Precision Targeting of KRAS-Mutant Cancers: Beyond G12C Toward G12D and Pan-RAS Therapeutic Strategies.

KRAS is one of the most frequently mutated oncogenes in human cancer and has long been considered an "undruggable" therapeutic target because of its high affinity for guanine nucleotides and limited druggable binding pockets. Recent advances in structural biology and molecular pharmacology have transformed this paradigm, leading to the successful development of KRAS G12C inhibitors such as sotorasib and adagrasib. These agents established proof-of-concept for direct KRAS inhibition and marked an important advance in precision oncology. However, intrinsic and acquired resistance mechanisms, ada

Inosine misincorporation into mRNA triggers the integrated stress response and activates an innate immune gene expression signature.

The inosine triphosphate pyrophosphatase (ITPase) enzyme restricts levels of the non-canonical nucleotides (deoxy)inosine triphosphate (dITP/ITP) and prevents their aberrant misincorporation into nucleic acids. ITPase deficiency is associated with dilated cardiomyopathy and epileptic encephalopathy in humans and is usually fatal in infancy. It leads to pronounced inosine misincorporation into RNA but the cellular consequences of this misincorporation are not well understood and the pathogenic basis of ITPase deficiency remains unknown. Here we show that cellular transfection of mRNA with inosi

Pillararene-based fluorescence indicator displacement assays for the ratiometric and turn-on detection of ATP.

For ATP detection, indicator displacement assays with multicationic macrocyclic hosts and anionic fluorescent dye guests represent an intuitive design strategy. However, such systems typically exhibit turn-off responses. For biological samples with strong background- and autofluorescence, ratiometric or turn-on signaling is preferred, which requires more sophisticated assay architectures. In the present work, the host-guest complexation of fluorescent pH indicator dye trisodium 8-hydroxypyrene-1,3,6-trisulfonic acid (HPTS) with a cationic ammonium-pillar[6]arene (AP6) was employed in the desig

Spiegelmer Aptamers: Innovative Approach in Breast Cancer.

Breast cancer remains one of the leading causes of cancer-related mortality among women worldwide, emphasizing the urgent need for improved diagnostic and therapeutic strategies. Aptamers which are synthetic oligonucleotides that specifically bind to target molecules, offer high affinity, low immunogenicity, and superior tumor penetration compared to antibodies. However, their clinical translation has been limited by instability in biological systems. Spiegelmers, a novel class of mirror-image aptamers composed of L-nucleotides, overcome this limitation through intrinsic nuclease resistance an

Mechanical Signatures of Nucleic Acid Trefoil Knot Topology.

Molecular knots represent a fundamental form of polymer topology, yet their mechanical behavior in nucleic acids remains largely unexplored. Here, we engineer a single-stranded DNA sequence that can fold into either a knot or a pseudoknot while maintaining identical base-pairing interactions. Using single-molecule force spectroscopy with optical tweezers, we show that molecular topology alone produces distinct mechanical behavior. Knotted ssDNA exhibits three characteristic signatures relative to the pseudoknot: higher unfolding forces, shorter unfolding extensions, and faster refolding kineti

5,10-Methylenetetrahydrofolate Reductase─the Key Allosteric Regulator in One-Carbon Metabolism.

Collectively known as one-carbon metabolism (OCM), both the folate and methionine cycles are highly regulated to meet cellular demands. These cycles are key in the production and recycling of methyl groups to be used in many essential cellular processes such as the production of nucleotides, as well as S-adenosyl-l-methionine (SAM) the global methyl donor for DNA, RNA, and post translational modifications. Within the folate cycle, 5,10-methylenetetrahydrofolate is the main species through which methyl groups enter OCM. Therefore, 5,10-methylenetetrahydrofolate reductase (MTHFR), which reduces

Genomic analysis and pathogenic characterization of novel PRRSV-1 strain HBEU-328 in North China.

