Fundamental insight into the effect of carbodiimide crosslinking on cellular recognition of collagen-based scaffolds.

UNLABELLED: Research on the development of collagen constructs is extremely important in the field of tissue engineering. Collagen scaffolds for numerous tissue engineering applications are frequently crosslinked with 1-ethyl-3-(3-dimethylaminopropyl-carbodiimide hydrochloride (EDC) in the presence of N-hydroxy-succinimide (NHS). Despite producing scaffolds with good biocompatibility and low cellular toxicity the influence of EDC/NHS crosslinking on the cell interactive properties of collagen has been overlooked. Here we have extensively studied the interaction of model cell lines with collage

"You Shall Not Pass"-tight junctions of the blood brain barrier.

The structure and function of the barrier layers restricting the free diffusion of substances between the central nervous system (brain and spinal cord) and the systemic circulation is of great medical interest as various pathological conditions often lead to their impairment. Excessive leakage of blood-borne molecules into the parenchyma and the concomitant fluctuations in the microenvironment following a transient breakdown of the blood-brain barrier (BBB) during ischemic/hypoxic conditions or because of an autoimmune disease are detrimental to the physiological functioning of nervous tissue

Defining the cellular and molecular mechanisms of toxicant action in the testis.

A symposium was held at the 41st annual meeting of the Society of Toxicology with presentations that emphasized novel molecular and cellular pathways that modulate the response to testicular toxicants. The first two presentations described cellular alterations after exposure to the Sertoli cell toxicant, mono-(2-ethylhexyl) phthalate (MEHP). The expression of flamingo1, a G protei coupled receptor family member that may couple cell-cell adhesion to G protein-dependent signaling in Sertoli cells, was rapidly altered after MEHP exposure. Sertoli cell associated flamingo1 immunostaining was redis

Biliary epithelial cells regulate autoreactive T cells: implications for biliary-specific diseases.

The biliary epithelial cell (BEC) is the target for several human immune mediated liver diseases, including primary biliary cirrhosis, but it is not always clear whether the BEC functions as an accessory cell or an antigen presenting cell, although it is well documented that BECs express high levels of human leukocyte antigen Class II, intercellular adhesion molecule-1, and lymphocyte function-associated antigen-3. To examine this issue, we established autoreactive T-cell clones from human leukocyte antigen-DR53 patients with primary biliary cirrhosis and characterized BEC function as a functi

Distinct homing pathways direct T lymphocytes to the genital and intestinal mucosae in Chlamydia-infected mice.

Immunity to genital tract infection with Chlamydia trachomatis is mediated by type 1 CD4+ T lymphocytes. To define the signals that govern lymphocyte trafficking to the genital mucosa, integrins expressed by infiltrating T cells and endothelial addressins displayed on local vasculature were characterized during the course of infection. All T cells expressed the alphaLbeta2 heterodimer that binds vascular ICAM-1, and most displayed enhanced levels of the alpha4beta1 integrin that interacts with VCAM-1. AlphaE and beta7(low) integrin chains were detected on approximately 15 and 30% of infiltrati

HIV infection of human brain capillary endothelial cells--implications for AIDS dementia.

We have demonstrated that human brain capillary endothelial (HBCE) cells, unlike umbilical or aortic endothelial cells are permissively infected by HIV. HIV infection of HBCE cells is noncytolytic and is mediated by a CD4- and GalCer-independent mechanism, implying that HBCE cell tropic strains utilize a unique receptor. The V3 loop of gp120 appears to be important in this reaction. T-cell tropic but not brain-derived macrophage tropic HIV strains selectively infect brain endothelium suggesting that T-cell tropism is important for HIV entry through the blood-brain barrier (BBB). The ability of

Magnetic brain stimulation upregulates adhesion and prevents Eae: MMP-2, ICAM-1, and VCAM-1 in the choroid plexus as a target.

