Poor drug penetration into tumor cells and tissues is a worldwide difficulty in cancer therapy. A strategy is developed for virion-like membrane-breaking nanoparticles (MBNs) to smoothly accomplish tumor-activated cell-and-tissue dual-penetration for surmounting impermeable drug-resistant cancer. Tailor-made dendritic arginine-rich peptide prodrugs are designed to mimic viral protein transduction domains and globular protein architectures. Attractively, these protein mimics self-assemble into virion-like nanoparticles in aqueous solution, having highly ordered secondary structure. Tumor-specif

Clinically, irreversible pulpitis is treated by the complete removal of pulp tissue followed by replacement with artificial materials. There is considered to be a high potential for autologous transplantation of human dental pulp stem cells (DPSCs) in endodontic treatment. The usefulness of DPSCs isolated from healthy teeth is limited. However, DPSCs isolated from diseased teeth with irreversible pulpitis (IP-DPSCs) are considered to be suitable for dentin/pulp regeneration. In this study, we examined the stem cell potency of IP-DPSCs. In comparison with healthy DPSCs, IP-DPSCs expressed lower

Dental pulp stem cells, or DPSC, are neural crest-derived cells with an outstanding capacity to differentiate along multiple cell lineages of interest for cell therapy. In particular, highly efficient osteo/dentinogenic differentiation of DPSC can be achieved using simple in vitro protocols, making these cells a very attractive and promising tool for the future treatment of dental and periodontal diseases. Among craniomaxillofacial organs, the tooth and salivary gland are two such cases in which complete regeneration by tissue engineering using DPSC appears to be possible, as research over the

Vitiligo is an acquired depigmenting disorder that affects 0.5% to 2% of the world population. Three different forms are classified according to the distribution of lesions; namely non-segmental, segmental and mixed vitiligo. Vitiligo is associated with polymorphisms in genes involved in the immune response and in melanogenesis. However, environmental factors are required for the development of manifest disease. In general, the diagnosis is clinical and no laboratory tests or biopsies are required. Metabolic alterations are central to current concepts in pathophysiology. They induce an increas

The success of the anti-CD20 monoclonal antibody rituximab in the treatment of lymphoid malignancies provided proof-of-principle for exploiting the immune system therapeutically. Since the FDA approval of rituximab in 1997, several novel strategies that harness the ability of T cells to target cancer cells have emerged. Reflecting on the promising clinical efficacy of these novel immunotherapy approaches, the FDA has recently granted 'breakthrough' designation to three novel treatments with distinct mechanisms. First, chimeric antigen receptor (CAR)-T-cell therapy is promising for the treatmen

For cancer therapy, optimization of carrier features is necessary to effectively deliver the targeting agents to tumor sites. Biodegradable poly(ether-anhydrides) micelles with filamentous, rod-like, and spherical shapes are fabricated. Their size and morphology are characterized by AFM and TEM. The encapsulation of doxorubicin hydrochloride (DOX) into the micelles does not impact their shape. The effect of micellar shape on the drug loading capacity and encapsulation efficiency, as well as in vitro drug release, is investigated. The cellular uptakes are evaluated using fluorescence microscopy

Chronic beryllium disease is an occupational lung disease that begins as a cell-mediated immune response to beryllium. Although respiratory and engineering controls have significantly decreased occupational beryllium exposures over the last decade, the rate of beryllium sensitization has not declined. We hypothesized that skin exposure to beryllium particles would provide an alternative route for sensitization to this metal. We employed optical scanning laser confocal microscopy and size-selected fluorospheres to demonstrate that 0.5- and 1.0- micro m particles, in conjunction with motion, as

BACKGROUND AIMS: Reference genes are an essential part of clinical assays such as droplet digital polymerase chain reaction (ddPCR), which measure the number of copies of vector integrated into genetically engineered cells and the loss of plasmids in reprogrammed cells used in clinical cell therapies. Care should be taken to select reference genes, because it has been discovered that there may be thousands of variations in copy number from genomic segments among different individuals. In addition, within the same person in the context of cancer and other proliferative disorders, substantial pa

Three-dimensional (3D) bioprinting of hydrogels offers great promise in tissue engineering, as it enables precise fabrication of cell-laden scaffolds with complex architectures. The formulation and rheological tuning of bioinks are critical for successful printing and scaffold performance. While considerable efforts have focused on optimizing composition and crosslinking chemistry, the influence of time on material behavior, such as pre-printing aging, remains insufficiently studied. In this study, we developed dual-crosslinked chitosan-polyacrylic acid (CSAA) hydrogels with varied aging times

Artificial intelligence (AI) strategies are revolutionizing genomics by extracting complex patterns that traditional statistical pipelines are likely to miss. This mini-review aims to provide a concise overview of how AI is transforming major genomic technologies including variant calling, gene expression analysis, single-cell transcriptomics, CRISPR-Cas9 optimization, and multi-omics integration. In genome sequencing, machine learning variant callers greatly improve the accuracy and the rate at which single nucleotide and structural variants are called. In bulk RNA-Seq, AI augmented quantific

