Blocking NS3-NS4B interaction inhibits dengue virus in non-human primates.

Dengue is a major health threat and the number of symptomatic infections caused by the four dengue serotypes is estimated to be 96 million1 with annually around 10,000 deaths2. However, no antiviral drugs are available for the treatment or prophylaxis of dengue. We recently described the interaction between non-structural proteins NS3 and NS4B as a promising target for the development of pan-serotype dengue virus (DENV) inhibitors3. Here we present JNJ-1802-a highly potent DENV inhibitor that blocks the NS3-NS4B interaction within the viral replication complex. JNJ-1802 exerts picomolar to low

Interaction Profiles of Central Nervous System Active Drugs at Human Organic Cation Transporters 1-3 and Human Plasma Membrane Monoamine Transporter.

Many psychoactive compounds have been shown to primarily interact with high-affinity and low-capacity solute carrier 6 (SLC6) monoamine transporters for norepinephrine (NET; norepinephrine transporter), dopamine (DAT; dopamine transporter) and serotonin (SERT; serotonin transporter). Previous studies indicate an overlap between the inhibitory capacities of substances at SLC6 and SLC22 human organic cation transporters (SLC22A1-3; hOCT1-3) and the human plasma membrane monoamine transporter (SLC29A4; hPMAT), which can be classified as high-capacity, low-affinity monoamine transporters. However,

Patient with H syndrome, cardiogenic shock, multiorgan infiltration, and digital ischemia.

BACKGROUND: H syndrome (HS) is a rare autoinflammatory disease caused by a mutation in the solute carrier family 29, member 3 (SCL29A3) gene. It has a variable clinical presentation and little phenotype-genotype correlation. The pathognomonic sign of HS is cutaneous hyperpigmentation located mainly in the inner thighs and often accompanied by other systemic manifestations. Improvement after tocilizumab treatment has been reported in a few patients with HS. We report the first patient with HS who presented cardiogenic shock, multiorgan infiltration, and digital ischemia. CASE PRESENTATION: 8-y

Non-structural protein 1 from Zika virus: Heterologous expression, purification, and potential for diagnosis of Zika infections.

Zika virus (ZIKV) infections were associated with neurological disorders only after the Brazilian outbreak in 2015. The lack of vaccines and precise diagnosis requires a precise method to detect ZIKV infection. This study aimed to evaluate three ZIKV recombinant proteins for the development of ZIKV infections. Here, it was purified stable recombinant ZIKV Capsid (r-ZIKV-c), non-structural proteins NS1 (r-ZIKV-NS1), and NS3 (r-ZIKV-NS3) for detection of the infection by ZIKV in blood sera of patients. A commercial polyclonal antibody recognized the r-ZIKV-NS1. Here, among three proteins, NS1 sh

Proline-Based Allosteric Inhibitors of Zika and Dengue Virus NS2B/NS3 Proteases.

The NS2B/NS3 serine proteases of the Zika and Dengue flaviviruses are attractive targets for the development of antiviral drugs. We report the synthesis and evaluation of a new, proline-based compound class that displays allosteric inhibition of both proteases. The structural features relevant for protease binding and inhibition were determined to establish them as new lead compounds for flaviviral inhibitors. Based on our structure-activity relationship studies, the molecules were further optimized, leading to inhibitors with submicromolar IC50 values and improved lipophilic ligand efficiency

Prevalence of naturally occurring protease inhibitor resistance-associated variants in hemodialysis and renal transplant patients with hepatitis C virus infection.

Background NS3 protease inhibitors (PIs) were the first direct antiviral agents used for the treatment of hepatitis C virus. The combination of second-wave PIs with other direct antiviral agents enabled the use of interferon-free regimens for chronic kidney disease patients on dialysis and renal transplant (RTx) recipients, populations in which the use of interferon and ribavirin is limited. However, the occurrence of PI resistance-associated variants (RAVs), both baseline and induced by therapy, has resulted in the failure of many treatment strategies. Methods The aim of this study was to est

In Vitro Antiviral Activity and Resistance Profile Characterization of the Hepatitis C Virus NS5A Inhibitor Ledipasvir.

