Multimodal explainable artificial intelligence identifies patients with non-ischaemic cardiomyopathy at risk of lethal ventricular arrhythmias.

The efficacy of an implantable cardioverter-defibrillator (ICD) in patients with a non-ischaemic cardiomyopathy for primary prevention of sudden cardiac death is increasingly debated. We developed a multimodal deep learning model for arrhythmic risk prediction that integrated late gadolinium enhanced (LGE) cardiac magnetic resonance imaging (MRI), electrocardiography (ECG) and clinical data. Short-axis LGE-MRI scans and 12-lead ECGs were retrospectively collected from a cohort of 289 patients prior to ICD implantation, across two tertiary hospitals. A residual variational autoencoder was devel

Accelerating diagnosis of Parkinson's disease through risk prediction.

BACKGROUND: Characterization of prediagnostic Parkinson's Disease (PD) and early prediction of subsequent development are critical for preventive interventions, risk stratification and understanding of disease pathology. This study aims to characterize the role of the prediagnostic period in PD and, using selected features from this period as novel interception points, construct a prediction model to accelerate the diagnosis in a real-world setting. METHODS: We constructed two sets of machine learning models: a retrospective approach highlighting exposures up to 5 years prior to PD diagnosis,

Radiomic Features From Multi-Parameter MRI Combined With Clinical Parameters Predict Molecular Subgroups in Patients With Medulloblastoma.

The 2016 WHO classification of central nervous system tumors has included four molecular subgroups under medulloblastoma (MB) as sonic hedgehog (SHH), wingless (WNT), Grade 3, and Group 4. We aimed to develop machine learning models for predicting MB molecular subgroups based on multi-parameter magnetic resonance imaging (MRI) radiomics, tumor locations, and clinical factors. A total of 122 MB patients were enrolled retrospectively. After selecting robust, non-redundant, and relevant features from 5,529 extracted radiomics features, a random forest model was constructed based on a training coh

Achalasia phenotypes and prediction of peroral endoscopic myotomy outcomes using machine learning.

OBJECTIVES: High-resolution manometry (HRM) and esophagography are used for achalasia diagnosis; however, achalasia phenotypes combining esophageal motility and morphology are unknown. Moreover, predicting treatment outcomes of peroral endoscopic myotomy (POEM) in treatment-naïve patients remains an unmet need. METHODS: In this multicenter cohort study, we included 1824 treatment-naïve patients diagnosed with achalasia. In total, 1778 patients underwent POEM. Clustering by machine learning was conducted to identify achalasia phenotypes using patients' demographic data, including age, sex, dis

Applying Machine Learning for Prescriptive Support: A Use Case with Unfractionated Heparin in Intensive Care Units.

Continuous unfractionated heparin is widely used in intensive care, yet its complex pharmacokinetic properties complicate the determination of appropriate doses. To address this challenge, we developed machine learning models to predict over- and under-dosing, based on anti-Xa results, using a monocentric retrospective dataset. The random forest model achieved a mean AUROC of 0.80 [0.77-0.83], while the XGB model reached a mean AUROC of 0.80 [0.76-0.83]. Feature importance was employed to enhance the interpretability of the model, a critical factor for clinician acceptance. After prospective v

Constrained IoT-Based Machine Learning for Accurate Glycemia Forecasting in Type 1 Diabetes Patients.

Individuals with diabetes mellitus type 1 (DM1) tend to check their blood sugar levels multiple times daily and utilize this information to predict their future glycemic levels. Based on these predictions, patients decide on the best approach to regulate their glucose levels with considerations such as insulin dosage and other related factors. Nevertheless, modern developments in Internet of Things (IoT) technology and innovative biomedical sensors have enabled the constant gathering of glucose level data using continuous glucose monitoring (CGM) in addition to other biomedical signals. A multi-task learning-based deep learning model for precise estimation of rib fracture age on chest CT.

Rib fractures are a common type of chest injury, and the estimation of the fracture age mainly relies on clinical or forensic imaging experts making rough judgments based on CT scans, which is highly subjective. In recent years, artificial intelligence (AI) models have performed exceptionally well in rib fracture detection tasks, providing a potential technical foundation for inferring the time of fracture formation. Therefore, it is necessary to develop deep learning model tools to assist human judgment. In this study, a multi-task deep learning model (based on the 3D-ResNet18) was developed

DARMN: Domain-Aware Residual Feature Modulation Network for Multidomain Protein Dynamic Inter-Residue Contact Prediction.

The regulation of conformational stability in multidomain proteins (MDP) is a central challenge in protein engineering with major implications for antigen optimization, enzyme activity modulation, and signal transduction. The function of these proteins depends on the properties of their constituent domains as well as on interdomain orientation and interface rearrangement during conformational transitions. Previous studies show that conformational transition is largely governed by changes in a small number of key residue pairs. However, such dynamic inter-residue contacts are typically sparse,

Predicting one-year left ventricular mass index regression following transcatheter aortic valve replacement in patients with severe aortic stenosis: A new era is coming.

Aortic stenosis (AS) is the most common valvular heart disease in the western world, particularly worrisome with an ever-aging population wherein postoperative outcome for aortic valve replacement is strongly related to the timing of surgery in the natural course of disease. Yet, guidelines for therapy planning overlook insightful, quantified measures from medical imaging to educate clinical decisions. Herein, we leverage statistical shape analysis (SSA) techniques combined with customized machine learning methods to extract latent information from segmented left ventricle (LV) shapes. This en
