Do patients with Prader-Willi syndrome have favorable glucose metabolism?

BACKGROUND: In recent years, more studies have observed that patients with Prader-Willi syndrome have lower insulin levels and lower insulin resistance than body mass index-matched controls, which may suggest protected glucose metabolism. METHOD: The PubMed and Web of Science online databases were searched to identify relevant studies published in the English language using the terms "Prader-Willi syndrome" with "glucose", "insulin", "diabetes mellitus", "fat", "adipo*", "ghrelin", "oxytocin", "irisin" or "autonomic nervous system". RESULTS: The prevalence of impaired glucose intolerance, ty

Inflammasome activation promotes venous thrombosis through pyroptosis.

Crosstalk between coagulation and innate immunity contributes to the progression of many diseases, including infection and cardiovascular disease. Venous thromboembolism (VTE), including pulmonary embolism and deep vein thrombosis (DVT), is among the most common causes of cardiovascular death. Here, we show that inflammasome activation and subsequent pyroptosis play an important role in the development of venous thrombosis. Using a flow restriction-induced mouse venous thrombosis model in the inferior vena cava (IVC), we show that deficiency of caspase-1, but not caspase-11, protected against

Predicting Vibrio cholerae Infection and Disease Severity Using Metagenomics in a Prospective Cohort Study.

BACKGROUND: Susceptibility to Vibrio cholerae infection is affected by blood group, age, and preexisting immunity, but these factors only partially explain who becomes infected. A recent study used 16S ribosomal RNA amplicon sequencing to quantify the composition of the gut microbiome and identify predictive biomarkers of infection with limited taxonomic resolution. METHODS: To achieve increased resolution of gut microbial factors associated with V. cholerae susceptibility and identify predictors of symptomatic disease, we applied deep shotgun metagenomic sequencing to a cohort of household c

Iron toxicity in yeast: transcriptional regulation of the vacuolar iron importer Ccc1.

All eukaryotes require the transition metal, iron, a redox active element that is an essential cofactor in many metabolic pathways, as well as an oxygen carrier. Iron can also react to generate oxygen radicals such as hydroxyl radicals and superoxide anions, which are highly toxic to cells. Therefore, organisms have developed intricate mechanisms to acquire iron as well as to protect themselves from the toxic effects of excess iron. In fungi and plants, iron is stored in the vacuole as a protective mechanism against iron toxicity. Iron storage in the vacuole is mediated predominantly by the va

Protective and therapeutic effects of fucoxanthin against sunburn caused by UV irradiation.

Mild exposure to ultraviolet (UV) radiation is also harmful and hazardous to the skin and often causes a photosensitivity disorder accompanied by sunburn. To understand the action of UV on the skin we performed a microarray analysis to isolate UV-sensitive genes. We show here that UV irradiation promoted sunburn and downregulated filaggrin (Flg); fucoxanthin (FX) exerted a protective effect. In vitro analysis showed that UV irradiation of human dermal fibroblasts caused production of intracellular reactive oxygen species (ROS) without cellular toxicity. ROS production was diminished by N-acety

Protective Effect of Zingiber Officinale against CCl4-Induced Liver Fibrosis Is Mediated through Downregulating the TGF-β1/Smad3 and NF-ĸB/IĸB Pathways.

No ideal hepatoprotective agents are available in modern medicine to effectively prevent liver disorders. In this study, we aimed at evaluating the potential of Zingiber officinale in the regression of liver fibrosis and its underlining mechanism of action. To induce liver fibrosis, male Wistar rats received CCl4 (2 ml/kg/2 times/week; i.p.), with and without 300 or 600 mg/kg Z. officinale extract daily through oral gavage. To assess the protective effect of Z. officinale, liver function parameters, histopathology, inflammatory markers and gene expression of transforming growth factor-beta 1 (

Inhibition of advanced glycation end products (AGEs): an implicit goal in clinical medicine for the treatment of diabetic nephropathy?

