Targeting CDK-2 With Novel Indole-Pyrazole Hybrids: Discovery of Potent Anticancer Agents Supported by Mechanistic and In Silico Studies.

The current study devised and synthesized a novel class of pyrazole derivatives based on indole as possible inhibitors of cyclin-dependent kinase-2 (CDK-2). 1H NMR, 13C NMR, NOESY, HMQC, and elemental analysis were used to confirm the structural integrity of the synthesized compounds. Promising CDK-2 inhibitory activity was observed in biological assays, and numerous compounds exhibited sub-micromolar IC50 values. Compound 5 d outperformed the reference inhibitor Roscovitine (IC50 = 0.716 µM) as the most potent inhibitor (IC50 = 0.536 µM), followed by compound 9 g (IC50 = 0.675 µM). SAR analys

Mechanistic anticancer effects of melittin across cancer hallmarks: A narrative evidence synthesis.

Venoms are complex natural secretions that contain bioactive molecules with diverse pharmacological effects. Melittin, a 26-amino-acid amphipathic peptide derived from the venom of the honeybee Apis mellifera, has been widely reported to exert cytotoxic effects in cancer models. Although melittin is best known for its membrane-disruptive and haemolytic properties, increasing evidence suggests that it also modulates intracellular signalling pathways involved in cancer progression. This review synthesised evidence on the mechanistic effects of melittin in cancer-related pathways. Articles were r

Multi-mechanistic effects of bisphenol A on testicular dysfunction and endocrine disruption in adult male Labeo bata: oxidative stress, inflammation, and dysregulated energy sensors.

Leaching of BPA, a potent endocrine disruptor, from microplastics in aquatic environments has garnered renewed interest in its impact on male reproduction. However, the mechanisms underlying BPA modulation of testicular metabolic, endocrine, and autocrine-paracrine axes in teleost remained relatively unexplored, prompting us to examine BPA-induced testicular dysfunction in adult male Labeo bata. Present results demonstrate that congruent with reduced gonadosomatic index (GSI), chronic BPA treatment at environmentally relevant concentrations enhanced ROS synthesis, oxidative stress, and testicu

BDNF improves fracture healing through promoting osteoblasts proliferation and migration via trkb/akt regulated NUCKS1 expression.

BACKGROUND: Bone nonunion is a major complication in fracture treatment. Despite considerable advancements in fracture care, 5-10% of fractures result in nonunion; This demonstrates the requirement for novel molecules to enhance fracture healing. Brain-derived neurotrophic factor (BDNF) has been increasingly recognized for its role in fracture healing; however, the underlying molecular mechanisms remain unclear. Osteoblast proliferation and migration are critical to this process. The nuclear casein kinase and cyclin-dependent kinase substrate 1 gene (NUCKS1), located on human chromosome 1 at 1

Mediterranean diet protects against a neuroinflammatory cortical transcriptome: Associations with brain volumetrics, peripheral inflammation, social isolation, and anxiety in nonhuman primates (Macaca fascicularis).

Mediterranean diets may be neuroprotective and prevent cognitive decline relative to Western diets; however, the underlying biology is poorly understood. We assessed the effects of Western versus Mediterranean-like diets on RNAseq-generated transcriptional profiles in lateral temporal cortex and their relationships with longitudinal changes in neuroanatomy, circulating monocyte gene expression, and observations of social isolation and anxiety in 38 socially-housed, middle-aged female cynomolgus macaques (Macaca fascicularis). Diet resulted in differential expression of seven transcripts (FDR <

SMTP-44D Inhibits Atherosclerotic Plaque Formation in Apolipoprotein-E Null Mice Partly by Suppressing the AGEs-RAGE Axis.

SMTP-44D has been reported to have anti-oxidative and anti-inflammatory reactions, including reduced expression of receptor for advanced glycation end products (RAGE) in experimental diabetic neuropathy. Although activation of RAGE with its ligands, and advanced glycation end products (AGEs), play a crucial role in atherosclerotic cardiovascular disease, a leading cause of death in diabetic patients, it remains unclear whether SMTP-44D could inhibit experimental atherosclerosis by suppressing the AGEs-RAGE axis. In this study, we investigated the effects of SMTP-44D on atherosclerotic plaque f

Rosai-Dorfman disease of the breast: a clinicoradiologic and pathologic study.

Rosai-Dorfman disease (RDD) is an uncommon histiocytic disorder typically involving lymph nodes and less frequently extranodal tissues. RDD involving the breast is rare and may clinically and radiologically mimic neoplastic and non-neoplastic disorders. We report seven patients with breast RDD, describe their clinicoradiologic and pathologic features, and discuss the differential diagnosis. Patients, ranging from 15 to 74 years of age, presented with unilateral and unifocal (5/7) or bilateral and multifocal (2/7) masses. RDD was either confined to the breast (6/7) or concurrently involved a ly
