Isolation of spheroplastic forms of Haemophilus influenzae from sputum in conventionally treated chronic bronchial sepsis using selective medium supplemented with N-acetyl-D-glucosamine: possible reservoir for re-emergence of infection.

The isolation rate of Haemophilus influenzae from patients with persistent production of purulent sputum has been increased by the routine use of a selective medium. Nevertheless, some purulent sputum still fails to yield a pathogen. The selective medium was supplemented with N-acetyl-D-glucosamine to encourage primary isolation of colony forming, spheroplastic H influenzae, which reverted to normal forms on subculture. On the basis of in vitro experiments it is postulated that these spheroplastic forms of H influenzae may be induced by inadequate antimicrobial chemotherapy and may be responsi

A clinical and pathological study of inherited glaucoma in New Zealand white rabbits.

The eyes of 7 rabbits with clinical megaloglobus, selected from a large colony, were studied with slit-lamp microscopy, gonioscopy, indirect ophthalmoscopy and tonometry. Selected light and scanning electron microscopy was also performed. There were 11 glaucomatous eyes among the 7 rabbits: gonioscopy demonstrated open iridocorneal angles with absence of pectinate fibres. Incomplete cleavage of the iridocorneal angles in glaucomatous eyes was noted histologically. Because of the similarity of the iridocorneal angles of glaucomatous rabbit eyes with those of human eyes with congenital glaucoma,

The effect of sulfite on the yeast Saccharomyces cerevisiae.

After a short period of tolerance, living cells of Saccharomyces cerevisiae were irreversibly damaged by low concentrations of sulfite. The length of the period of tolerance and the rate of the damaging effect depended on the concentration on sulfite, pH-value, temperature, the physiological state of the cells, and incubation time. Inhibitors of protein synthesis and mitochondrial ATP synthesis did not alter the deleterious effect of sulfite on living cells. Furthermore, cell damage leading to inhibition of colony formation occured under aerobic as well as under anaerobic conditions. Prior to

Syndrome of polydactyly, cleft lip/palate or lingual lump, and psychomotor retardation in endogamic gypsies.

Six children in an inbred isolate (a gypsy colony) were found to have a syndrome of reduplication of the big toes, supernumerary fingers on the hands, cleft lip/palate or lingual nodule, and somatic and psychomotor retardation. Other features sometimes present were absence of olfactory bulbs and tracts, cryptorchidism, inguinal hernia, and congenital heart disease. The disorder has overlapping features with several previously delineated syndromes, but is most similar to the anomalies of trisomy 13 Mand ohr's syndrome. Our patients had a normal karyotype. The mode of inheritance of this lethal

Intracellular multiplication of Legionella pneumophila in cultured human embryonic lung fibroblasts.

Cell cultures of normal human embryonic lung fibroblasts were inoculated with two strains of Legionella pneumophila, the etiological agent of Legionnaires disease. Large numbers of intact organisms, including many dividing forms, were seen within fibroblasts by light and electron microscopy. Intracellular multiplication of organisms was demonstrated by the progressive increase in bacterial colony counts from plated extracts of infected fibroblast cultures in which extracellular multiplication had been eliminated by bactericidal concentrations of antibiotics. It is evident that L. pneumophila c

Further observations on infections of guinea pigs with Venezuelan encephalitis virus strains.

Guinea pigs from a Guatemalan colony died after subcutaneous inoculating of moderately small doses of equine-benign strains of Venezuelan encephalitis (VE) virus of hemagglutination-inhibition subtype I-E from enzootic habitats in Mexico and Guatemala. Thus these guinea pigs were unlike English short hair and inbred 13 guinea pigs, which usually survive infections with equine-benign VE strains of subtype I-E. We therefore caution others that not all strains of guinea pigs can be used to evaluate the potential equine virulence of VE viruses.

Immunogenicity of a chaotropically extracted protective antigen(s) of Pasteurella multocida type A (bovine origin) against experimental pasteurellosis in mice.