Ledipasvir (LDV; GS-5885), a component of Harvoni (a fixed-dose combination of LDV with sofosbuvir [SOF]), is approved to treat chronic hepatitis C virus (HCV) infection. Here, we report key preclinical antiviral properties of LDV, including in vitro potency, in vitro resistance profile, and activity in combination with other anti-HCV agents. LDV has picomolar antiviral activity against genotype 1a and genotype 1b replicons with 50% effective concentration (EC50) values of 0.031 nM and 0.004 nM, respectively. LDV is also active against HCV genotypes 4a, 4d, 5a, and 6a with EC50 values of 0.11

Inhibition of adenosine kinase attenuates inflammation and neurotoxicity in traumatic optic neuropathy.

Traumatic optic neuropathy (TON) is associated with apoptosis of retinal ganglion cells. Local productions of reactive oxygen species and inflammatory mediators from activated microglial cells have been hypothesized to underlie apoptotic processes. We previously demonstrated that the anti-inflammatory effect of adenosine, through A2A receptor activation had profound protective influence against retinal injury in traumatic optic neuropathy. This protective effect is limited due to rapid cellular re-uptake of adenosine by equilibrative nucleotside transporter-1 (ENT1) or break down by adenosine

Lineage-specific purification of neural stem/progenitor cells from differentiated mouse induced pluripotent stem cells.

Since induced pluripotent stem (iPS) cells have differentiation potential into all three germ layer-derived tissues, efficient purification of target cells is required in many fields of iPS research. One useful strategy is isolation of desired cells from differentiated iPS cells by lineage-specific expression of a drug-resistance gene, followed by drug selection. With this strategy, we purified neural stem/progenitor cells (NSCs), a good candidate source for regenerative therapy, from differentiated mouse iPS cells. We constructed a bicistronic expression vector simultaneously expressing blast

Bcr-Abl dependent post-transcriptional activation of NME2 expression is a specific and common feature of chronic myeloid leukemia.

We have previously identified NME2 (Nm23-H2) as a tumor antigen in a patient with chronic myeloid leukemia (CML). Here we investigated the association between NME2 and Bcr-Abl. NME2 protein was highly overexpressed in the cytoplasm of peripheral blood mononuclear cells from 29/30 patients with CML at diagnosis and 10/10 patients resistant to imatinib. Protein was overexpressed in the absence of increased levels of mRNA and was limited to Bcr-Abl + populations, being absent from Bcr-Abl - patient cells, normal donors and 14/15 acute myeloid leukemia (AML) samples. Furthermore, the Bcr-Abl depen

Nonconsensus West Nile virus genomes arising during mosquito infection suppress pathogenesis and modulate virus fitness in vivo.

West Nile virus (WNV) is similar to other RNA viruses in that it forms genetically complex populations within hosts. The virus is maintained in nature in mosquitoes and birds, with each host type exerting distinct influences on virus populations. We previously observed that prolonged replication in mosquitoes led to increases in WNV genetic diversity and diminished pathogenesis in mice without remarkable changes to the consensus genome sequence. We therefore sought to evaluate the relationships between individual and group phenotypes in WNV and to discover novel viral determinants of pathogene

Viral protease inhibitors.

This review provides an overview of the development of viral protease inhibitors as antiviral drugs. We concentrate on HIV-1 protease inhibitors, as these have made the most significant advances in the recent past. Thus, we discuss the biochemistry of HIV-1 protease, inhibitor development, clinical use of inhibitors, and evolution of resistance. Since many different viruses encode essential proteases, it is possible to envision the development of a potent protease inhibitor for other viruses if the processing site sequence and the catalytic mechanism are known. At this time, interest in develo

Poly(adenosine diphosphate-ribosylation) of nuclear matrix proteins in alkylating agent resistant and sensitive cell lines.

Using Walker 256 breast carcinoma cell lines either with or without acquired resistance to alkylating agents, the structural framework proteins of the nucleus, the nuclear matrix proteins, were found to be effective acceptors for poly(ADP-ribose). Incubation of isolated nuclei with nicotinamide adenine [32P] dinucleotide ([32P] NAD), followed by the isolation of the nuclear matrix, demonstrated that two polypeptides of approximate molecular weight (Mr) 105 000 and 116 000 were extensively poly(ADP-ribosylated). By an in vitro [32P] NAD assay, the nuclear matrix fraction was found to maintain a

In silico design and molecular docking evaluation of antisense oligonucleotides targeting IDO1 mRNA in the context of depression.