Clinical signs appearance and significant increases of ICAM-1 and MMP-2 expressions with the clusters of VCAM-1(+) immunoreactivity in the choroids plexus epithelium to transferred anti-myelin oligodendroglial antibodies into the third brain ventricle, indicate important role of choroids plexus in the induction of acute experimental autoimmune encephalomyelitis (EAE). Magnetic brain stimulation with AKMA micro-magnet flux density of 60 miliTesla, 5 mm in diameter, implanted upon the pineal gland (PG), immediately after antibody injection, significantly decreases the expression of MMP-2 and ICA

CUB-domain-containing protein 1 overexpression in solid cancers promotes cancer cell growth by activating Src family kinases.

The transmembrane glycoprotein, CUB (complement C1r/C1s, Uegf, Bmp1) domain-containing protein 1 (CDCP1) is overexpressed in several cancer types and is a predictor of poor prognosis for patients on standard of care therapies. Phosphorylation of CDCP1 tyrosine sites is induced upon loss of cell adhesion and is thought to be linked to metastatic potential of tumor cells. Using a tyrosine-phosphoproteomics screening approach, we characterized the phosphorylation state of CDCP1 across a panel of breast cancer cell lines. We focused on two phospho-tyrosine pTyr peptides of CDCP1, containing Tyr707

Triple Helical-Peptides: Collagen-Mimicking Tools for Tissue Engineering.

Reproducing the biological cues naturally provided by collagen is a promising yet challenging approach to influence cell behavior in the field of tissue engineering. To mimic collagen, biomimetic peptides that self-assemble into the characteristic triple helix structure of collagen have been functionalized to biomaterials designed for tissue repair. These triple-helical peptides (THPs) contain recognition motifs for collagen-binding proteins (receptors expressed on the cell surface or secreted proteins), thus replicating the collagen interactions occurring in native tissues and triggering spec

Single-cell transcriptomics reveals functionally specialized vascular endothelium in brain.

The blood-brain barrier (BBB) limits the entry of leukocytes and potentially harmful substances from the circulation into the central nervous system (CNS). While BBB defects are a hallmark of many neurological disorders, the cellular heterogeneity at the neurovascular interface, and the mechanisms governing neuroinflammation are not fully understood. Through single-cell RNA sequencing of non-neuronal cell populations of the murine cerebral cortex during development, adulthood, ageing, and neuroinflammation, we identify reactive endothelial venules, a compartment of specialized postcapillary en

The hallmarks of CMV-specific CD8 T-cell differentiation.

Upon cytomegalovirus (CMV) infection, large T-cell responses are elicited that remain high or even increase over time, a phenomenon named memory T-cell inflation. Besides, the maintained robust T-cell response, CMV-specific T cells seem to have a distinctive phenotype, characterized by an advanced differentiation state. Here, we will review this "special" differentiation status by discussing the cellular phenotype based on the expression of CD45 isoforms, costimulatory, inhibitory and natural killer receptors, adhesion and lymphocyte homing molecules, transcription factors, cytokines and cytot

Preclinical evaluation of the antineoplastic efficacy of 7-(2-hydroxyethyl)theophylline on melanoma cancer cells.

Differentiation-based therapeutics are an underutilized but potentially a significant option for cancer treatment. The effect of methylxanthines on melanoma cell differentiation has been well documented. We report the in-vitro and in-vivo anticancer potential of a theophylline analogue, 7-(2-hydroxyethyl)theophylline (HET), on murine B16-F10 and human Sk-Mel 110 metastatic melanoma cell lines. The effects on cell proliferation were related to the induction of differentiation, demonstrated as increased intracellular transglutaminase activity. The involvement of this methylxanthine in the contro

Sickle cell disease at the dawn of the molecular era.

Recent investigations have identified a syndrome of hemolysis-associated vasculopathy in patients with sickle cell disease (SCD), which features severe hemolytic anemia that leads to scavenging of nitric oxide (NO) and its biochemical precursor arginine. This diminished bioavailability of NO promotes several clinical sequelae, which includes pulmonary hypertension, cutaneous leg ulceration, priapism, and ischemic stroke. Additional correlates of this vasculopathy include activation of endothelial cell adhesion molecules and leukocytes, as well as oxidative stress-related pathways. Some known r

Production of recombinant extracellular domains of canine desmoglein 1 (Dsg1) by baculovirus expression.

