Alternative insecticides for larval control of the dengue vector Aedes aegypti in Lao PDR: insecticide resistance and semi-field trial study.

BACKGROUND: The mosquito Aedes aegypti is the primary vector of several arboviruses, such as dengue, chikungunya and Zika, and represents a major public health problem in Southeast Asia. In Laos, where dengue is reemerging, several Ae. aegypti populations from the capital Vientiane have shown resistance to the organophosphate temephos, a commonly-used larvicide for public health interventions. METHODS: Here, we tested the insecticide susceptibility of a wild larval population of Ae. aegypti against Bacillus thuringiensis israelensis (Bti), diflubenzuron, pyriproxyfen and spinosad. Residual ef

Successes and failures of sixty years of vector control in French Guiana: what is the next step?

Since the 1940s, French Guiana has implemented vector control to contain or eliminate malaria, yellow fever, and, recently, dengue, chikungunya, and Zika. Over time, strategies have evolved depending on the location, efficacy of the methods, development of insecticide resistance, and advances in vector control techniques. This review summarises the history of vector control in French Guiana by reporting the records found in the private archives of the Institute Pasteur in French Guiana and those accessible in libraries worldwide. This publication highlights successes and failures in vector con

Zika puzzle in Brazil: peculiar conditions of viral introduction and dissemination - A Review.

This article discusses the peculiar conditions that favoured the unexpected introduction of Zika virus into the poorest northeastern region of Brazil in 2015, its speed of transmission to other Brazilian states, other Latin American countries and other regions, and the severity of related neurological disorders in newborns and adults. Contrasting with evidence that Zika had so far caused only mild cases in humans in the last six decades, the epidemiological scenario of this outbreak in Brazil indicates dramatic health effects: in 2015, an increase of 20-fold in notified cases of microcephaly a

[New and reemerging viral fever carriers--a threat of epidemic complications in southern Europe and Russia].

Issues of emergence and spread of viral fever and malaria carriers in non-endemic areas are discussed. The role of asian tiger mosquito global spread--a dangerous arbovirus carrier--is noted. Epidemic consequences of emergence of West Nile fever virus in the New World that is presumably connected with Culex pipiens complex mosquitoes are discussed. Rigorous monitoring and prophylaxis measures are suggested.

Mutations in the Methyltransferase Motifs of L Protein Attenuate Newcastle Disease Virus by Regulating Viral Translation and Cell-to-Cell Spread.

The large (L) polymerase proteins of most nonsegmented, negative-stranded (NNS) RNA viruses have conserved methyltransferase motifs, (G)-G-G-D and K-D-K-E, which are important for the stabilization and translation of mRNA. However, the function of the (G)-G-G-D and K-D-K-E motifs in the NNS RNA virus Newcastle disease virus (NDV) remains unclear. We observed G-G-D and K-D-K-E motifs in all NDV genotypes. By using the infection cloning system of NDV rSG10 strain, recombinant NDVs with a single amino acid mutated to alanine in one motif (G-G-D or K-D-K-E) were rescued. The intracerebral pathogen

Orthoflavivirus circulation in mosquitoes (Diptera: Culicidae) from an Atlantic Forest remnant and adjacent urban areas, in São Paulo, Brazil.

Orthoflaviviruses cause large epidemics in Brazil, such as those caused by dengue (DENV), yellow fever (YFV), and Zika viruses (ZIKV). Since they are transmitted by mosquitoes, entomo-virological surveillance is recommended in many areas of the country. In addition to medically important orthoflaviviruses, insect-specific viruses (ISVs), which replicate only in insect cell lines, have been detected in mosquitoes collected in this country. Parque Ecológico do Tietê (PET) is an Atlantic forest remnant located within a large urban area in the city of São Paulo, State of São Paulo, Brazil. Since P

Projections of Aedes and Culex mosquitoes across North and South America in response to climate change.

INTRODUCTION: Climate and land use change may cause the geographical range of mosquitoes to expand, shift, or contract, ultimately changing what communities are at risk for contracting mosquito-borne diseases. Across North and South America, mosquitoes from the Aedes and Culex genera are vectors for numerous diseases, including chikungunya, dengue, various equine encephalitis viruses, Saint Louis encephalitis virus, West Nile virus, yellow fever virus, and Zika virus. The goal of our study was to project the geographical distributions of important mosquito vectors across North and South Americ

A computational tool for the design of live attenuated virus vaccine based on microRNA-mediated gene silencing.

