The proliferative response of B cell precursor acute lymphoblastic leukemia (BCP-ALL) cells to IL-3 is dependent on the expression of functional IL-3 receptors (IL-3R). Here we report that CD40 ligand (CD40L) in the presence of recombinant IL-3 increased proliferation of BCP-ALL cells by upregulating expression of IL-3R. Upregulation of IL-3R in BCP-ALL cells was observed as early as 1 h after treatment with CD40L, and a 50- to 500-fold increase of IL-3R expression after 24 h was detected in all 12 cases studied. Moreover, expression of receptors for IL-7 (IL-7R) and stem cell factor (SCF-R, c

The purpose of this study was to analyse the proliferative fraction with the monoclonal antibody M1-R-R to M1-subunit ribonucleotide reductase and with MIB-1 to Ki-67 antigen in relation to p53 protein expression in fine needle aspirates from B-cell non-Hodgkin's lymphomas. One hundred and thirty-seven cases, previously diagnosed and sub-typed according to the Kiel classification and characterized by immunophenotyping, were included in the study. The M-1 subunit ribonucleotide reductase (M1-R-R), Ki-67 and p53 antigens were detected using monoclonal antibodies on stored cytospin preparations.

BACKGROUND: In rodents, CsA has been shown to affect T-cell development, giving rise to an abnormal production of mature T cells and the absence of many T-cell subsets as well as to autoimmunity. Surprisingly, only a few studies investigated the effect of the immunosuppressive drug on the immune system of the human fetus.
METHODS: We examined six infants born to female kidney transplant recipients who had received cyclosporine and methylprednisolone throughout their pregnancies. Peripheral blood was obtained 1 day and 2, 4, 6, and 12 months after birth, and two-color flow cytometric immunophe

The t(12;21)(p13;q22) is the most common translocation in childhood B-precursor ALL. It results in a TEL-AML1 rearrangement and is associated with a good prognosis. Because many chromosomal alterations in leukemia are associated with distinct cell surface phenotypes, we investigated whether there was an association seen between surface marker expression and the TEL-AML1 rearrangement. Of 166 unselected cases of B-precursor ALL studied by Southern hybridization, 45 cases (27%) showed TEL rearrangement. Blasts of patients with TEL rearrangement were much more likely to be CD9-negative, CD45-posi

CD30(Ki-1) positive anaplastic large cell lymphoma (ALCL) is a distinct entity, in which the monoclonal antibody-positivity against the CD30(Ki-1) antigen of tumour cells has a diagnostic value. The histological subtypes of ALCL show also certain clinical differences. Except for some pediatric cases and cutaneous forms clinical outcome is very unfavourable despite of the various treatment methods. In this prospective study (follow-up of 11-60, median 16 months) clinicopathological data and treatment results of fifteen adult patients with ALCL were analysed, Mean age was 46 (16-69) ys with a bi

We have sought the presence of rearrangements of the immunoglobulin heavy chain gene locus in 13 patients with chronic myeloid leukemia (CML) in lymphoid blastic transformation (L-BT) using the polymerase chain reaction (PCR). The lymphoid nature of the transformation was confirmed by immunophenotyping and/or Southern blot hybridization with a J(H) probe. Clonal rearrangements were detected in 85% of cases and two or more rearrangements were visible in 64% of informative cases. The pattern of V(H) gene family utilization revealed an apparent reduction in V(H)4 family gene usage but otherwise r

One of the goals of the study NHL-BFM 90 was to investigate the distribution and prognosis of the different subtypes of Non-Hodgkin's Lymphoma (NHL) in children and adolescents according to histological, cytomorphological and immunological characteristics. From 4/1990 to 12/1992, 346 patients (pts) (84 females, 262 males) were enrolled (median age: 9.1 years; range: 0.8-17.9 years). Histology was available from 290 pts (84%), cytomorphology from 155 (44%), and immunophenotyping from 245 (70%). Cases with L1 oder L2 cytomorphology according to the French-American-British Classification were cla

When immunophenotyping is used for diagnosis, it is essential to use panels of antibodies, rather than a single marker for a specific tumor type. There is virtually no single marker that is absolutely diagnostic and completely reliable for any particular neoplasm. For the most important clinical differential diagnoses--reactive versus neoplastic lymphoid lesions and lymphoma versus nonlymphoid tumor--relatively simple panels of antibodies are usually sufficient. In the former situation, immunoglobulin heavy and light chains (G, M, kappa, lambda) and a pan-T-cell antibody (Leu-4) will usually p

Hepatoid carcinoma of the ovary (HCO) exhibits morphological and immunophenotypic features highly similar to those of hepatocellular carcinoma. Immunohistochemical staining for alpha-fetoprotein, hepatocyte paraffin antigen 1, and glypican-3 is an important auxiliary approach for the clinical diagnosis of HCO. Currently, most treatment regimens for this disease are developed with reference to clinical management protocols for high-grade epithelial ovarian cancer. Hepatoid carcinoma of the ovary (HCO) is an extremely rare, distinct histological subtype of primary ovarian adenocarcinoma that exh

