Discovery of 9-arylamino-2,3-dihydro-[1,4]dioxino[2,3-g]quinoline-8-carbonitriles: Potent inducers of immunogenic cell death via colchicine site-targeted microtubule polymerization inhibition.

A series of microtubule polymerization inhibitors that target the colchicine site on the basis of a new scaffold have been developed. Among them, compound 17 exhibited outstanding comprehensive antitumor efficacy against a panel of cancer cell lines (with an average IC50 value of 17.7 nM). Paclitaxel resistance was successfully overcome in a xenograft model in which paclitaxel-resistant A549/TxR cells were used in BALB/c nude mice, as expected. Moreover, the outstanding antitumor efficacy of compound 17 is attributed not only to its chemical toxicity but also to its ability to induce immunogen

Structure-Based Drug Design of Novel Indazole/Indolinone-Based Biaryl Derivatives as Oral IRAK4 Inhibitors for the Treatment of Inflammatory Diseases.

Pathological inflammation in multiple diseases arises from dysregulated innate immune signaling, wherein the interleukin-1 receptor-associated kinase 4 (IRAK4) acts as a critical node that mediates pro-inflammatory cascades and cytokine release downstream of toll-like and interleukin-1 receptors (TLR/IL-1R). Consequently, the therapeutic inhibition of IRAK4 using small molecules has emerged as a promising strategy for these conditions, particularly, rheumatoid arthritis. In this study, we identified a series of novel IRAK4 inhibitors featuring an indazole/indolinone-based biaryl scaffold, and

Rationally designed small peptides targeting DPP-4: Identification of potent anti-diabetic agents.

DPP-4 inhibition is a pioneering strategy to subdue type 2 diabetes. In this context, we designed and synthesized small peptide-based DPP-4 inhibitors. Eighteen peptides were synthesized, and their inhibition activity was determined. Compound 8 emerged as the most potent DPP-4 inhibitor (IC50 0.12 nM) with desirable drug likeness properties including solubility, plasma stability, kinetic study, and ADME properties. The in vivo anti-diabetic potency was evaluated in a diabetic model of STZ/NA-induced Wistar rats. Compound 8 not only lined up glucose levels to normal but also improved serum's li

Ellagic acid C and aspergone R, two undescribed pyrones from the endophytic fungus Diaporthe sp. CB10100: biological evaluation, molecular docking and molecular dynamics studies.

Two new compounds, ellagic acid C (5) and aspergone R (6), along with four previously identified compounds (7-10), were successfully isolated from the fungus Diaporthe sp. CB10100. Ellagic acid C (5) contains an uncommon tricyclic 5/6/6 ring system. A biosynthetic pathway for 6 was proposed, although additional investigations are needed to elucidate the ethyl biosynthesis mechanism. Both aspergone R (6) and (-)-terpestacin (9) decreased lipopolysaccharide (LPS)-induced protein expression of iNOS and COX-2. Molecular docking analysis revealed that 6 and 9 potentially interact with the active si

Design and synthesis of novel sulfanilamide derivatives as aminopeptidase N inhibitors.

Guided by the structural architecture of the aminopeptidase N (APN) active site, we designed and synthesized a series of novel APN inhibitors featuring sulfanilamide scaffold coupled with hydroxamate zinc-binding motifs. Among the series, compound 2k exhibited the inhibitory activity (IC50 = 4.3 μM) as effectively as a positive control drug Bestatin. Notably, our compounds exhibited pronounced selectivity against zinc-dependent metallopeptidase MMP-2. The SAR research indicated that ortho-disubstitution in the phenyl group could lead to an order of magnitude improvement. A molecular docking st

Discovery of Novel NO-Donor Containing hCA II Inhibitors with Retinal Ganglion Cell-Protective Effects for the Efficient Treatment of Glaucoma.

Glaucoma-induced blindness primarily results from RGC apoptosis. A medication capable of effectively reducing IOP while simultaneously preventing RGC apoptosis could offer a holistic approach to glaucoma treatment. In this study, novel NO-donor containing hCA II inhibitors were developed to achieve dual functionality: lowering IOP by decreasing aqueous humor production (via hCA II inhibition) and enhancing outflow (via NO release). The experimental results indicated that the dual-target compound A1 outperformed its single-target counterpart and the reference drug brinzolamide in both acute and

Synthesis, Structure-Activity Relationships, and Antitumor Activities of Quinoxiline-Containing Inhibitors of the Protein-Protein Interactions between Transcription Coactivator AF9/ENL and DOT1L/AF4.

