The cycling B precursor cells in rat bone marrow (BM) that carry the B220 antigen and no surface Ig daily produce 780 million new cells. The pool of recirculating B lymphocytes in the rat, however, renew at a rate of only about 40 million cells/day. To analyze at which stages in B lymphocyte genesis the cell loss occurs, we identified post-mitotic cells in the rat BM B lineage, and determined their renewal rates. We used 5-bromo-deoxyuridine (BrdUrd) to label DNA-synthesizing cells, identifying incorporated BrdUrd with the mouse monoclonal antibody BU-1. B lineage cell subsets were identified

OBJECTIVES: Chronic intervillositis of unknown etiology (CIUE) is the pathological influx of maternal inflammatory cells into the intervillous space of the placenta without demonstrable cause. CIUE is associated with placental damage and increased risk of pregnancy loss, fetal demise, and growth restriction. The pathogenic mechanism is unclear, and we lack biomarkers for detection, and treatments to prevent recurrence. DNA methylation (DNAme) is a biomarker altered in association with many disease states. We hypothesized that CIUE may be associated with a distinct signature placental DNAme sig

Neurogenesis is a physiological response after cerebral ischemic injury to possibly repair the damaged neural network. Therefore, promoting neurogenesis is very important for functional recovery after cerebral ischemic injury. Our previous research indicated that hyperbaric oxygen therapy (HBOT) exerted neuroprotective effects, such as reducing cerebral infarction volume. The purposes of this study were to further explore the effects of HBOT on the neurogenesis and the expressions of cell migration factors, including the stromal cell-derived factor 1 (SDF1) and its target receptor, the CXC che

Replacement of pancreatic β-cells is one of the most promising treatment options for treatment of type 1 diabetes (T1D), even though, toxic immunosuppressive drugs are required. In this study, we aim to deliver allogeneic β-cell therapies without antirejection drugs using a bioengineered hybrid device that contains microencapsulated β-cells inside 3D polycaprolactone (PCL) scaffolds printed using melt electrospin writing (MEW). Mouse β-cell (MIN6) pseudoislets and QS mouse islets are encapsulated in alginate microcapsules, without affecting viability and insulin secretion. Microencapsulated MI

Vaccinia virus (VACV) is a notorious virus for a number of scientific reasons; however, most of its notoriety comes from the fact that it was used as a vaccine against smallpox, being ultimately responsible for the eradication of that disease. Nonetheless, many different vaccinia virus strains have been obtained over the years; some are suitable to be used as vaccines, whereas others are virulent and unsuitable for this purpose. Interestingly, different vaccinia virus strains elicit different immune responses in vivo, and this is a direct result of the genomic differences among strains. In ord

BACKGROUND: NASP (Nuclear Autoantigenic Sperm Protein) is a linker histone chaperone required for normal cell division. Changes in NASP expression significantly affect cell growth and development; loss of gene function results in embryonic lethality. However, the mechanism by which NASP exerts its effects in the cell cycle is not understood. To understand the pathways and networks that may involve NASP function, we evaluated gene expression in HeLa cells in which NASP was either overexpressed or depleted by siRNA.
METHODS: Total RNA from HeLa cells overexpressing NASP or depleted of NASP by s

Injury to the adult CNS often involves death of motoneurons, resulting in the paralysis and progressive atrophy of muscle. There is no effective therapy to replace motoneurons in the CNS. Our strategy to replace neurons and to rescue denervated muscles is to transplant dissociated embryonic day 14-15 (E14-15) ventral spinal cord cells into the distal stump of a peripheral nerve near the denervated muscles. Here, we test whether long-term delivery of two pharmacological inhibitors to denervated muscle, FK506 or SB203580, enhances reinnervation of muscle from embryonic cells transplanted in the

This in vitro study evaluated the cytotoxic effects of a restorative resin composite applied to an immortalized odontoblast-cell line (MDPC-23). Seventy-two round resin discs (2-mm thick and 4 mm in diameter) were light-cured for 20 or 40 seconds and rinsed, or not, with PBS and culture medium. The resin discs were divided into four experimental groups: Group 1: Z-100/20 seconds; Group 2: Z-100/20 seconds/rinsed; Group 3: Z-100/40 seconds; Group 4: Z-100/40 seconds/rinsed. Circular filter paper was used as a control material (Group 5). The round resin discs and filter papers were placed in the

Fluorescent dyes were used in the present study to map three rostrocaudal levels of the primitive streak of mouse embryos at 7.5 days of gestation. Injections at the most rostral level, the 1/4 streak level, principally labeled cells of the paraxial mesoderm, including mesenchyme in the head, and somites and segmental plate mesoderm in the trunk. Injections at the intermediate level, the 1/2 streak level, principally labeled cells of the lateral plate mesoderm of the trunk. Injections at the most caudal level, the 3/4 streak level, principally labeled cells of the extraembryonic mesoderm at th

1. A method is described which permitted stable extracellular recordings to be made from 115 neurones in nucleus interpositus of the cerebellum in unanaesthetized free-to-move cats. At least 95% of the neurones were cerebellar efferent cells since they were antidromically invaded following electrical stimulation of the brachium conjunctivum in the region of the contralateral red nucleus. 2. In cats in a state of quiet wakefulness the majority of interpositus neurones were tonically active at rates ranging from 12 to 77 impulses/sec (over-all mean 34/sec). The remaining neurones were silent or

