Tapinanthus globiferus mitigates arsenic-induced oxidative stress and genetic dysregulation In vivo.

Environmental toxins such as sodium arsenite induce oxidative stress, disrupt redox homeostasis, and trigger oncogenic signaling, providing a model for studying natural redox modulators. This study investigated the protective effects of Tapinanthus globiferus fractions (butanol, methanol, and ethyl acetate) on sodium arsenite-exposed Drosophila melanogaster. Biochemical, behavioral, and molecular assays were employed to assess oxidative stress markers, enzyme activities, locomotor performance, cell viability, and gene expression. Sodium arsenite exposure significantly decreased acetylcholinest

Ovalbumin oxidative modification fingerprints depend on gas plasma-driven reactive species profiles.

OBJECTIVE: Oxidative protein modifications have been linked to several diseases, but the variety and diversity of modifications are less studied. METHODS: We used the chicken egg protein ovalbumin and gas plasma technology, a potent source of various reactive species, for protein oxidation. Using high-resolution mass spectrometry and an in-house workflow, over 80 distinct oxidative protein modifications were mapped at per-amino-acid resolution. To examine how modification profiles depend on changes in reactive species types and concentrations, we generated 12 distinct argon gas plasmas by sys

Celastrol mitigates diabetic kidney disease by inhibiting ferroptosis via the SIRT1/NRF2/GPX4 pathway.

BACKGROUND: Diabetic kidney disease (DKD) is a leading cause of end-stage renal failure. Ferroptosis, an iron-dependent form of lipid peroxidation, contributes to DKD pathogenesis in diabetic mice. Celastrol, a bioactive compound derived from Tripterygium wilfordii, mitigates DKD-related renal injury, however, its effects and mechanisms concerning diabetes-associated ferroptosis remain undefined. METHODS: Using a db/db mice model, we compared celastrol, losartan, and ferrostatin-1 (Fer-1) in mitigating renal damage via the ferroptosis pathway. We assessed biochemical indices, renal histopathol

Oleanolic acid prevents ferroptosis and enhances oocyte competence during in vitro maturation in a porcine model.

The in vitro production of porcine embryos is a valuable model for livestock biotechnology and human reproductive research. However, oocyte quality is often compromised during culture by ferroptosis, which is an iron-dependent form of cell death driven by lipid peroxidation and redox imbalance. This study induced ferroptosis using RSL3 during porcine oocyte maturation to evaluate the protective potential of oleanolic acid (OA), an antioxidant triterpenoid compound. Exposure to RSL3 increased Fe2+ accumulation (as detected by FerroOrange staining), lipid peroxidation (as determined by the MDA a

Vitamin C-Assisted Fenton Oxidation Improves Oxidative Killing of Helicobacter pylori In Vitro.

Helicobacter pylori infection remains a major global health burden, and the increasing prevalence of antibiotic resistance underscores the need for alternative antibacterial strategies. This study examined whether vitamin C dosing could modulate Fenton oxidation and enhance oxidative antibacterial activity against H. pylori under defined acidic in vitro conditions. Oxidative activity was assessed using Acid Orange 7 decolorization as an indirect chemical readout and a terephthalic acid fluorescence assay as a relative indicator of hydroxyl radical-associated product formation in iron-peroxide

Andrographolide Suppresses Head and Neck Squamous Cell Carcinoma Progression via EGR1-ACSL4 Axis-Mediated Ferroptosis.

Head and neck squamous cell carcinoma (HNSCC) is a highly aggressive malignancy with high rates of recurrence and metastasis that result in poor clinical outcomes. Ferroptosis, an iron-dependent, lipid peroxidation-driven regulated cell death pathway, offers a therapeutic strategy to enhance antitumor effects by exploiting cancer cell vulnerability to iron-mediated lipid peroxidation. Andrographolide (ADE), the major bioactive component extracted from Andrographis paniculata, demonstrates extensive antitumor activities. However, its specific role and mechanisms in HNSCC are unclear. The in vit

Biological Consequences of Single and Combined Exposure to Magnetite-Chitosan Nanocomposite with Adsorbed Cobalt (II) in Danio rerio.

