new therapeutic approaches to manage PAD which can be applied to patients unlikely to ben

In Vivo Models to Study the Pathogenesis of Extra-Respiratory Complications of Influenza A Virus Infection.

Animal models are an inimitable method to study the systemic pathogenesis of virus-induced disease. Extra-respiratory complications of influenza A virus infections are not extensively studied even though they are often associated with severe disease and mortality. Here we review and recommend mammalian animal models that can be used to study extra-respiratory complications of the central nervous system and cardiovascular system as well as involvement of the eye, placenta, fetus, lacteal gland, liver, pancreas, intestinal tract, and lymphoid tissues during influenza A virus infections.

Models and mechanisms of vascular dementia.

Vascular dementia (VaD) is the second leading form of dementia after Alzheimer's disease (AD) plaguing the elderly population. VaD is a progressive disease caused by reduced blood flow to the brain, and it affects cognitive abilities especially executive functioning. VaD is poorly understood and lacks suitable animal models, which constrain the progress on understanding the basis of the disease and developing treatments. This review article discusses VaD, its risk factors, induced cognitive disability, various animal (rodent) models of VaD, pathology, and mechanisms of VaD and treatment option

Cortical brain stimulation: a potential therapeutic agent for upper limb motor recovery following stroke.

There is emerging interest in the use of cortical electrical stimulation as a novel physical agent for the treatment of upper limb motor impairment after stroke. Evidence from animal models suggests that both motor learning and cortical stimulation alter intracortical inhibitory circuits and can facilitate long-term potentiation and cortical remodeling. This article reviews current research on the physiology and use of cortical stimulation animal models and in humans with stroke related hemiplegia. Both transcranial cortical stimulation techniques and targeted epidural cortical stimulation are

Measurement and evaluation of surgical models of spinal fusion in mice, rats, and rabbits.

This study aims to establish a theoretical foundation for selecting and constructing animal models in clinical research on spine-related diseases. By establishing surgical models in three experimental animal species-mice, rats, and rabbits-we compare their respective advantages and disadvantages. First, three experimental animals of each of the three species were selected in good health. Subsequently, the experimental animals were dissected and several indices of the designated intervertebral discs and the height of the adjacent vertebrae were measured. At the same time, the ease of the surgic

Profiling blood-based neural biomarkers and cytokines in experimental autoimmune encephalomyelitis model of multiple sclerosis using single-molecule array technology.

Experimental autoimmune encephalomyelitis (EAE) is a preclinical animal model widely used to study multiple sclerosis (MS). Blood-based cytokines and neural biomarkers are predictors of neurodegeneration, disease activity, and disability in patients with MS. However, understudied confounding factors cause variation in reports on EAE across animal strains/studies, limiting the utility of these biomarkers for predicting disease activity. In this study, we investigated blood-based analyte profiles, including neural markers (NFL and GFAP) and cytokines (IL-6, IL-17, IL-12p70, IL-10, and TNF-α), in

Intermittent hypoxia and respiratory recovery in pre-clinical rodent models of incomplete cervical spinal cord injury.

Impaired respiratory function is a common and devastating consequence of cervical spinal cord injury. Accordingly, the development of safe and effective treatments to restore breathing function is critical. Acute intermittent hypoxia has emerged as a promising therapeutic strategy to treat respiratory insufficiency in individuals with spinal cord injury. Since the original report by Bach and Mitchell (1996) concerning long-term facilitation of phrenic motor output elicited by brief, episodic exposure to reduced oxygen, a series of studies in animal models have led to the realization that acute

NLRP3 inflammasome priming and activation is involved in depression: A systematic review and meta-analysis.

Depression is characterised by a low mood, loss of interest or pleasure, pessimism, impaired concentration, decreased energy, and fatigue. A strong relationship between inflammatory processes and the pathophysiology of depression had been identified. Studies have suggested that the NLRP3 inflammasome is a key contributor to the pathogenesis of depression. This systematic review aimed to synthesise evidence from animal models of depression to evaluate both priming and activation of the inflammasome, with a specific focus on clarifying the role in depressive pathology. Comparison of industrial- and medical-grade silicone phantoms for surgical laser training in dental students.

OBJECTIVE: This study explored the potential of industrial- and medical-grade silicone phantoms as alternatives to animal models for surgical laser training in dental students. METHODS: Tongue molds with exophytic lesions were created using 3D-printed polylactic acid molds. Industrial-grade and medical-grade silicone were injected into the molds to create two distinct educational models. Forty-five sixth-year dental students participated in the study. Students first practiced with an 810-nm diode laser on a sheep tongue and then resected the lesions on the silicone models. Afterwards, they com

Fetal Surgery for Gastroschisis-A Review with Emphasis on Minimally Invasive Procedures.

(1) Background: The morbidity of gastroschisis is defined by exposure of unprotected intestines to the amniotic fluid leading to inflammatory damage and consecutive intestinal dysmotility, the viscero-abdominal disproportion which results in an abdomen too small to incorporate the herniated and often swollen intestine, and by associated pathologies, such as in complex gastroschisis. To prevent intestinal damage and to provide for growth of the abdominal cavity, fetal interventions such as amnio exchange, gastroschisis repair or covering have been evaluated in several animal models and human tr