Several factors are incriminated in the genesis of diabetic nephropathy (DN). To elucidate their interplays, we utilized a diabetic rat model with nephropathy (SHR/NDmcr-cp). This model is characterized by hypertension, obesity with the metabolic syndrome, diabetes with insulin resistance, and intrarenal AGE accumulation. Various therapeutic approaches were used to achieve renoprotection. Caloric restriction corrects metabolic abnormalities and protects the kidney without correcting hypertension. Anti-hypertensive agents, angiotensin II receptor blocker (ARB) and calcium channel blocker, lower

Upregulation of glial cell line-derived neurotrophic factor (GDNF) in the rat cochlea following noise.

There are endogenous intracellular mechanisms that provide cells with protection from stress, as well as repair from damage. These pathways often involve stress proteins and neurotrophic factors. The present study used Western blot analysis to examine changes in glial cell line-derived neurotrophic factor (GDNF) following noise overstimulation. A noise exposure was utilized which causes a temporary threshold shift and has been previously shown to upregulate heat shock protein 72 in the rat cochlea. This noise exposure also provides protection from a second noise exposure that would otherwise c

Factors associated with gender and sex differences in anxiety prevalence and comorbidity: A systematic review.

Background: The prevalence and comorbidity of anxiety disorders are significantly different between women and men, with research showing a greater impact on women. The aim of this review was to identify the psychosocial and biological factors that have been considered to explain this gender and sex difference in prevalence and determine whether these factors are related to any anxiety comorbidity differences between men and women. Methods: Following the PRISMA guidelines, we carried out a systematic review of studies published between 2008 and 2021 in PsycINFO and PubMed databases. Empirical a

Blood type susceptibility to SARS-CoV-2 at a tertiary hospital in Accra, Ghana.

UNLABELLED: Pandemics from viral outbreaks, such as that caused by SARS-CoV-2, have significant impacts worldwide. The factors that underlie differential susceptibility to severe COVID-19 outcomes are not fully understood. The role of the ABO blood group in the outcome of SARS-CoV-2 infections remains to be clarified in different populations. This study described the SARS-CoV-2 seroprevalence and examined the association of the ABO blood group with COVID-19 disease among apparently healthy and COVID-19 patients at the Korle Bu Teaching Hospital, Accra. The study involved 277 participants compr

SARS-CoV-2 Antibody Prevalence Across Unvaccinated Health Care Workers During the COVID-19 Pandemic in Yemen: Cross-Sectional Study.

BACKGROUND: The COVID-19 pandemic presented significant challenges to health care centers across Yemen. The lack of access to COVID-19 vaccines and limited availability of personal protective equipment greatly increased the risk of SARS-CoV-2 exposure among health care workers (HCWs). Only a few studies have examined the seroprevalence and burden of SARS-CoV-2 among Yemeni HCWs. OBJECTIVE: This study aimed to assess the seroprevalence of SARS-CoV-2 and associated risk factors among a cohort of unvaccinated HCWs in Ibb City, the capital of Ibb Governorate, located in the highlands of southwest

Immunologic response after laparoscopic colon cancer operation within an enhanced recovery program.

OBJECTIVE: It has been demonstrated that colon operation combined with fast-track (FT) surgery and laparoscopic technique can shorten the length of hospital stay, accelerate recovery of intestinal function, and reduce the occurrence of post-operative complications. However, there are no reports regarding the combined effects of FT colon operation and laparoscopic technique on humoral inflammatory cellular immunity. METHODS: This was a prospective, controlled study. One hundred sixty-three colon cancer patients underwent the traditional protocol and open operation (traditional open group, n=42

Ancestral Wuhan SARS-CoV-2 anti-spike CD4+ T cells predict protection from symptomatic omicron breakthrough infection.

Neutralizing antibodies are considered the best surrogate markers of protection against a SARS-CoV-2 infection. However, the predictive value of these antibodies overlaps only slightly with other variants. In a case-control study, in pre-infected vaccinated patients, polyfunctional anti-Wuhan CD4+ T cell responses predicted protection against both the ancestral Wuhan strain and the distant Omicron variant, whereas CD4+ T cell responses to unrelated viruses (e.g., CMV, influenza) did not. Conversely, neutralizing antibodies to the Wuhan spike correlated with protection from Wuhan infection, but

Brimonidine Therapy for Protection From Noise-Induced Hearing Loss.