Magnetic nanocomposite sorbents are increasingly explored for the remediation of metal-contaminated waters; however, high abiotic removal efficiency may not always translate into biological safety. The present study evaluated the single and combined effects of dissolved cobalt (II) ions and magnetite-chitosan nanocomposites (MCN) in zebrafish (Danio rerio) embryos and larvae. MCN (30 wt.% Fe3O4) were synthesized via co-precipitation and crosslinking and physiochemically characterized. Adsorption experiments conducted in fish incubation medium demonstrated the efficacy of divalent Co removal an

Remodeling TME via feedback-driven photothermal-ferroptosis-immune cascade.

Triple-negative breast cancer (TNBC) is recognized as one of the most aggressive breast cancer subtypes, with poor responsiveness to current immunotherapies primarily due to its strongly immunosuppressive tumor microenvironment (TME). Here, we report a multifunctional nanoplatform, AIF NPs that integrates near-infrared (808 nm) photothermal therapy (PTT), ferroptosis induction, and immunosuppressive axis blockade into a feedback-amplified therapeutic system. The silicon-based photothermal agent IR1024 demonstrates an exceptionally high photothermal conversion efficiency of 93.4%, enabling effi

Ferroptosis: a promising therapeutic target for periodontitis.

Periodontitis is a chronic inflammatory disease driven by dysbiotic biofilm and abnormal host adaptive immune responses, characterized by irreversible destruction of periodontal tissues. Recent studies indicate that ferroptosis-a form of iron-dependent lipid peroxidation-mediated cell death-is not a passive outcome of the disease process but rather an important active inducer and pathological amplifier in the development of periodontitis. This review synthesizes existing evidence to propose that ferroptosis acts as an important regulatory node within the pathological network of periodontitis,

Eco-friendly stability-indicating HPLC method development, validation, and LC-MS-based characterization of degradation products of the polar drug imeglimin.

Imeglimin (IMG) is a recently developed cyclic biguanide compound used for the management of hyperglycaemia. To comprehend the stability behavior of the polar drug IMG under various degradation environments, an HPLC method was developed, and the major degradation products (DPs) were characterized using mass-spectrometric studies. The mobile phase comprising a 20 mM potassium dihydrogen phosphate (KH2PO4) buffer (pH 3.1) along with ethanol in a simple isocratic mode (98:02, v/v) was employed for optimum resolution of the drug and its DPs using a reverse-phase Luna C18(2) (250 × 4.6 mm, 5 µm) co

Metabolic sovereignty through oxidative hostility: a mechanistic perspective on how cancer engineers stromal dependency via ROS-mediated lysosomal reprogramming.

Cancer cells orchestrate a profound remodeling of their microenvironment to suppress immune surveillance and create metabolic dependency in surrounding stroma. We propose a mechanistic hypothesis in which this transformation is driven by a coordinated reactive oxygen species (ROS) signaling cascade. Cancer cells generate superoxide (O2•-) through NADPH oxidase (NOX) upregulation and mitochondrial respiration. Superoxide is rapidly converted to hydrogen peroxide (H2O2), a stable, membrane-diffusible ROS species that crosses stromal cell membranes via aquaporin channels. Within cancer-associated

Provoking Potent Cellular Ferroptosis in the Thermo-Sensitive S-Phase via CPT-11 Loaded Janus Nanomedicine for Enhanced Cancer Therapy.

