Dissecting the effect of blood colony-stimulating factors and receptors on Alzheimer's disease: the role of myeloid traits and inflammation.

BACKGROUND: Research has suggested a potential link between colony-stimulating factors (CSFs) and Alzheimer's disease (AD), but the findings have been inconsistent, and the causal relationship remains uncertain. METHODS: We performed a Mendelian Randomization (MR) analysis to explore the association between blood levels of CSFs and their receptors with AD and its biomarkers. The study utilized summary-level data on blood levels of granulocyte colony-stimulating factor (G-CSF), granulocyte-macrophage colony-stimulating factor, macrophage colony-stimulating factor, and their receptors from deCO

Expression of stearoyl coenzyme a desaturase in neuronal cells facilitates pancreatic cancer progression.

BACKGROUND: Pancreatic adenocarcinoma (PDAC) is the most fatal malignant tumor that focuses on men and the elderly (40-85 years) and is aggressive. Its surgical resection rate is only 10-44%, and the rate of local recurrence in the retroperitoneum 1 year after surgery is as high as about 60%. The main reason for local recurrence in the majority of patients is that PDAC is perineural invasion (PNI) and the cancer cells infiltrate and grow along the peripancreatic nerve bundles. The identification of biomarkers associated with the diagnosis of PDAC may help to improve the current difficulty in e

Causal relationship between circulating cytokines and follicular lymphoma: a two-sample Mendelian randomization study.

Follicular lymphoma (FL), derived from germinal centre (GC) B cells, is a kind of systemic neoplasm. Even though FL is indolent, it remains an incurable haematology Neoplasm. Accumulating evidence has suggested that the circulating cytokine is associated with the development of FL, yet the causal relationship between FL and circulating cytokines remains undetermined. Therefore, we conducted a two-sample Mendelian randomization (MR) to confirm the causal link between FL and levels of circulating cytokines with the use of summary data on circulating cytokines and FL. All these data from genome-w

Association between genetically proxied glucosamine and risk of cancer and non-neoplastic disease: A Mendelian randomization study.

INTRODUCTION: Observational investigations have examined the impact of glucosamine use on the risk of cancer and non-neoplastic diseases. However, the findings from these studies face limitations arising from confounding variables, reverse causation, and conflicting reports. Consequently, the establishment of a causal relationship between habitual glucosamine consumption and the risk of cancer and non-neoplastic diseases necessitates further investigation. METHODS: For Mendelian randomization (MR) investigation, we opted to employ single-nucleotide polymorphisms (SNPs) as instruments that exh

Mendelian randomization supports causality between gut microbiota and chronic hepatitis B.

BACKGROUND: Observational studies have provided evidence of a close association between gut microbiota and the progression of chronic hepatitis B (CHB). However, establishing a causal relationship between gut microbiota and CHB remains a subject of investigation. METHODS: Genome-wide association study (GWAS) summary data of gut microbiota came from the MiBioGen consortium, while the GWAS summary data of CHB came from the Medical Research Council Integrative Epidemiology Unit (IEU) Open GWAS project. Based on the maximum likelihood (ML), Mendelian randomization (MR)-Egger regression, inverse v

Smoking, alcohol consumption, diabetes, body mass index, and peptic ulcer risk: A two-sample Mendelian randomization study.

Background: Observational evidence has shown that smoking, alcohol consumption, type 2 diabetes, and body mass index (BMI) are risk factors for peptic ulcer disease (PUD), including gastric ulcer (GU) and duodenal ulcer (DU). However, the observed associations may be confounding factors. Herein, we use Mendelian randomization (MR) to examine causal associations such as smoking, alcohol, type 2 diabetes, BMI, and risks of PUD. Methods: We used 8,17,41,325,82, 231, and 616 identified genetic variants as proxies for age of smoking initiation (AgeSmk), smoking cessation (SmkCes, current/former), n

Placenta mediates the effect of maternal hypertension polygenic score on offspring birth weight: a study of birth cohort with fetal growth velocity data.

BACKGROUND: Low birth weight (LBW) and fetal growth restriction are associated with the development of cardio-metabolic diseases later in life. A recent Mendelian randomization study concluded that the susceptibility of LBW infants to develop hypertension during adulthood is due to the inheritance of hypertension genes from the mother and not to an unfavorable intrauterine environment. Therein, a negative linear association has been assumed between genetically estimated maternal blood pressure (BP) and birth weight, while the observed relationship between maternal BP and birth weight is substa

Novel immune-related susceptibility loci associated with pediatric steroid-sensitive nephrotic syndrome identified by a transethnic genome-wide association study.