Porcine reproductive and respiratory syndrome virus 1 (PRRSV-1) is an emerging threat to the Chinese swine industry, with limited pathogenic data available on the BJEU06-1-like subgroup in North China. In this study, we isolated PRRSV-1 strain HBEU-328 from an unvaccinated pig farm in Hebei Province, 2022. Whole-genome sequencing showed that its genome is 15,068 nucleotides long and that it belongs to the BJEU06-1-like subgroup, clustering with Henan isolates, indicating its cross-regional transmission. HBEU-328 has characteristic deletions in Nsp2 (4 + 1 amino acids), GP3 (unique 4 + 1 amino

Novel Roussin's red salt esters as cardiovascular protectors: "Structure-activity" correlations, properties in solid state and solutions.

A novel binuclear neutral tetranitrosyl iron complexes (NICs) with phenylmethanethiolyl and phenylethanethiolyl ligands were synthesized, and their properties were compared with known phenylthiolyl NIC. It has been shown that in this series of compounds, the complex with phenylethanethiolyls demonstrates a prolonged release of nitric oxide (NO) in aqueous solutions, which is unusual for the class of Roussin's red salt esters. The combination of data on NO generation with the results of biological activity (based on changes in the level of cyclic nucleotides and their cytotoxicity on Vero cells

Marek's disease virus-encoded microRNA-M6-5p suppresses viral replication by targeting UL42.

Marek's disease virus (MDV) is a highly contagious and oncogenic avian alphaherpesvirus that causes severe economic losses on the global poultry industry. MicroRNAs (miRNAs), a class of endogenous noncoding RNAs typically 20-25 nucleotides in length, are crucial regulators of post-transcriptional genes expression. Our previous study found that MDV-encoded miR-M6-5p suppresses viral replication in cells. However, the underlying molecular mechanisms remain incompletely understood. In this study, we identified the viral UL42 transcript, bioinformatically predicted to encode a subunit of the viral

Pivotal role of long non-coding RNA in the development of new therapeutic approaches for plural mesothelioma.

Pleural mesothelioma is a rare aggressive tumor that affects the parietal layer of the pleura. It constitutes 90% of mesotheliomas. Usually, the diagnosis takes place 30 to 40 years after asbestos exposure. Thus, patients are diagnosed at a very late stage, leading to a poor prognosis, with a low median survival rate of approximately one year and a 5-year survival rate of only 5%. The available treatment usually consists of a multimodal approach, including chemotherapy (platinum-based drugs and antifolates), surgery, and radiotherapy. Yet, new therapeutic approaches are still needed to achieve

Molecular dynamics simulations to investigate Mycobacterium tuberculosis DNA-gyrase-fluoroquinolone complexes.

The widespread use of fluoroquinolone antibiotics to treat Mycobacterium tuberculosis (Mtb) infections has led to a rise in fluoroquinolone-resistant Mtb strains, mainly due to specific mutations in the target DNAgyrase. To overcome this resistance and develop treatment alternatives, a deeper understanding of fluoroquinolone's mechanism of action is necessary. In this study we performed molecular dynamics (MD) simulations on experimentally derived complexes of wild-type and Ala90Ser mutated Mtb DNA gyrase with three fluoroquinolones - moxifloxacin, gatifloxacin, and levofloxacin. The differenc

UV signature mutation hyperhotspots as genomic dosimeters.

Screening for UV-sensitive DNA mutation sites as dosimeters for individual sun exposure, using high-fidelity Duplex Sequencing to reduce false positives and a stringent genome alignment protocol to reduce false negatives, revealed: (i) Sun-exposed human skin contained UV signature mutations (C → T at a diypyrimidine) >1000-fold more frequently than unexposed neonatal skin, making these mutations sensitive genomic dosimeters for UV exposure. (ii) Tumor suppressor gene hotspot nucleotides (nt) reached variant allele frequency (VAF) 0.1%-10% via 30-3000 fold clonal expansion in occasional biopsie

BASELINE: a CRISPR base editing platform for mammalian-scale single-cell lineage tracing.