BACKGROUND: The microRNA-based gene-silencing machinery has been recognized as a promising approach to control viral replication and used for improving safety for the live attenuated virus vaccines. The effective host microRNA response elements (MREs) have been incorporated into a virus sequence mainly based on the experimental trials for identifying both microRNA binding sites and effective mutations. The design of MREs for viral genomes or with multiple host microRNAs of interest, then, will be time and cost consuming. RESULTS: In this paper, we introduced a computational flow that could be

BACKGROUND: Aedes aegypti and Ae. albopictus are the most important vector mosquito species globally and are capable of transmitting various viral diseases, such as dengue fever, zika virus disease, and chikungunya fever. Although they overlap in terms of ecological niches and geographical distribution, their virus carriage and transmission capacities differ significantly. Metavirome studies can provide new perspectives for understanding these differences. METHODS: In this study, next-generation sequencing (NGS) was used to analyze the epidemiologically significant metaviromes of Ae. aegypti

Nicotinamide metabolism is essential for Hepatitis C Virus replication and the production of infectious Lipo-Viro-Particles.

BACKGROUND & AIMS: Hepatitis C virus (HCV) has the unique characteristic of forming lipo-viro-particles (LVPs), which are lipid-rich virions containing both the viral components and host apolipoproteins such as ApoB and E. This unique composition gives to LVPs a low buoyant density, facilitates their entry into the hepatocyte, and is a hallmark of highly-infectious HCV particles. Although recent studies have shown that inhibiting NAD biosynthesis can both disrupt central carbon metabolism and thereby interfere with the replication of hepatotropic viruses such as dengue virus (DENV) and hepatit

STAT2-Dependent Immune Responses Ensure Host Survival despite the Presence of a Potent Viral Antagonist.

A pathogen encounter induces interferons, which signal via Janus kinases and STAT transcription factors to establish an antiviral state. However, the host and pathogens are situated in a continuous arms race which shapes host evolution toward optimized immune responses and the pathogens toward enhanced immune-evasive properties. Mouse cytomegalovirus (MCMV) counteracts interferon responses by pM27-mediated degradation of STAT2, which directly affects the signaling of type I as well as type III interferons. Using MCMV mutants lacking M27 and mice lacking STAT2, we studied the opposing relations

Novel multiplex family-wide PCR and Nanopore sequencing of amplicons (FP-NSA) approach for surveillance of influenza- and coronaviruses in humans and animals.

BACKGROUND: Recent outbreaks of zoonotic diseases like Ebola, Mpox, dengue fever, and COVID-19 highlight gaps in surveillance and early detection at disease hotspots. Virus family-wide diagnostic assays offer a cost-effective and sensitive alternative to metagenomics for initial virus identification. This study introduces a multiplex family-wide PCR coupled with Nanopore sequencing of amplicons (FP-NSA) for surveillance of novel and known zoonotic respiratory viruses, including influenza A and D viruses (IAV and IDV), alpha (α-), beta (β-), and gamma (γ-) coronaviruses (CoVs). Cellular Responses to Flavivirus Infections: Stress Signaling at the Crossroads of Host Defense and Virus Infection.

Flaviviruses, encompassing notable pathogens, like Dengue, Zika, West Nile, and tick-borne encephalitis viruses, elicit complex cellular stress responses, involving pathways such as the unfolded protein response (UPR), integrated stress response (ISR), apoptosis, autophagy, and the antiviral immune response. These pathways regulate cell fate by either promoting survival to counteract virus-induced damage or triggering cell death programs under prolonged and irreparable stress. Therefore, the primary aim of flavivirus-induced cellular responses is to protect cells and hinder viral propagation.

CRISPR/Cas13a-based colorimetric biosensing platform for point-of-care detection of viral nucleic acids.

Rapid and accurate diagnosis is important in preventing and effectively combating infectious disease outbreaks. The CRISPR/Cas13a-based Specific High-sensitivity Enzymatic Reporter UnLOCKing (SHERLOCK) platform possesses the advantages of high efficiency, good specificity and sensitivity, and it has been widely adopted in molecular diagnostics. However, the traditional SHERLOCK platform requires dual-labeled RNA probes for fluorescence detection or lateral flow assay, which entail tedious modification procedures and sophisticated optical instruments, limiting its broad applications. Herein, we