Mixed lineage leukemia (MLL) gene rearrangements cause ∼75% of acute leukemia in infants and 5-10% in children and adults with poor clinical outcomes. Protein-protein interactions (PPI) between frequent MLL fusion partners AF9/ENL and AF4 or histone methyltransferase DOT1L are drug targets for MLL-rearranged (MLL-r) leukemia. Sixty-seven quinoxiline compounds were synthesized and tested for their ability to inhibit such PPIs. Compounds 16, 17, 59, and 63 were found to be potent inhibitors with IC50 values of 0.35-1.5 μM. Structure-activity relationships are discussed. Potent inhibitors can sup

Recommended Tool Compounds for Modifying the Cystic Fibrosis Transmembrane Conductance Regulator Channel Variants.

Cystic fibrosis (CF) is a genetic disorder arising from variations in the cystic fibrosis transmembrane conductance regulator (CFTR) gene, leading to multiple organ system defects. CFTR tool compounds are molecules that can modify the activity of the CFTR channel. Especially, patients that are currently not able to benefit from approved CFTR modulators, such as patients with rare CFTR variants, benefit from further research in discovering novel tools to modulate CFTR. This Review explores the development and classification of CFTR tool compounds, including CFTR blockers (CFTRinh-172, GlyH-101)

A Divergent Synthesis of Numerous Pyrroloiminoquinone Alkaloids Identifies Promising Antiprotozoal Agents.

On the basis of a streamlined route to the pyrroloiminoquinone (PIQ) core, we made 16 natural products spread across four classes of biosynthetically related alkaloid natural products, and multiple structural analogs, all in ≤8 steps longest linear sequence (LLS). The strategy features a Larock indole synthesis as the key operation in a five-step synthesis of a key methoxy-PIQ intermediate. Critically, this compound was readily diverged via selective methylation of either (or both) of the imine-like or pyrrole nitrogens, which then permitted further divergence by either O-demethylation to o-qu

Structure-Activity Relationship of Fluorinated Benzenesulfonamides as Inhibitors of Amyloid-β Aggregation.

Amyloid-beta aggregation is considered one of the factors influencing the onset of the Alzheimer's disease. Early prevention of such aggregation should alleviate disease condition by applying small molecule compounds that shift the aggregation equilibrium toward the soluble form of the peptide or slow down the process. We have discovered that fluorinated benzenesulfonamides of particular structure slowed the amyloid-beta peptide aggregation process by more than three-fold. We synthesized a series of ortho-para and meta-para double-substituted fluorinated benzenesulfonamides that inhibited the

Small molecule protein assembly modulators with pan-cancer therapeutic efficacy.

Two structurally unrelated small molecule chemotypes, represented by compounds PAV-617 and PAV-951, with antiviral activity in cell culture against Mpox virus (formerly known as monkeypox virus) and human immunodeficiency virus (HIV) respectively, were studied for anti-cancer efficacy. Each exhibited apparent pan-cancer cytotoxicity with reasonable pharmacokinetics. Non-toxicity is demonstrated in a non-cancer cell line and in mice at doses achieving drug exposure at active concentrations. Anti-tumour properties of both chemotypes were validated in mouse xenografts against A549 human lung canc

Design and Synthesis of Novel Indole Ethylamine Derivatives as a Lipid Metabolism Regulator Targeting PPARα/CPT1 in AML12 Cells.

Peroxisome proliferator-activated receptor alpha (PPARα) and carnitine palmitoyltransferase 1 (CPT1) are important targets of lipid metabolism regulation for nonalcoholic fatty liver disease (NAFLD) therapy. In the present study, a set of novel indole ethylamine derivatives (4, 5, 8, 9) were designed and synthesized. The target product (compound 9) can effectively activate PPARα and CPT1a. Consistently, in vitro assays demonstrated its impact on the lipid accumulation of oleic acid (OA)-induced AML12 cells. Compared with AML12 cells treated only with OA, supplementation with 5, 10, and 20 μM o

Benzene Amide Ether Scaffold is Active against Non-replicating and Intracellular Mycobacterium tuberculosis.

New drugs to treat tuberculosis which target intractable bacterial populations are required to develop shorter and more effective treatment regimens. The benzene amide ether scaffold has activity against intracellular Mycobacterium tuberculosis, but low activity against extracellular, actively replicating M. tuberculosis. We determined that these molecules have bactericidal activity against non-replicating M. tuberculosis but not actively replicating bacteria. Exposure to compounds depleted ATP levels in non-replicating bacteria and increased the oxygen consumption rate; a subset of molecules
