Analysis of promoters from tyrosine/dihydroxyphenylalanine decarboxylase and berberine bridge enzyme genes involved in benzylisoquinoline alkaloid biosynthesis in opium poppy.

Tyrosine/dihydroxyphenylalanine decarboxylase (TYDC) and the berberine bridge enzyme (BBE) represent the entry point and a key branch point, respectively, in the biosynthesis of benzylisoquinoline alkaloids in select species of the Papaveraceae and Fumariaceae. Genomic clones for tydc7 and bbe1 from opium poppy (Papaver somniferum L.) were isolated. Deletion analysis of tydc7 and bbe1 5'-flanking regions revealed the location of putative regulatory domains necessary for expression of the beta-glucuronidase (gus) reporter gene in a transient assay system based on the microprojectile bombardment

Insulin gene: organisation, expression and regulation.

Insulin, a major hormone of the endocrine pancreas, plays a key role in the control of glucose homeostasis. This review discusses the mechanisms of cell-specific expression and regulation of the insulin gene. Whereas expression is restricted to islet beta-cells in adults, the insulin gene is more widely expressed at several embryonic stages, although the role of extrapancreatic expression is still unclear. beta-cell-specific expression relies on the interactions of 5'-flanking sequence motifs of the promoter with a number of ubiquitous and islet-specific transcription factors. IEF1 and IPF-1,

A GC-rich region containing Sp1 and Sp1-like binding sites is a crucial regulatory motif for fatty acid synthase gene promoter activity in adipocytes. Implication In the overactivity of FAS promoter in obese Zucker rats.

We have previously shown that the proximal 2-kb sequence of the fatty acid synthase (FAS) promoter transfected into rat adipocytes was highly sensitive to the cellular context, displaying an overactivity in obese (fa/fa) versus lean Zucker rat adipocytes. Using deletional analysis, we show here that FAS promoter activity mainly depends on a region from -200 to -126. This sequence exerts a strong negative effect on FAS promoter in adipocytes from lean rats but not in those from obese rats, resulting in a marked overtranscriptional activity in the latter cells. This region, fused to a heterologo

Transactivation of human T-cell leukemia virus type 1 by herpes simplex virus type 1.

HTLV-1 transcription depends upon activation by the HTLV-1 tax gene product. In addition, various substances and cellular transcription factors are also known to activate the HTLV-1 long terminal repeat (LTR)-mediated transcription in the absence of Tax. In this work we demonstrate that infection of either Jurkat or 293 cell lines with herpes simplex I (HSV-1), a widespread infectious virus of humans, activates HTLV-1 LTR-mediated gene expression. Further investigation revealed that each of the immediate-early (IE) gene products--ICPO, ICP4, and ICP27--of HSV-1 transactivates the HTLV-1 LTR-me

[Therapeutic strategy in anaphylactoid shock during general anesthesia. Etiologic agents and diagnostic evaluation].

In 1983, the risk of developing severe shock during anaesthesia was estimated to be 1 case in 4600 procedures with an associated mortality rate of 6 per cent. The incidence of severe accidents has further increased since this time. This review focuses on the aspects of anaphylactoid shock (AS) in the context of iatrogenic responsibility involving drugs or substances used during anaesthesia or placed in contact with viscera. The elements predictive of anaphylactoid shock are described together with the arguments involved in a biochemical conception and a physiological conception. The authors em

Cross-reactivity of human nickel-reactive T-lymphocyte clones with copper and palladium.

Twenty Ni-reactive T-lymphocyte clones were obtained from eight different donors and analyzed for their ability to cross-react with other metals. All Ni-reactive T-lymphocyte clones were CD4+CD8- and recognized Ni in association with either HLA-DR or -DQ molecules. Based on the periodic table of the elements, the metals Cr, Fe, Co, Cu, and Zn from the same horizontal row as Ni, and Pd and Pt from the same vertical row, were selected to study T-lymphocyte clone cross-reactivity. Distinct cross-reactivity patterns were found that could be divided into three major groups: Ni-reactive T-lymphocyte

The 5'-sequence of the murine Hox-b3 (Hox-2.7) gene and its intron contain multiple transcription-regulatory elements.

