Natriuretic peptide responses to exercise in HFpEF diagnosis: a pilot study.

BACKGROUND: Diagnosing heart failure with preserved ejection fraction (HFpEF) can be challenging because natriuretic peptide (NP) plasma levels and diastolic function parameters during rest echocardiography may be normal or marginally abnormal in patients displaying exercise-induced symptoms suggestive of HF. In these patients exercise echocardiography is recommended. We wanted to assess whether directly measured NP levels post-exercise had added diagnostic value beyond rest plasma N-terminal pro-brain NP (NT-proBNP) levels for diagnosing HFpEF. METHODS: Participants with left ventricular dias

Two hours of urine output are a reliable parameter for intravenous furosemide adequacy and the need for doubling dose in acute heart failure: a prospective study.

OBJECTIVES: This study compared urine output at 2 and 6 h post-furosemide and explored factors influencing diuretic efficacy in acute heart failure (AHF). METHODS: A prospective study (June-August 2023) included AHF patients (≥18 years) with NT-proBNP ≥300 pg/mL. Urinary spot sodium and urine output were measured at baseline, 2, and 6 h after 40 mg IV furosemide. RESULTS: Among 32 patients, 26 (81.25 %) had satisfactory urine output, and 30 (93.75 %) met the target urinary sodium concentration at 2 h. No significant difference in mean urine output was found between the 2 and 6 h intervals (2

ESA VIVALDI (1 & 2) campaigns: sex-specific bone responses to dry immersion-induced unloading.

Skeletal unloading during microgravity rapidly disrupts bone and metabolic homeostasis, yet sex-specific physiological responses remain poorly characterized. Dry immersion (DI), a ground-based analogue of microgravity, provides a model to investigate early determinants of unloading-induced bone deconditioning. We assessed sex differences in serum biochemical markers of bone and energy metabolism during two 5-day European Space Agency DI campaigns (VIVALDI 1 and 2) involving healthy males (n = 19) and females (n = 18). DI induced rapid alterations in bone remodeling in both sexes, characterized

Randomized Comparison of Cardiotoxicity With 60 Versus 90 mg Daunorubicin in AML Induction Therapy.

Anthracyclines are an essential component of induction therapy for acute myeloid leukemia (AML), but their optimal dosing and the associated risk for cardiotoxicity remain under debate. The DaunoDouble trial compared daunorubicin at 60 (Dauno60) versus 90 mg/m2 (Dauno90) in combination with cytarabine (100 mg/m2 for 7 days) in newly diagnosed AML patients aged 18-65 years. Cardiac function was assessed by left ventricular ejection fraction (LVEF) and cardiac biomarkers (high-sensitivity troponin T [hsTnT], NT-proBNP) before treatment and on Day 15 in 317 randomized patients. Median LVEF declin

High-intensity interval training versus mesenchymal stem cells in amyloid-β-induced Alzheimer's disease: comparative effects on cognitive function, hippocampal inflammation, oxidative balance, and BDNF.

Alzheimer's disease (AD) is a neurodegenerative disorder characterized by progressive cognitive decline. Although high-intensity interval training (HIIT) and mesenchymal stem cells (MSCs) have demonstrated neuroprotective effects, their comparative efficacy has not been compared. Male Wistar rats were assigned to five groups: Control, Vehicle, AD, AD + HIIT, and AD + MSCs. AD was induced by intracerebroventricular (ICV) injection of amyloid-beta (Aβ1-42). HIIT was performed for six weeks, and MSCs were injected via ICV. Learning and memory were evaluated using the Morris water maze (MWM), and

ALKBH5-mediated m6A demethylation of PRMT6 inhibits neuronal apoptosis and ferroptosis in Alzheimer's disease via reducing transcription of ACSL4.

BACKGROUND: Protein arginine methyltransferase 6 (PRMT6) is confirmed to regulate Alzheimer's disease (AD) process. Acyl-CoA synthetase long-chain family member 4 (ACSL4)-mediated ferroptosis is involved in regulating AD progression. However, whether PRMT6 regulates AD progression through ACSL4-mediated ferroptosis is unknown. METHODS: AD mice were injected with adeno-associated virus to explore the role of PRMT6 and AlkB homologue 5 (ALKBH5) in vivo. Neuronal cells were treated with amyloid-β protein fragment 1-42 (Aβ1-42) to mimic AD cell models. The expression levels of PRMT6, ALKBH5, ACSL

Can Biomarker-Based Monitoring Detect the Early Development of Diastolic Dysfunction During Chemotherapy?

Background and Objectives: Although modern oncologic therapies have substantially improved cancer survival and overall prognosis, they have also led to an increasing burden of cardiovascular toxicity. Preventing severe complications such as heart failure and death remains a major priority, while maintaining the need for effective and potentially curative cancer therapy. Careful patient monitoring and early detection of cardiovascular complications may allow the timely implementation of cardioprotective strategies and treatments that could delay or prevent myocardial toxicity. Gastrodin alleviates Aβ25-35-induced glycolytic dysfunction via activating PI3K/AKT/BACH1 signaling in Alzheimer's disease models.

Cerebral glycolytic alteration has been identified as an important contributor to the pathological progress of Alzheimer's disease (AD). Research has shown that gastrodin (GAS) possesses neuroprotection in various experimental models of AD, but its specific mechanism remains unclear. In this study, we determined whether GAS exerted neuroprotective effects on AD models through regulating PI3K/AKT/BACH1 signaling axis. Eight-week-old C57BL/6 J male mice were intracerebroventricularly injected with Aβ25-35, to establish an AD model, followed by the administration of GAS (30, 60 mg·kg-1·d-1, i.g.)

Activation of Epac2 improves Aβ-induced impairment of memory retrieval in an acute model of Alzheimer's disease.

Impaired memory retrieval is one of the cognitive markers in the early stage of Alzheimer's Disease (AD). Previous studies report that exchange protein directly activated by cAMP 2 (Epac2) plays a specific and time-limited role in promoting memory retrieval. In this study, we investigated the effect of a novel Epac2 activator, S220, on neuronal and synaptic activities, and memory impairment in an acute AD mouse model. S220 treatment increased the firing rate of action potential and intracellular calcium in primary neuronal cultures. Moreover, S220 treatment increased synaptic currents in CA1 n

Association of Brain and Ventricular Boundary Shift Integral Measurements with CSF Biomarkers: A Case-Control Study.

AIMS: This study seeks to examine the relationship between cerebrospinal fluid (CSF) biomarkers (Aβ1-42, Phospho-Tau181p, Total-Tau) and brain volumetric changes measured by Brain Shift Integral (BSI) in Alzheimer's disease (AD) spectrum. We explore the potential of BSI as a complementary, non-invasive tool for early diagnosis and progression monitoring of AD. BACKGROUND: AD is a neurodegenerative disorder marked by amyloid plaques and tau tangles, leading to cognitive decline. CSF biomarkers are key indicators of AD pathology, but their integration with imaging metrics like BSI could enhance

Brain Connectivity Gradients Alterations in Discordant Cerebrospinal Fluid Profile for Alzheimer's Disease Biomarkers.

Alzheimer's disease (AD) is a heterogeneous disorder characterized by brain accumulation of amyloid-beta (Aß, simplified as A for the AD biological model) and tau (T) proteins, with Aß emerging first. However, a significant proportion of individuals exhibit discordant biomarkers' profiles, such as elevated phosphorylated tau181 (p-tau181) with normal Aß42 from cerebrospinal fluid (CSF), posing diagnostic and mechanistic challenges. This study investigated whether functional and structural brain connectivity can distinguish individuals with discordant CSF profiles (A-T+) from those with concord
