Management of Levonorgestrel and Etonogestrel Adsorption to Polypropylene During in Vitro Release from Long-Acting Release Dosage Forms.

Adsorption of active pharmaceutical ingredients (APIs) to various surfaces of labware can skew data during drug development. We observed substantial loss of two clinically significant hydrophobic contraceptive steroids, levonorgestrel (LNG) and etonogestrel (ENG), during in vitro release evaluation from poly(lactic-co-glycolic acid) (PLGA) microspheres and microneedles, leading to incomplete drug recovery. Our investigation identified the adsorption of these APIs to polypropylene (PP) as a key factor adversely affecting both the drug loss and sensitivity/linearity of ultra performance liquid c

Colonic Barrier Dysfunction and Inflammation Induced by Nano-/Micro-Plastics Fibers in Mice via the FAK/NF-κB/iNOS and TLR4/NF-κB/iNOS Pathways.

As emerging environmental contaminants, nano- and micro-plastics (NMPs) have drawn increasing research attention due to potential hazards to organisms. Particle shape is a key factor influencing the toxic effects of NMPs, and NMPs of different shapes cause varying degrees of tissue damage. We investigated the specific mechanisms of polystyrene NMPs (PS-NMPs) with different shapes underlying damage to the colonic intestinal barrier. We divided 120 five-week-old male C57BL/6J mice into seven groups that were orally exposed to different doses of PS-NMPs for 5 weeks. Hematoxylin and eosin staining

Selenite modulates phenotype-dependent epithelial-mesenchymal plasticity in pancreatic ductal adenocarcinoma: integrated in vitro analyses and patient-derived ex vivo tissue-slice cultures.

BACKGROUND: Pancreatic ductal adenocarcinoma (PDAC) is characterized by profound therapy resistance, desmoplasia, and phenotypic plasticity. While sodium selenite is a redox-active compound with reported tumor-selective cytotoxicity, its impact on epithelial-mesenchymal transition (EMT) states in PDAC remains insufficiently defined. We investigated whether selenite modulates EMT-associated phenotypes in a dose- and context-dependent manner using complementary in vitro and patient-derived ex vivo models and linked marker shifts to histological tumor regression in PDAC tissue slices. METHODS: T

Sustainable Conversion of Plastic and Corrugated Paper into Ni@C Integrated N/S Dual-Doped Carbon Membranes for Accelerated Polysulfide Redox in Lithium Polysulfide Batteries.

Rechargeable systems based on sulfur cathodes have attracted considerable interest as promising candidates for advanced energy storage owing to their exceptionally large theoretical energy density. However, widespread implementation of these batteries is still hindered by several intrinsic challenges. In particular, soluble lithium polysulfide intermediates tend to dissolve into the electrolyte and migrate between electrodes, which leads to active material loss and severe capacity decay. In addition, the redox conversion reactions involving these sulfur species proceed relatively slowly, resul

Therapeutic potential of nasal kisspeptin-54 for reducing α-synuclein accumulation and restoring hippocampal synaptic plasticity in a 6-hydroxydopamine-induced rat model of Parkinson's disease.

BACKGROUND/AIM: Parkinson's disease (PD) is characterized by progressive degeneration of dopaminergic neurons and pathological aggregation of α-synuclein (α-syn), leading to significant motor and cognitive impairments. Kisspeptin-54 (KP-54), a neuropeptide primarily involved in reproductive regulation, has recently been implicated in neurorestorative processes, with emerging evidence suggesting beneficial effects on locomotor activity and cognitive function. However, its therapeutic potential in PD has not yet been fully investigated. This study aimed to evaluate the therapeutic efficacy of ch

Enhanced expression of ADAMTS1 in ovarian carcinomas: loss of ADAMTS1 expression instigates cellular reprogramming of extracellular matrix ensuing altered plasticity, augmented migration and attenuated adhesion.