Ferroptosis is a promising anticancer strategy, yet its efficacy is limited by insufficient cellular hydrogen peroxide (H2O2) levels and intrinsic antioxidant defenses. Meanwhile, mild photothermal therapy (mPTT) enhances ferroptosis by accelerating lipid peroxidation. Herein, we designed a Janus nanoplatform that specifically arrests cells in thermo-vulnerable S-phase to maximize the ferroptosis-sensitizing capacity of mPTT. A Janus architecture with a unique morphology was engineered to incorporate three functional modules: (1) A photothermal half-bowl composed of polydopamine (PDA) that gen

[Elucidation of a Novel Protective Function of AADAC Against Drug-induced Liver Injury through Ferroptosis Suppression].

Drug-induced liver injury (DILI) remains a major cause of drug withdrawal from the market, and safety concerns in clinical settings. Reactive metabolites, primarily produced by cytochrome P450 enzymes, are known to induce oxidative stress and hepatotoxicity through covalent binding to macromolecules and the generation of reactive oxygen species. Recent evidence suggests that ferroptosis, an iron-dependent form of lipid peroxidation-mediated cell death, is involved in DILI triggered by compounds such as carbon tetrachloride (CCl4) and acetaminophen (APAP). Arylacetamide deacetylase (AADAC), a h

Reducing agents for induction and maintenance therapy achieve long-term remission of refractory ulcerative colitis: A case report and review of literature.

BACKGROUND: Ulcerative colitis (UC) is a chronic and debilitating inflammatory bowel disease. Cumulative evidence indicates that excess hydrogen peroxide, a potent neutrophilic chemotactic agent, produced by colonic epithelial cells has a causal role leading to infiltration of neutrophils into the colonic mucosa and subsequent development of UC. This evidence-based mechanism identifies hydrogen peroxide as a therapeutic target for reducing agents in the treatment of UC. CASE SUMMARY: Presented is a 41-year-old female with a 26-year history of refractory UC. Having developed steroid dependence

Cooperative inhibition of mTORC1 disrupts iron-redox homeostasis and triggers ferritinophagy-dependent ferroptosis in hepatocellular carcinoma.

Hepatocellular carcinoma (HCC) remains a leading cause of cancer mortality, and the clinical utility of β-lapachone (β-Lap) is limited by dose-dependent toxicity. Here, we identify cryptotanshinone (CPT), a bioactive compound derived from Salvia miltiorrhiza, as a synergistic partner that enables dose reduction while enhancing antitumor efficacy. Integrated transcriptomic profiling, biochemical analyses, target engagement assays, and in vivo validation support that β-Lap and CPT cooperatively induce ferroptosis through disruption of iron-redox homeostasis. Mechanistically, β-Lap and CPT engage

Ferroptosis regulation in doxorubicin-induced cardiotoxicity: multi-mechanism interventions and translational strategies.

Doxorubicin (DOX), a highly effective anthracycline chemotherapy drug, has its clinical application severely restricted by dose-dependent doxorubicin-induced cardiotoxicity (DIC). Ferroptosis, an iron-dependent, lipid peroxidation-driven regulated cell death, has been confirmed as a core pathological mechanism in DIC, where it synergistically participates with multiple cell death modalities in myocardial injury. This review systematically elaborates the multidimensional molecular mechanisms underlying DOX-induced myocardial ferroptosis, including: lipid peroxidation cascade self-amplification,

Acid-responsive cobalt-doped ZIF-8 nanoplatform potentiates ferroptosis in osteosarcoma by disrupting the PSMD14/SLC7A11 axis.

Ferroptosis induction represents a promising antitumor strategy, yet osteosarcoma cells frequently resist ferroptosis through the PSMD14-mediated stabilization of SLC7A11. Here, we report an acid-responsive cobalt-doped ZIF-8 nanoplatform loaded with 1, 10-phenanthroline and coated with PEG (denoted as CPP) that simultaneously amplifies oxidative stress and dismantles the PSMD14/SLC7A11 antioxidant axis. Upon exposure to the acidic tumor microenvironment, the ZIF-8 framework degrades, releasing Co2+ ions that catalyze Fenton-like reactions to generate hydroxyl radicals (•OH) and initiate lipid
