Natural and artificial enzymes have emerged as promising candidates for biomedical applications, possessing the potential to address redox imbalances and metabolic disorders in liver diseases. However, their clinical translation remains limited by challenges in biocompatibility, targeted delivery, dose-response balance, and long-term stability. Nanozymes, novel nanomaterials with potent enzyme-like catalytic activity, exhibit promising capacities to overcome these challenges. In this review, we discuss nanozymes applied to liver diseases, including metabolic disorders, tissue repair, hepatic t

Endogenous retroviruses (ERVs), the genomic remnants of ancient retroviral infections, constitute a significant and dynamic component of livestock and poultry genomes. This review synthesizes current knowledge on the dual roles of ERVs, from drivers of genomic instability and disease to essential agents of evolution and innovation in domesticated species. We detail the classification, structure, and life cycle of ERVs, highlighting the distinct genomic landscapes and host-silencing strategies that differentiate avian and mammalian lineages, with poultry genomes maintaining lower ERV loads thro

The human T-cell leukaemia virus type 1 (HTLV-1) is an oncogenic retrovirus responsible for severe diseases, including adult T-cell leukaemia/lymphoma (ATLL) and HTLV-1-associated myelopathy/tropical spastic paraparesis (HAM/TSP). Despite its significant global prevalence, specific direct-acting antiviral therapies remain unavailable. This review comprehensively surveys the HTLV-1 protease (HTLV-1 PR), a pivotal homodimeric aspartic protease essential for viral maturation and infectivity, as a critical antiviral drug target. A detailed analysis of HTLV-1 PR's narrow substrate specificity, inf

INTRODUCTION: Latin America is a paradox in hepatitis B virus (HBV) elimination: aggregate prevalence is low, yet the region is far from WHO and PAHO 2030 targets because the burden is structurally concealed within underserved populations and fragmented health systems.
AREAS COVERED: This review examines regional epidemiology, structural and programmatic barriers to elimination, and a pragmatic framework for populations of concentrated unmet need (indigenous communities, incarcerated and displaced persons, and people living with HIV), alongside perinatal prevention; hepatitis D, a defective s

Japanese encephalitis virus (JEV) remains a major health threat across Asia, yet the contribution of genotype-specific variation in the multifunctional NS5 protein to viral fitness is not fully resolved. This study evaluated how sequence differences among JEV genotypes G1-G5 shape NS5 stability and, in turn, replication potential. A unified in-silico workflow combined physicochemical profiling, residue-level substitution mapping, and atomistic molecular dynamics to compare structural stability and conformational behavior across genotypes, with a focus on substitutions predicted to modulate enz

Hepatitis C Virus (HCV) is a blood borne pathogen that affects around 200 million individuals worldwide. Immunizations against the Hepatitis C Virus are intended to enhance T-cell responses and have been identified as a crucial component of successful antiviral therapy. Nevertheless, attempts to mediate clinically relevant anti-HCV activity in people have mainly failed, despite the vaccines present satisfactory progress. In this study, we used an array of immunoinformatics approaches to design a multiepitope peptide-based vaccine against HCV by emphasizing 6 conserved epitopes from viral prote

Severe acute respiratory syndrome coronavirus type 2 (SARS-CoV-2), first identified in late 2019 as the causative agent of the COVID-19 pandemic, has triggered a global public health crisis. Transmembrane protease serine 2 (TMPRSS2) is one of the key host factors mediating SARS-CoV-2 infection and invasion. The inhibitors against TMPRSS2 emerge as a promising therapeutic strategy for COVID-19 and other potential viral infections. However, the precise mechanism of action for TMPRSS2 inhibitors remains unclear, with studies on structural optimization being notably scarce. In this work, multiple

Commercial development of gene therapeutics often requires transitioning to human payload genes as initial proof-of-concept studies in animal models often use taxa-specific orthologs. Such transitions also provide opportunities to address potential secondary structure and immune-related subsequences as with human Smad7 cDNA, which was optimized by removing several repeats, potential hairpins and negative cis elements. Thermodynamic modeling at or above minimal free energy states revealed substantial improvements in secondary structure with fewer hairpins and improved diversity scores. Serotype

Amid the current multi-country mpox outbreak, analyzing monkeypox virus (MPXV) and vaccinia virus (VACV) genomes is vital for understanding evolutionary processes that may impact vaccine efficacy and design. This study aimed to elucidate the phylogenetic relationships and structural features of viral antigens, which are crucial for developing effective vaccines. By aligning 1903 MPXV genomes from the NCBI Virus repository (released between 2022 and 2024), an increase in phylogenetic diversity was observed compared to previous studies. These genomes were grouped into Clade I (25 genomes) and Cl

The role that neurotrophins, such as nerve growth factor, play in the pathogenesis of neurodegenerative diseases has long been appreciated. However, the neuregulin (NRG) family of growth factors and/or their v-erb-B2 avian erythroblastic leukemia viral oncogene homolog (ERBB) receptors have also been implicated in the pathogenesis of conditions, such as Alzheimer disease (AD), frontotemporal lobar degeneration (FTLD), and amyotrophic lateral sclerosis (ALS). In this review, we consider the structural variability of NRG isoforms generated by alternative RNA splicing, the use of multiple promote

Skin damage caused by excessive UV exposure has gradually become one of the most common skin diseases, leading to desquamation, scab formation, inflammation and even skin cancer. Animal-derived hydrolyzed collagen peptides have been developed to treat UV-damaged skin; however, they have raised severe concerns such as potential viral transmission, random sequences and the lack of a triple helix structure. Nano collagen, a novel type of short collagen, has attracted increasing attention in the mimicking of natural collagen, while its applications in UV-damaged skin treatment remains unexplored.