We sought to clone and characterize the murine Hox-b3 gene. In Xenopus embryos, the homologous gene has been shown to be responsive to retinoic acid, an agent which has profound effects on tissue growth and development. By plaque hybridization, using a partial, murine Hox-b3 cDNA as a probe, we screened a genomic library and isolated a series of overlapping clones. Restriction fragments from positive clones were sequenced by the dideoxy method on an automated DNA sequencer. We report the genomic sequence of the murine Hox-b3 gene. The sequence has been submitted to the GenBank database (access

Transcriptional regulation by triiodothyronine requires synergistic action of the thyroid receptor with another trans-acting factor.

Human placental lactogen B (hCS-B) promoter activity is strongly stimulated by triiodothyronine (T3) in pituitary GC cells through interaction between the thyroid receptor and a thyroid receptor-binding element (TBE) spanning coordinates -67 to -41. This TBE is adjacent to the binding site for pituitary factor GHF1 (-95 to -68) which seems necessary for T3 stimulation of hCS-B promoter activity (M. L. Voz, B. Peers, A. Belayew, and J. A. Martial, J. Biol. Chem. 266:13397-13404, 1991). We here demonstrate actual synergy between the thyroid receptor and GHF1. Indeed, in placental JEG-3 cells dev

Transcription and cancer.

The normal growth, development and function of an organism requires precise and co-ordinated control of gene expression. A major part of this control is exerted by regulating messenger RNA (mRNA) production and involves complex interactions between an array of transcriptionally active proteins and specific regulatory DNA sequences. The combination of such proteins and DNA sequences is specific for given gene or group of genes in a particular cell type and the proteins regulating the same gene may vary between cell types. In addition the expression or activity of these regulatory proteins may b

Excitation-contraction coupling in hypertrophied myocardium.

In hypertensive cardiac hypertrophy, inotropic responsiveness of alpha and beta adrenergic stimuli is reduced. We have previously shown that hearts from two-kidney, one-clip renal hypertensive rats (RHR) have increased beta-adrenergic receptor density and a defect in the guanine nucleotide regulatory protein, leading to decreased adenylate cyclase activity. In spontaneously hypertensive rats (SHR), beta-receptor density was decreased with no change in adenylate cyclase. In these present experiments, we have shown that the alpha 1-adrenergic receptor changes are in the opposite direction, decre

Reciprocal transfer of abnormalities in clonable B lymphocytes and myeloid progenitors between NZB and DBA/2 mice.

Previous studies in this laboratory revealed that NZB mice have abnormalities of myeloid progenitor populations from an early age such that they are poorly responsive to a particular type of colony-stimulating activity (CSA). In addition, these mice develop abnormally high numbers of B cells that can be cloned in semisolid agar cultures and that are atypical in resisting inhibition by anti-mu antibodies. To investigate whether these abnormalities, like other autoimmune phenomena studied previously, are transferrable with hemopoietic cell grafts, we performed reciprocal bone marrow transplants

The I--R system of hybrid dysgenesis in Drosophila melanogaster: are I factor insertions responsible for the mutator effect of the I--R interaction?

When Drosophila melanogaster males coming from a class of strains known as inducer are crossed with females from the complementary class (reactive), a quite specific kind of sterile female (SF) is obtained that exhibits other dysgenic traits such as non-disjunctions and non-randomly distributed mutations. This syndrome is caused by the interaction of the 'I factor' linked to inducer chromosomes with the maternally inherited reactive state 'R'. This I--R interaction is also responsible for 'chromosomal contamination' that is likely to result from very frequent I factor insertions into reactive

Renal chemoreceptors in the rat.

There are afferent nerve fibers responsive to alterations of the kidney's chemical environment in the renal nerves of the rat. In anesthetized, artificially ventilated, male Sprague-Dawley rats, single unit recordings were prepared by dissection of the centrally cut nerves of the right kidney. The stimuli used included occlusion of the renal artery, systemic asphyxia, changes in renal arterial and venous pressures, changes in ureteral pressure, and cyanide infusion. We found a population of sensory nerve fibers whose endings are activated only during markedly impaired renal blood flow (produce

Au nanoparticle-loaded mixed-valence MnOₓ nanosheets as an oxidase-like nanozyme for rapid colorimetric determination of leucomalachite green in fish and water.

