Revisiting endometriosis management: Polyphenols as alternative therapeutic candidates.

Endometriosis (EM) is a common gynecological disorder imposing a considerable burden on the quality of life of women through chronic pain, infertility, and other clinical manifestations. The effectiveness of management options for EM is limited by substantial side effects associated with current treatment options, including hormonal therapies and surgery, increasing the need for an alternative approach associated with fewer adverse effects. Polyphenols, a wide class of naturally occurring bioactive compounds, have generated considerable attention due to their antioxidant, anti-inflammatory, an

Steroid-sparing strategies in polymyalgia rheumatica: a systematic review and meta-analysis of tocilizumab with practical guidance for tapering.

Polymyalgia rheumatica (PMR) is an inflammatory rheumatic condition primarily managed with glucocorticoids (GCs), but prolonged GC use carries significant adverse effects. Tocilizumab, an interleukin−6 receptor inhibitor, has emerged as a promising steroid-sparing therapy for patients with relapsing or GC-dependent PMR. We conducted a systematic review and meta-analysis according to PRISMA guidelines to assess the efficacy and safety of tocilizumab in PMR. Four randomized controlled trials (RCTs) involving 317 patients were included in the meta-analysis, and one multicentre observational cohor

Casdatifan shows durable response linked to HIF-2α biology in kidney cancer.

Clear cell renal cell carcinoma (ccRCC) is largely driven by the transcription factor hypoxia-inducible factor 2α (HIF-2α)1. Here we show that monotherapy with casdatifan-an orally bioavailable, potent and selective HIF-2α inhibitor2-produces meaningful, durable antitumour activity with manageable safety in individuals with refractory metastatic ccRCC. Dose-expansion data from the ARC-20 study ( NCT05536141 ) are presented, including for the 100 mg once daily (QD) cohort (n = 32) and the total cohort (n = 127). Treatment discontinuation from casdatifan-related adverse events was infrequent (3%

Autologous cryo-shocked neutrophils enable targeted therapy of sepsis via broad-spectrum neutralization of pro-inflammatory cytokines and endotoxins.

Background: Sepsis is a life-threatening disease characterized by multiple organ failure due to excessive activation of the inflammatory response and cytokine storm. Despite recent advances in the clinical use of anti-cytokine biologics, sepsis treatment efficacy and improvements in mortality remain unsatisfactory, largely due to the mechanistic complexity of immune regulation and cytokine interactions. Methods: In this study, a broad-spectrum anti-inflammatory and endotoxin neutralization strategy was developed based on autologous "cryo-shocked" neutrophils (CS-Neus) for the management of sep
