A New Simplified Coralmycin Derivative with Potent Anti-Gram-Negative Activity and Its Congeners Produced by the Myxobacterium Corallococcus coralloides M23.

Multidrug-resistant (MDR) Gram-negative bacterial infections urgently require new antibacterial agents with novel chemical scaffolds. In our continued investigation of minor metabolites produced by the myxobacterium Corallococcus coralloides M23, we isolated three new coralmycin derivatives, coralmycins J (1), K (2), and L (3), from large-scale fermentation cultures. Spectroscopic analyses revealed that these compounds represent structurally simplified derivatives of coralmycin with varying degrees of truncation. Compounds 1-4 (coralmycins J, K, L, and A) are new natural products isolated in o

Structure-resolved tannin interactions in food systems: nutritional effects, digestion, and applications.

Tannins are structurally diverse food polyphenols whose behavior cannot be predicted from total tannin content alone. This review uses tannin class, linkage chemistry, degree-of-polymerization metrics, galloylation and hydroxylation, conformational flexibility, and oxidation state as a common framework for interpreting interactions with food proteins, digestive enzymes, starch, plant cell-wall polysaccharides, minerals, and salivary constituents. These descriptors explain differences in digestibility, mineral bioaccessibility, astringency, interfacial behavior, and crosslinking. Many apparentl

Topology-controlled polymer brushes on LDPE for enhanced cellulase immobilization and lignocellulose conversion.

By providing a flexible microenvironment, immobilization of cellulase on polymer brushes represents an effective strategy to improve the interactions of cellulase with the substrate in biomass conversion. However, manipulation of polymer brush morphology remains largely unexplored, owing to the lack of an effective synthesis strategy. Herein, we developed a versatile visible-light-induced graft polymerization strategy to precisely fabricate three types of polymer brushes with well-defined topologies on low-cost low-density polyethylene (LDPE) films. Benefiting from its unique three-dimensional

Long-term capsaicin intake and gut inflammation: microbial alterations, metabolic mechanisms, and intervention effects.

The effects of capsaicin consumption on gut health and its mechanisms remain uncharacterized. This study integrated human cohort and colonic simulation fermentation found that long-term capsaicin intake induced gut dysbiosis, characterized by an increase in Klebsiella and a decrease in Lactobacillus, which correlated significantly with gut inflammation and the dysregulation of key metabolites (including bile acids, short-chain fatty acids derivatives and steroids). Capsaicin metabolites, such as dihydrocapsaicin, ferulic acid, and veratric acid were identified. The study also enriched and iden

Gut Microbiota-Derived Short-Chain Fatty Acids in Inflammatory Bowel Disease: Mechanistic Insights into Gut Inflammation, Barrier Function, and Therapeutic Potential.

This review delves into the complex relationship between short-chain fatty acids (SCFAs) produced by the gut microbiota and inflammatory bowel disease (IBD). IBD, which includes Crohn's disease and ulcerative colitis, is a group of chronic gastrointestinal disorders with an increasing global incidence. Despite extensive research, the exact etiopathogenesis remains elusive, although a complex interplay involving genetic predisposition, environmental influences, and abnormal immune responses against commensal gut microbes is widely recognized. SCFAs, primarily acetate and butyrate, emerge as key

Enterococcus faecalis NBRC 100481 Protects the Intestinal Barrier via α-catenin/HMP-1 in Caenorhabditis elegans.

Intestinal barrier disruption is a hallmark of gastrointestinal disorders and is also linked to metabolic diseases, posing serious health risks. Probiotic-based interventions have thus attracted growing attention. This study isolated Enterococcus faecalis NBRC 100481 from recovered colitis mice after dietary intervention with pentagalloyl glucose (PGG), and evaluated its protective effects and PGG's regulatory role. Fermentation revealed that PGG enhanced metabolic activity of E. faecalis NBRC 100481 and promoted production of bioactive metabolites such as N-acetylglutamic acid and 4-hydroxybe

Metabolic strategies of Enterobacteriaceae gut colonization.

Enterobacteriaceae are facultative anaerobic bacteria that commonly colonize the gut and include both commensals and diverse pathogens. They can flexibly adapt their metabolic strategy to grow in the intestinal environment. As Enterobacteriaceae blooms are linked to enteric disease and the dissemination of antibiotic resistance, a deeper understanding of the factors that drive their expansion is needed. In the healthy intestine, Enterobacteriaceae subsist on low concentrations of diet-derived and mucus-derived monosaccharides that feed glycolysis and mixed-acid fermentation. Inflammation resha

Occurrence of anticancer drugs and widely used pharmaceuticals in sewage sludge, compost, and river sediment.

Pharmaceuticals enter the environment mainly via municipal wastewater treatment plants, where some compounds bind to sewage sludge during treatment processes. The contaminated sludge often undergoes fermentation and composting and is later used as a soil amendment. This study aims to investigate the fate of five pharmaceuticals letrozole, tamoxifen, carbamazepine, diclofenac, and naproxen, in environmental solid matrices, including sewage sludge, compost, and sediment. An analytical method based on microwave-assisted extraction, SPE, and UPLC-MS/MS was developed and validated. The most abundan

Gut Microbiota and Metabolic Syndrome: A Narrative Review.

Obesity is a major global health problem and is closely associated with a broad range of metabolic disorders, including metabolic syndrome (MetS), dyslipidemia, hypertension, atherosclerosis, type 2 diabetes mellitus, and cardiovascular disease. The gut microbiota plays a central role in maintaining intestinal epithelial integrity, regulating glucose and lipid metabolism, and modulating immune function. Through the gut-brain axis, it also contributes to appetite regulation and energy homeostasis by influencing the release of anorexigenic hormones. Dysbiosis, including alterations in the relati

Unlocking the nutritional benefits of dietary fiber in sow feed: a short review.