BACKGROUND: Steroid-sensitive nephrotic syndrome (SSNS) is a relatively rare glomerular disease with considerable clinical and genetic heterogeneity. Although efforts have been made to explore the genetic determinants of SSNS, the underlying genetic architecture has still been hampered by limited sample size and remains to be elucidated. METHODS: We conducted a genome-wide association study involving 510 sporadic pediatrics of SSNS and 1062 controls with Chinese ancestry, followed by a transethnic meta-analysis involving 2942 cases and 37 041 controls. We evaluated the colocalization probabil

Genetically predicted circulating polyunsaturated fatty acids and Crohn's disease susceptibility: A two-sample mendelian randomization study.

Crohn disease (CD) remains an increasing global health burden. Omega-3 and omega-6 polyunsaturated fatty acids participate in lipid metabolism and inflammatory regulation, but previous observational and interventional studies have produced inconsistent findings. This study aimed to assess whether genetically predicted circulating omega-3, omega-6, and the omega-6/omega-3 ratio are associated with CD susceptibility using a two-sample Mendelian randomization (MR) design. Genome-wide association study summary data for circulating omega-3, omega-6, and the omega-6/omega-3 ratio were obtained from

Causal Effects of Gut Microbiota and Associated Metabolites on Retinal Diseases and Visual Impairment: A Mendelian Randomization Study.

BACKGROUND: Previous observational study findings have indicated a vital association between gut microbiota features and retinal diseases based on the "gut-retina" axis. However, whether their relationships underlie causal effects remains to be established. METHODS: Instrumental variables of 211 gut microbiota taxa were obtained from a genome-wide association study (GWAS), and 28 gut-associated metabolites and pathways were included as exposures. A two-sample Mendelian randomization (MR) study was carried out to estimate gut microbiota effects on diabetic retinopathy (DR), early age-related m

CD4+CD8+ double-positive T cells are associated with severity of tuberculosis.

BACKGROUND: Tuberculosis (TB) remains a global public health burden, and how immune cell subsets regulate host anti-TB immunity and disease progression remains incompletely understood. While previous studies have focused on single-positive (SP) T cells (CD4+ or CD8+) in TB pathogenesis, the association between CD4+CD8+ double-positive (DP) T cells and TB susceptibility, severity, and treatment outcomes have not been fully elucidated. This study aimed to investigate the relationship between DP T cells and other immune cell subsets with TB, and to explore the potential diagnostic and prognostic

Dietary Vitamin C Intake Affects Lung Function Through White Blood Cell.

As an antioxidant, vitamin C has been increasingly used in the treatment of various pulmonary diseases in recent years. However, the mechanism by which vitamin C affects lung function remains unclear to this day. Given its low cost and low risk, vitamin C is highly suitable for widespread use as a conventional treatment, making research into its mechanisms of influencing lung function necessary. Considering the potential association between vitamin C and white blood cells (WBCs), it may influence lung function by affecting white blood count (WBC). The potential impacts of WBCs on the lungs may

Integrated multi-omics and causal inference framework with experimental validation reveals key drivers of air pollution-induced acute kidney injury.

BACKGROUND: Air pollution has emerged as a significant risk factor for acute kidney injury (AKI), yet the molecular mechanisms underlying this association remain poorly defined. This study aimed to elucidate the nephrotoxic effects of representative air pollutants and identify molecular targets involved in pollutant-induced AKI. METHODS: We developed a multi-layered computational and experimental framework integrating omics-based target prediction, network toxicology, machine learning, Mendelian randomization (MR), single-cell profiling, molecular docking with dynamic simulations, and analysi

Unraveling the causal role of TGF-βRII in osteoporosis and the potential of its associated differential genes as novel targets.

BACKGROUND: Osteoporosis (OP) is a common disorder associated with reduced density of the bones, increasing the likelihood of fractures and deformities. The global incidence of OP is rising rapidly due to aging populations and lifestyle changes, affecting not only older adults but, increasingly, younger and middle-aged individuals. The long-term care and rehabilitation required for OP-induced fractures and complications impose significant economic burdens on both individuals and society, as well as reducing their quality of life. METHODS: Recent studies have demonstrated an association betwee
