Stereotactic ventralis intermedius (Vim) thalamotomy is effective for essential tremor (ET) of the limb, but the effect on the activity of the sensorimotor cortex is still unclear. The functional changes in this cortical area of patients with ET after Vim thalamotomy were investigated using functional magnetic resonance (fMR) imaging. Six patients underwent Vim thalamotomy for medically intractable ET, predominantly in the right hand. 1.5-tesla fMR imaging was performed using the blood oxygenation level-dependent sequence, before and after Vim thalamotomy, during passive movements with right w

Dynamic facial expressions of emotion constitute natural and powerful media of communication between individuals. However, little is known about the neural substrate underlying the processing of dynamic facial expressions of emotion. We depicted the brain areas by using fMRI with 22 right-handed healthy subjects. The facial expressions are dynamically morphed from neutral to fearful or happy expressions. Two types of control stimuli were presented: (i) static facial expressions, which provided sustained fearful or happy expressions, and (ii) dynamic mosaic images, which provided dynamic inform

One characteristic of focal dystonia is the sensory trick, by which sensory input to a certain area of the body can reduce abnormal contractions in muscles nearby. This suggests that adjusting the link between sensory input and movement allows motor commands to be issued more effectively from the brain. To explore this sensorimotor link, we studied the attenuation (gating) of somatosensory evoked potentials (SEPs) before and during hand movements in patients with writer's cramp. For premovement gating, 10 patients and 11 age-matched normal subjects were given a warning sound followed 1s later

When manipulating objects with both hands, the corpus callosum (CC) is of paramount importance for interhemispheric information exchange. Hence, CC damage results in impaired bimanual performance. Here, healthy young adults performed a complex bimanual dial rotation task with or without augmented visual feedback and according to five interhand frequency ratios (1:1, 1:3, 2:3, 3:1, 3:2). The relation between bimanual task performance and microstructural properties of seven CC subregions (i.e., prefrontal, premotor/supplementary motor, primary motor, primary sensory, occipital, parietal, and tem

A better understanding of the central control of the physiology of deglutition is necessary for devising interventions aimed at correcting pathophysiological conditions of swallowing. Positive modulation of the cortical swallowing network can have clinical ramifications in dysphagia due to central nervous system deficits. Our aim was to determine the effect of nutritive sensory input on the cortical swallowing network. In 14 healthy right-handed volunteers, we utilized a paradigm-driven protocol to quantify the number of activated voxels and their signal intensity within the left hemispheric c

The combination of electrical deep brain stimulation (DBS) with functional imaging offers a unique model for tracing brain circuitry and for testing the modulatory potential of electrical stimulation on a neuronal network in vivo. We therefore applied parametric positron emission tomography (PET) analyses that allow characterization of rCBF responses as linear and nonlinear functions of the experimentally modulated stimulus (variable stimulator setting). In patients with electrodes in the thalamic ventrointermediate nucleus (VIM) for the treatment of essential tremor (ET) here we show that var

Functional disintegration has been observed in schizophrenia during task performance. We sought to investigate functional disintegration during rest because an intrinsic functional brain organization, including both "task-negative" (i.e., "default mode") and "task-positive" networks, has been suggested to play an important role in integrating ongoing information processing. Additionally, the brain regions that are involved in the intrinsic organization are believed to be abnormal in schizophrenia. Patients with paranoid schizophrenia (N=18) and healthy volunteers (N=18) underwent a resting-sta

Prefrontal cortex (PFC) activation during manual performance is often associated with motor control efficiency; however, its functional meaning depends on task demands and prior experience. Manual dexterity tasks differ in temporal structure, with discrete tasks involving clearly defined actions and continuous tasks requiring sustained visuomotor control. This study aimed to characterize task-dependent modulation of prefrontal metabolic activity during discrete and continuous manual dexterity tasks and to examine its association with prior experience in manual coordination activities and motor

BACKGROUND: Without visual feedback, the central nervous system must rely on the integration of internal models and proprioceptive information. While the dorsolateral prefrontal cortex (DLPFC) is involved in motor control and sensorimotor monitoring, whether it causally influences force steadiness and whether any such influence is dependent on corticospinal excitability remains unclear.
OBJECTIVE: This study aimed to investigate whether modulating DLPFC activity using theta burst transcranial stimulation (TBS) influences force steadiness during submaximal contractions performed without visual

Dysfunction of the cerebellum is increasingly recognised as a contributor to schizophrenia. As one of the most ubiquitous voltage-gated potassium channels in the brain, the expression and function of Kv2.1 remains underexplored in the cerebellum, and its potential involvement in cerebellar pathophysiology in schizophrenia is unknown. Here, we characterised the expression pattern of Kv2.1 in the cerebellar cortex and examined its changes, along with cerebellar cytoarchitecture, in a subchronic phencyclidine male mouse model of schizophrenia. Behavioural performance was assessed throughout phenc

BACKGROUND: Infantile esotropia inaugurates a series of preternatural eye movements that include dissociated vertical divergence (DVD), primary oblique muscle overaction, and latent nystagmus. Congenital binocular visual loss leads to infantile nystagmus, a seemingly unrelated condition. These disorders seem to defy clinical explanation or nosologic classification. So where do these conditions come from and why are their neurologic pathways so elusive?
METHODS: Analysis of these pediatric ocular motor disorders from the perspective of evolutionary biology and its applicability to humans.

