Pentobarbital-activated Cl(-) channels in cultured adult and embryonic human DRG neurons.

Developmental differences in pentobarbital-activated Cl(-) currents were studied in adult and embryonic human dorsal root ganglia (DRG) neurons using whole-cell patch-clamp recordings. Pentobarbital-induced Cl(-) conductance was significantly greater in adult DRGs (28.4 pS) than in embryonic cells (19.1 pS). Fluctuation analysis of the spectral density plots of Cl(-) channel activation by pentobarbital showed age differences in the length and number of open time constants (adult cells, tau(1), tau(2), tau(3) were 224, 8. 4, 1.5 ms, respectively; embryonic cells tau(1) and tau (2) were 165 and

Genetic epidemiological approaches to the search for osteoporosis genes.

Important progress has been made in the identification of specific environmental factors and estimation of hereditary components in bone density, quantitative ultrasound (QUS), and bone turnover indices. By contrast, the search for specific genes that regulate bone mass has progressed rather slowly, and the results are more difficult to interpret and reproduce. This article reviews the genetics of osteoporosis and problems plaguing genetic research. It is argued that the search for genes involved in the expression of osteoporotic phenotypes should be based on linkage studies in relatively homo

Impact of Spherix (Bacillus sphaericus B-101, serotype H5a, 5b) spraying on the control of mosquito breeding in rural areas of Farrukhabad District, Uttar Pradesh.

Spray impact of Spherix (Bacillus sphaericus B-101, Serotype H5a, 5b) was assessed against larval and adult populations of mosquito species in rural areas of Farrukhabad district, Uttar Pradesh. High mortality (> 95%) was observed against anopheline and culicine larvae within 48 h of spray @ 1 g/sq m. But the biolarvicidal effect declined within a week which indicated nonrecycling capacity and weekly reapplications were required. The encouraging results obtained through weekly sprays were short-lived because of decline in efficacy in some perennial intradomestic Culex breedings after 15 rounds

NGF prevention of neurotoxicity induced by cisplatin, vincristine and taxol depends on toxicity of each drug and NGF treatment schedule: in vitro study of adult rat sympathetic ganglion explants.

The simultaneous administration of nerve growth factor (NGF) has been found to prevent experimental neuropathies induced by anti-cancer drugs such as cisplatin, vincristine and taxol. However, it is clinically important to know whether NGF is beneficial once the neuropathy is already manifest. We established a bioassay system to examine the preventive effects of NGF in various treatment schedules. NGF significantly prevented the inhibition of neurite outgrowth by vincristine and taxol regardless of treatment schedules. The pre-treatment and co-treatment schedules were effective against cisplat

Helper activity for antibody synthesis encoded by mRNA extracted from human peripheral blood mononuclear cells.

Translation products in Xenopus laevis of mRNA from human peripheral blood mononuclear cells were tested for their capacity to replace T cells in the anti-SRBC response of nude spleen cells. When the starting material came from PHA or FCS-stimulated lymphocytes, the translated lymphokines, displaying such a B cell helper activity were found to be encoded by mRNA sedimenting at 6-7S and 13S on a sucrose density gradient. 6-7S mRNA from control, non-stimulated lymphocytes was also able to code for B cell helper activity. Thorough T-cell depletion of mouse responder cell populations left unchange

Murine typhus in Kuwait in 1978.

Murine typhus occurred in 254 individuals in Kuwait between April and August 1978; 81% of patients were aged between 15 and 44 years, and 63% were male. The highest attack rate occurred among people in the lowest socioeconomic class. The outbreak coincided with a period of high rat and flea density. A study of the first 104 cases suggested that infection was acquired in the home, but a case - control study revealed no significant differences between patients and control subjects in terms of exposure to rats or domestic animals, and other factors. This suggests that murine typhus is hyperendemi

G-protein-coupled receptor sensing of farnesol controls fungal dormancy and is susceptible to bacterial interference.

