Comprehensive Evaluation of Lipids, Lipoproteins, and Metabolites Associated with Gastrointestinal Cancer Risk.

BACKGROUND: Emerging evidence indicates that lipid metabolism plays a crucial role in gastrointestinal (GI) cancers. METHODS: This prospective cohort study included 112,655 cancer-free adults from the UK Biobank with metabolomics data at baseline. Multivariate Cox proportional hazards regression models were used to evaluate the associations between metabolites and GI cancers. LASSO regression was used to create metabolite risk scores, and their predictive performance for GI cancers was assessed using the C-statistic derived from the time-dependent ROC curves. RESULTS: A total of 93, 95, 33,

Biochemical composition of prostate core-needle biopsies before and after a single fraction of 13.5 Gy: a Raman spectroscopy-based study.

High dose rate brachytherapy (HDR-BT) is an attractive option for patients with favourable-risk intermediate-grade prostate cancer. However the relationship between radiation dose and the biochemical response of tumours remains poorly understood. The aim of this study was to investigate the tissue composition of prostate core-needle biopsies, before and after a single dose (13.5 Gy) of HDR-BT using Raman Spectroscopy (RS) and Group and Basis Restricted Non-Negative Matrix Factorisation (GBR-NMF) modelling. RS was used to measure the spectral profiles of benign and malignant regions of prostate

Ethanolamine plasmalogens increase luteinising hormone pulsatility and are associated with GPR61 expression in bovine gonadotrophin-releasing hormone neurons.

Pulsatile secretion of luteinising hormone (LH) reflects episodic activity of hypothalamic gonadotrophin-releasing hormone (GnRH) neurons and is essential for reproductive function. Plasmalogens are ether phospholipids enriched in neuronal membranes; however, their role in neuroendocrine regulation of reproduction remains unclear. We examined the effects of ethanolamine and choline plasmalogens on LH pulsatility in vivo and investigated GPR61 expression in bovine hypothalamic tissues and relevant cell lines. The oestrous cycles of 15 Holstein heifers were synchronised, and blood samples were c

Stool Microbiota, Metabolites, and Fecal Incontinence in Women.

IMPORTANCE: Stool metabolites influencing gut motility and sensation may contribute to fecal incontinence (FI). OBJECTIVE: The objective of this study was to test whether FI is associated with higher levels of butyrate and Clostridiales taxa in stool. STUDY DESIGN: Stool metabolites and microbiota were compared between 96 women with FI and 42 controls. FI frequency and subtype (urgency vs insensible) were measured using a 14-day bowel diary, St. Mark's score, and the Accidental Bowel Leakage questionnaire. Metabolite analysis included targeted analysis (8 short-chain fatty acids including bu

Liver specific-inhibition of LPCAT3 ameliorates metabolic dysfunction-associated steatotic liver disease.

The membrane unsaturation in the liver is associated with abnormal biosynthesis and accumulation of cholesterol and triglycerides, which is a hallmark of metabolic dysfunction-associated steatotic liver disease (MASLD). The lysophosphatidylcholine acyltransferase 3 (LPCAT3) incorporates arachidonic acid (AA) into the membrane phospholipids and increases the degree of unsaturation. The metabolites of AA are involved in the lipogenesis under high nutrient conditions. However, the mechanistic insights by which LPCAT3 contribute to lipogenesis and MASLD remain unknown. This study found that LPACT3

Regulation of lipid oxidation and ferroptosis by a tunable axis of vitamin E, high-density lipoprotein, and scavenger receptor class B type 1.

Ferroptosis is caused by lethal peroxidative damage to cell membrane phospholipids (PL) containing polyunsaturated fatty acid (PUFA) tail groups. Mechanisms to control cell membrane PL oxidation and ferroptosis include cellular expression of glutathione peroxidase 4 (GPx4), which repairs oxidized cell membrane PL, and the lipophilic antioxidant alpha-tocopherol (α-toc), an active form of Vitamin E. Data show that α-toc can rescue cells from ferroptosis in vitro and in vivo even in the absence of GPx4. Yet, pathways by which α-toc is transported in blood, for instance in high- and low-density l

Microbial physiological trait shifts link heavy metal remediation to enhanced soil carbon storage potential.