The brain operates through large-scale, distributed networks, which are disrupted after stroke. Cortico-cortical paired associative stimulation (ccPAS) is a novel neuromodulation technique designed to induce spike-timing-dependent plasticity in specific pathways. However, its effects on large-scale resting-state networks (RSNs) in stroke patients remain unclear. In this randomized, double-blind, sham-controlled study, 31 chronic stroke patients (mean age: 49.4 ± 11.8 years) with unilateral supratentorial lesions and upper limb motor dysfunction were recruited. Participants received a 12-day in

INTRODUCTION: Parkinson's disease (PD) is characterized by enhanced beta-band activity (13-30 Hz) in the motor control regions. Simultaneously, cortico-muscular (CM) connectivity in the beta-band during iso-metric contractions tends to decline with age, in various diseases, and under dual-task conditions.
OBJECTIVE: This study aimed to characterize electroencephalograph (EEG) and electromyograph (EMG) power spectra during a motor task, assess CM phase connectivity, and explore how these measures are modulated by an additional cognitive task. Specifically, we focused on the beta-band to explor

BACKGROUND: Unilateral cerebral palsy (UCP) presents a greater functional alteration of the upper limb. Among the recommended interventions are certain therapeutic tools, such as transcranial direct current stimulation (tDCS) that could increase the therapeutic window and enhance the effect of interventions.
AIMS: To determine the effectiveness of a 3-weeks intervention of cathodal tDCS applied over the motor cortex of the less affected hemisphere combined with a manual function intensive training program in the upper limbs on quality of movement and the spontaneous use of upper limb in child

BACKGROUND: Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are part of a well-established disease continuum, underpinned by TDP43-pathology. In contrast, the clinical manifestations of Tau-linked disorders are typically limited to cognitive phenotypes or atypical parkinsonism, although few reports describe motor neuron involvement associated with MAPT (microtubule-associated protein Tau) mutations. This study aimed to investigate the contribution of MAPT to the ALS phenotype.
METHODS: We analyzed a whole-exome sequencing database comprising 470 ALS patients and explored

The cerebellar cortex is organized into discrete regions populated by molecularly distinct Purkinje cells (PCs), the sole cortical output neurons. While studies in animal models have shown that PC subtypes differ in their vulnerability to disease, our understanding of human PC subtype and vulnerability remains limited. Here, we demonstrate that human cerebellar regions specialized for motor vs cognitive functions (lobule HV vs Crus I) contain distinct PC populations characterized by specific molecular and anatomical features, which show selective vulnerability in essential tremor (ET), a cereb

BACKGROUND: Alzheimer's disease (AD) is characterized by cognitive decline, with pathological features including amyloid β (Aβ) plaques and inflammation. Despite recent approvals of anti-amyloid antibodies, there remains a need for disease-modifying and easily accessible therapies. The endocannabinoid system presents a promising target for AD treatment, as it regulates various processes implicated in AD pathogenesis.
AIMS: This study assesses the effects of the synthetic cannabinoid WIN 55,212-2 on AD pathology and behavior deficits in aged 5XFAD mice, a well-established AD model.

BACKGROUND: Progressive supranuclear palsy (PSP) is characterized by the presence of hyperphosphorylated and misfolded tau aggregates in neurons and glia. Recent studies have illuminated the prion-like cell-to-cell propagation of tau via exosomes. Recognizing the potential significance of excretion through urine as a crucial pathway for eliminating pathological tau from the central nervous system, this study aimed to investigate whether exosomes derived from the urine of PSP-Richardson's syndrome (PSP-RS) patients can elicit tau pathology and PSP-like symptoms in mice.
METHODS: Urinary exosom

INTRODUCTION: The a-Synuclein Origin and Connectome (SOC) model of Lewy body diseases postulates that a-syuclein will be asymmetrically distributed in some patients with Lewy body diseases, potentially leading to asymmetric neuronal dysfunction and symptoms.
METHODS: We included two patient groups: 19 non-demented Parkinson's disease (nPD) patients with [18F]FDG PET and motor symptoms assessed by UPDRS-III, and 65 Lewy body dementia (LBD) patients with [18F]FDG PET and dopamine radioisotope imaging. Asymmetry indices were calculated for [18F]FDG PET by including the cortex for each hemisphere