Dietary fiber is a critical nutrient in sow diet and was addressed in several studies in the past decades. It plays a key role in improving digestive health, supporting metabolic functions, and enhancing the overall well-being of sows. Fiber, a plant-based feed ingredient is classified into soluble and insoluble fibers. Soluble fibers, such as pectin, dissolve in water and can form gels, influencing the fermentation process in the gut. Insoluble fibers, like cellulose, do not dissolve in water and contribute to the bulk of fecal matter, promoting intestinal motility. In sow nutrition, dietary

Biocontrol potential of endophytic Sphingobium xenophagum J-1 against Fusarium oxysporum causing root rot in Paris polyphylla var. yunnanensis.

Paris polyphylla var. yunnanensis (PPY) is a valuable medicinal plant frequently threatened by root rot caused by pathogenic fungi such as Fusarium oxysporum. Traditional chemical control methods pose environmental and resistance risks, highlighting the need for sustainable alternatives. This study investigated the antifungal potential of the endophytic bacterium Sphingobium xenophagum J-1, isolated from healthy PPY, against F. oxysporum in vitro. The ethyl acetate crude extract of J-1 fermentation broth exhibited significant concentration-dependent inhibition of spore germination, with a 94.6

Butyrate in ulcerative colitis and pouchitis: friend or foe?

It is widely known that the short chain fatty acid (SFA) butyrate, generated mainly through fermentation of dietary fibre by anaerobic bacteria, has beneficial effects on health. Less widely known is that butyrate, at the concentration (c20 mM) typically found in the healthy colonic lumen, is toxic to colon epithelial cells. It is probably the presence of the continuous adherent mucus layer in the healthy colon that ensures that butyrate concentrations in contact with the epithelium fall below the levels (c > 3 mM) that are consistently toxic in in vitro or ex vivo studies. In active ulcerativ

Gellan Gum Modification and Structural Regulation for Food Applications: Delivery Systems and Digestive Behavior.

Gellan gum (GG) is an anionic microbial polysaccharide widely used in foods because of its efficient gelation, thermal stability, and ion-responsive structuring. However, the contrasting limitations of high- and low-acyl GG, together with their sensitivity to molecular weight, ions, pH, thermal history, and food-matrix composition, restrict their performance in complex products. This review distinguishes four intervention levels of GG modification and structural regulation: covalent modification, chain-architecture regulation, supramolecular regulation, and food-matrix composite structuring. Isolation and biocontrol potential of Bacillus amyloliquefaciens against asparagus stem blight.

Garden asparagus (Asparagus officinalis L.) is an economically important vegetable crop widely cultivated for its nutritional value and health-promoting properties. Asparagus stem blight, caused by Phomopsis asparagi (Sacc.) Bubák, is one of the most destructive diseases affecting asparagus production worldwide, leading to significant yield losses. In this study, a pathogenic strain of P. asparagi was isolated and characterized, serving as the target for screening antagonistic microorganisms from the rhizosphere soil of healthy asparagus plants. Among 79 isolates, one strain, identified as Bac

Functional characterization of fungal laccases via siRNA silencing enables targeted biodegradation.

Edible fungi serve as efficient bioconverters, transforming lignocellulosic wastes into valuable fungal proteins by extracellular enzyme systems (e.g., laccases). However, the functional characterization of these lignocellulolytic enzymes is still a major challenge, limiting the strategic application of fungal systems in targeted biodegradation. Here, we developed a versatile, non-transformative siRNA delivery platform for rapid gene function analysis in the edible fungus Pleurotus ostreatus. Using spray-induced (SIGS) in mycelia, injection-induced (IIGS) in fruiting bodies, and co-culture-ind

Coupling Electrochemical CO2 Reduction With Ethanol Oxidation for Acetate Production in a Dual-Electrolyzer System.

Bioethanol production from biomass is suggested as a strategy for greener fuel production. However, an equimolar amount of CO2 is released during fermentation, making the process less efficient. Utilizing this released CO2 directly in an electrolyzer can further reduce carbon emissions, ultimately reaching a CO2-negative carbon balance. We coupled the electrochemical CO2 reduction reaction with ethanol oxidation as the anode reaction for acetate production. A nickel foam-supported cobalt selenium catalyst, which is highly active for ethanol oxidation, reached a Faradaic efficiency of nearly 96

Enhancing myrcene biosynthesis in yeast through nuclear compartmentalization.

Myrcene is a high-value monoterpene extensively applied in the fragrance, flavor, and agricultural industries, yet its efficient microbial production remains challenging due to pathway competition and limited metabolic flux. Compartmentalization offers a unique strategy to spatially organize heterologous metabolic pathways in Saccharomyces cerevisiae, enabling improved pathway efficiency through physical separation from competing cytosolic metabolism. In this study, we engineered the S. cerevisiae nucleus as a synthetic metabolic compartment for myrcene biosynthesis. Screening of myrcene synth

Impact of waste-derived ethanol on growth and metabolic allocation in black soldier fly larvae: Implications for organic waste treatment.

Insects possess chemosensory signalling systems that elicit coordinated behavioural and physiological responses to chemical stimuli. This study investigates the effects of ethanol (EtOH) exposure on black soldier fly larvae (BSFL) and evaluates its relevance to organic waste valorisation. Behavioural assays show that BSFL are attracted to EtOH and exhibit a stronger preference for direct contact over volatilised EtOH. Dietary EtOH supplementation enhances BSFL growth at concentrations of 1-5%, with larval dry weight increasing by up to 12% relative to the control. Further, EtOH supplementation
