Growth failure in aggrecan deficiency is due to decreased extracellular matrix and impaired growth plate chondrocyte hypertrophy.

Heterozygous loss-of-function mutations in the aggrecan (ACAN) gene cause autosomal dominant short stature often associated with advanced bone age, early-onset osteoarthritis and intervertebral disc disease (SSOAOD). These mutations are relatively common in patients with idiopathic short stature. However, the pathogenic mechanism of growth failure in this condition is not fully understood. Here, we studied the heterozygous cartilage matrix deficiency mouse (Acan+/-), which harbors a 7 bp microdeletion in aggrecan and develops postnatal growth cessation despite being born of normal size. Using

Glucagon-like peptide-1 receptor agonists rescued diabetic vascular endothelial damage through suppression of aberrant STING signaling.

Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) protect against diabetic cardiovascular diseases and nephropathy. However, their activity in diabetic retinopathy (DR) remains unclear. Our retrospective cohort study involving 1626 T2DM patients revealed superior efficacy of GLP-1 RAs in controlling DR compared to other glucose-lowering medications, suggesting their advantage in DR treatment. By single-cell RNA-sequencing analysis and immunostaining, we observed a high expression of GLP-1R in retinal endothelial cells, which was down-regulated under diabetic conditions. Treatment of GLP-1

Metformin rescues migratory deficits of cells derived from patients with periventricular heterotopia.

Periventricular neuronal heterotopia (PH) is one of the most common forms of cortical malformation in the human cortex. We show that human neuronal progenitor cells (hNPCs) derived from PH patients with a DCHS1 or FAT4 mutation as well as isogenic lines had altered migratory dynamics when grafted in the mouse brain. The affected migration was linked to altered autophagy as observed in vivo with an electron microscopic analysis of grafted hNPCs, a Western blot analysis of cortical organoids, and time-lapse imaging of hNPCs in the presence of bafilomycin A1. We further show that deficits in auto

Landscape of T Cells in Tuberculous Pleural Effusion.

The distribution and the function of T lymphocyte subsets in pleural effusion (PE) and peripheral blood (PB) in tuberculous pleural effusion (TPE) patients remain unclear. In this study, we aimed to explore the expression patterns, regulatory mechanisms, and functions of T lymphocyte subsets in TPE patients, especially the distribution of T lymphocyte subsets at the single cell level. The CD3+ T cells were isolated from PE and PB of four TPE patients for single-cell RNA sequencing (scRNA-seq) to screen T cell subsets. MAPKAP1 orchestrates macrophage polarization and lipid metabolism in fatty liver-enhanced colorectal cancer.

Various factors, including fatty liver and macrophage alterations, influence colorectal cancer (CRC). This study explores the mechanistic role of fatty liver in CRC progression, focusing on macrophage polarization and lipid metabolism. A murine fatty liver model was created with a high-fat diet (HFD), and CRC was induced using AOM and DSS. Single-cell transcriptome sequencing (scRNA-seq) identified MAPKAP1 as a critical gene promoting CRC via M2 macrophage polarization and lipid metabolism reprogramming. Prognosis analysis on the TCGA-CRC dataset confirmed MAPKAP1's significance. In vitro and

IFNγ and proinflammatory cytokines cooperatively license astrocytes to support antigen-dependent CD4+ T cell restimulation.

We report that type 2 interferon with proinflammatory cytokines induces astrocytes to express multiple components required for attraction of and antigen presentation to CD4+ T cells via MHC-II molecules. Re-analysis of astrocyte-enriched single-cell sequencing from bacterial mimetic-injected mice revealed a transcriptionally distinct interferon-responsive astrocyte population enriched for antigen-presentation-associated genes 24 hours after peripheral immune challenge. In vivo, we identify MHC-II+GFAP+ astrocytes localized to the surface of the brain across multiple mouse models of neuroinflam

Decoding the Spatiotemporal Dynamics of ENTPD1 in Diabetic Foot Ulcer Through Multi-Omic Profiling.