A cell's fate is shaped by its inherited state, or lineage, and the ever-shifting context of its environment. CRISPR-based recording technologies are a promising solution for mapping the lineage of a developing system; however, challenges remain regarding single-cell recovery, engineering complexity, and scale. Here, we introduce BASELINE, which uses base editing to generate high-resolution lineage trees in conjunction with single-cell profiling. BASELINE uses the Cas12a adenine base editor to irreversibly edit nucleotides across target arrays built from 50 synthetic target sites, which are in

Mechanisms behind the LncRNAs-mediated regulation of paclitaxel (PTX) resistance in human malignancies.

Paclitaxel (PTX) is extensively used to treat various cancers, including those of the breast, ovary, lung, esophagus, stomach, pancreas, and neck. However, despite its effectiveness in clinical settings, patients often experience cancer recurrence due to the emergence of resistance to PTX. The mechanisms underlying this resistance in cancer cells exposed to PTX involve modifications in β-tubulin, the primary target molecule associated with mitosis, the activation of pathways that facilitate drug efflux, and the dysregulation of apoptosis-related proteins. Long non-coding RNAs (lncRNAs), which

A Comprehensive Characterization of the Differences in Meat Quality, Nonvolatile and Volatile Flavor Substances Between Taoyuan Black and Duroc Pigs.

To compare the differences in meat quality between obese-type Chinese pig breeds and lean-type foreign pig breeds, we selected Taoyuan Black (TB) pigs and Duroc pigs at 180 and 210 days of age and analyzed their meat quality, chemical composition, and flavor compounds using an electronic tongue, chromatographic techniques, and two-dimensional gas chromatography-time-of-flight-mass-spectrometry (GC×GC-TOF-MS). A total of 16 main fatty acids, 18 main free amino acids, and 249 flavor compounds were identified. The results showed that TB pigs exhibited redder meat color, higher intramuscular fat,

In vivo mapping of mutagenesis sensitivity of human enhancers.

Distant-acting enhancers are central to human development1. However, our limited understanding of their functional sequence features prevents the interpretation of enhancer mutations in disease2. Here we determined the functional sensitivity to mutagenesis of human developmental enhancers in vivo. Focusing on seven enhancers that are active in the developing brain, heart, limb and face, we created over 1,700 transgenic mice for over 260 mutagenized enhancer alleles. Systematic mutation of 12-base-pair blocks collectively altered each sequence feature in each enhancer at least once. We show tha

Epigenetic background of lineage-specific gene expression landscapes of four Staphylococcus aureus hospital isolates.

Bacteria with similar genomes can exhibit different phenotypes due to alternative gene expression patterns. In this study, we analysed four antibiotic-resistant Staphylococcus aureus hospital isolates using transcriptomics, PacBio genome sequencing, and methylomics analyses. Transcriptomic data were obtained from cultures exposed to gentamicin, the iodine-alanine complex CC-196, and their combination. We observed strain-specific expression patterns of core and accessory genes that remained stable under antimicrobial stress - a phenomenon we term the Clonal Gene Expression Stability (CGES) that

Derivatization of pBACpAK entrapment vectors for enhanced mobile genetic element transposition detection in multidrug-resistant Escherichia coli.

Aim. Antimicrobial resistance poses a critical global health threat, driven by the dissemination of resistance genes via mobile genetic elements (MGEs). This study aims to enhance the detection of MGE insertions in multidrug-resistant Escherichia coli by derivatizing the pBACpAK entrapment vector. Methods and results. Three derivatives were constructed with additional nucleotides upstream of the cI repressor gene, based on conserved regions identified from GenBank sequences containing known IS26 and IS1 insertions. Using colony PCR, intracellular transposition screening was performed on 194 te