Widespread and chronic heavy metal pollution resulting from industrial activities has compromised the sustainability of soil ecosystems. Increasing and stabilizing soil carbon storage is central to soil development, but how remediation reshapes soil carbon cycling processes during the mitigation of heavy-metal contamination remains unclear. Here, we tracked genome-scale microbial metabolism, community turnover, and phenotype-level physiological responses during a 120-day remediation of heavy metal-contaminated soils with several decades of pollution histories and resolved their dynamic interpl

OBJECTIVE: Methotrexate (MTX) is a clinically approved, potent disease-modifying antirheumatic drug used to manage rheumatoid arthritis. However, its systemic administration is associated with severe toxicity and bioavailability challenges. Localized MTX therapy holds significant potential by enabling site-specific action while diminishing the risk of systemic toxicity. METHODS: In this study, we investigated the therapeutic potential of systematically designed methotrexate-loaded high permeation vesicles (MTX-HPVs) in rheumatoid arthritis. HPVs comprise biocompatible phospholipids and a syne

Loss of Mtarc1 Protects Against Steatotic Liver Disease in Mice.

BACKGROUND & AIMS: Metabolic dysfunction-associated steatotic liver disease (MASLD) spans from simple steatosis to metabolic dysfunction-associated steatohepatitis (MASH) and can progress to cirrhosis or hepatocellular carcinoma. Despite its prevalence, effective therapies are lacking. Recent genome-wide association studies identified a common missense variant (rs2642438) in the Mitochondrial Amidoxime Reducing Component 1 (MTARC1) gene that protects against liver cirrhosis without increasing cardiovascular disease risk. Biochemical and disease risk signatures associated with carriers of this

Lipoprotein X as a source of analytical and interpretative interference in routine laboratory testing.

Lipoprotein X (LpX) is an abnormal lipoprotein-like particle occurring mainly in cholestasis, lecithin-cholesterol acyltransferase deficiency, post-transplant cholestatic liver dysfunction, hepatic graft-versus-host disease, and selected intravenous lipid emulsion or parenteral nutrition settings. Enrichment in phospholipids and unesterified cholesterol, paucity of cholesteryl esters and triglycerides, and usual absence of apolipoprotein B (apoB) challenge assumptions underlying routine laboratory testing. This review distinguishes analytical interference, in which LpX biases measurement proce

Effects of α-Synuclein on the Lipid Phenotype of SZ95 Human Sebocytes: A Preliminary Study in the Context of Parkinson's Disease.

Pathological aggregation of α-synuclein (αSyn) is a hallmark of Parkinson's disease (PD), but increasing evidence indicates that αSyn deposits extend beyond the central nervous system to peripheral tissues, including the skin. In PD patients, cutaneous αSyn accumulation has been associated with seborrheic dermatitis (SD), a chronic inflammatory condition linked to sebocyte dysfunction and altered sebum production. Marked differences in sebum composition between PD patients and healthy controls have been described. We aimed to investigate whether pathological αSyn could contribute to dysregulat

Urotensin II Receptor Deletion Mitigates Kidney Injury and Renal Lipid Remodeling in Mice Fed a Long-Term High-Fat Diet.

Urotensin II (UII) signaling through its Gq-coupled receptor (UT) has been implicated in metabolic and vascular dysfunction; however, its contribution to diabetic kidney disease (DKD) and renal lipid remodeling remains incompletely defined. We examined kidney injury and lipidomic alterations in wild-type (WT) and UT knockout (UT KO) mice fed a long-term high-fat diet (HFD). Male WT and UT KO mice were fed HFD (60% kcal fat) or low-fat diet (LFD; 10% kcal fat) for 33 weeks. Despite comparable HFD-induced obesity, hyperglycemia, and hyperinsulinemia, UT deletion significantly attenuated albuminu

Bioactive compounds in milk/dairy foods and their value to health at key life stages: Functionality beyond nutrient supply.

