Desensitization, inactivation, and the tension-proof safety mechanism of inactivated MscS.

MscS is the main low-threshold tension-activated osmolyte release valve in bacteria. Working alone or together with high-threshold MscL, it regulates turgor and protects cells from mechanical rupture during osmotic down-shock. The channel exhibits complex adaptive behavior, including desensitization and full inactivation, both of which occur at relatively low sub-lytic tension. There is debate over whether the commonly observed non-conductive state of MscS with the tension-sensing helices splayed away from the gate corresponds to the closed or inactivated state. In this work, using specialized

Red Blood Cell Positional History, Sphericity, and Network Architecture Influence Lingering and Partitioning.

The transport and distribution of red blood cells (RBCs) through microvascular networks is central to diverse applications in drug delivery, tissue engineering, and the study of pathological mechanisms. Recent findings highlight two critical phenomena: (i) a history-dependent partitioning effect, wherein the RBC partitioning at each bifurcation is influenced by the positional and configurational state inherited from previous bifurcations and (ii) RBC lingering at the bifurcation apex, which strongly affects downstream flow distribution. However, the interplay between network geometry, RBC sphe

Serum metabolomic profiling by 1H NMR reveals distinct metabolic signatures in diabetic and non-diabetic chronic kidney disease.

Chronic kidney disease (CKD) and Type 2 diabetes mellitus (DM) are global health burdens, with diabetes being the leading cause of end-stage renal disease, both marked by systemic metabolic disruption. This study applied 1H NMR-based serum metabolomics to characterize metabolic alterations in CKD only (n = 31), DM (n = 33), and their comorbid state (CKD_DM; n = 45). A total of 45 polar metabolites and seven lipid signals were quantified, and statistical analysis was performed using MetaboAnalyst 6.0. The CKD_DM profile was not a linear combination of the CKD and DM profiles. Compared with DM,

Effects of adhesion pattern of 2D substrate on cell morphology and migration.

Focal adhesions play critical roles in a variety of cellular behaviors and physiological processes, including cell migration, proliferation, wound healing, and tumor invasion. Although focal adhesions are recognized as key protein signaling and mechanosensory hubs that mediate interactions between the cell and the extracellular matrix (ECM), the mechanisms by which cells sense and respond to ECM geometry at the subcellular level, and how these cues are translated into cell-scale behaviors, remain unclear. In this study, we develop a computational cell model to investigate the effects of adhesi

Core physical principles of allostery.

Allostery is a basic property of proteins that represents the functional coupling of distantly positioned amino acids. In different manifestations, this property underlies signal transmission, cooperative binding, regulation, catalysis, and evolvability-the elementary features of molecular processes in living cells. While thermodynamic models have provided excellent abstract phenomenological descriptions and classifications of allostery, comparable descriptions and classifications at a more microscopic level, where the nature of physical interactions is specified, are lacking. Yeast biomimetic iron‑titanium oxides for electrochemical biosensing.

A yeast cell template technology is described to biomimetically synthesize FeTi oxides. The resulting samples, yeast Fe2O3 and yeast Fe2O3/TiO2, are then employed as active materials for the electrochemical detection of cancer-associated miRNAs. For the perfect matched cancer miR-141, electrochemical analysis shows a main oxidation peak at about +0.4 V, along with a reduction peak at about +0.30 V. For the detection of cancer miR-29c, the introduction of the Ti element is found to stabilize the structure of the resulting materials. These results suggest a potential of the biomimetic FeTi oxide

Transient perinuclear actin rings prevent cell aging and apoptosis via nuclear mechanical protection.

Epithelial tissues in vivo are often subjected to the challenge of mechanical forces and osmotic shock. However, how epithelial cells prevent DNA damage and reduce aging and apoptosis in the presence of these intense external stimuli remains unclear. Here, we found a reversible perinuclear actin ring can be assembled after rapid and significant hypotonic shock or mechanical forces. Combining experiments and simulations, we demonstrate it is merely a transient response of actin network that is regulated by Ca2+-mediated actin disassembly and Arp2/3-dependent actin polymerization. The mechanical

G12 mutations rewire allosteric communication at the Ras-RalGDS interface.

The Ras subfamily is the most extensively studied branch of the Ras superfamily, with 20% of all human tumors having activating mutations in one of the RAS genes. Recent studies have shown that the Ras/RalGDS/Ral pathway plays a more significant role in the progression of Ras-driven colon and pancreatic cancers than the Ras/Raf and Ras/PI3K pathways. In this study, we investigated the interaction between Ras and the Ras/Rap binding domain (RBD) of RalGDS using long-timescale molecular dynamics simulations. The binding free energy of dimerization showed that Rap1-RBD has the strongest interacti

Water concentrations across lipid bilayers by HYSCORE spectroscopy of nitroxides.

Structural organization of cellular membranes and membrane proteins is governed by the hydration water. Distribution of water molecules across lipid bilayers is highly heterogeneous due to the amphiphilic nature of this barrier, consisting of a 3- to 4-nm-thick hydrophobic core and a hydrophilic region formed by lipid polar head groups. Hyperfine sublevel correlation (HYSCORE) spectroscopy is capable of selective detection of hyperfine interactions originating from water molecules H-bonded directly to the N‒O⋅ group of the nitroxide. Here, we describe a high-spatial-resolution method to measur

AFM-Fold: Rapid reconstruction of protein conformations from AFM images.

High-speed atomic force microscopy (HS-AFM) enables direct visualization of protein dynamics under near-physiological conditions, yet its intrinsic limitation to surface topography prevents atomic-level structural characterization. We present AFM-Fold, a generative AI-based framework that reconstructs three-dimensional protein conformations directly from AFM images. AFM-Fold combines a group-equivariant convolutional neural network, which extracts low-dimensional collective variables (CVs) from AFM images, with a guided diffusion process that generates conformations consistent with the inferre

Identification of hot spot residues in the FusB:EF-G binding interface.

