Combination therapy is rapidly becoming the cornerstone of hepatocellular carcinoma (HCC) treatment. Immune checkpoint inhibitors (ICIs) have emerged as a central strategy in systemic therapy, yet their efficacy as monotherapies remains limited. Consequently, combinatorial approaches, such as ICIs-Tyrosine kinase inhibitors (TKIs), ICIs-chemotherapy, and dual ICI regimens, are gaining momentum. While clinical trials have established efficacy benchmarks, mechanistic insights remain scarce, partly due to the limitations of current preclinical models in mimicking the complex tumor microenvironmen

Lipid nanoparticles (LNPs) are pivotal for mRNA delivery, yet predominant hepatic accumulation limits their therapeutic application in immunologically active sites like the spleen. Engineering LNPs for splenic mRNA delivery is therefore essential for advancing mRNA-based immunotherapies. Here, we rationally designed a series of LNPs by modulating the alkyl chain length of PEGylated lipids (fast-shedding C14 DMG-PEG vs. slow-shedding C18 DSG-PEG) and their molar ratios (0.75% and 1.5%). This systematic variation precisely controlled nanoparticle physicochemical properties and protein corona com

BACKGROUND: In our previous study, we demonstrated the colitis-alleviating effects of Centella asiatica hot-water extract (CA-HE) in an inflammatory bowel disease mouse model. In this study, we aimed to demonstrate its potential to activate intestinal immunity via Peyer's patch (PP)-mediated mechanisms and serve as a functional agent for maintaining intestinal immune homeostasis.
RESULTS: Four-week oral administration of CA-HE induced no toxicological effects and significantly upregulated hematopoietic (interleukin-4, -6, and granulocyte-macrophage colony-stimulating factor) and immunoglobuli

In clinical practice, for patients with autoimmune hepatitis (AIH), appropriate therapeutic measures focus on addressing the progression of liver fibrosis (LF) and promptly assessing its severity. This study aims to identify and characterize Periplaneta americana glycoproteins (PAG) and explore their potential mechanisms of action in the treatment of immunological liver fibrosis (ILF) by establishing an in vivo ILF model. PAG was confirmed to be a glycoprotein through Periodic acid-Schiff and Coomassie brilliant blue staining, with protein and total sugar contents of 68.35 ± 0.01 % and 17.66 ±

Pseudomonas aeruginosa is an environmentally resilient bacterium and an important cause of both acute and chronic infections in people with impaired natural barriers or immunological defences. Chronic respiratory infection with P. aeruginosa is a major cause of morbidity and mortality in people with airway diseases, including cystic fibrosis (CF) and non-CF bronchiectasis. Chronic airway infection is characterized by periods of relative stability punctuated by pulmonary exacerbations, during which times rapid bacterial outgrowth necessitates intense antimicrobial chemotherapy. The periods of s

Opioids remain a mainstay for managing moderate-to-severe cancer pain, yet accumulating evidence suggests that opioid analgesics can impair antitumor immunity. In their recent study, McIlvried et al propose a mechanistic framework for how morphine administration compromises response to anti-programmed-death-1 immunotherapy in a syngeneic mouse model of oral squamous cell carcinoma. They identify mu-opioid receptor signaling on CD8+ T cells as the main conduit for morphine-induced immunosuppression, observing reduced T-cell infiltration, enhanced expression of inhibitory receptors, and diminish

Tuberculosis (TB) relapse after appropriate drug treatment is poorly understood but critical to developing shorter treatment regimens. Using a cynomolgus macaque model of human TB, macaques with active TB disease were treated with a short course of isoniazid and rifampin and subsequently infected with SIV. Serial clinical, microbiologic, immunologic and position emission and computed tomography (PET CT) assessments were performed to identify risk factors of relapse. Of the 12 animals, eight developed radiologically defined relapse including four that had clinical and/or microbiologic signs. Gr

The pathophysiological relationship between wound healing impairment and diabetes is an intricate process. Burn injury among diabetes patients leads to neurological, vascular, and immunological abnormalities along with impaired activities of cell proliferation, collagen production, growth factors, and cytokine activities with huge bacterial infestation. In our study, we aimed to achieve a burn wound dressing material with the help of electrospun Chitosan/Polyethylene oxide/Rosmarinic acid (CS/PEO/RA) nanofibers. Chitosan is known for its biocompatibility and anti-bacterial properties; however,

Tissue exposure to implanted biomaterials triggers a foreign body response (FBR), which is a stepwise immunological process involving innate immune cells and tissue repair cells. Although the regulatory T (Treg) cells play a crucial role in inflammation and tissue repair, their function in the process of FBR has not been well investigated. In this study, as titanium (Ti) exhibits better biocompatibility and induces milder FBR than polymethyl methacrylate (PMMA), we analyzed the characteristics of Treg cells during FBR caused by the two types of biomaterials. In a rat femur implantation model,