Noise exposure is a known cause of hearing loss, and only a few effective preventive drugs are available. Therefore, in this study, we aimed to investigate the protective effects of brimonidine on noise-induced inner ear hearing impairment in mice and explore its underlying mechanisms and long-term outcomes. Mice were randomly divided into control, noise exposure, and brimonidine groups. A 62-week follow-up was conducted after noise exposure. Brimonidine inhibited the noise-induced increase in inner ear glutamate concentration and downregulated inflammatory factors and immunoglobulins. Brimoni

Advancing protective effects of maternal antibodies in neonates through animal models.

Maternal immunity is critical for neonatal defense during early life, when infants are highly susceptible to infection. Antibodies transferred across the placenta or through breast milk not only provide immediate protection but also shape immune development. Defining the mechanisms and outcomes of this passive protection is essential for guiding maternal vaccination strategies and reducing the global burden of early-life infections. Animal models are indispensable for studying maternal antibody effects on neonatal immunity because they allow controlled investigation of transfer and protective

Microbiota-mediated protection against antibiotic-resistant pathogens.

Colonization by the microbiota provides one of our most effective barriers against infection by pathogenic microbes. The microbiota protects against infection by priming immune defenses, by metabolic exclusion of pathogens from their preferred niches, and through direct antimicrobial antagonism. Disruption of the microbiota, especially by antibiotics, is a major risk factor for bacterial pathogen colonization. Restoration of the microbiota through microbiota transplantation has been shown to be an effective way to reduce pathogen burden in the intestine but comes with a number of drawbacks, in

Mechanism of Aloe Vera extract protection against UVA: shelter of lysosomal membrane avoids photodamage.

The premature aging (photoaging) of skin characterized by wrinkles, a leathery texture and mottled pigmentation is a well-documented consequence of exposure to sunlight. UVA is an important risk factor for human cancer also associated with induction of inflammation, immunosuppression, photoaging and melanogenesis. Although herbal compounds are commonly used as photoprotectants against the harmful effects of UVA, the mechanisms involved in the photodamage are not precisely known. In this study, we investigated the effects of Aloe Vera (Aloe barbadensis mil) on the protection against UVA-modulat

Vaccination with leishmania hemoglobin receptor-encoding DNA protects against visceral leishmaniasis.

Leishmaniasis is a severe infectious disease. Drugs used for leishmaniasis are very toxic, and no vaccine is available. We found that the hemoglobin receptor (HbR) of Leishmania was conserved across various strains of Leishmania, and anti-HbR antibody could be detected in kala-azar patients' sera. Our results showed that immunization with HbR-DNA induces complete protection against virulent Leishmania donovani infection in both BALB/c mice and hamsters. Moreover, HbR-DNA immunization stimulated the production of protective cytokines like interferon-γ (IFN-γ), interleukin-12 (IL-12), and tumor

Exercise training plus calorie restriction causes synergistic protection against cognitive decline via up-regulation of BDNF in hippocampus of stroke-prone hypertensive rats.

One of the important organ damage of hypertension is cognitive decline. Cognitive function is determined by the function of hippocampus, and previous studies have suggested that the decrease in brain-derived neurotrophic factor (BDNF) in the hippocampus causes cognitive decline. Protection against cognitive decline is reported not only in pharmacological therapy but also in exercise training or calorie restriction. The aim of the present study was to determine whether exercise training plus calorie restriction cause synergistic protection against cognitive decline via BDNF in the hippocampus o

Heat shock proteins and their role in heart injury.

Heat shock protein (HSP) synthesis arises transiently as a tool to protect cellular homeostasis after exposure to heat and a wide spectrum of stressful and potentially deleterious stimuli. HSPs are "molecular chaperones" that recognize and form a complex with incorrectly folded or denatured proteins, which ultimately leads to correct folding, compartmentalization, or degradation. Accumulating evidence has implicated HSPs as mediators of myocardial protection, particularly in experimental models of ischemia and reperfusion injury. Impaired myocardial performance, which results from many factors