Primary immune regulatory disorders (PIRDs), autoinflammatory, and hyperinflammatory syndromes can be life-threatening and/or significantly impact quality of life. Targeted immune modulation may control symptoms for some, but allogeneic hematopoietic stem cell transplantation (alloHCT) remains the only cure for many patients. Despite advances in alloHCT, outcomes for this population continue to need optimization. Focus areas include referral timing, patient selection, multidisciplinary evaluation, targeted pretransplant immune suppression (PTIS), graft selection, model- and exposure-based cond

Lactate, traditionally regarded as a metabolic byproduct of glycolysis, is now recognized as a critical signaling molecule in the central nervous system. Emerging evidence indicates that lactate participates in a dynamic metabolic-epigenetic regulatory network through protein lactylation, a post-translational modification capable of modulating chromatin structure and gene transcription. We summarize the physiological roles of lactate in neuronal-glial metabolic coupling and highlight cell-type-specific functions of the lactate-lactylation axis under both normal and pathological conditions. The molecular engineering of Y6 has marked a paradigm shift in the development of non-fullerene acceptors (NFAs) for organic solar cells (OSCs), enabling remarkable enhancements in power conversion efficiencies (PCEs) through its distinct A-DA'D-A architecture and optimized intermolecular packing. However, further advancement is hindered by persistent non-radiative recombination energy losses (ΔE3), which predominantly originate from molecular relaxation and sub-gap charge recombination pathways. Among various structural optimization strategies, the implementation of molecular asymmetry has re

The matrix protein 2 (M2) is one of the most representative and widely studied ion channel proteins, which plays broad biological roles in the life cycle of the influenza A virus. The M2 protein affects the function of host cells and the regulation of cellular homeostasis in an ion channel activity-dependent or-independent manner and it is also the target for anti-influenza virus drugs, such as amantadine and rimantadine. The focus of this review is to summarize the research progress on the M2 protein, such as structure and function, vaccine and drug development as well as antagonist of host i

Cancer functional genomics using CRISPR base editors (BEs) holds great promise for molecular characterization and new target discovery. However, traditional BEs, using intact DNA deaminases as mutators, are often constrained by limited control and nonspecific toxicities. Here we developed a small-molecule-controllable system using split-engineered BEs (seBEs). By placing deaminase activity under small-molecule control, seBEs significantly reduced cellular toxicity and enabled robust and inducible in vivo functional genomics screens. High-density seBE genetic screens using ~11,000 single guide

Cancer is a multifaceted disease that places a significant burden on healthcare systems and economies, driving the urgent need for novel therapeutic agents to curb its prevalence and impact. Thirty-three analogues of N-substituted-7-and 8-methyl-4-hydroxy-2-quinolone-3-carboxamides were synthesized and characterized using FT-IR, NMR (1H and 13C), HRMS, and elemental analysis. The analogues displayed potential cytotoxicity against human colon cancer (HCT-116) and epithelial colorectal adenocarcinoma (Caco-2) cells. Compounds bearing a phenyl ring tailored with o-CF3 (26) and a phenyl ring subst

Postoperative cognitive dysfunction (POCD) is a frequent neurological complication in older surgical patients, leading to substantial declines in quality of life and placing a considerable burden on society. This review provides a comprehensive overview of how mitochondrial dysfunction contributes to POCD, including disrupted energy metabolism, excessive reactive oxygen species production, calcium imbalance, and abnormalities in mitochondrial dynamics and quality control processes. These mitochondrial impairments further trigger neuroinflammation and activate multiple cell death pathways. In a

Estrogen deficiency is a major risk factor for degenerative disorders of the musculoskeletal system in postmenopausal women, while ferroptosis, an iron-dependent form of programmed cell death, has been implicated in the pathogenesis of various osteoarticular diseases. Recent studies indicate an interplay between estrogen signaling and ferroptosis, with evidence relatively well supported in postmenopausal osteoporosis (PMOP), emerging in osteoarthritis (OA), and still limited in intervertebral disc degeneration (IVDD). Estrogen deficiency may disrupt iron homeostasis, lipid metabolism, and anti

BACKGROUND: Ewing sarcoma belongs to the family of undifferentiated small round cell sarcomas of bone and soft tissue. It is characterized by a gene fusion involving EWSR1 and an ETS-family transcription factor gene. In 85-96% of cases, a specific chromosomal translocation results in the EWSR1-FLI1 fusion gene, whose product functions as an oncogene essential for tumorigenesis. Ewing sarcoma is most common in adolescents and young adults. It primarily affects the diaphyses of long bones, the pelvis, and the axial skeleton, although extraosseous involvement is not uncommon. This is a highly mal

Vascular endothelial dysfunction is a central pathophysiological mechanism underlying a broad spectrum of metabolic diseases, including type 2 diabetes mellitus, atherosclerosis, obesity, and non-alcoholic fatty liver disease. As a metabolically active interface, the endothelium regulates vascular tone, inflammation, oxidative stress, and nutrient exchange; its impairment contributes to the initiation and progression of these disorders. This review synthesizes current understanding of the molecular mechanisms linking endothelial dysfunction to metabolic disease progression. Particular focus is
