Low-frequency oscillations in cortical level to help diagnose task-specific dystonia.

Task-specific dystonia is a neurological movement disorder that abnormal contractions of muscles result in the twisting of fixed postures or muscle spasm during specific tasks. Due to the rareness and the pathophysiology of the disease, there is no test to confirm the diagnosis of task-specific dystonia, except comprehensive observations by the experts. Evidence from neural electrophysiological data suggests that enhanced low frequency (4-12 Hz) oscillations in the subcortical structure of the globus pallidus were associated with the pathological abnormalities concerning β and γ rhythms in mot

The median preoptic nucleus is a key site for estradiol regulation of sleep-wake behaviors in females rats.

STUDY OBJECTIVES: Women experience more sleep disruptions than men, particularly during hormonal transitions such as puberty, pregnancy, and menopause. This study investigated the role of estradiol (E2) in regulating sleep-wake behavior in female rats and identified the brain regions involved. METHODS: Using an exogenous E2 replacement model in ovariectomized rats, we assessed changes in sleep-wake patterns via electroencephalographic/electromyogram telemetry. The effects of E2 and progesterone, selective estrogen receptor agonists, and direct brain infusions of E2 and receptor antagonists we

Optogenetic Inhibition of Striatal Parvalbuminergic Interneurons Unmasks Impaired GABA and Adenosine Signaling in DYT1 Knock-In Mice.

Neurochemical imbalances in the striatum are thought to contribute to the pathophysiology of DYT1 dystonia (TOR1A), a severe movement disorder. Parvalbumin-positive GABAergic fast-spiking interneurons (PV+ FSI) exert a powerful inhibition within the striatal microcircuitry. To elucidate the impact of PV+ FSI on striatal neurotransmitter dynamics in a DYT1 knock-in (KI) mouse model, we combined optogenetic inhibition of PV+ FSI with in vivo microdialysis (optodialysis) and LC-MS/MS analysis. Dialysates were collected across baseline (light off), stimulation (light on, 595 nm), and post-stimulat

Gene supplementation therapy restores motor deficits in Gnao1[C215Y] mice and proves broad efficacy in C. elegans model.

The heterotrimeric G proteins are ubiquitous membrane-bound complexes specialized in the transduction of receptor-mediated extracellular signals into intracellular responses. Mutations in the G protein subunit alpha O1 (Gαo), encoded by GNAO1, have been associated with neurodevelopmental disorders characterized by prominent movement disorder with or without epilepsy. The Gnao1[C215Y]/+ mouse model recapitulates key features of the human disease, with a relatively mild phenotype, normal viability and late onset movement abnormalities, detectable in specific motor tasks. Here we report functiona

RETRACTED: Optimizing the impact of time domain segmentation techniques on upper limb EMG decoding using multimodal features.

Источник пометил публикацию как отозванную

Neurological disorders, such as stroke, spinal cord injury, and amyotrophic lateral sclerosis, result in significant motor function impairments, affecting millions of individuals worldwide. To address the need for innovative and effective interventions, this study investigates the efficacy of electromyography (EMG) decoding in improving motor function outcomes. While existing literature has extensively explored classifier selection and feature set optimization, the choice of preprocessing technique, particularly time-domain windowing techniques, remains understudied posing a significant knowle

Quantitative assessment of dynamic movement reveals deficits due to hemiparetic stroke.

The absence of sensitive tools for quantifying movement dysfunction hinders our ability to study the underlying causes of motor impairments and makes it difficult to demonstrate the effectiveness of therapeutic approaches. Consequently, it slows down progress in developing novel treatment protocols, including personalized impairment-targeted interventions. While there exist well-established metrics of static and quasi-static motion, such as reaching range, little emphasis has been placed on quantifying dynamic response-controlled and timing-sensitive movements where the continuous modulation o

Effect of proprioceptive neuromuscular facilitation on balance, strength, cognition and ADL'S IN elderly - A randomized control study.

BACKGROUND: A common and significant worry for people in their later year falls. Falls are frequently caused by residual impairments, including decreasing strength, poor sensory processing, balance issues, and cognitive gait inadequacies. Deficits in movement and balance can persist for years and are associated with a high fall rate (>6 months). Neuromuscular re-education by proprioceptive neuromuscular facilitation involves stimulating sensory receptors to provide feedback on incorrect movement and posture. OBJECTIVE: To Identify the effects of PNF on Balance, Strength, cognition and ADL. M

Dynamic Digital Radiography in Ehlers-Danlos Syndrome: Visualizing Diaphragm Motility Impairment and Its Influence on Clinical Management.