Immunization of mice with a potassium thiocyanate extract of Pasteurella multocida type A (bovine strain) imparted complete protection against an experimental challenge infection with 1.6 x 10(3) colony-forming units of the homologous strain. Formalin-treated vaccine (one or two doses) was much less effective. Significantly higher bactericidal antibody titres were observed in the sera of mice immunized with the extract compared with those given the vaccine. The bactericidal activity was localized in the IgM and IgG fractions. The extract consisted of protein(s) and carbohydrate(s) and on immun

Trisomy of 1q in preleukaemia with progression to acute leukaemia.

Marrow cells from an untreated man with preleukaemia were found to have trisomy for the long arm of chromosome 1 which was translocated to the end of the long arm of number 11, i.e., 46,XY, -11,+t (1:11) (q11 or q12; q25). The same abnormality was found in metaphases from 8 individual granulocytic colonies. With development of acute myelomonocytic leukaemia, in addition to the basic chromosome abnormality, trisomy of chromosome 8 and an extra chromosome number 19 with partial deletion were found. Fibroblasts grown from marrow biopsy material showed a normal diploid complement, 46,XY.

Diseases of guinea-pigs.

Information on diseases of guinea-pigs is generally available in research institutes and breeding establishments. When dealing with an individual patient it is tempting to extrapolate from this pool of information, without realising that a number of factors can affect the disease situation in a colony which need not apply to the individual animal kept as a pet--or vice versa. Thus diseases may appear as an explosive outbreak in a group of cavies kept under uniform conditions while a single animal may be exposed to amateur management (Townsend 1975) and suffer in consequence. The diseases gener

[On the growth of various Enterobacteriaceae, Pseudomonas aeruginosa and Alkaligenes spec. in distilled water, de-ionized water, tap water, and mineral salt solution (author's transl)].

The behaviour of 12 strains of gram-negative bacteria was analysed in media having a low content of nutrients. Sterile tap water, de-ionized water, aqua bidestillata and mineral salt solution were used. It was found that Serratia marcescens, Pseudomonas aeruginosa, Enterobacter, Klebsiella and Citrobacter are those with the most frequently observed growth. In the mineral salt solution survived also Providencia, E. coli, Erwinia, Hafnia, and Alkaligenes and reached max. colony numbers of some 100000/ml. When the salts used were added stepwise potassium phosphate and especially magnesium sulfate

Eperythrozoon coccoides. I. Effect on the interferon response in mice.

Eperythrozoon coccoides is a common blood parasite of rodents and the etiological agent of a chronic infection present in many mouse colonies. After primary infection, mice develop a parasitemia and anemia followed by a chronic, latent infection. During the acute phase of infection, mice manifest a striking suppression of interferon production in response to induction with Newcastle disease virus, Chikungunya virus, and poly I:C. These data suggest that the reticuloendothelial system involvement with this agent is associated with impairment of the interferon response. The enhanced susceptibili

Hidden partnerships in the dark: Cold-water coral-fish associations in Fiordland, New Zealand.

The ichthyofauna of the Fiordland ecosystems of southern Aotearoa New Zealand was documented during four remotely operating vehicle (ROV) dives between 100 and 350 m depth. A total of 26 fish species were documented within two fiord basins. An association between cold-water corals, such as the stony coral Madrepora oculata and black coral Antipathella fiordensis, and several fish species was observed. In particular, the observed association between M. oculata and the wavyline perch (Lepidoperca tasmanica) highlighted the use of M. oculata colonies not only as sheltering habitats but also as nu

Current Status and Future Directions in the Pharmacologic Management of Cirrhosis.