Au nanoparticle-loaded mixed-valence MnOₓ nanosheets were prepared as an oxidase-like nanozyme for colorimetric determination of leucomalachite green (LMG). The nanozyme oxidized colorless LMG to the strongly absorbing malachite green (MG) chromophore using dissolved molecular oxygen without externally added H2O2. The resulting MG produced a characteristic absorption band at 620 nm. Electron microscopy, elemental mapping, X-ray diffraction, and X-ray photoelectron spectroscopy confirmed crystalline Au nanodomains dispersed on a low-crystallinity mixed-valence MnOx matrix. A Metamaterial Buckling-Assisted Surface-Acoustic-Wave Enabled Sensor (mBASES) for Versatile and Ultrasensitive Biomarker Detection.

Hydrogel metamaterials with programmable microarchitectures offer a promising route for converting molecular recognition into mechanical responses. However, efficiently transducing weak biomolecular perturbations into detectable signals without relying on molecular amplification remains a fundamental challenge. Here, we report a metamaterial Buckling-Assisted Surface-acoustic-wave Enabled Sensor (mBASES), which introduces a structure-triggered physical amplification mechanism by integrating bio-crosslinked hydrogel metamaterials with a surface acoustic wave (SAW) device. The Influence of Different Starting Block Distances on Internal Tibial Load in Elite Sprinters.

In sprint running, the starting block distance (SBD) directly influences an athlete's start performance, yet there is a lack of research exploring how block distance affects internal tibial load. Therefore, this study examined internal tibial loading in 28 elite sprinters using finite element analysis. Maximum principal stress, shear stress, total deformation, and bending moments were assessed under 3 front block distances (50%, 60%, and 70% of leg length). A 2-way analysis of variance evaluated the effects of gender, SBD, and their interaction. The results showed a significant main effect of

Finite Element Model Updating for Rotating Machinery: Methods, Applications, and Future Directions-A Review.

Finite element model updating (FEMU) provides a physics-based approach for reducing discrepancies between numerical models and measured responses by estimating uncertain physical parameters. Its application to rotating machinery is challenging because rotor dynamics are strongly affected by rotational speed, bearing and support properties, nonlinear interactions, operating conditions, measurement limitations, and parameter correlation. This review examines FEMU methods and applications for rotating machinery, distinguishing direct FEMU studies from nonlinear, uncertainty, surrogate, and AI-bas

Rice LSD1-like Genes: Genome-Wide Characterization and Evidence Linking OsLSD3 to Plant Height.

LSD1-like zinc-finger proteins participate in programmed cell death, redox homeostasis, and stress responses in plants, but their functional diversification and contributions to agronomic variation in rice remain poorly defined. This study aimed to characterize the rice LSD1-like gene family and evaluate the potential agronomic roles of selected members, with particular emphasis on OsLSD3. Genome-wide analyses were integrated with OsLSD3 natural variation and haplotype analyses in 4666 rice accessions, CRISPR/Cas9 mutant phenotyping in the ZH11 background, and subcellular localization assays.

Tip-Functionalized Gold Nanorod Micropillar Arrays for Millisecond-Resolved Photothermal Neuromodulation Toward Retinal Prosthetic Applications.

Photothermal stimulation using plasmonic nanomaterials presents a promising alternative to electrical stimulation in retinal prostheses, addressing various functional limitations associated with electrical methods. However, many photothermal strategies utilize dispersed nanoparticles randomly accumulated on target locations and do not demonstrate millisecond-scale thermal dynamics. Herein, a flexible polydimethylsiloxane (PDMS) micropillar array with gold nanorods (GNRs), immobilized at the pillar tips, is introduced to create a deterministic and conformal platform that spatially localizes pho

Laser-Synthesized Carbon-Coated Elemental Bismuth Nanoparticles for Sustained Reactive Oxygen Species Generation Under Visible-Light Irradiation.

Owing to its visible light-responsive electronic properties, redox activity, and potential biocompatibility, elemental bismuth is a promising candidate for photodynamic and chemodynamic therapies primarily via reactive oxygen species generation. However, accelerated surface oxidation of zero-valent bismuth suppresses photoinduced charge transfer, limiting its practical application. Here we introduce metallic bismuth core-shell nanoparticles produced by femtosecond laser ablation in acetone, featuring a crystalline Bi° core encapsulated within a conformal amorphous carbon shell (∼5 nm thick), p