A 40-year-old woman with a known diagnosis of Ehlers-Danlos syndrome (EDS) began experiencing progressive shortness of breath and reduced exercise tolerance following her second pregnancy. The patient underwent an unenhanced computed tomography (CT) scan that showed a marked elevation of the left diaphragm. Suspecting diaphragm dysfunction, the patient underwent Dynamic Digital Radiography (DDR) that confirmed a reduction in left diaphragm motility, indicative of impaired diaphragm function. Based on the DDR findings, which demonstrated reduced but preserved diaphragmatic motion without parado

Position Statement of the American Society for Stereotactic and Functional Neurosurgery on Focused Ultrasound Lesioning of the Brain by Non-neurosurgeons.

Since its US Food and Drug Administration approval in 2016, magnetic resonance-guided focused ultrasound (MRgFUS) thalamotomy has grown into one of the procedures of choice among patients with essential tremor (ET). Approved applications for the procedure have expanded over time from unilateral thalamotomy to treat ET and Parkinson disease tremor to bilateral staged thalamotomy for ET. As the procedure expands to healthcare environments beyond large academic centers, guidance is required to ensure that the responsible clinicians are appropriately trained to undertake this operative procedure.

Cholinergic Basal Forebrain Integrity and Cognition in Parkinson's Disease: A Reappraisal of Magnetic Resonance Imaging Evidence.

Cognitive impairment is a well-recognized and debilitating symptom of Parkinson's disease (PD). Degradation in the cortical cholinergic system is thought to be a key contributor. Both postmortem and in vivo cholinergic positron emission tomography (PET) studies have provided valuable evidence of cholinergic system changes in PD, which are pronounced in PD dementia (PDD). A growing body of literature has employed magnetic resonance imaging (MRI), a noninvasive, more cost-effective alternative to PET, to examine cholinergic system structural changes in PD. This review provides a comprehensive di

Objective Depression Detection Using EEG and Eye Movement Signals Induced by Oil Paintings.

Depression is a mental disorder characterized by persistent sadness and loss of interest, which has become one of the leading causes of disability worldwide. There are currently no objective diagnostic standards for depression in clinical practice. Previous studies have shown that depression causes both brain abnormalities and behavioral disorders. In this study, both electroencephalography (EEG) and eye movement signals were used to objectively detect depression. By presenting 40 carefully selected oil paintings-20 positive and 20 negative-as stimuli, we were able to successfully evoke emotio

Alpha and Beta Synucleins: From Pathophysiology to Clinical Application as Biomarkers.

The synuclein family includes three neuronal proteins, named α-synuclein, β-synuclein, and γ-synuclein, that have peculiar structural features. α-synuclein is largely known for being a key protein in the pathophysiology of Parkinson's disease (PD) and other synucleinopathies, namely, dementia with Lewy bodies and multisystem atrophy. The role of β-synuclein and γ-synuclein is less well understood in terms of physiological functions and potential contribution to human diseases. α-synuclein has been investigated extensively in both cerebrospinal fluid (CSF) and blood as a potential biomarker for

Consensus on Virtual Management of Vestibular Disorders: Urgent Versus Expedited Care.

The virtual practice has made major advances in the way that we care for patients in the modern era. The culture of virtual practice, consulting, and telemedicine, which had started several years ago, took an accelerated leap as humankind was challenged by the novel coronavirus pandemic (COVID19). The social distancing measures and lockdowns imposed in many countries left medical care providers with limited options in evaluating ambulatory patients, pushing the rapid transition to assessments via virtual platforms. In this novel arena of medical practice, which may form new norms beyond the cu

A description of novel variants and review of phenotypic spectrum in UBA5-related early epileptic encephalopathy.

Early infantile epileptic encephalopathy-44 (EIEE44, MIM: 617132) is a previously described condition resulting from biallelic variants in UBA5, a gene involved in a ubiquitin-like post-translational modification system called UFMylation. Here we report five children from four families with biallelic pathogenic variants in UBA5 All five children presented with global developmental delay, epilepsy, axial hypotonia, appendicular hypertonia, and a movement disorder, including dystonia in four. Affected individuals in all four families have compound heterozygous pathogenic variants in UBA5 All hav

Characterization of the Cerebrospinal Fluid Proteome in Patients with Fragile X-Associated Tremor/Ataxia Syndrome.

Fragile X-associated tremor/ataxia syndrome (FXTAS), first described in 2001, is a neurodegenerative and movement disorder, caused by a premutation in the fragile X mental retardation 1 (FMR1) gene. To date, the biological mechanisms causing this condition are still not well understood, as not all premutation carriers develop FXTAS. To further understand this syndrome, we quantitatively compared the cerebrospinal fluid (CSF) proteome of FXTAS patients with age-matched controls using mass spectrometry. We identified 415 proteins of which 97 were altered in FXTAS patients. These proteins suggest
