Loss of hepatic circadian organization in fatty liver syndrome identifies CLOCK as a regulator of lipid homeostasis in laying hens.

Fatty liver syndrome (FLS) is a major metabolic disorder in laying hens, but how hepatic lipid dysregulation is linked to circadian disruption remains unclear. Here, we investigated the role of circadian regulation in FLS pathogenesis and focused on the core clock gene Clock Circadian Regulator (CLOCK). In a high-fat diet-induced FLS model in Lohmann pink layers, affected birds showed hepatomegaly, hepatic lipid accumulation, elevated serum total cholesterol, triglycerides, alanine aminotransferase, and aspartate aminotransferase, increased hepatic pro-inflammatory gene expression, and reduced

A high-carbohydrate diet-induced lipid deposition is alleviated by sodium pyruvate via suppression of the cPLA2-arachidonic acid-PPARγ axis.

BACKGROUND: Pyruvate is a critical node linking glycolysis and lipid metabolism. However, its regulatory role and mechanism in fish fed high-carbohydrate diets (HCD) remain obscure. OBJECTIVES: In this study, we determined the role of sodium pyruvate (SP) in metabolic disorders induced by HCD and elucidated its underlying mechanisms using in vivo and in vitro models. METHODS: All-male 4-mo-old AB strain zebrafish (Danio rerio) (0.356 ± 0.02 g) were fed a control diet, HCD, or HCD supplemented with 2% or 4% SP for four weeks (3 tanks/treatment, 30 fish/tank). Juvenile all-male 3-mo-old Nile t

The differential expression of lipocalin-2 in pig tissues and its molecular mechanism of regulating cell viability, differentiation, adipogenesis, and glycometabolism in porcine intramuscular preadipocytes.

Lipocalin-2 (LCN-2) is an adipocytokine secreted mainly by adipose tissue. Increasing evidences suggest that LCN-2 is an inflammatory factor associated with insulin resistance, obesity, and its complications. The precise mechanism of the development of obesity-related disorders induced by LCN-2 is not very clear. This study evaluated the expression of LCN-2 in pig tissues and its molecular mechanism of regulating preadipocyte differentiation in porcine intramuscular preadipocytes. LCN-2 expression in tissues of Tongcheng pigs, intramuscular adipose tissue of Tongcheng pigs (fat type) and Landr

Metabolic signatures and machine learning identify gut-liver-heart axis dysfunction as a potential link to major adverse cardiovascular events in coronary artery disease.

BACKGROUND: Traditional risk factors do not fully account for the residual cardiometabolic risk of major adverse cardiovascular events (MACE) in coronary artery disease (CAD). We aimed to identify circulating metabolic signatures associated with MACE susceptibility and uncover potential pathobiological mechanisms underlying the gut-liver-heart axis. METHODS: In this retrospective case-control study, untargeted high-performance liquid chromatography-mass spectrometry (HPLC-MS) was performed on fasting serum from 200 patients with CAD and MACE, 200 with CAD without MACE, and 400 matched non-CAD

Cell-Specific Roles of Angiopoietin-2 in CKD.

KEY POINTS: Global deletion of angiopoietin-2 protected against the progression of CKD. Endothelial cell-derived angiopoietin-2 drove early inflammation by promoting endothelial activation. Tubular epithelial cell-derived angiopoietin-2 contributed to functional decline and kidney fibrosis in advanced CKD. BACKGROUND: CKD is driven by inflammation, vascular dysfunction, and fibrosis, with emerging evidence implicating angiopoietin-2 as a key mediator. While angiopoietin-2 is produced by endothelial cells and upregulated in injured tubular epithelial cells (TECs), the distinct contributions of

Role of primary protectors of plant cells in salinity tolerance: molecular mechanisms and adaptive strategies.

Salinity stress is among the most pervasive abiotic factors limiting plant growth and agricultural productivity worldwide, currently affecting more than 20% of irrigated croplands. Elevated salt concentrations induce osmotic imbalance, ionic toxicity, and oxidative stress, leading to disruptions in photosynthesis, metabolism, and plant development. To counteract these deleterious effects, plants have evolved intricate, highly coordinated defense systems comprising structural and functional cellular protectors. At the structural level, the cell wall, plasma membrane, vacuole, and peroxisome pla

Mechanistic insights into EOC-PMCs-Induced macrophage polarization via the Galnt15-PPARγ pathway.

BACKGROUND: Ascites represents an extremely unique liquid immunosuppressive microenvironment of epithelial ovarian cancer (EOC), and it contains a substantial quantity of macrophages; however, these macrophages fail to exhibit effective anti-tumor activity. Peritoneal mesothelial cells (PMCs) constitute a critical component of ascites. Whether PMCs can modulate macrophages function through some mechanisms, thereby influencing the tumor immune microenvironment, remains to be elucidated. METHODS: Co-culture models of EOC-educated PMCs (EOC-PMCs) and macrophages in human and mouse were respectiv

Pemafibrate is associated with greater reductions in MDA-LDL and small dense LDL fractions with exploratory PBMC gene-expression changes: a prespecified subanalysis of the PARADISE in Kagoshima study.

BACKGROUND: Pemafibrate, a selective peroxisome proliferator-activated receptor alpha (PPARα) modulator, lowers triglycerides and improves lipid parameters. However, its effects on oxidized LDL-related markers, including malondialdehyde-modified low-density lipoprotein (MDA-LDL), and associated in vivo gene-expression changes in humans remain limited. In addition, whether pemafibrate-associated changes in oxidized LDL-related biomarkers have clinical implications for atherosclerotic progression or cardiovascular risk remains uncertain. METHODS: In this prespecified subanalysis of the randomiz

Effects of Intake Levels of High-Protein and High-Rumen Undegradable Protein Concentrate on Carcass Characteristics of 26-Month-Slaughtered Hanwoo Steers.

Shortened 26-month feeding program in Hanwoo steers often impair intramuscular fat (IMF) accumulation and carcass marbling. Therefore, this study examined whether increasing the intake level of a high-crude protein (CP) and high-rumen undegradable protein (RUP) concentrate could overcome these marbling limitations. Thirty-two Hanwoo steers (initial body weight (BW) 217 ± 14 kg; 8 months of age) were randomly assigned to low (LI; n = 16) or high (HI; n = 16) nutrient intake groups. Both groups were fed a high-CP and high-RUP concentrate diet at differing intake levels. During the growing stage