Conidial dormancy is a critical survival strategy for fungi under stress, yet the mechanisms governing the switch between dormancy and germination remain unclear. Here we identify the G-protein-coupled receptor, Rsd1, as a farnesol-responsive sensor controlling this dormancy switch in the plant pathogen Fusarium pseudograminearum, which causes Fusarium crown rot. Under high population density and nutrient limitation, endogenous farnesol activates Rsd1 and downstream signalling pathways to suppress conidial germination. Rsd1 also responds to extracellular farnesol, linking environmental signals

Targeting of PCSK9 in the Management of Alzheimer's Disease: Expanding the New Potential Key Player.

Alzheimer's disease (AD) represents the most common cause of dementia in the elderly population worldwide. However, most of the anti-AD medications did not resolve the underlying neuropathology. Consequently, targeting other signaling pathways may be helpful in the management of AD. Particularly, preprotein convertase subtilisin/kexin type 9 (PCSK9), which is a regulator protein of low-density lipoprotein (LDL), is intricate in the pathogenesis of AD. Normally expressed PCSK9 in the brain plays a critical role in the regulation of neuronal differentiation and apoptosis, and degradation of LDL

Understanding the Personal Networks of People Experiencing Homelessness in King County, WA with Aggregate Relational Data.

Personal networks among people experiencing homelessness are vital but often overlooked. Drawing on a unique three-year dataset (2022-2024) from over 3,000 unhoused individuals in King County, Washington, this study analyzes the structure and change of four different personal networks: acquaintances, close friends, kinship (household), and peer referrals. Using this respondent-driven sample data set, we find that the mean size of the acquaintance networks ranged from 22 to 27 people, while the mean number of close friendships declined from 4.9 to 4.19. This drop, despite population growth, sug

Dopant distributions and band-edge positions in Sr-doped NaTaO3: a first-principles study.

Sr doping is known to enhance the water-splitting activity of NaTaO3 photocatalysts and is often accompanied by surface segregation of Sr, yet, the electronic-structural origin remains unclear. Here, we employ first-principles calculations with explicit enumeration of all Sr configurations in bulk NaTaO3 (NTO) and TaO2-terminated (001) surfaces. In the bulk, the enumeration shows that shorter Sr-Sr separations are energetically preferred. At surfaces, Sr segregates and accumulates strongly. Layer-resolved local density of states indicates that in-gap states are confined to the outermost TaO2 l

Pervasive sublethal impacts of heat exposure revealed by telomere dynamics.

Predicting species responses to anthropogenic environmental change remains one of the most pressing and complex challenges in contemporary biology. While mass mortality events and localized extinctions have been linked to global warming, the more elusive and probably common, effects of sublethal heat are less well understood. In a cooperatively breeding bird from the Australian tropical savannah, we identified that hotter conditions accelerated telomere attrition through two critical life-history stages: juveniles transitioning to nutritional independence and adults transitioning into a reprod

Machine learning-based high-resolution estimation and explainable analysis of road transport CO2 emissions across South Korea.

To support targeted mitigation of road-transport carbon dioxide (CO2) emissions, this study constructs a machine-learning-based high-resolution CO2 emissions inventory for South Korea at the hourly road-link level. The results show that roads with similar monthly total emissions can exhibit substantially different hourly emission patterns, which were systematically classified into three major types-Daytime-Steady, Commuting-Peak, and Mixed/Weekend-High-according to road class and regional characteristics. To examine regional predictive associations, shapley additive explanations (SHAP) analysi

Hypoxia-activated scleraxis a mediates epicardial progenitor differentiation into a unique cardiac perivascular cell type.

The epicardium provides progenitor cells and paracrine signals essential for heart development and regeneration, yet the mechanisms regulating epicardial cell fate remain poorly understood. Here, we identify the transcription factor Scleraxis a (scxa) as a key regulator of epicardial progenitor differentiation in zebrafish. Single-cell transcriptomics, genetic lineage tracing, and cardiac injury models reveal transient scxa expression in activated epicardial progenitor cells (aEPCs) during developmental coronary angiogenesis and heart regeneration. scxa+ epicardial cells predominantly differen

Network based statistics associates increased connectivity to REM sleep disorder and hallucinations in early DLB.

