Reverse-tracing unravels microbial succession and flavor stratification driven by vertical physicochemical gradients in Zaopei fermentation.

Chinese Baijiu brewing relies on the vertical layering of Zaopei, yet its effect on microbial-flavor interactions remain unclear. This study employs result-oriented backward analysis of distinct flavor patterns to decipher how spatial differences govern metabolic networks and flavor distribution. GC-IMS/GC-MS revealed spatial divergence: the surface layer concentrated 62.89 % of total volatile flavors, significantly higher than the bottom layer (18.88 %) and the middle/upper layers (8.67-9.55 %). Fungal communities played a key role in layer differentiation (p = 0.002) through temperature and

Simultaneous reduction of odorous and greenhouse gases emissions by thermophilic microbial agents during chicken manure composting.

Odorous and greenhouse gases emissions from animal manure composting cause air pollution and nutrient loss. This research examined the effects of thermophilic fungal agent (F) and bacterial agent (B) on the emissions of odorous and greenhouse gases during chicken manure composting and explored the underlying mechanisms. The results indicated that the cumulative emission of ammonia (NH3), hydrogen sulfide (H2S), methane (CH4) and nitrous oxide (N2O) in F treatment decreased by 20.1 %, 34.2 %, 8.3 % and 26.1 %, respectively, in comparison to 25.3 % reduction in H2S and 9.3 % reduction in N2O in

Molecular signaling in biomaterial-induced foreign body response: current perspectives.

The foreign body response (FBR) is an inevitable host response to implanted materials, initiated by tissue injury and marked by a cascade of inflammatory and fibrotic processes. Following implantation, local tissue damage triggers acute inflammation, characterized by immune cell recruitment and activation. Over time, this response advances to a chronic fibrotic phase marked by dense extracellular matrix deposition and fibrous capsule formation, which can encapsulate and, in some cases, functionally isolate the implant. Both the early inflammatory and late fibrotic stages of FBR can severely im

Patterned wireless transcranial optogenetics generates artificial perception.

Synthesizing perceivable artificial neural inputs independent of typical sensory channels remains a fundamental challenge in developing next-generation brain-machine interfaces. Establishing a minimally invasive, wirelessly effective and miniaturized platform with long-term stability is crucial for creating research methods and clinically meaningful biointerfaces capable of mediating artificial perceptual feedback. Here we demonstrate a miniaturized, fully implantable transcranial optogenetic neural stimulator designed to generate artificial perceptions by patterning large cortical ensembles w

A comprehensive study on Herba Epimedium-derived extracellular nanovesicles as a prospective therapy for alveolar bone regeneration in postmenopausal osteoporosis.

Natural products rich in phytoestrogens, particularly those derived from traditional Chinese medicine (TCM) herbs, have garnered increased attention. Plant-derived extracellular vesicles are emerging as a promising strategy in cell communication and disease defense. Here, a comprehensive study on Herba Epimedium-derived extracellular nanovesicles (EELNs) for postmenopausal osteoporosis treatment was conducted. The results showed that EELNs exhibit a typical exosome morphology with an average diameter of 130 nm and are rich in specific small-molecule metabolites and miRNAs. Network pharmacology

Research progress on the kidney-gut-brain axis in brain dysfunction in maintenance hemodialysis patients.

Maintenance hemodialysis (MHD) has become the primary renal replacement therapy for patients with end-stage renal disease. The kidney-gut-brain axis represents a communication network connecting the kidney, intestine and brain. In MHD patients, factors such as uremic toxins, hemodynamic changes, vascular damage, inflammation, oxidative stress, and intestinal dysbiosis in MHD patients refers to a range of clinical syndromes, including brain injury, and is manifested by conditions such as white matter disease, brain atrophy, cerebrovascular disease, cognitive impairment, depression, anxiety, and

Self-supervised dynamic learning for long-term high-fidelity image transmission through unstabilized diffusive media.

Multimode fiber (MMF) which supports parallel transmission of spatially distributed information is a promising platform for remote imaging and capacity-enhanced optical communication. However, the variability of the scattering MMF channel poses a challenge for achieving long-term accurate transmission over long distances, of which static optical propagation modeling with calibrated transmission matrix or data-driven learning will inevitably degenerate. In this paper, we present a self-supervised dynamic learning approach that achieves long-term, high-fidelity transmission of arbitrary optical

Understanding the Tumor Microenvironment in Melanoma Patients with In-Transit Metastases and Its Impacts on Immune Checkpoint Immunotherapy Responses.

Melanoma is the leading cause of global skin cancer-related death and currently ranks as the third most commonly diagnosed cancer in Australia. Melanoma patients with in-transit metastases (ITM), a type of locoregional metastasis located close to the primary tumor site, exhibit a high likelihood of further disease progression and poor survival outcomes. Immunotherapies, particularly immune checkpoint inhibitors (ICI), have demonstrated remarkable efficacy in ITM patients with reduced occurrence of further metastases and prolonged survival. The major challenge of immunotherapeutic efficacy lies

Social Protections and Climate Migration: Service Navigation among Puerto Ricans Displaced by Hurricane Maria.

As climate change continues to displace greater numbers of people, transnational ties are important sources of social protection for climate migrants. Migrants assemble unique configurations of formal and informal social protections depending on the resources available within their sending and receiving communities. However, the specific constellations of social protections that climate migrants use following disaster and displacement remain underexamined. Authors conducted semistructured interviews with Puerto Ricans who migrated in the aftermath of Hurricane Maria (N = 41) and used qualitati

The role of exosomes and exosomal microRNA in diabetic cardiomyopathy.