Non-structural protein 3 (Nsp3) is the largest protein encoded by the coronavirus (CoV) genome. It consists of multiple domains that perform critical functions during the viral life cycle. CoV-Y is the most C-terminal domain of Nsp3, and it exhibits evolutionary conservation across diverse CoVs; however, the exact biological function of CoV-Y remains unclear. Here, we determined the crystal structure of CoV-Y of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) Nsp3 using the single-wavelength anomalous diffraction method. We revealed the interaction between CoV-Y and the host BRCA1

BACKGROUND: Long non-coding RNA (lncRNA) plays a crucial role in various biological processes, and mutations or imbalances of lncRNAs can lead to several diseases, including cancer, Prader-Willi syndrome, autism, Alzheimer's disease, cartilage-hair hypoplasia, and hearing loss. Understanding lncRNA-protein interactions (LPIs) is vital for elucidating basic cellular processes, human diseases, viral replication, transcription, and plant pathogen resistance. Despite the development of several LPI calculation methods, predicting LPI remains challenging, with the selection of variables and deep lea

Coronavirus disease 2019 is a respiratory infectious disease caused by the severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2). Evidence on the pathogenesis of SARS-CoV-2 is accumulating rapidly. In addition to structural proteins such as Spike and Envelope, the functional roles of non-structural and accessory proteins in regulating viral life cycle and host immune responses remain to be understood. Here, we show that open reading frame 8 (ORF8) acts as messenger for inter-cellular communication between alveolar epithelial cells and macrophages during SARS-CoV-2 infection. Mechanistic

LCB1 is a computationally designed 56-mer miniprotein targeting the spike (S) receptor-binding motif of SARS-CoV- 2 with high potent activity (Science, 2020; Cell host microbe, 2021); however, recent studies have demonstrated that emerging SARS-CoV-2 variants are highly resistant to LCB1's inhibition. In this study, we first identified a truncated peptide termed LCB1v8, which maintained the high antiviral potency. Then, a group of lipopeptides were generated by modifying LCB1v8 with diverse lipids, and of two lipopeptides, the C-terminally stearicacid-conjugtaed LCB1v17 and cholesterol-conjuga

Wolbachia are obligate intracellular bacteria that occur in insects and filarial worms. Strains that infect insects have genomes that encode mobile genetic elements, including diverse lambda-like prophages called Phage WO. Phage WO packages an approximately 65 kb viral genome that includes a unique eukaryotic association module, or EAM, that encodes unusually large proteins thought to mediate interactions between the bacterium, its virus, and the eukaryotic host cell. The Wolbachia supergroup B strain, wStri from the planthopper Laodelphax striatellus, produces phage-like particles that can be

BACKGROUND: While antiretroviral therapy (ART) has improved outcomes for people with HIV (PWH), brain dysfunction is still evident. Immune activation and inflammation remain elevated in PWH receiving ART, thereby contributing to morbidity and mortality. Previous studies demonstrated reduced functional and structural changes in PWH; however, underlying mechanisms remain elusive.
METHODS: Our cohort consisted of PWH with ART adherence and viral suppression ( < 50 copies/mL; N = 173). Measurements included immune cell markers of overall immune health (CD4/CD8 T-cell ratio) and myeloid inflammati

SARS-CoV-2 remains a major global public health concern. Antibody waning and immune escape variant emergence necessitate the development of next generation vaccines that induce cross-reactive durable immune responses. T cell responses to SARS-CoV-2 demonstrate higher conservation, antigenic breadth, and longevity than antibody responses. Therefore, we sought to identify pathogen-derived T cell epitopes for a potential peptide-based vaccine. We pursued an approach leveraging: 1) liquid chromatography and tandem mass spectrometry (LC-MS/MS)-based identification of peptides from ancestral SARS-Co

BACKGROUND: Genomic characterization of the dengue virus (DENV) is useful for understanding its molecular evolution, transmission, pathogenicity and infectivity. The DENV genomic RNA encodes three structural proteins, capsid (C) envelope (E) and membrane (M) proteins mediating viral entry and assembly during host infection. The current study aims to explore the DENV serotypes and mutations in the E and M proteins.
METHODS: Twenty-three samples of DENV-positive patients were processed and selected for whole genome sequencing (WGS) from the Punjab Province of Pakistan.
RESULTS: Among the 23 WG

The Lassa virus (LASV) causes Lassa fever, a highly infectious and lethal agent of acute viral hemorrhagic fever. At present, there are still no effective treatments available, creating an urgent need to develop novel therapeutics. Some benzimidazole compounds targeting the arenavirus envelope glycoprotein complex (GPC) are promising inhibitors of LASV. In this study, we synthesized two series of LASV inhibitors based on the benzimidazole structure. Lentiviral pseudotypes bearing the LASV GPC were established to identify virus entry inhibitors. Surface plasmon resonance (SPR) was further used