This review aims to highlight some of the key bioactive functionalities that a range of components in milk can potentially make to reduce risks to health at key life stages. Such functionalities cannot be adequately explained by traditional estimates of nutrient supply. The benefits of gangliosides from the milk fat globule membrane (MFGM) for neonatal neurodevelopment are examined along with milk proteins for growth stimulation of children via the proteins' ability to stimulate insulin-like growth factor-1 (IGF-1). Exosomes in bovine milk and their miRNA are bioavailable, with an animal study

Conformational features of guinea pig apolipoprotein E offer insights into functioning of human apolipoprotein E.

Apolipoprotein (apo) E is a major cholesterol transport protein in the plasma and brain of humans, with the APOE ε4 allele (coding for R112) associated with a higher risk for cardiovascular and Alzheimer's diseases (CVD and AD, respectively) compared to APOE ε3 (coding for C112). The molecular basis underlying the link between APOE ε4 and CVD/AD is poorly understood. Here apoE from Cavia porcellus (guinea pig, GP), which is 72 % identical to human apoE4 but lacking residues 193-197 and 246-252, a feature noted in all hystricomorph apoE, was used as a model to understand the role of apoE4. n - 3 polyunsaturated fatty acids mediate hyodeoxycholic acid-FXR signaling to ameliorate metabolic dysfunction-associated fatty liver disease.

Accumulating evidence has shown that n - 3 polyunsaturated fatty acids (PUFA) intervention contributes to ameliorating metabolic dysfunction-associated fatty liver disease (MAFLD); however, the underlying mechanism through which n - 3 PUFA alleviate MAFLD remains elusive. Critical gut microbiota-modified metabolites and host targets have been implicated in the initiation and development of MAFLD. Here, a case-control study was performed which indicated that gut microbiota-modified bile acids, notably serum hydeoxycholic acid (HDCA) species, were significantly lower in MAFLD subjects compared w

Functional characterization of TMEM86A and TMEM86B mutants by a novel lysoplasmalogenase assay.

Plasmalogens are an abundant class of glycero-phospholipids with a characteristic 1-O-alk-1'-enyl double bond. While their synthesis has been extensively investigated, their degradation remains understudied. Plasmalogen deficiencies are associated with severe disorders in humans and interfering with their degradation would be a treatment option, but it remains out of reach due to limited knowledge. The plasmalogen double bond is degraded either directly by a plasmalogenase or by conversion to the 2' lyso forms by phospholipase and subsequent cleavage by lysoplasmalogenase (E.C. 3.3.2.2). Two l

Alteration in hippocampal mitochondria ultrastructure and cholesterol accumulation linked to mitochondrial dysfunction in the valproic acid rat model of autism spectrum disorders.

RATIONALE: Autism spectrum disorders (ASD) are a group of neurodevelopmental and multifactorial conditions with cognitive manifestations. The valproic acid (VPA) rat model is a well-validated model that successfully reproduces the behavioral and neuroanatomical alterations of ASD. Previous studies found atypical brain connectivity and metabolic patterns in VPA animals: local glucose hypermetabolism in the prefrontal cortex, with no metabolic changes in the hippocampus. AIM: This study aimed to explore mitochondrial structural features, lipid content, and functionality in the hippocampus and c

Cinnamaldehyde and its combination with deferoxamine ameliorate inflammation, ferroptosis and hematoma expansion after intracerebral hemorrhage in mice.

Intracerebral hemorrhage (ICH) is a most serious type of hemorrhagic stroke with a continuously rising incidence globally, without effective cure available. The underlying mechanisms driving brain injury are complex and include inflammation, oxidative stress, glutamate excitotoxicity, membrane damage, lipid peroxidation, ferroptosis and other cellular death modes. Hematoma clearance is the key to limit brain damage and foster the recovery process. The quest for effective ICH remedies is continuing and strategically evolving with the expansion of knowledge and understanding of target mechanisms