The aim of this study was to generate a recombinant protein to represent the entire extracellular domain of canine desmoglein 1 (Dsg1), a desmosomal cell-cell adhesion molecule, by the baculovirus expression system. Cotransfection of a baculovirus transfer vector containing the cDNA for the entire extracellular domain of canine Dsg1 with baculovirus DNA into insect cells resulted in the secretion of soluble canine Dsg1 into insect culture supernatants. Immunoreactivity of 11 human pemphigus foliaceus (PF) sera against the cell surface of canine keratinocytes was completely removed when the ser

Effects of ethanol on the cytoskeleton of migrating and differentiating neural crest cells: possible role in teratogenesis.

Neural crest mesenchyme participates in the formation of craniofacial structures that are malformed in the fetal alcohol syndrome (FAS). We studied the effects of continuous ethanol treatment (0.05%, 0.10%, 0.15%, 0.20%) on developing neural crest cells in vitro. These cells migrate, but many fail to develop their usual arborized dendrites. Exposure of well differentiated dendritically arborized cells to ethanol only on day 6 for 2 hr and 20 min results in rapid cell retraction and alteration in cell-to-cell contacts. Longer treatment causes loss of substratum adhesion. Monoclonal antibodies a

Anemoside B4 upregulates DDAH2 and attenuates oxidative stress induced endothelial dysfunction.

Endothelial dysfunction, involving diminished nitric oxide (NO) bioavailability and vascular inflammation, plays a pivotal role in cardiovascular disease progression. Anemoside B4 (AB4), a major bioactive component extracted from Pulsatilla chinensis, has been reported to exert anti-inflammatory, anti-tumor, and antiviral effects. However, its potential role in regulating endothelial function remains largely unexplored. Integrating RNA-seq analysis, we revealed that AB4 may exert anti-atherosclerotic effects by upregulating DDAH2 expression and improving endothelial NO production. Using an H2O

ITGA11 regulates lens epithelial-mesenchymal transition by modulating PTGS2-mediated lipid metabolism.

PURPOSE: Anterior subcapsular cataract (ASC) is driven by lens epithelial cell (LEC) epithelial-mesenchymal transition (EMT), yet the upstream regulators and metabolic programs remain incompletely understood. This study aimed to identify key drivers of lens fibrosis and define their downstream signaling and metabolic mechanisms. METHODS: Transcriptomic datasets from human ASC lens capsules and TGF-β2-induced LEC EMT models were re-analyzed, together with targeted lipidomic profiling. ITGA11 was silenced in LECs using siRNA, and PTGS2(COX-2) activity was inhibited using the COX-2 inhibitor cel

Circulating Endothelial-Associated Biomarker Responses During Beating-Heart Partial Versus Cardioplegic Total Cardiopulmonary Bypass in Juvenile Pigs: An Exploratory Comparative Study.

Cardiopulmonary bypass (CPB) alters blood flow patterns and may induce endothelial activation; however, the effects of different CPB configurations on endothelial-associated biomarkers remain unclear. In this exploratory pilot study, we compared circulating biomarkers and tissue immunohistochemical findings between beating-heart partial CPB and cardioplegic total CPB in juvenile pigs. Ten 8-week-old female crossbred piglets underwent partial CPB with continued cardiac beating (n = 5) or total CPB with aortic cross-clamping and cardioplegic arrest (n = 5) for 60 min. Investigating the Cellular Activity and Differential Gene Expression of Human Astrocytes in Interaction with Protein Composite Nanofibers.

Collagen is a major extracellular matrix component, and soy protein has been reported to influence cellular and immune-related processes. Nanofiber scaffolds incorporating collagen and soy protein isolate (SPI) may provide a platform for modulating cell-material interactions in neural systems. In this study, we fabricated electrospun nanofibers composed of collagen (CO), SPI, and polycaprolactone (PCL) and investigated the cellular and transcriptional responses of human astrocytes to these scaffolds in vitro. The nanofibers were characterized by scanning electron microscopy (SEM), Fourier tran