Depression is a prevalent mental disorder worldwide, often accompanied by severe complications that significantly impair patients' daily lives. Indoleamine-2,3-dioxygenase 1 (IDO1), a key metabolic enzyme in the kynurenine pathway, is considered a promising therapeutic target for depression treatment. Antisense oligonucleotides (ASOs) are small nucleic acid drugs characterized by high specificity, high potency, minimal drug-drug interactions, a prolonged duration of action, and a low risk of off-target activity, making them effective tools for targeting molecules previously considered undrugga

Fabrication, spectral characterisation and exploration of DNA/RNA binding affinity and biological potential of transition metal complexes incorporating isoniazid and aromatic nitrogen-donor ligands.

Four mixed-ligand transition metal(II) complexes, [ML(L1)Cl2] (where M = Co, Ni, Cu, and Zn; L = Schiff base derived from isoniazid and 3-nitrobenzaldehyde; L1 = 1,10-phenanthroline), were synthesised and characterised using elemental analysis, FT-IR, UV-Vis, 1H NMR, EPR (for Cu), and molar conductance measurements. Physicochemical and spectroscopic data suggested an octahedral geometry for all complexes. Antimicrobial assays revealed that the complexes exhibited greater activity than the free Schiff base ligand, with inhibition zones of 14-22 mm against Staphylococcus aureus and Escherichia c

Design, synthesis, and biological properties of novel metabolizable pH-responsive lipids for use in lipid nanoparticles for siRNA delivery and mRNA vaccination.

We developed pH-responsive lipids, LIPIDs-1 and LIPIDs-2, which feature an N,N-dimethylaminopropyl head and 2 hydrophobic tails connected by carbonate esters. These lipids are metabolizable owing to acetyl ester structures that can be cleaved by esterases, reducing their affinity for biological membranes after hydrolysis of the esters. We evaluated both lipids in vivo using siRNA-loaded lipid nanoparticles (LNPs) targeting Factor VII, which resulted in significant knockdown activity for up to 6 days. Toxicological studies of LNPs using LIPIDs-1 and LIPIDs-2 in rats indicated minimal liver toxi

USP53 regulates glycolytic reprogramming and accelerates Abdominal aortic aneurysm progression and rupture.

Abdominal aortic aneurysm (AAA) rupture remains a fatal vascular event with limited mechanistic understanding beyond diameter-based risk assessment. Emerging evidence suggests that metabolic reprogramming contributes to aneurysm progression, yet regulators linking metabolism to rupture remain poorly defined. An AngII/BAPN-induced mouse model generated intact (iAAA) and ruptured AAA (rAAA) tissues. Bulk RNA sequencing and untargeted metabolomics profiled transcriptomic and metabolic alterations across sham, iAAA, and rAAA groups. Candidate ubiquitin-specific proteases (USPs) were screened by in

Post-COVID increase in N-methyl-2-pyridone-5-carboxamide (Met2PY) and N-methyl-4-pyridone-3-carboxamide (Met4PY) associates with inflammatory and endothelial activation markers.

COVID-19 is associated with long-term vascular complications, but the underlying mechanisms remain incompletely understood. During infection, NAD+ homeostasis becomes dys-regulated, with excessive NAD+ consumption and enhanced catabolic flux through nicotinamide methylation pathways. This imbalance leads to NAD+ depletion accompanied by accumulation of pyridone metabolites, including N-methyl-2-pyridone-5-carboxamide (Met2PY) and N-methyl-4-pyridone-3-carboxamide (Met4PY). These derivatives have been linked to oxidative stress, endothelial dysfunction, and cardiovascular risk, yet their role i

"Aptamer-guided therapeutics and diagnostics for metabolic syndrome: Design, delivery, and translation of aptamer-based systems".

Metabolic syndrome (MetS) is a composite cardiometabolic condition defined by visceral obesity, dyslipidemia, hypertension, and insulin resistance, and it promotes the worldwide prevalence of type 2 diabetes and atherosclerotic cardiovascular disease. Nucleic-acid aptamers have developed as programmable recognition ligands that integrate antibody-like specificity with chemical synthesis, modular functionalization, low batch variability, and usually a low immunogenicity. Their translational significance in MetS is not solely in target binding, but in the capacity to establish disease-relevant a