Diabetic cardiomyopathy, a formidable cardiovascular complication linked to diabetes, is witnessing a relentless surge in its incidence. Despite extensive research efforts, the primary pathogenic mechanisms underlying this condition remain elusive. Consequently, a critical research imperative lies in identifying a sensitive and dependable marker for early diagnosis and treatment, thereby mitigating the onset and progression of diabetic cardiomyopathy (DCM). Exosomes (EXOs), minute vesicles enclosed within bilayer lipid membranes, have emerged as a fascinating frontier in this quest, capable of

Regulation of cell distancing in peri-plaque glial nets by Plexin-B1 affects glial activation and amyloid compaction in Alzheimer's disease.

Communication between glial cells has a profound impact on the pathophysiology of Alzheimer's disease (AD). We reveal here that reactive astrocytes control cell distancing in peri-plaque glial nets, which restricts microglial access to amyloid deposits. This process is governed by guidance receptor Plexin-B1 (PLXNB1), a network hub gene in individuals with late-onset AD that is upregulated in plaque-associated astrocytes. Plexin-B1 deletion in a mouse AD model led to reduced number of reactive astrocytes and microglia in peri-plaque glial nets, but higher coverage of plaques by glial processes

Single-Cell RNA Sequencing in Organ and Cell Transplantation.

Single-cell RNA sequencing is a high-throughput novel method that provides transcriptional profiling of individual cells within biological samples. This method typically uses microfluidics systems to uncover the complex intercellular communication networks and biological pathways buried within highly heterogeneous cell populations in tissues. One important application of this technology sits in the fields of organ and stem cell transplantation, where complications such as graft rejection and other post-transplantation life-threatening issues may occur. In this review, we first focus on researc

Effects of Eribulin on the RNA Content of Extracellular Vesicles Released by Metastatic Breast Cancer Cells.

Extracellular vesicles (EVs) are small lipid particles secreted by almost all human cells into the extracellular space. They perform the essential function of cell-to-cell communication, and their role in promoting breast cancer progression has been well demonstrated. It is known that EVs released by triple-negative and highly aggressive MDA-MB-231 breast cancer cells treated with paclitaxel, a microtubule-targeting agent (MTA), promoted chemoresistance in EV-recipient cells. Here, we studied the RNA content of EVs produced by the same MDA-MB-231 breast cancer cells treated with another MTA, e

The role of the extracellular matrix in cardiac regeneration.

The extracellular matrix (ECM) is a complex and dynamic three-dimensional network that functions as an architectural scaffold to maintain cardiac homeostasis. Important biochemical and mechanical signals associated with cell‒cell communication are provided via the reciprocal interaction between cells and the ECM. By converting mechanical cues into biochemical signals, the ECM regulates many cell processes, including migration, adhesion, growth, differentiation, proliferation, and apoptosis. Moreover, the ECM facilitates the replacement of dead cells and preserves the structural integrity of th

PIBF1 regulates trophoblast syncytialization and promotes cardiovascular development.

Proper placental development in early pregnancy ensures a positive outcome later on. The developmental relationship between the placenta and embryonic organs, such as the heart, is crucial for a normal pregnancy. However, the mechanism through which the placenta influences the development of embryonic organs remains unclear. Trophoblasts fuse to form multinucleated syncytiotrophoblasts (SynT), which primarily make up the placental materno-fetal interface. We discovered that endogenous progesterone immunomodulatory binding factor 1 (PIBF1) is vital for trophoblast differentiation and fusion int

A spatiotemporal atlas of mouse liver homeostasis and regeneration.

The mechanism by which mammalian liver cell responses are coordinated during tissue homeostasis and perturbation is poorly understood, representing a major obstacle in our understanding of many diseases. This knowledge gap is caused by the difficulty involved with studying multiple cell types in different states and locations, particularly when these are transient. We have combined Stereo-seq (spatiotemporal enhanced resolution omics-sequencing) with single-cell transcriptomic profiling of 473,290 cells to generate a high-definition spatiotemporal atlas of mouse liver homeostasis and regenerat

Genome-wide variation study and inter-tissue communication analysis unveil regulatory mechanisms of egg-laying performance in chickens.

Egg-laying performance is of great economic importance in poultry, but the underlying genetic mechanisms are still elusive. In this work, we conduct a multi-omics and multi-tissue integrative study in hens with distinct egg production, to detect the hub candidate genes and construct hub molecular networks contributing to egg-laying phenotypic differences. We identifiy three hub candidate genes as egg-laying facilitators: TFPI2, which promotes the GnRH secretion in hypothalamic neuron cells; CAMK2D, which promotes the FSHβ and LHβ secretion in pituitary cells; and OSTN, which promotes granulosa

Updated Toolbox for Assessing Neuronal Network Reconstruction after Cell Therapy.

Cell therapy has proven to be a promising treatment for a range of neurological disorders, including Parkinson Disease, drug-resistant epilepsy, and stroke, by restoring function after brain damage. Nevertheless, evaluating the true effectiveness of these therapeutic interventions requires a deep understanding of the functional integration of grafted cells into existing neural networks. This review explores a powerful arsenal of molecular techniques revolutionizing our ability to unveil functional integration of grafted cells within the host brain. From precise manipulation of neuronal activit

Surface remodeling and inversion of cell-matrix interactions underlies community recognition and dispersal in Vibrio cholerae biofilms.

Biofilms are ubiquitous surface-associated bacterial communities embedded in an extracellular matrix. While it is commonly assumed that biofilm-dwelling cells are glued together by the matrix, how the cell-matrix interaction depends on the specific biochemistry of the matrix components and how this interaction varies during biofilm growth remains unclear. Here, we investigated cell-matrix interactions in Vibrio cholerae ( Vc ), the causative agent of cholera. We combine genetics, microscopy, simulation, and biochemical tools to show that Vc cells are not attractive to V ibrio p oly s accharide