The global incidence of diabetic foot ulcers (DFUs) has increased in parallel with the global incidence of diabetes mellitus, leading to a range of pathophysiologic conditions and a poor prognosis. ENTPD1, also known as CD39, is an extracellular nucleotidase associated with immunogenic cell death (ICD), but its role in DFUs remains unclear. Through single-cell RNA sequencing, we obtain the genetic, functional, trajectory, and communication differences between DFUs and healthy subjects. Then, we selected vascular endothelial cells (Vasendo) for further re-clustering to identify DFU-healing-prom

Development of Novel Monoclonal Antibodies for Evaluation of Transmembrane Prostate Androgen-Induced Protein 1 (TMEPAI) Expression Patterns in Gastric Cancer.

Transmembrane prostate androgen-induced protein 1 (TMEPAI) is a single-span membrane protein, functionally involved in transforming growth factor beta signaling pathway. The particular protein presented in cells in three isoforms, which differs in the length of the soluble N-terminal extracellular domain, making it challenging for the immunochemical recognition. By using quantitative real-time polymerase chain reaction, we identified significant upregulation of PMEPA1 gene expression in malignant tissues of patients with gastric adenocarcinoma. The main part of commercially available anti-TMEP

Heterogeneity of peripheral immune cell landscape in systemic lupus erythematosus patients after belimumab treatment.

OBJECTIVES: The diversity and heterogeneity of circulating immune cells have been extensively investigated in systemic lupus erythematosus (SLE). However, little is known about the influence of belimumab, an anti-BAFF (B cell-activating factor) monoclonal antibody, on the heterogeneity of peripheral immune cell landscape in SLE patients. In this study, we aimed to investigate the altering effect of belimumab on autoimmunity in SLE. METHODS: The single-cell RNA sequence revealed a total of 11 cell clusters by comparing the transcriptome profiles of 24.869 peripheral blood mononuclear cells (PB

Integrative analysis of adenosine-related RNA modifications defines molecular subtypes of bladder cancer and identifies CES1 as a driver of tumor progression and immunotherapy resistance.

BACKGROUND: Adenosine-related RNA modifications, including N6-methyladenosine (m6A), N1-methyladenosine (m1A), alternative polyadenylation (APA), and adenosine-to-inosine (A-to-I) editing, critically regulate transcriptional programs in cancer. However, their coordinated contribution to bladder cancer biology and response to immunotherapy remains poorly understood. METHODS: We systematically characterized four classes of RNA modification “writer” genes in bladder cancer, assessing somatic mutations, copy-number variations, and mRNA expression patterns across seven independent cohorts, includin

Involvement of both T cell receptor V alpha and V beta variable region domains and alpha chain junctional region in viral antigen recognition.

We have studied the lymphocytic choriomeningitis virus (LCMV)-specific cytotoxic T cell response in transgenic mice expressing either the T cell receptor (TcR) alpha (V alpha 2/J alpha TA31) or the corresponding TcR beta (V beta 8.1/D beta/J beta 2.4) chain originally isolated from the LCMV glycoprotein specific (residues 32-42), H-2Db-restricted T cell clone P14. The expression of single transgenic TcR chains did not influence the corresponding endogenous TcR V gene usage in unstimulated T cells indicating that one particular TcR alpha or beta chain can randomly pair with different V beta or

Development and validation of an interpretable machine learning model identify the lactylation-related protein SUSD3 as a prognostic and therapeutic biomarker for breast cancer.

BACKGROUND: Breast cancer is one of the most prevalent malignancies and a leading cause of cancer-related mortality among women. Lactylation, a recently recognized post-translational modification, has emerged as a significant factor in tumor biology, with increasing evidence linking it to cancer progression and immune modulation. However, the role of lactylation in tumorigenesis remains ambiguous. This raises questions about whether it serves as a primary driver or a secondary regulator during cancer development, as well as its influence on the tumor immune microenvironment and prognostic impl

Photobiomodulation is more effective than long-term scrotal hyperthermia in improving testis tissue and spermatogenesis in mice with busulfan-induced azoospermia.

OBJECTIVE: The use of photobiomodulation (PBM) for tissue repair has gained acceptance. This study investigated and compared the effects of PBM on mice exposed to scrotal hyperthermia and busulfan. METHODS: Forty 8-week-old adult mice were divided into five groups: (I) control, (II) hyperthermia, (III) busulfan, (IV) hyperthermia+PBM, and (V) busulfan+PBM. To induce azoospermia in groups II and IV, the scrotum of the mice was exposed to water at 43 °C every other day for 5 weeks. In groups III and V, a single dose of busulfan (45 mg/kg) was administered intraperitoneally. Mice in groups IV an

A rare olive compound oleacein functions as a TrkB agonist and mitigates neuroinflammation both in vitro and in vivo.

