Imatinib induces autophagy through BECLIN-1 and ATG5 genes in chronic myeloid leukemia cells.

Imatinib is a chemotherapeutic drug used for the treatment of chronic myeloid leukemia (CML). Recent data showed imatinib-induced cell death in various types of cancers. Autophagy is the physiological process in which cellular components are broken down by the lysosomal activation. In this study, we aimed to examine the effects of imatinib on autophagy in addition to apoptosis in CML cells. Results suggested that imatinib induces autophagy in CML cells through inducing over-expression of BECLIN-1 and ATG5 genes with the statistical significance. Our results demonstrated that autophagy might be

Apoptosis and the assessment of radiation injury.

Apoptosis is a necessary form of cell death for all multicellular organisms and is controlled by the action of genes that initiate, facilitate or inhibit the process. In addition to physiological triggers, apoptosis can follow cellular injury, such as exposure to ionizing radiation. Cell death following radiation serves as a means by which the possibility of cells surviving with potentially harmful genomic damage can be reduced. Apoptosis can be identified in tissues shortly after whole body radiation; but apoptotic cells are rapidly cleared by phagocytosis, and apoptosis is unlikely to serve

Ethanol-induced disturbances of gliogenesis and neuronogenesis in the developing murine brain: an in vitro and in vivo immunohistochemical and ultrastructural study.

The effects of ethanol on murine brain development were studied in a model of whole embryo culture and in vivo. Ethanol locally enhances cell death in the primitive neuroepithelium. During neuronal migration, ethanol induces a premature transformation of the radial glial guides into astrocytes. Ethanol also inhibits the late gliogenesis. The resulting postmigratory neocortex displays an abnormal neuronal pattern almost completely deprived of vertical columnization. These glial-neuronal disturbances can explain neuropathological and clinical features of the fetal alcohol syndrome. Our study pro

Temporal Tracking of Cell Cycle Progression Using Flow Cytometry without the Need for Synchronization.

This protocol describes a method to permit the tracking of cells through the cell cycle without requiring the cells to be synchronized. Achieving cell synchronization can be difficult for many cell systems. Standard practice is to block cell cycle progression at a specific stage and then release the accumulated cells producing a wave of cells progressing through the cycle in unison. However, some cell types find this block toxic resulting in abnormal cell cycling, or even mass death. Bromodeoxyuridine (BrdU) uptake can be used to track the cell cycle stage of individual cells. Cells incorporat

Malaria, mummies, mutations: Tutankhamun's archaeological autopsy.

The cause of death of the Egyptian pharoah Tutankhamun has now for decades been matter of speculation and various hypotheses. A recent article in the Journal of the American Medical Association (JAMA) provided new evidence and suggested malaria, together with Köhler’s disease, as the most probable cause of death of the boy king. We are sceptical towards this elucidation of the cause of death of King Tut and discuss alternative and differential diagnoses, among them, in particular, sickle cell disease and Gauche’s disease.

Nanocomposites integrating antigen release and presentation for treatment of sarcoma.

Soft-tissue sarcomas present a significant clinical challenge owing to their high postoperative recurrence rates and poor responsiveness to radiotherapy and chemotherapy. The insufficient infiltration of immune cells into the tumor microenvironment further limits the efficacy of immunotherapy. Photodynamic therapy (PDT) can induce immunogenic cell death (ICD) in tumor cells by releasing tumor-associated antigens. These antigens can be used to activate dendritic cells (DCs) in vitro and extract DC vesicles. These vesicles can activate T cells in a more effective and specific manner, promoting t

PlatinumIV Complex Enabling Multiple and Potent Tumor Microenvironment Remodeling for Cancer Chemo-Immunotherapy.

Tumor microenvironment not only compromises the therapeutic efficacy of chemotherapy but also weakens chemotherapy-induced immunogenic cell death. Herein, we report novel platinumIV prodrugs that integrate two clinically approved drugs with different but complementary mechanisms, where cisplatin provides tumor cytotoxicity while the pirfenidone analogue contributes to tumor microenvironment modulation. In vitro evaluations suggested their superior anticancer activity, improved resistance profiles, and favorable selectivity across multiple cancer cell lines. Mechanistic studies revealed that th

Neuro-ophthalmic Complications of Immune-Checkpoint Inhibitors.

Immune checkpoint inhibitors (ICIs) have revolutionized the field of oncology by modulating the immune cell-cancer cell interaction and thereby promoting immune system disinhibition in order to target several types of malignancies. There are three classes of immune checkpoint inhibitors (ICIs): anti-cytotoxic T-lymphocyte associated antigen 4 (CTLA-4), anti-programmed cell death protein-1 (PD-1), and anti-programmed cell death ligand-1 (PD-L1).It is not uncommon for physicians across all specialties to encounter a patient with a history of malignancy and ICI exposure, necessitating familiarity

Differential impact of bortezomib on HL-60 and K562 cells.

Bortezomib (PS-341, or Velcade), reversible inhibitor of 20S proteasome approved for the treatment of multiple myeloma and mantle cell lymphoma, exhibited a cytotoxic effect toward other malignancies including leukaemia. In this study, we have documented that incubation of both HL-60 and K562 leukaemia cells with nanomolar concentrations of bortezomib is associated with the death of HL-60 cells observed within 24 hours of incubation with bortezomib and the death of K562 cells that were observed after 72 hours of incubation with bortezomib. The relative resistance of K562 cells to bortezomib co

E3 ubiquitin ligase-mediated necroptosis: regulatory mechanisms and therapeutic implication in cancer.