A 15-year-old girl who had previously undergone surgery and chemotherapy for non-metastatic chondroblastic osteosarcoma of the left fibula was evaluated with 18F-fluorodeoxyglucose positron emission tomography/computed tomography (18F-FDG PET/CT) during routine follow-up while in clinical remission. Imaging demonstrated a 36 × 17 mm hypermetabolic lesion in the right occipital condyle maximum standard uptake value [(SUVmax): 16.5], with adjacent bone and soft-tissue extension, together with a concurrent 18F-FDG-avid pulmonary nodule (SUVmax: 3.4). Shortly after imaging, the patient developed m

Solid pseudopapillary neoplasm (SPN) of the pancreas is a rare epithelial tumor with low malignant potential, predominantly affecting young women and only rarely encountered in the pediatric population. Although the prognosis is generally favorable following complete surgical excision, diagnostic challenges may arise due to unusual histomorphology or aberrant immunohistochemical expression. We report the case of a seven-year-old female patient who presented with abdominal pain and vomiting and was found to have a large intra-abdominal mass initially suspected to be of mesenteric origin on imag

Sézary syndrome (SS) is an aggressive cutaneous T-cell lymphoma, characterized by erythroderma, lymphadenopathy and circulating malignant CD4+ T cells. Despite therapeutic advances, SS remains an incurable disease. Early diagnosis is therefore essential for timely therapeutic intervention; however, reliable biomarkers to identify circulating SS cells are still lacking. Recent studies highlighted dysregulation of the ectoenzymes CD39, CD73 and CD38 in SS, suggesting a potential role for these molecules in the disease. Thirty-three patients were enrolled in this study, and CD39/CD73/CD38 express

Despite advances in systemic therapies for patients with (locally) advanced stage lung cancer, response rates remain poor, underscoring the urgent need for predictive models to guide therapy selection. Patient-derived ex vivo models are promising, however, their clinical utility is restricted by the need for surgical biopsies, low establishment rates, and culture durations exceeding the clinically-relevant therapeutic decision-making window. Here, we developed and clinically validated a rapid, high-throughput ex vivo 3D tumor replica platform that enables functional drug testing from small dia

AIMS: Acinar cell carcinoma (ACC) is a lineage-specific carcinoma that occurs nearly exclusively in the pancreas. We report, to our knowledge, the first series of gallbladder carcinoma demonstrating extensive acinar differentiation in the absence of a pancreatic primary.
METHODS: Three gallbladder carcinomas with extensive acinar differentiation were identified from an international cohort. Clinical, radiologic, macroscopic, histologic, immunophenotypic and molecular features were reviewed.
RESULTS: All tumours lacked an associated pancreatic primary on imaging and clinical evaluation. Two t

Inflammatory and nested testicular sex cord tumor (IN-TSCT) is a recently characterized malignant neoplasm within the spectrum of testicular sex cord-stromal tumors. Previously misclassified as Sertoli cell tumor, not otherwise specified, or as seminoma, this entity has emerged as a distinct clinicopathologic and molecular subtype defined by recurrent EWSR1::ATF1 gene fusions and a potentially aggressive clinical course. Patients most commonly present with unilateral painless testicular enlargement, and radiologic findings are typically nonspecific. Histologically, tumors demonstrate solid and

Bulk sequencing of relapsed tumors reveals mutations associated with resistance to cancer therapy but is insufficient to fully assess all causes of relapse. Because of inherent tumor heterogeneity, on-treatment tumor evolution may select genetically distinct clones or shifts in malignant transcriptional states not resolvable by bulk sequencing. We performed multiomic single-cell DNA/protein and RNA/protein profiling of a clinical trial cohort of patients with acute myeloid leukemia (AML) treated on the phase 1b clinical trial of the BCL2 inhibitor venetoclax and the FLT3 inhibitor gilteritinib

Urine-derived stem cells (UDSCs) have emerged as a promising cell source for regenerative medicine due to their non-invasive procurement, high proliferative capacity, and potential relevance to kidney-specific repair. Unlike conventional mesenchymal stem cells (MSCs) obtained from bone marrow or adipose tissue, UDSCs originate from multiple regions of the urinary tract and exhibit a unique biological profile that combines MSC characteristics with features of renal progenitor populations. This review provides a comprehensive overview of the current understanding of UDSC biology, including their

With the widely increasing implementation of modern next generation sequencing (NGS) technologies in routine molecular pathology practice, the fraction of cancers with a definite or probable hereditary background has been steadily increasing. Currently, it is assumed that 5-10% of all malignancies develop in the context of germline predisposition diseases. Not rarely, the diagnosis and recognition of cancer predisposition syndromes rely on distinctive histopathological and/or immunophenotypical findings that proved to be highly reliable and reproducible in uncovering hereditary neoplastic dise

Understanding how metabolism governs human hematopoietic stem cells (HSCs) function is essential for advancing regenerative therapies, yet direct metabolic profiling of human HSCs has been limited by their extreme scarcity and the technical limitations of conventional methods. Here, we applied a low-input mass spectrometry-based metabolomics platform, optimized for rare cell populations, to generate metabolic profiles of 13 immunophenotypically defined hematopoietic cell types from adult human bone marrow. Using as few as ∼10 000 cells per sample, we detected >80 metabolites and uncovered both
