Eriocitrin Attenuates Diabetic Cardiomyopathy by Regulating Copper Homeostasis and Cuproptosis-Associated Pathways via SIRT7/YAP/ATP7A Signalling.

Diabetic cardiomyopathy (DCM) is a major complication of diabetes mellitus, with limited therapeutic strategies available. Cuproptosis, a novel form of copper-dependent cell death, has been implicated in cardiac dysfunction but remains unexplored in DCM. Here, we investigated the effects of Eriocitrin, a natural flavonoid, on DCM and examined the potential involvement of copper homeostasis, cuproptosis-associated molecular alterations and SIRT7/YAP/ATP7A signalling. Using high-fat diet and streptozotocin-induced diabetic mice, along with high-glucose-treated cardiomyocytes, we demonstrated tha

Steam explosion modified pea peptides alleviates hepatosteatosis by regulating lipid metabolism pathways and promoting autophagy.

Pea peptides (PP) are natural compounds with multiple biological activities. The purpose of this study was to explore the effect and mechanisms of steam explosion (SE)-modified PP on lipid metabolism in vivo and in vitro. The findings demonstrated that SE-modified PP treatment significantly inhibited lipid accumulation in HepG2 cells induced by free fatty acids (FFA). In addition, SE-modified PP treatment significantly alleviated liver index, improved biochemical parameters in high-fat diet (HFD) mice. SE-modified PP prevented lipid accumulation through regulating AMPK activity and decreased l

Microbial diversity and fitness in the gut-brain axis: influences on developmental risk for Alzheimer's disease.

The gut-brain axis (GBA) denotes the dynamic and bidirectional communication system that connects the gastrointestinal tract and the central nervous system (CNS). This review explored this axis, focusing on the role of microbial diversity and fitness in maintaining gastrointestinal health and preventing neurodegeneration, particularly in Alzheimer's disease (AD). Gut dysbiosis, characterized by the imbalance in populations of beneficial and harmful bacteria, has been associated with increased systemic inflammation, neuroinflammation, and the progression of AD through pathogenic mechanisms invo

Serum- and glucocorticoid-induced kinase 3 orchestrates glucocorticoid signaling to facilitate chromatin remodeling during murine adipogenesis.

Elevated glucocorticoid levels are common in conditions such as aging, chronic stress, Cushing syndrome, and glucocorticoid therapy. While glucocorticoids suppress inflammation through the glucocorticoid receptor (GR), they also cause metabolic side effects. Investigating alternative pathways beyond GR activation is crucial for reducing these side effects. Our phosphoproteomics analysis revealed that glucocorticoid exposure promotes phosphorylation at the RxxS motifs of multiple proteins in preadipocytes, including those mediated by serum- and glucocorticoid-induced kinase 3 (SGK3). SGK3 is a

Iron overload exacerbates metabolic dysfunction-associated steatohepatitis via the microbiota-gut-liver axis through lipopolysaccharide-mediated Akr1b8 activation.

Iron homeostatic is closely linked to the development of metabolic dysfunction-associated steatohepatitis (MASH). However, the underlying mechanisms remain poorly understood. HFE knockout (KO) mice were used to generate mild iron-overload models. MASH was induced by feeding mice a methionine- and choline-deficient (MCD) diet for 4 weeks. Iron overload significantly exacerbated the pathologies of MCD-induced MASH, including liver injury, hepatic lipid accumulation, inflammation, and fibrosis. Additionally, iron overload reshaped the composition of gut microbiota, and fecal microbiota transplant

Low risk, high cost: challenging the role of gastric cancer screening in low-prevalence countries.

Gastric cancer is the second commonest gastrointestinal (GI) neoplasm, with a high mortality rate and highly variable regional incidence data associated with factors such as diet, genetics, and infection with Helicobacter pylori. In high-risk countries as Japan and South Korea endoscopic screening has shown its potential to reduce this cancer's mortality. However, in low-risk countries such as Spain, where population screening is not cost-effective, a selective approach is advised focused on high-risk groups, including patients with a family history of gastric cancer, or in the form of opportu

Ilexgenin A inhibits lipid accumulation in macrophages and reduces the progression of atherosclerosis through PTPN2/ERK1/2/ABCA1 signalling pathway.

Macrophage lipid accumulation indicates a pathological change in atherosclerosis. Ilexgenin A (IA), a pentacyclic triterpenoid compound, plays a role in preventing inflammation, bacterial infection, and fatty liver and induces a potential anti-atherogenic effect. However, the anti-atherosclerotic mechanism remains unclear. The present study investigated the effects of IA on lipid accumulation in macrophage-derived foam cells and atherogenesis in apoE-/- mice. Our results indicated that the expression of adenosine triphosphate-binding cassette transporter A1 (ABCA1) was up-regulated by IA, prom

β-Hydroxybutyrate ameliorates osteoarthritis through activation of the ERBB3 signaling pathway in mice.

The ketogenic diet (KD) has demonstrated efficacy in ameliorating inflammation in rats with osteoarthritis (OA). However, the long-term safety of the KD and the underlying mechanism by which it delays OA remain unclear. We found that while long-term KD could ameliorate OA, it induced severe hepatic steatosis in mice. Consequently, we developed 2 versions of ketogenic-based diets: KD supplemented with vitamin D and intermittent KD. Both KD supplemented with vitamin D and intermittent KD effectively alleviated OA by significantly reducing the levels of inflammatory cytokines, cartilage loss, sen