Dementia with Lewy Bodies (DLB) is a heterogeneous disease characterized by four core clinical features: visual hallucinations, REM sleep behaviour disorder (RBD), cognitive fluctuations and Parkinsonism. We study the relationship between these features and functional connectivity from high-density electroencephalographic data of 33 subjects with early-stage DLB and 21 healthy controls. We used two connectivity metrics averaged on the individual theta and alpha bands, defined by determining the individual theta-to-alpha transition frequency through the package transfreq. After showing that DLB

Green space exposure predicts manic episodes in bipolar disorder: one year longitudinal analysis using smartphone-derived GPS data.

Urban living is linked to increased psychiatric morbidity, while green spaces may protect mental health via stress-buffering effects. Yet, the role of green space exposure in the onset of affective episodes in bipolar disorder (BD) remains unclear. Using 12 months of GPS data from 29 participants with BD-I/II, we quantified daily exposure to green spaces (green area & NDVI; covariates: population density & imperviousness). Affective status (euthymia, depressive or (hypo)manic episode) was assessed via SCID-based biweekly clinician interviews (26 assessments per participant). Multilevel logisti

Interkingdom sensing of fungal tyrosol promotes bacterial antifungal T6SS activity in the murine gut.

Type VI secretion systems (T6SSs) are molecular machines used by bacteria to release effectors that target either host cells, competing bacteria or fungi. Regulatory mechanisms underlying antifungal T6SS activity remain unexplored. Here we show, using mouse infection with wild-type and T6SS mutant bacteria, that T6SS activity of the enteropathogen, Yersinia pseudotuberculosis (Yptb), reduces fungal prevalence in the gut microbiota and has direct activity on Candida albicans. Screening of bacterial effector mutant strains, and structural and biochemical analyses identify TfeC as an antifungal c

Beyond homogeneity: charting the landscape of heterogeneity in neurodevelopmental and psychiatric electroencephalography.

Electroencephalography (EEG) has been thoroughly studied for decades in neurodevelopmental and psychiatric research. Yet its integration into clinical practice as a diagnostic/prognostic tool remains unachieved. We hypothesize that a key reason is the underlying patient's heterogeneity, overlooked in EEG research relying on a case-control approach. We combine high-density EEG with normative modeling to quantify this heterogeneity using two well-established and extensively investigated EEG characteristics -spectral power and functional connectivity- across a cohort of 1674 patients with attenti

Mechanistic Insights into the Role of Extracellular Vesicles at the Bacterial-Host Interface.

Bacterial and human cells produce extracellular vesicles (EVs) in response to diverse stimuli, e.g., toxins, oxidative stress, nutrient depletion, or high cell density. Here, we describe a cocultivation platform that allows recovery of mixed extracellular vesicles (mix-EVs) produced simultaneously by both cell types. We investigated interactions between Gram-positive and Gram-negative bacteria (Streptococcus pyogenes, Staphylococcus aureus, Escherichia coli, and Neisseria meningitidis) and human peripheral blood mononuclear cells (PBMCs). The production of the mix-EVs population decreased with

Mitigation of sulfide adsorption in natural gas by silanized stainless steel: insights from density functional theory.

This study employs Density Functional Theory (DFT) to investigate the inhibition of sulfide adsorption in natural gas through the use of silanized stainless steel. Various adsorption models were developed to explore the interactions between hydrogen sulfide, thionyl carbon, methyl mercaptan, and ethyl mercaptan with silanized stainless steel surfaces. Calculations revealed positive adsorption energies of 1.55 eV, 1.87 eV, 1.56 eV, and 1.60 eV, respectively, indicating a non-spontaneous adsorption process. Furthermore, the adsorption configurations of these sulfur compounds on the surface close

Non-coding RNAs in osteoarthritis and osteoporosis-related hip fractures: molecular biomarkers for precision diagnosis and prognosis.

As individuals age, bone density declines and the likelihood of fractures increases, particularly in women experiencing menopause. Hip fractures, the most severe consequence of osteoporosis, are becoming more common due to the aging global population and a 1-3% annual increase in hip fractures in most regions. The specific epigenetic mechanisms underlying the onset of osteoporosis and its related fractures remain predominantly unexamined. Research indicates that epigenetic modifications can elevate the risk of osteoarthritis, osteoporosis and bone fractures, especially hip fractures, by linkin
