Extended follow-up of in vivo BCMA CAR-T therapy in relapsed/refractory multiple myeloma.

Data on long-term outcomes of in vivo chimeric antigen receptor T cell (CAR‑T) therapy remain scarce. We previously reported preliminary safety and efficacy data of in vivo B-cell maturation antigen (BCMA) CAR‑T (ESO-T01), in four patients with relapsed/refractory multiple myeloma, without leukapheresis and lymphodepletion, with up to 3 months of follow-up. The primary endpoints included safety and tolerability, while the secondary endpoints comprised efficacy, pharmacokinetics and pharmacodynamics. All patients developed dual-phase cytokine release syndrome (grades 1-3) and hematotoxicities (

Envafolimab combined with chemotherapy in advanced thymic carcinoma - first clinical experience with a novel subcutaneous programmed death-ligand 1 inhibitor.

Thymic carcinoma is rare with poor prognosis, and novel treatments are needed. We report two cases of advanced thymic carcinoma treated with the programmed death-ligand 1 inhibitor envafolimab with chemotherapy (liposomal paclitaxel and cisplatin). Case 1: A 59-year-old woman with metastatic thymic carcinoma (suspected neuroendocrine carcinoma) involving liver and bone received radiotherapy for spinal metastasis followed by eight cycles of envafolimab with liposomal paclitaxel and cisplatin. She achieved a partial tumor response with lesion shrinkage and normalization of liver function and tum

The glutathione transferase omega 1-1 GSTO1∗A140D polymorphism is associated with unfavourable prognosis in patients with thymoma and myasthenia gravis.

PURPOSE: Thymomas are rare organotypic tumors frequently associated with immune-related paraneoplastic syndromes. The genetic alterations of thymomas are largely unknown and rely on general transcription factor I (GTF2I) and Harvey rat sarcoma (HRAS) mutations. Glutathione transferase omega-1-1 (GSTO1-1) is an atypical member of the glutathione transferase superfamily involved in regulating pro-survival and anti-apoptotic pathways. The association between GSTO1-1 polymorphisms and the increased risk of different types of cancer has been investigated, but no study has focused on thymomas to dat

Targeting the Extracellular Signal-Regulated Kinase 5-Cellular Jun-Vimentin Axis to Inhibit Epithelial-Mesenchymal Transition and Metastasis in Patients with Triple-Negative Breast Cancer.

Breast cancer is the second most common cancer worldwide and remains the leading cause of cancer-related deaths among women. Triple-negative breast cancer (TNBC) represents approximately 15-20% of all breast cancer cases and is characterized by an aggressive clinical course and a high risk of metastasis. Extracellular signal-regulated kinase 5 (ERK5) is a critical biomarker that promotes tumor progression through mechanisms involving cell proliferation, invasion, and metastasis; however, its precise role in epithelial-mesenchymal transition (EMT) in TNBC remains unclear. In this study, we anal

Assessing time to symptomatic progression, a patient-relevant efficacy endpoint, in the MARIPOSA study in non-small cell lung cancer.

INTRODUCTION: In the phase 3 randomized MARIPOSA study, amivantamab and lazertinib combination therapy demonstrated improved progression-free survival (PFS) and overall survival (OS) versus osimertinib in participants with previously untreated, epidermal growth factor receptor-mutated advanced non-small cell lung cancer. Time to symptomatic progression (TTSP) was introduced to assess clinical worsening and complement endpoints that investigate radiographic disease progression and patient-reported outcomes. TTSP provides an easily interpretable measure of disease-specific symptom worsening to f

Comprehensive evaluation of the PCDHB family in gastric cancer: Prognostic significance, immune infiltration, and experimental validation of PCDHB5's oncogenic role.

The protocadherin B family genes (PCDHBs) play a significant role in tumor progression, yet their specific functions in gastric cancer remain insufficiently understood. This study aims to investigate the expression patterns of PCDHBs in gastric cancer, explore their influence on immune infiltration and patient prognosis, and preliminarily validate the functional role and underlying mechanisms of PCDHB5 through in vitro experiments. Using multi-omics data from the TCGA database, we found that most PCDHBs are significantly upregulated in gastric cancer, with missense mutations being the most com

Outcome of osimertinib-treated patients with epidermal growth factor receptor mutation-positive nonsmall cell lung cancer requiring dose reduction: a secondary analysis of the Reiwa study.