BACKGROUND: ADAMTS1, a disintegrin and metalloproteinase with thrombospondin motif 1, plays a role in inflammation, organogenesis, and ovulation. Up or downregulation of ADAMTS1 has been implicated in tissue remodelling leading to cancer. We analysed the expression of ADAMTS1 in different stages/grades of primary and metastatic serous ovarian tumours and ascites-derived tumour cells from patients and assessed the functional role of ADAMTS1 in ovarian cancer (OC) cell lines. METHODS: The expression and localisation of ADAMTS1 was assessed by immunohistochemistry (IHC) and Opal Multiplex IHC st

Contralesional cTBS promotes motor function by enhancing synaptic plasticity and axonal remodeling via TGF-β/Smad3/Foxp3 signaling axis in ischemic stroke rats.

BACKGROUND: Intracortical microstimulation (ICMS), an emerging therapeutic neuromodulation strategy, has shown promise in improving stroke recovery. However, the mechanisms of ICMS in promoting neurological restoration remain elusive. METHODS: We assessed motor dysfunction in middle cerebral artery occlusion/reperfusion (MCAO/R) rats following 3-week contralesional intermittent versus continuous theta-burst stimulation (iTBS vs. cTBS) in the subacute phase. Subsequently, we evaluated cTBS efficacy with or without TGF-β1 inhibitor (SB431542). We investigated structural and molecular alteration

The Effects of In-Phase Bilateral Upper-Limb Exercises on Corticospinal Plasticity in People with Progressive Multiple Sclerosis: A Pilot Randomized Single-Case Concurrent Multiple-Baseline Design.

Background: Corticospinal plasticity is impaired in MS. In-phase bilateral upper-limb exercises have been shown to improve corticospinal excitability in people with relapsing-remitting multiple sclerosis, but there is limited literature that supports the same conclusion for progressive multiple sclerosis. Therefore, our aim was to investigate whether a 12-week in-phase bilateral upper-limb exercise protocol could enhance corticospinal plasticity and improve clinical outcomes in people with progressive multiple sclerosis. Methods: Five participants (two females; age = 61 ± 12.04 years) with sec

Electroacupuncture ameliorates learning and memory impairment by inhibiting inflammation and promoting synaptic plasticity via inhibition of the NF-KB/NLRP3 signaling pathway in cerebral ischemic rats.

OBJECTIVE: Electroacupuncture (EA) has a protective effect on cerebral ischemic injury. However, the specific mechanism of action of EA has not been studied. In this study, we investigated whether EA was involved in the treatment of learning and memory impairment in rats with cerebral ischemia‒reperfusion injury (CIRI) through the NF-KB/NLRP3 signaling pathway. METHODS: Ninety-five male Sprague-Dawley (SD) rats were randomly divided into five groups, each consisting of 19 rats. A rat model of cerebral ischemia was established using transient middle cerebral artery occlusion (tMCAO) combined w

Dissipation fate of non-phthalate plastic additives in well water using UHPLC-Q-Orbitrap-HRMS and GC-Q-Orbitrap-HRMS: A comparison with pure water.

Non-phthalate plastic additives (NPPAs) are increasingly being used as alternatives to phthalates, which are avoided due to health and environmental risks. However, the behaviour and hazards of many NPPAs remain poorly understood. To better characterise their fate in aquatic environments, two representative NPPAs, 1-hydroxycyclohexyl phenyl ketone (HCPK) and acetyl triethyl citrate (ATEC), were selected for dissipation and degradation studies in deionized water under three environmental conditions: exposure to sunlight, darkness, and sunlight in the presence of 10 mg/L of nitrate. Bisphenol A in the Urine: Association with Urinary Creatinine, Impaired Kidney Function, Use of Plastic Food and Beverage Storage Products but Not with Serum Anti-Müllerian Hormone in Ovarian Malignancies.