With a lack of new antimicrobials recently entering the clinic, another tactic to combat antimicrobial resistance is to inhibit the resistance mechanisms to existing antibiotics, prolonging the usefulness of these important drugs. One key clinical antibiotic is fusidic acid which inhibits bacterial protein synthesis by binding to EF-G on the ribosome and preventing its release, stalling protein synthesis. The FusB-family of proteins bind to EF-G and confer fusidic acid resistance by promoting the dissociation of these stalled complexes. This resistance mechanism could be inhibited by preventin

Chlorpromazine inhibits EAG1 channels by altering the interdomain coupling.

EAG1 depolarization-activated potassium selective channels are important targets for treatment of cancer and neurological disorders. EAG1 channels are formed by a tetrameric subunit assembly with each subunit containing an N-terminal Per-Arnt-Sim (PAS) domain and C-terminal cyclic nucleotide-binding homology (CNBH) domain. The PAS and CNBH domains from adjacent subunits interact and form an intracellular tetrameric ring that regulates the EAG1 channel gating, including the movement of the voltage sensor domain (VSD) from closed to open states. Small-molecule ligands can inhibit EAG1 channels b

Optimized tissue sample preparation for cryo-electron tomography using serial lift-out.

Cryo-electron tomography (cryo-ET) can provide invaluable insights into subcellular structures in their native environment at molecular resolution. Cryo-focused ion beam (cryo-FIB) has been widely adopted to obtain thin lamellae of cells suitable for tomographic data acquisition. However, its application to tissue samples faces significant limitations. The larger sample size extends milling time, reducing sample preparation throughput. Furthermore, the need for repeated, intricate lift-out procedures adds considerable time and complexity to the workflow. The serial lift-out technique, which ca

Tau protein differentially affects Piezo1 and Kir2.1 channels in brain capillary endothelial cells.

Accumulation of amyloid-β (Aβ) peptides and Tau proteins in the brain is a hallmark of neurodegeneration. Such build-up forms Aβ plaques and Tau neurofibrillary tangles, both of which are associated with synaptic loss, cognitive decline, and reduced cerebral blood flow in Alzheimer's disease (AD). Two ion channels in brain capillary endothelial cells (ECs)-the inwardly rectifying potassium channel Kir2.1 and the mechanosensitive channel Piezo1-are critical regulators of cerebral blood flow, and both display impaired activity in AD. Whether Aβ and Tau affect these channels remains incompletely

Mechanisms of thrombin inhibition by protein S and the TFPIα-fVshort-protein S complex.

Protein S (PS) is an important anticoagulant implicated in both bleeding and thrombotic disorders, making it a promising drug target. The anticoagulant function of PS arises in part because PS enhances the anticoagulant function of tissue factor pathway inhibitor α (TFPIα). PS has been proposed to circulate in the bloodstream together with TFPIα and a truncated form of factor V (fV-short) in the trimolecular complex, TFPIα-PS-fV-short, which we call protein S complex (PSC). PSC has been proposed to strongly inhibit thrombin production by enhancing the ability of TFPIα to inhibit clotting facto

Invasion dynamics of multifocal breast tumor spheroids in three-dimensional extracellular matrix.

Breast cancer in human patients often occurs as multifocal lesions, and the mechanisms through which primary tumors in proximity interact with each other is not well understood. Here, we study an in vitro model of bifocal breast tumors by embedding tumor spheroids of MDA-MB-231 cells in 3D extracellular matrices (ECMs) made of type I collagen at different concentrations. Combining quantitative experiment, mathematical modeling, and numerical simulation, we show that collective traction force from the spheroids creates dipolar deformation in the ECM. As a result, in the inter-spheroid region, w

Protein structure and coupled solvent dynamics in α-synuclein fibrils under controlled confinement.

The formation of amyloid fibrils by the intrinsically disordered protein α-synuclein in human brain neurons and extracellular tissue is a hallmark of Parkinson disease. Although the stable protein β-structure in α-synuclein fibrils has been determined at high resolution, the status of the dynamically disordered domains and their responses to in vivo confinement and small-molecule interactions remain elusive. We use electron paramagnetic resonance spectroscopy of the spin probe, TEMPOL, colocalized with α-synuclein fibrils, and temperature-controlled, ice boundary confinement to characterize th

Quantifying species-specific binding affinities of transthyretin aggregation inhibitors.

Transthyretin (TTR) amyloidosis is one of the most common forms of systemic amyloidosis, involving deposits of pathogenic TTR aggregates in tissues and organs throughout the body. TTR aggregation is initiated by the dissociation of a native TTR tetramer into a monomeric intermediate, which subsequently misfolds and assembles into insoluble aggregates. Using an efficient 19F-NMR aggregation assay, we previously showed that designed peptide inhibitors can interact with multiple TTR species. However, quantifying species-specific binding affinities has remained difficult because the populations of

Polymorphic structures of rapidly twisting 40-residue amyloid-β fibrils.

Fibrils formed by 40- and 42-residue amyloid-β peptides (Aβ40 and Aβ42) are polymorphic, containing molecular structures that vary with growth conditions in ways that are not fully understood. Here, we use cryoelectron microscopy to characterize the structure of rapidly twisting Aβ40 fibrils, for which the distance between apparent width minima in electron microscope images ("cross-over distances") is approximately 25 nm. From samples grown under a single set of growth conditions, we obtain high-resolution structures for three different rapidly twisting polymorphs. Although their cross-over di