In recent years, the necessity for pharmacologic treatments to mitigate the risk of hepatic decompensation has been highlighted, with non-selective beta-blockers identified as the most effective option. However, a significant therapeutic gap persists, and additional or alternative treatments have been proposed, including statins, rifaximin, albumin, anticoagulants, angiotensin receptor blockers, granulocyte-colony stimulating factor, and fecal microbial transplant. According to the data from limited clinical trials, these pharmacotherapies offer promising possibilities; nonetheless, additional

Challenging frontiers: Canine sporotrichosis due to Sporothrix brasiliensis, first case in Chile.

We report the first case of Sporothrix brasiliensis in a dog at Santiago, Chile. From an initial biopsy obtained from the patient, a histopathological study was performed that oriented a fungal infection. The molecular diagnosis from the isolated colony confirmed the identity of the S. brasiliensis agent. Although the literature indicates itraconazole as the first treatment option, the patient was treated with terbinafine followed by itraconazole, without achieving a clinical cure. S. brasiliensis that spread rapidly in Latin America, hence highlight the importance of timely diagnosis that con

Myotonic dystrophy Type 1 (DM1) is a severe inherited neuromuscular disease and is the most prevalent form of muscular dystrophy in adults. DM1 involves not only the striated muscles including skeletal, and cardiac but also other organs such as the eye, brain and gonads. We have generated and characterized 4 adult heterozygous DM1 iPSC lines carrying between 1300 and 1600 CTG repeat expansion in the DM1 protein kinase gene, and a control from an apparently healthy individual. They all show strong pluripotency markers, differentiation capacity, the absence of residual viral vectors as well as n

Chaetomium globosum: Spotting a scarce saprophyte in bacterial-fungal co-infection of the sinuses in a post-COVID-19 patient.

Atypical fungal co-infections in post-COVID-19 patients may have been underreported due to limited diagnostic methods. We present a case of Chaetomium globosum sinusitis in a 55-year-old post-COVID-19 patient with pain in the left side of the face, mimicking rhino-cerebral mucormycosis. CT-paranasal sinuses showed mucosal thickening of left paranasal sinuses, biopsy of which grew a velvety, white colony. It was confirmed as Chaetomium globosum. The patient responded to oral Posaconazole therapy for three months. Prompt identification of atypical fungal agents is critical for appropriate treatm

Generation of an iPSC line (UMGWi001-B) from a patient with Floating-Harbor Syndrome (FLHS) carrying a heterozygous SRCAP mutation (p.Arg2444).

Floating-Harbor syndrome (FLHS) is a rare genetic disease caused by mutations in the SRCAP gene. Here, we generated an induced pluripotent stem cell line from gingival fibroblasts of a male patient with a heterozygous mutation in exon 34 of the SRCAP gene (c.7330C > T, p.Arg2444*). The iPSC colonies have an atypical morphology with diffuse borders and disintegrate quickly upon touch. Still, the cell line expresses pluripotency markers and differentiates into three germ layers. The cell line can be used as patient-specific disease model and help elucidate the molecular mechanisms involving SRCA

Generation of Neural Crest Progenitors from Human Pluripotent Stem Cells.

There are a vast range of diseases and disorders that are neurocristopathic in origin, including Hirschsprung's disease, pheochromocytoma, familial dysautonomia, craniofacial disorders, and melanomas. Having a source of human neural crest cells is highly valuable for investigating potential treatments for such diseases. This chapter describes a robust and well-characterized protocol for deriving neural crest from human pluripotent stem cells (hPSCs), which can then be differentiated to neuronal and non-neuronal lineages. The protocol is adapted to suit hPSC maintenance as a monolayer bulk cult

White Men in Quarantine: Disease, Race, Commerce and Mobility in the Pacific, 1872.

In July 1872, the steamship Hero underwent quarantine at Sydney's North Head after a case of smallpox was diagnosed. This article brings together the histories of quarantine, white subjectivity and Pacific mobility through an analysis of the Loganiana newspaper produced by the passengers of the Hero during their confinement. The Loganiana provides a unique insight into the formation of white identities through discussions of race, commerce, science and inter-colonial politics. The case provides an important perspective on a transformative period in Australia's border history, and also illumina
