We compared a lateral flow device to galactomannan and (1-->3)-beta-D-glucan assays to detect invasive aspergillosis in an established guinea pig model of pulmonary disease. The lateral flow device became positive earlier (day 3) than the (1-->3)-beta-D-glucan and galactomannan assays (day 5), with all samples positive by each assay on day 7.

A study was made of the biological properties of the spore culture of anthrax vaccine strain STI-1 lyophilized in 1944 and kept for 30 years without any passages. This dry culture contained not less than 29% of live spores; the culture growth in the nutrient broth and on the agar medium was typical for the strain. Immunogenicity tested in experiments on guinea pigs and rabbits was not reduced and corresponded to that of the reference STI-1 vaccine.

We have studied the origin of tri-iodothyronine (T3) secreted by human and sheep thyrocytes in primary culture and also the expression of type-I thyroidal iodothyronine deiodinase (ID-I) in the thyroid and liver of man and various other animals. Inhibitors of ID-I reduced T3 secretion from human but not sheep thyrocytes. When studied in vivo (in guinea pigs) with the use of electron microscopy, B. mallei (strains C-5, 10230) were found to form a capsule. In the subacute course of infection, the encapsulated forms of B. mallei parasitized mainly in the cells of the system of mononuclear phagocytes in the liver, the spleen and the lungs. The capsule formed by B. mallei was shown to be one of the factors facilitating its persistence in the body.

Synovial fluids from rabbits with antigen-induced arthritis were investigated for the presence of slow-reacting substance of anaphylaxis (SRS-A) using biological assay in guinea-pig ileum. Inflamed joints from rabbits sacrificed within eight hours of the articular challenge disclosed SRS-A activity, indicating that peptidic leukotrienes may also be important in the physiopathology of acute articular inflammatory processes.

The occurrence of contact urticaria was studied in 101 laboratory technicians investigated for allergy to laboratory animals. Fourteen cases of contact urticaria caused by rat were found. Other animals causing contact urticarial reactions were mouse (7), guinea-pig (4), and cat (2). Hand eczema was not found in laboratory technicians with contact urticaria and IgE-antibodies to laboratory animals.

Triphenyl tin fluoride (TPTF) is a bioactive organo-tin compound used in concentrations 2-12% as anti-foulants in boat paints. The chemical is moderately toxic to the skin. An occupational irritant contact folliculitis from TPTF in a marine paint plant worker is described. Contact allergy was excluded by patch testing. An attempt to reproduce the follicular reaction in man, rabbit and guinea pig was unsuccessful.

The beta-adrenergic receptors in the taenia isolated from the guinea pig caecum were irreversibly inactivated by photoaffinity labeling with 1-isoprenaline and 2-(2-hydroxy-3-isopropylaminopropoxy)iodobenzene. The photoinactivation was eliminated in the presence of protectors of the receptors. Photoinactivation caused a considerable parallel shift in the concentration-action curve of 1-isoprenaline. The results possibly point to a certain receptor reserve for 1-isoprenaline.

The experiments on guinea pigs showed that arbidol administered orally in a single dose 24 hours prior vaccination with TEOVAC and ridostin administered in a single dose intranasally on the 4th day after the vaccination lowered the vaccine virus accumulation in the animal organs and tissue without any effect on the vaccine immunogeneity. The results are someway indicative of the possible use of the interferon inductors for prevention of postvaccinal reactions to TEOVAC.

Experimental infections were produced in guinea pigs, rabbits and dogs with lesions similar to those seen in human seborrheic dermatitis and otitis externa by cutaneous application of cultures of Malassezia furfur and M. pachydermatis. Infected animals were treated by topical application of a concentrated yeast killer toxin (Hansenula anomala UCSC 25F). Clinical recovery as well as negative mycological test cultures of infected animals proved to the clearly associated with the treatment by the killer toxin.

The effects of pentacaine and heptacaine were compared with that of procaine on the isolated tracheal smooth muscle of guinea-pigs and anococcygeal muscle of rats. Pentacaine increased the initial tone of the tracheal smooth muscle. In higher concentration however pentacaine and heptacaine induced smooth muscle relaxation. Pentacaine diminished the action of histamine on the tracheal smooth muscle and both pentacaine and heptacaine depressed the effects of noradrenaline and acetylcholine on anococcygeal muscle.

Guinea pigs were sensitized with different doses of homogenous Candida albicans cells introduced by 3 subcutaneous injections at intervals of 2--3 days. The allergic rearrangement in the animal body thus induced was found to depend on the dose of the preparation injected into the animals: the lowest dose of the antigen (0.1 mg) induced immediate allergic reaction, and doses 5--10 times higher induced double rearrangement i.e. immediate and delayed reactions.

Brush border sucrase and alkaline phosphatase activities are considerably enhanced in the intestine of ascorbic acid deficient guinea-pigs. Similar increase in the uptake of D-glucose and L-alanine also occurs in chronic vitamin C deficiency. However the permeability of D-glucose and L-alanine in the intestine of animals fed with large doses of vitamin C is severely depressed, with a reduction in the levels of sucrase and alkaline phosphatase activities.

The immunogenic properties of the freeze-dried preparation of fraction I tisolated from the culture of avirulent Pasteurella pestis strain were studied in experiments on hamadryas baboons, guinea-pigs and white mice. The preparation was found to have a pronounced protective effect in hamadryas baboons and white mice, especially when the preparation was used in mixture with incomplete Freund adjuvant. In hamadryas baboons resistance to infection was shown to correlate with hemagglutionation antibody titres.

Fifteen cases of contact dermatitis and three of irritant contact conjunctivitis due to multifunctional acrylic monomers in printing inks are described. Seven with dermatitis were sensitized to pentaerythritol triacrylate but evidenced no cross-reactivity to methyl methacrylate. The other eight cases of dermatitis were irritant. Pentaerythritol triacrylate has been shown experimentally to be a cutaneous sensitizer for guinea-pigs and man. Multi-functional acrylates should be handled with minimal skin contact, to avoid cutaneous problems.

Inoculation into foot-pads of guinea-pigs of a living suspension of Candida albicans together with Complete Freund's Adjuvant, provoked a hypersensitive state, characterized by: 1) positive skin tests; 2) direct degranulation of peritoneal mast cells when exposed in vitro to the antigen; and 3) homologous "mast cell sensitizing antibodies" detected by the "indirect mast cell degranulation test". The mast cell sensitizing antibodies were heat labile, and their activity could not be restored by complement.

Samples from concentrated aqueous solutions of β-lactam antibiotics were examined by gel filtration. The eluates were monitored by ultraviolet (uv) absorption and by passive cutaneous anaphylaxis (PCA) in guinea-pigs sensitized with appropriate antiserum raised in rabbits. The appearance of uv absorbing material, giving a positive PCA reaction, in fractions from the high molecular weight regions of the eluates from gel filtration of aged solutions was taken as evidence of antigenic polymer formation.

OBJECTIVES: Antiviral vaccines are usually created by inactivating viruses using physical or chemical methods. Inactivation of poliovirus with β-propiolactone (BPL) has advantages, including the absence of a requirement for long-term incubation with the inactivating agent, which minimizes the risk of negative effects of the potentially dangerous substance on the virus and reduces the duration of the inactivation process.
