CD4+ and CD8+ T-cell clones from congenital rubella syndrome patients with IDDM recognize overlapping GAD65 protein epitopes. Implications for HLA class I and II allelic linkage to disease susceptibility.

To fully characterize human glutamic acid decarboxylase (GAD)65 protein T-cell epitopes associated with insulin-dependent diabetes mellitus (IDDM), CTL clones specific to GAD65 protein antigens were isolated from two congenital rubella syndrome (CRS)-associated IDDM patients. Overlapping nonamer T-cell epitopes recognized by both CD4+ or CD8+ CTL clones within peptides GAD65(252-266) and GAD65(274-286) were identified as sequences bounded by GAD65(255-266) with 6/9 overlapping residues, and GAD65(276-285) with 8/9 overlapping residues, respectively, using two panels of overlapping peptide anal

Induction of cytotoxic T-cell activity by the protective antigen of Schistosoma mansoni Sm28GST or its derived C-terminal lipopeptide.

In a previous work the authors demonstrated that immunization with Schistosoma mansoni 28-kDa glutathione-S-transferase (Sm28GST) was able to reduce hepatic damage in infected mice and that the adoptive transfer of Sm28GST-specific T cells reproduced the protective effect obtained with the recombinant molecule. In the present paper, the authors show that Sm28GST is also able to stimulate an antigen-specific, cytotoxic T-cell response against Sm28GST-pulsed P815 target cells in normal mice and that effector cells induced in vivo were classical Class I MHC-restricted CD8+ lymphocytes. Association of eplet mismatch load and allograft injury in kidney transplant recipients.

INTRODUCTION: Eplet mismatch load is a molecular measure of donor-recipient histocompatibility that has been associated with alloimmune outcomes after kidney transplantation. However, its relationship with comprehensive markers of early allograft injury remains incompletely defined. METHODS: In this single-center retrospective cohort study, we evaluated 657 adult kidney transplant recipients who underwent transplantation between January 2021 and December 2024. Eplet mismatch load was calculated at the HLA-A, HLA-B, HLA-C, HLA-DRB1, HLA-DQA1, and HLA-DQB1 loci using the HLA Eplet Registry. The

IL-2 complex therapy prolongs fully MHC-mismatched murine cardiac allograft survival.

Regulatory T cell (Treg)-biased IL-2/anti-IL-2 monoclonal antibody complexes (IL-2c) can preferentially deliver IL-2 to CD25+ Tregs, causing proliferation of Tregs that is potentially advantageous in transplantation. We tested the ability of IL-2c to prolong murine cardiac allograft survival. C57BL/6 (H-2b) mice received fully major histocompatibility complex-mismatched BALB/c (H-2d) or hemi-allogeneic B6 × BALB/c (H-2b/d) F1 allografts. Recipients were treated prior to transplantation with IL-2c or control. Graft survival, anti-donor T cell priming, and donor-specific antibody production were

Neutrophil Antigen Presentation Reprograms the Tumor Microenvironment and Elicits Durable and Broad Antitumor Immunity.

UNLABELLED: Immunotherapy often fails in solid tumors due to an immunosuppressive tumor microenvironment, driven in part by dysfunctional dendritic cells (DC) impairing antigen presentation and suppressive neutrophils. Recent studies revealed a noncanonical role for neutrophils in antigen presentation (nAPC), whose frequency in tumors correlates with improved prognosis in several human cancers. Here, we show that an intravenously delivered antibody-antigen-conjugate (AAC) targeting neutrophil FcγRIIIB reprograms neutrophils into nAPCs that drive robust CD8+ T-cell activation. This response req

CAR-NK Cell Biology and Engineering for Solid Tumors, With a Focus on Lung Cancer.

Over the past decade, chimeric antigen receptor (CAR) T-cell therapy has revolutionized cancer immunotherapy, demonstrating remarkable efficacy in treating relapsed or refractory hematologic malignancies across both pediatric and adult populations. In parallel, CAR-engineered natural killer (CAR-NK) cells have emerged as a complementary and promising alternative to CAR-T therapy, offering several inherent advantages. Unlike CAR-T cells, CAR-NK cells operate independently of major histocompatibility complex (MHC) compatibility and are associated with a lower risk of adverse immune reactions. Th

Effects of Combining Immune-Priming Sub-Lethal Low-Dose Radiation with 4-1BB Activation and Gal-3 Blockade in In Vitro and Preclinical Group-3 Medulloblastoma Models.

BACKGROUND/OBJECTIVES: Pediatric group 3 (G3) medulloblastomas (MB) are therapy resistant and have a significantly worse prognosis than the other MB subtypes. Aggressive radiation/chemotherapy improves survival, but potential long-term comorbidities include neurocognitive deficits. In previous work, we demonstrated that low-dose X-ray radiation (LDXR) acts as an immunological adjuvant. Recent studies have demonstrated that galectin-3 (Gal-3) expression in MB tumors accelerates M2 macrophage infiltration and restricts T cell receptor (TCR)-mediated signaling. Cross-Presentation and Activation of CD8+ T Cells: The Role of Pannexin-1 in Dendritic Cells.

Cross-presentation of exogenous antigens by dendritic cells (DCs) relies on the cytosolic pathway, enabling proteasomal processing and subsequent loading of antigenic peptides onto major histocompatibility complex class I (MHC-I) molecules. Although this pathway is central to CD8+ T-cell activation, the molecular mechanisms that regulate intracellular antigen processing and redistribution during cross-presentation remain incompletely defined. In this study, we investigated the contribution of the large-pore channel Pannexin-1 (Panx1) to antigen handling during cross-presentation. Using confoca

The ubiquitination of CD274 at MERCs enhances the anti-tumor immunity of cervical cancer.