Mesenchymal stem cell-derived exosomes (Exo) have emerged as potent therapeutic agents for skin regeneration; however, their clinical translation is often impeded by poor colloidal stability and the formidable barrier of the stratum corneum. In this study, we developed a novel "bio-shuttle" system by engineering chitosan-shielded exosomes (cExo) via precise electrostatic surface assembly. The optimized cExo exhibited a uniform hydrodynamic diameter of 216 nm and a robust positive surface charge of +30.7 mV, ensuring superior structural integrity. To evaluate their transdermal potential, we emp

A major limitation across nanoparticle delivery platforms is inefficient intracellular delivery following cellular uptake, often resulting from sequestration within endosomal compartments. Here, we show that peptide architecture can influence intracellular trafficking and reduce accumulation within early endosomal compartments in lipid-protein nanocarriers. Specifically, we fuse R6W3 (RRWWRRWRR), an amphipathic cell penetrating peptide, to the N- or C-terminus of the nanodisc scaffold proteins and systematically evaluate its impact on membrane interactions and cellular behavior. Structural and

OBJECTIVE: There is presently no cure for osteoarthritis (OA). Despite the promise of mesenchymal stem cell (MSC) transplantation, issues such as low cell survival and engraftment have shifted research toward MSC-derived exosomes (MSC-EXOs). Because they mirror the immunomodulatory and tissue-regenerative actions of MSCs without the safety and logistical issues of cell-based therapies, MSC-EXOs constitute an emerging class of medicines. This paper compares and contrasts MSCs and MSC-EXOs for OA treatment.
MATERIALS AND METHODS: A literature search was conducted in PubMed, Google Scholar, Web

Corneal blindness remains a major clinical challenge, yet donor grafts are scarce. Bioprinting has emerged in recent decades to potentially overcome donor shortage. In bioprinting, collagen is a common biomaterial. However, one alternative biomaterial, which has shown promising results in corneal tissue engineering, is keratin. Therefore, human hair keratin was investigated in this study as a bioink component for stroma bioprinting. Two keratin preparations, an aqueous extraction and an alkaline extraction, were incorporated into a methacrylated hyaluronic acid bioink and compared with a colla

Pyroptosis is a proinflammatory form of regulated cell death across the tree of life. The gasdermin (GSDM) protein family functions as an evolutionarily conserved executioner of pyroptotic cell death by forming membrane pores. As the canonical activation mechanism, proteolytic cleavage drives the formation of GSDM pores. In contrast to the well-studied modern GSDM-directed pyroptotic pathway, biochemical mechanisms of primitive GSDM pores are poorly understood. Here, we functionally characterize two primitive GSDM variants with cytotoxicity, one of which is a bacterial homolog (termed bGSDM) f

Although lifespan has long been the focus of ageing research, preventing functional decline late in life is a more pressing societal need. Here, we investigate the basis of senescence and declining fitness during replicative ageing in budding yeast, and describe a metabolic perturbation that preserves late-life fitness even on an unrestricted glucose diet. We show that senescence can be prevented by constitutive activation of AMPK, though only for approximately half the ageing population, and use genetic and functional assays to link this heterogeneous response with differences in cytosolic ac

BACKGROUND AND OBJECTIVE: In vitro culture systems are widely used in cell-based research, yet they predominantly rely on flat substrates that fail to capture the curved mechanical landscapes encountered by cells in vivo. While curved substrates offer enhanced physiological relevance, their adoption is often limited by fabrication complexity and cost. Here, we investigate whether flat, soft substrates can reproduce the nuclear stress profiles induced by rigid curved surfaces during cell adhesion.
METHODS: Using an axisymmetric finite element model of a human mesenchymal stem cell (hMSC), we s

Despite the potential of current cancer immunotherapies, tumor cells frequently evade immune surveillance by forming an immunosuppressive microenvironment, leading to treatment resistance. Current inquiry positions the sympathetic nervous system (SNS) at the forefront of tumor immunology as a critical driver of this immune evasion. This review delineates the cellular pharmacology of SNS-mediated immune regulation across the tumor ecosystem. Operating predominantly through the cyclic adenosine monophosphate-protein kinase A (cAMP-PKA) signaling axis, the SNS engages in bidirectional regulation

Ischemic brain injury (IBI), including stroke and cardiac arrest-induced global cerebral injury, presents a significant clinical challenge due to its high morbidity and incidence of neurological deficits. Currently, effective strategies for neurological repair remain limited. Extracellular vesicles (EVs) are a diverse group of cell-derived, lipid-bound nanoparticles that encapsulate RNAs, proteins, lipids, metabolites, growth factors, and cytokines. EVs play an essential role in intercellular communication and are involved in various physiological and pathological processes. Stem cell-derived