Bisphenol A (BPA) is a high-production-volume industrial chemical and component of commonly used plastic products. However, it is also an endocrine-disrupting chemical that can negatively affect human health. It is not yet known whether it is associated with the development of epithelial ovarian cancer (EOC), a severe and highly fatal human disease. Therefore, the purpose of this study was to determine the concentrations of BPA in the urine of women with EOC or epithelial borderline ovarian tumors (EBOTs) using gas chromatography tandem mass spectrometry (GC-MS/MS) and find their possible asso

Electroacupuncture induces analgesia by regulating spinal synaptic plasticity via the AMPA/NMDA receptor in a model of cervical spondylotic radiculopathy: secondary analysis of an experimental study in rats.

OBJECTIVE: Cervical spondylotic radiculopathy (CSR) is characterized by neuropathic pain (NP). Although the analgesic effect of electroacupuncture (EA) has been widely recognized in clinical practice, the mechanism of EA in the treatment of CSR remains unknown. We previously reported that 7 days of EA improved behavioral markers of NP, attenuated increases in α-synuclein, synapsin 1 and 2, postsynaptic density (PSD)-95 and growth-associated protein (GAP)-43, and improved ultrastructural changes within synapses in a rat model of CSR. Herein, we present supplemental data from the same cohort of

Low-frequency repetitive transcranial magnetic stimulation alleviates abnormal behavior in valproic acid rat model of autism through rescuing synaptic plasticity and inhibiting neuroinflammation.

Autism is a complex neurodevelopmental disorder with no effective treatment available currently. Repetitive transcranial magnetic stimulation (rTMS) is emerging as a promising neuromodulation technique to treat autism. However, the mechanism how rTMS works remains unclear, which restrict the clinical application of magnetic stimulation in the autism treatment. In this study, we investigated the effect of low-frequency rTMS on the autistic-like symptoms and explored if this neuroprotective effect was associated with synaptic plasticity and neuroinflammation in the hippocampus. Mechanisms of exacerbation of Th2-mediated eosinophilic allergic asthma induced by plastic pollution derivatives (PPD): A molecular toxicological study involving lung cell ferroptosis and metabolomics.

Polystyrene microplastics (PS-MP) and dibutyl phthalate (DBP) are plastic pollution derivatives (PPDs) commonly found in the natural environment. To investigate the effects of PPD exposure on the risk of allergic asthma, we established a PPD exposure group in a mouse model. The dose administered for PS-MP was 0.1 mg/d and for DBP was 30 mg/kg/d, with a 5-week oral administration period. The pathological changes of airway tissue and the increase of oxidative stress and inflammatory response confirmed that PPD aggravated eosinophilic allergic asthma in mice. The mitochondrial morphological chang

Disturbances in fear extinction learning after mild traumatic brain injury in mice are accompanied by alterations in dendritic plasticity in the medial prefrontal cortex and basolateral nucleus of the amygdala.

Mild traumatic brain injury (mTBI) and post-traumatic stress disorder (PTSD) have emerged as the signature injuries of the U.S. veterans who served in Iraq and Afghanistan, and frequently co-occur in both military and civilian populations. To better understand how fear learning and underlying neural systems might be altered after mTBI, we examined the acquisition of cued fear conditioning and its extinction along with brain morphology and dendritic plasticity in a mouse model of mTBI. To induce mTBI in adult male C57BL/6J mice, a lateral fluid percussive injury (LFP 1.7) was produced using a f

Modulation of Human Motor Cortical Excitability and Plasticity by Opuntia Ficus Indica Fruit Consumption: Evidence from a Preliminary Study through Non-Invasive Brain Stimulation.

Indicaxanthin (IX) from Opuntia Ficus Indica (OFI) has been shown to exert numerous biological effects both in vitro and in vivo, such as antioxidant, anti-inflammatory, neuro-modulatory activity in rodent models. Our goal was to investigate the eventual neuro-active role of orally assumed fruits containing high levels of IX at nutritionally-relevant amounts in healthy subjects, exploring cortical excitability and plasticity in the human motor cortex (M1). To this purpose, we applied paired-pulse transcranial magnetic stimulation and anodal transcranial direct current stimulation (a-tDCS) in b