Mitochondria-ER contact sites (MERCs) are crucial signaling hubs, but their role in anti-tumor immunity is unclear. This study revealed that the mitophagy regulator PRKN ubiquitinated CD274 at MERCs in human cervical cancer cells, a key mechanism for anti-tumor immunity. CD274 expression inversely correlated with PRKN in cervical cancer. Upon mitophagy activation, CD274 was recruited from ER to MERCs by PINK1, enhancing its interaction with PRKN. PRKN then ubiquitinated CD274 at residues K89 and K105 within its extracellular domain. Functionally, a ubiquitination-deficient CD274 mutant promote

Primary and acquired resistance to immunotherapy in NSCLC.

Non-small cell lung cancer (NSCLC) is one of the leading causes of cancer incidence and mortality worldwide. In recent years, immune checkpoint inhibitors (ICIs), particularly those targeting the programmed cell death protein 1/programmed death-ligand 1 (PD-1/PD-L1) axis, have significantly improved survival outcomes in a subset of patients. However, the magnitude and durability of clinical benefit vary considerably according to PD-L1 expression, treatment setting, histological subtype, oncogenic driver status, and whether ICIs are administered as monotherapy or in combination regimens. Spatially resolved single-cell landscape of tumor immunotypes reveals the central role of interferon signaling and plasmacytoid dendritic cells in triple-negative breast cancer.

BACKGROUND: Tumor-infiltrating lymphocytes (TILs) are established determinants of clinical outcomes in breast cancer.1-6 While stromal TILs (sTILs) are commonly evaluated due to easier scoring and higher interobserver reproducibility, intraepithelial TILs may provide additional insights into anti-tumor immunity.7 We aimed to comprehensively characterize immunotype-associated cellular landscapes in triple-negative breast cancer (TNBC) and identify features associated with clinical outcomes. METHODS: 212 TNBC tumors were classified into three immunotypes based on immune infiltrate location and

A Complete Genome for the Common Marmoset.

The common marmoset is a New World monkey (NWM) commonly used as a model organism to investigate questions in primate evolution and human disease, including Alzheimer's and other neurodegenerative diseases, as well as neuropsychiatric disorders. Here we present the first telomere-to-telomere (T2T) reference genome for the common marmoset, adding over 88 Mb of sequence and resolving challenging genomic regions. An additional near-T2T assembly from a second unrelated individual yields a total of four high-quality haplotypes for analysis. The improved contiguity and accuracy of these assemblies e

Opposing Ageing-Related Transcriptomic Profiles Distinguish Opioid Exposure Liability From Dependence Liability.

Opioid use disorder (OUD) involves separable stages of exposure and dependence, but many genetic studies have treated these liabilities as a single phenotype. In this study, transcriptome-wide association study (TWAS) methods were applied to the Psychiatric Genomics Consortium Substance Use Disorders 2020 opioid meta-analysis summary statistics to examine ageing-related gene pathways across brain tissues. Genetic liability for opioid exposure was strongly associated with reduced predicted expression of complement components and synaptic-pruning genes, together with negative signals in astrocyt

Virtual single-cell perturbation and genetic causal inference reveal CSF1R-dependent immunometabolic communication in iron metabolism-associated osteoarthritis.

Iron dysregulation has emerged as a contributor to osteoarthritis (OA), yet the cell-communication mechanisms connecting iron-related genetic signals to joint degeneration remain insufficiently defined. Here, we established an integrative computational framework combining transcriptomic screening, machine learning, Mendelian randomization, immune and metabolite mediation analysis, single-cell transcriptomics, virtual gene perturbation, molecular docking, molecular dynamics simulation, and experimental validation to identify signaling regulators involved in iron metabolism-associated OA. Distinct Effects of Complement C4A and C4B Copy Numbers in Systemic Sclerosis Serological and Clinical Subtypes.

OBJECTIVE: Complement component 4 (C4), encoded by C4A and C4B within the major histocompatibility complex (MHC) on chromosome 6, regulates the immune response and clears immune complexes. The variable copy number (CN) of C4 genes and retroviral human endogenous retrovirus K (HERV-K) element influence its function. Given the relationship of C4 CN with systemic sclerosis (SSc) risk, we assessed associations with SSc clinical and serologic subtypes. METHODS: We compared imputed C4 CNs across SSc subgroups (4,049 anticentromere positive [ACA+]; 2,200 anti-topoisomerase I [ATA+]; 577 anti-RNA pol

Synergizing radiotherapy and immunotherapy for locally advanced gastric cancer: evolving paradigms and future directions.

This article innovatively reviews and unveils the synergistic mechanisms, clinical research directions, and future challenges of the combination of preoperative radiotherapy (RT) and immunotherapy (especially the most popular belonging to immune checkpoint inhibitors, ICIs) in the treatment of locally advanced gastric cancer and gastroesophageal junction adenocarcinoma (GC/GEA). The integration of RT and ICIs represents a promising therapeutic strategy for locally advanced, even unresectable, GC/GEA. RT potentiates antitumor immunity by inducing immunogenic cell death (ICD) and targeting iron

Autophagy functions in lung macrophages and dendritic cells to regulate allergen-dependent inflammatory responses.

Asthma affects 260 million people worldwide, with severe asthma cases that are associated with TH17 and TH1 responses and neutrophil-dominated inflammation being the most difficult to treat due to corticosteroid insensitivity. Single nucleotide polymorphisms in the ATG5 gene, which codes for a protein required for the cellular recycling process of macroautophagy/autophagy, are associated with higher risk for developing severe asthma. We explored the mechanistic basis for the genetic association of ATG5 variants with severe asthma. We identified an autophagy-dependent role for ATG5 in lung macr