BACKGROUND: Neuroinflammation is widely acknowledged as a characteristic feature of almost all neurological disorders and specifically in depression- and anxiety-like disorders. In recent years, there has been significant attention on natural compounds with potent anti-inflammatory effects due to their potential in mitigating neuroinflammation and neuroplasticity. METHODS: In the present study, we aimed to evaluate the neuroprotective effects of oleacein (OC), a rare secoiridoid derivative found in extra virgin olive oil. Our goal was to explore the BDNF/TrkB neurotrophic activity of OC and s

Toll-like receptor 9-positive plasmacytoid dendritic cells promote Th17 immune responses in oral lichen planus stimulated by epithelium-derived cathepsin K.

Oral lichen planus (OLP) is a chronic inflammatory disease associated with T cell infiltration. The crosstalk between oral epithelium and mucosal T cells was considered to be crucial in the pathogenesis of OLP. Here, we selectively extracted the normal epithelium (NE) and lesional epithelium (LE) of buccal mucosa specimens from three patients with OLP by laser capture microdissection due to identify the pathogenic factors. Cathepsin K (CTSK) was identified as one of common upregulated genes in the LE by DNA microarray. Immunohistochemically, CTSK was distinctly detected in and around the LE, w

Dominant effect of a single amino acid mutation in the motor domain of myosin VI on hearing in mice.

An unconventional myosin, myosin VI gene (MYO6), contributes to recessive and dominant hearing loss in humans and mice. The Kumamoto shaker/waltzer (ksv) mouse is a model of deafness resulting from a splice-site mutation in Myo6. While ksv/ksv homozygous mice are deaf due to cochlear hair cell stereocilia fusion at the neonatal stage, the hearing phenotypes of ksv/+ heterozygous mice have been less clear. Due to this splicing error, most MYO6 protein expression is lost in ksv mice; however, MYO6 with a single amino acid mutation (p.E461K) remains expressed. In this study, we investigated the h

Single-cell profiling of immune activation, dysregulation, and reconstitution in rhesus macaques after measles virus infection.

Protective immunity is induced by both, wild-type measles virus (WT MeV) and live-attenuated (LA) measles virus vaccine (LAMV), affording the opportunity to compare common and selective immunological underpinnings of immune protection. Here, peripheral blood mononuclear cells (PBMCs) were collected from rhesus macaques at several timepoints after infection with either virus for single-cell RNA/VDJ sequence analysis. Data revealed acute immune activation at 10/11 days post infection (dpi) with WT MeV, with increased frequencies of CD8 T cells, monocytes and NK cells, and B and T cell expansion

CEBPB is associated with active tumor immune environment and favorable prognosis of metastatic skin cutaneous melanoma.

Metastatic skin cutaneous melanoma (SKCM) is a common malignancy that accounts for low morbidity but high mortality of skin cancer. SKCM is characterized by high lymphocytic infiltration, whereas the states of infiltrated cells are variable in patients leading to a heterogeneous prognosis and hindering appropriate clinical decisions. It is therefore urgent to identify markers associated with lymphocytic infiltration, cellular conditions, and the prognosis of SKCM. In this study, we report that CEBPB, a transcriptional factor, is mainly expressed in macrophages in metastatic SKCM and associated

Runx2 drives Schwann cells repair phenotype switch through chromatin remodeling and Sox2 activation after nerve injury.

BACKGROUND: The states of Schwann cells undergo significant shifts during nerve regeneration. Previous studies have shown the expression of Runx2 is locally upregulated within the affected areas. However, the regulatory mechanisms underlying its epigenetic control remain unclear. METHODS: To investigate the epigenetic mechanisms through which Runx2 influences the phenotypic transition of repair Schwann cells. Runx2 siRNA fragments and Runx2 overexpression plasmids were constructed. Healthy adult Sprague-Dawley (SD) rats weighted 100-150 g, regardless of sex, were randomly selected. Following