BACKGROUND: Osimertinib is effective in patients with epidermal growth factor receptor (EGFR) mutation-positive nonsmall cell lung cancer (NSCLC). However, some patients require osimertinib dose reduction because of adverse events. This study assessed the characteristics of osimertinib dose reduction and compared the efficacies of reduced-dose and regular-dose osimertinib. METHODS: This multicenter, prospective, observational study enrolled patients with EGFR mutation-positive NSCLC who started first-line osimertinib treatment between September 2018 and August 2020. We categorized the patient

Real-world outcomes of second-line carboplatin plus pemetrexed after first-line osimertinib in EGFR-mutant advanced NSCLC: An international multicentre cohort study.

BACKGROUND: First-line osimertinib is one of the standards of care for EGFR-mutant advanced non-small cell lung cancer (NSCLC), but most patients eventually progress. After progression, carboplatin plus pemetrexed remains the most widely used second-line therapy, yet robust real-world data in this setting are scarce. METHODS: We conducted a retrospective multicentre cohort study from four European countries in patients with EGFR-mutant advanced NSCLC who received second-line carboplatin plus pemetrexed following osimertinib. The primary endpoints were progression-free survival (PFS) and overa

Association of lysyl oxidase expression with clinicopathological features in colorectal adenocarcinomas.

PurposeColorectal adenocarcinoma (CRC) is one of the leading causes of cancer-related mortality worldwide. Within the tumor microenvironment, neoplastic cells, along with tumor-promoting fibroblasts, contribute to the progression of CRC. Lysyl oxidase (LOX), an enzyme involved in this process facilitates collagen cross-linking within the extracellular matrix and plays a crucial role in remodeling the tumor microenvironment (TME) and promoting metastasis through epithelial-mesenchymal transition (EMT). This study investigates LOX expression in both tumor cells and the tumor stroma in relation w

[Palliative surgery for metastatic prostate cancer].

Androgen deprivation in combination with novel hormonal agents, docetaxel, or in combination with abiraterone/prednisone plus docetaxel or darolutamid plus docetaxel represent the standard therapeutic approach in metastatic hormone-sensitive prostate cancer (mHSPC). Patients with low-risk prostate cancer also benefit from additional radiation therapy or radical prostatectomy in terms of progression-free and overall survival. Despite favorable response rates, basically all patients will develop castration resistant prostate cancer (CRPC) within 2.5 to 4 years. In addition to systemic chemothera

Efficacy and safety of SBRT combined with sintilimab and IBI305 in patients with advanced HCC and previously failed immunotherapy: study protocol of a phase 2 clinical trial.

INTRODUCTION: Hepatocellular carcinoma (HCC) is a leading cause of cancer-related death in China. The combination of immune checkpoint inhibitors (ICIs) and antiangiogenic drugs, such as bevacizumab and tyrosine kinase inhibitors, has been recommended as first-line treatment for advanced HCC. However, two-thirds of patients did not benefit from this form of immunotherapy. Currently, data on the subsequent regimen for patients previously treated with ICIs are lacking. Studies have shown that the combination of radiotherapy (RT) and ICIs is a potentially effective second-line therapy for HCC. Th

COL6A1 expression as a potential prognostic biomarker for risk stratification of T1 high grade bladder cancer: Unveiling the aggressive nature of a distinct non-muscle invasive subtype.

PURPOSE: T1 high grade (T1HG) bladder cancer (BC) is a type of non-muscle invasive BC (NMIBC) that is recognized as an aggressive subtype with a heightened propensity for progression. Current risk stratification methods for NMIBC rely on clinicopathological indicators; however, these approaches do not adequately capture the aggressive nature of T1HG BC. Thus, new, more accurate biomarkers for T1HG risk stratification are needed. Here, we enrolled three different patient cohorts and investigated expression of collagen type VI alpha 1 (COL6A1), a key component of the extracellular matrix, at dif