Necroptosis, a form of regulated cell death, has attracted significant attention as a potential therapeutic target for cancer. Although necroptosis is regulated at various levels, ubiquitination is key to post-translational regulation of necroptotic cell death. E3 ubiquitin ligases are the most remarkable enzymes in the ubiquitination modification, responsible for recognizing and ubiquitinating specific substrates. In this review article, we will explain how E3 ubiquitin ligases influences necroptosis, as well as to discuss what is the present state of the knowledge about the role of E3 ubiqui

Brachial Plexitis during Maintenance Atezolizumab after Palliative Radiotherapy for Cervical Spine Metastasis in Lung Cancer.

We present the case of a patient with lung cancer who developed brachial plexitis following treatment with immunochemotherapy, which included atezolizumab and palliative radiotherapy for a metastatic cervical spine tumor. This case highlights the importance of considering immune-related adverse events (irAEs) when initiating immune checkpoint inhibitor therapy, in addition to other recognized causes such as radiation, tumor compression and invasion, and paraneoplastic syndromes. While brachial plexitis has been reported as an irAE associated with programmed cell death 1 (PD-1) antibodies, this

Pembrolizumab-Induced Pancreatitis: Take It With a Grain of Salt.

Keytruda (pembrolizumab) is an immunomodulator that prevents the interaction between programmed cell death protein (PD-1) and programmed death ligand (PD-L1/2) on immune cells and tumour cells, thereby preventing T cell dysfunction. At times, mounting a strong immune response against tumour cells may not spare normal cells, leading to a variety of multisystemic adverse effects. With this, we present a case of a 64-year-old male who developed acute pancreatitis after completing eight cycles of Keytruda for castrate-resistant metastatic prostate cancer for six months, after all other causes of p

Hypothyroidism After Use of Immune Checkpoint Inhibitor Therapy in Patient With Graves' Disease: Cure?

Immune checkpoint inhibitors (ICIs) are frequently used as treatment for many malignancies. Immune-related adverse events (irAEs) due to use of ICIs are common. Thyroid involvement is the most common endocrine irAE. Here, we present an unusual case of Graves' disease potentially cured due to destructive thyroiditis caused by inflammation due to ICIs. Thyroid irAEs are more common with programmed cell death protein-1 (PD-1) inhibitor or programmed cell death-ligand 1 (PD-L1) inhibitors than cytotoxic T-lymphocyte-associated protein 4 (CTLA-4) inhibitors. Baseline and serial monitoring of thyroi

Detecting Necroptosis in Virus-Infected Cells.

Necroptosis has been implicated as a critical cell death pathway in cancers, Alzheimer's and other neurodegenerative diseases, and virus-infected cells. Necroptosis occurs when mixed-lineage kinase domain-like protein (MLKL) punctures the cytoplasmic membrane allowing a rapid influx of water leading to a loss of cellular integrity. As its role in human disease becomes apparent, methods identifying necroptosis will need to be further developed and optimized. Here we describe identification of necroptosis through quantifying cell death with pathway inhibitors and using western blots to identify

De Novo Myasthenia Gravis Induced by Atezolizumab in a Patient with Urothelial Carcinoma.

The programmed cell death ligand-1 antibody, atezolizumab, is an immune checkpoint inhibitor approved for the treatment of various cancers. Herein, we describe a case of an 87-year-old man with advanced urothelial carcinoma. After surgery, atezolizumab was given. Subsequently, he developed generalized myasthenia gravis (MG) with elevated creatinine kinase and positive anti-striated muscle antibody. Although intravenous immunoglobulin was started, the patient developed cardiac arrhythmia and arrest. Our case not only reported MG as an immune-related adverse event of atezolizumab but also emphas

Neurodegenerative disease: amyloid pores from pathogenic mutations.

Alzheimer's and Parkinson's diseases are associated with the formation in the brain of amyloid fibrils from beta-amyloid and alpha-synuclein proteins, respectively. It is likely that oligomeric fibrillization intermediates (protofibrils), rather than the fibrils themselves, are pathogenic, but the mechanism by which they cause neuronal death remains a mystery. We show here that mutant amyloid proteins associated with familial Alzheimer's and Parkinson's diseases form morphologically indistinguishable annular protofibrils that resemble a class of pore-forming bacterial toxins, suggesting that i

Immune classification of advanced melanoma identifies non-responders to anti-PD1 therapy.

BACKGROUND: Immunotherapy based on anti-PD1 inhibitors has significantly improved survival in advanced melanoma. However, a significant proportion of patients do not benefit, and predicting response to immunotherapy remains an area of unmet need. Our group previously defined an immune signature able to predict response to anti-PD1 inhibitors in this scenario. METHODS: In this study, we analyzed two cohorts of patients with advanced melanoma treated with anti-PD1 inhibitors: the GEM cohort, previously used to validate our immune signature, and Campbell's cohort, which contains data about diffe

Understanding the Therapeutic Potential of PD-1 Agonism in Inflammatory and Autoimmune Disorders.

The therapeutic potential of programmed cell death-1 (PD-1) agonism is increasingly recognized as a cornerstone for restoring immune tolerance in autoimmune diseases. However, while PD-1 antagonism has revolutionized oncology, a critical knowledge gap remains regarding optimal strategies to therapeutically harness PD-1 activation. This review provides a comprehensive analysis of the mechanical and molecular requirements for inducing PD-1 signaling, addressing the current lack of a unified framework for agonistic drug development. We first delineate the fundamental biology of PD-1 signaling in