Miscarriage and preterm birth (PB) remain major challenges in obstetric care and are often associated with excessive inflammation at the feto-maternal interface. Although the role of progesterone (P4) in maintaining pregnancy is well known, its anti-inflammatory effects in immune-mediated pregnancy complications remain poorly understood. In this study, we investigated the impact of prophylactic P4 administration on miscarriage using a mouse model induced by α-galactosylceramide (αGC), a potent activator of invariant natural killer T (iNKT) cells. Prophylactic, but not therapeutic, P4 administr

Systemic lupus erythematosus (SLE) is a complex autoimmune disease that poses serious long-term patient burdens. (1) Background: SLE patient classification and care are often complicated by case heterogeneity (diverse variations in symptoms and severity). Large language models (LLMs) and generative artificial intelligence (genAI) may mitigate this challenge by profiling medical records to assess key medical criteria. (2) Methods: To demonstrate genAI-based profiling, ACR (American College of Rheumatology) 1997 SLE classification criteria were used to define medically relevant LLM prompts. Reco

BACKGROUND: Mitochondrial dysfunction is a well-recognized pathological feature of Duchenne Muscular Dystrophy (DMD). The potential regulatory role of mitochondria-related genes (MRGs) in DMD remains to be further explored.
METHODS: GEO datasets and MRGs were used to analysis mitochondrial scores and evaluate patients' immunological characteristics. Weighted gene co-expression network analysis, differentially expressed genes (DEGs) and MRGs were used to identify hub genes. A specific hub gene was selected, and the effects of this gene overexpression on a horse serum (HS) treated C2C12 cell in

The therapeutic clotting factor VIII (FVIII) is known for its particular immunogenicity, with nearly 30% of hemophilic patients developing neutralizing antibodies against the infused protein. The root cause of this immunogenicity is still not well understood, but intrinsic factors, such as FVIII byproducts, have been linked to the immunological response elicited. Bioengineering of the FVIII molecule has been improving its recombinant (rhFVIII) production in many aspects, mainly enhancing its expression and stability. Assessment of immunogenicity for novel recombinant isoforms is crucial for fu

INTRODUCTION: Beyond the role of Bacillus Calmette-Guérin (BCG) for tuberculosis prevention, BCG has demonstrated heterologous protective effects. The global health crisis caused by the SARS-CoV-2 virus led to research on whether BCG-induced trained immunity could strengthen antiviral defenses. However, studies reported quite different results on its effect against COVID-19.
METHODS AND RESULTS: In this study, we evaluated the impact of pre-existing trained immunity induced by a BCG-derived Mycobacterium bovis strain (dpB), in both live and inactivated forms, in combination with SARS-CoV-2 va

Giardia duodenalis as a common zoonotic parasite is a public health problem infecting humans and a wide range of animals, including horses. This systematic review and meta-analysis was conducted to estimate the global pooled prevalence of Giardia infection in horses. Literature searches were conducted using databases including Scopus, ScienceDirect, PubMed, Web of Science and ProQuest to December 2023. A random-effect model was performed to calculate the pooled prevalence and corresponding 95% confidence interval (CI) according to the degree of heterogeneity in the included studies. Of 2661 ar

Background: Dyslipidemia and schistosomiasis are major public health challenges, particularly in endemic regions where their coexistence may influence host metabolism and immune responses. This study aimed to evaluate visceral adipose tissue (AT) remodeling in a murine model of acute Schistosoma mansoni infection combined with diet-induced dyslipidemia. Methodology: Female Swiss Webster mice were fed either a standard or high-fat diet (HFD) for 29 weeks and infected with S. mansoni at week 20. Nine weeks after infection, biochemical, morphometric, histopathological, and immunological analyses

Coccidioidomycosis, or Valley fever, is an emerging respiratory disease caused by soil-dwelling fungi of the Coccidioides genus that is expected to spread from the southwest into the central U.S. by 2050. While 60% of infections are asymptomatic, the other 40% of patients experience a range of symptoms, from self-limiting pneumonia to life-threatening disseminated disease. The immunological events that underlie the progression to severe disease remain underdefined. Here, we probed the early immune response to Coccidioides using a high dose of an attenuated strain of Coccidioides posadasii in a

Visceral leishmaniasis (VL), caused by Leishmania donovani, is a neglected tropical disease with limited therapeutic options and increasing drug resistance. This study investigates the immunological mechanisms and antiparasitic efficacy of imidazoquinoline-based Toll-like receptor 7/8 (TLR7/8) agonists as host-directed agents in an in vitro VL model. Using RAW 264.7 macrophages and L. donovani promastigotes and amastigotes, we examined macrophage activation, nitric oxide (NO) induction, and cell cycle disruption in parasites. The lead compounds (5 and 10) significantly enhanced NO production i

Polyomaviruses (PyVs) are widespread commensals among vertebrates, including humans, where they silently persist lifelong in healthy hosts. Polyomavirus infection in immunocompromised individuals can cause life-threatening diseases. Of the 14 human polyomaviruses discovered to date, resurgent infections by the JC and BK PyVs are responsible for high morbidity and mortality in individuals with certain inherited or acquired immune perturbations. JCPyV causes several brain disorders, the most fully characterized and of highest (albeit rare) incidence being Progressive Multifocal Leukoencephalopat
