Steroid hormone receptors: interaction with deoxyribonucleic acid and transcription factors.

Gene regulation by steroid hormones is mediated by binding of the hormone ligand to the corresponding receptor that triggers a complex set of interactions of the hormone receptors with each other, with DNA in chromatin, and with a variety of other proteins. In this review we attempt to summarize what is known about these interactions using as the main example the regulation of mouse mammary tumor virus transcription by glucocorticoids and progestins. We describe in some detail the interaction of monomers and homodimers of the steroid receptors with their recognition sequences, and the molecula

Experimental superficial siderosis of the central nervous system: biochemical correlates.

The pathogenesis of superficial siderosis of the central nervous system (CNS) may be examined by the repeated intracisternal injection of washed autologous red blood cells (RBC). In rabbits, the injections cause the accumulation of iron in the cytoplasm of microglial cells and astrocytes of cerebellar and cerebral cortices. Immunocytochemistry for ferritin reveals enhanced reaction product mainly in microglia but hemosiderin occurs only after extending the injections to 6 months. In an effort to determine the biochemical correlates of these morphological changes, iron, ferritin, ferritin subun

CD3- T cells with cis- or trans-acting mutations affecting expression of T cell receptor beta-chain mRNA.

Mutants of an untransformed T cell clone that no longer respond to TCR/CD3 stimulation have been derived using a selection procedure based on the loss of functional response to Ag. This functional selection gives rise to clones of several different phenotypes. We have previously described mutants with a TCR/CD3+ cell surface phenotype whose TCR are uncoupled from cellular responses. We describe six additional mutants that do not express TCR/CD3 at the cell surface. One of the CD3- clones contains a deletion in the successfully rearranged TCR-alpha gene, whereas another carries a deletion in th

Evaluation of a silicon-substrate modiolar eighth nerve implant in a guinea pig.

The availability of thin-film multichannel electrodes provides new possibilities for implantation and direct stimulation of the modiolar portion of the auditory nerve. Electrodes in direct contact with the auditory nerve should be functional at lower thresholds and might require fewer remaining neurons for stimulation compared to electrodes in the scala tympani. This strategy would also provide close contact with neural elements subserving a greater frequency range. Implantation of the eighth nerve may also be advantageous in profoundly deaf subjects who lack an implantable scala tympani. In t

Psoriasis. Leukocytes and cytokines.

Elements of the immune system must take their place alongside other potential mechanisms of psoriasis, such as psoriatic epidermal keratinocytic hyperproliferation, endothelial cell and fibroblast activation and proliferation, abnormal lipid regulation, and transmembrane signalling abnormalities. These data provide support for the concept that cellular immunologic processes are active in a manner that further promotes the patho-physiologic events observed in psoriasis. Thus, therapies useful for psoriasis may have activity on immunologic processes in addition to more traditional mechanistic co

Post-translational regulation of Lcr plasmid-mediated peptides in pesticinogenic Yersinia pestis.

The low calcium response of wild type Yersinia pestis, the causative agent of bubonic plague, and of enteropathogenic Yersinia pseudotuberculosis and Yersinia enterocolitica is known to be mediated by a shared Lcr plasmid of about 70 kb. At 37 degrees C in Ca2+-deficient medium, this element promotes restriction of growth with concomitant production of virulence functions including the common V antigen and a set of yersiniae outer membrane peptides termed YOPs (Lcr+). The latter are expressed by the enteropathogenic species but not by wild type Y. pestis which possesses a unique 10 kb Pst plas

Induction of B2 RNA polymerase III transcription by heat shock: enrichment for heat shock induced sequences in rodent cells by hybridization subtraction.

When hybridization subtraction was used to enrich for sequences induced by heat shock in Chinese hamster cells, B2 sequences were found to be one of the major sequences enriched. With cloned B2 probes, we found that the level of the short, 0.1 to 0.6 kb, polyadenylated RNA polymerase III transcripts of this repetitive genetic element increased approximately 10 to 20 fold after heat shock. Transcription of B2 RNA by RNA polymerase III was rapidly induced after heat shock based on time course studies and nuclear runoff experiments. The induction of B2 RNA was not a nonspecific response to lethal

Comparative pathology of the enlarged carotid body.

A histological study was made of the carotid bodies of man and various animal species from low and high altitudes. The animals studied were the alpaca, llama, cattle, guinea-pig, rabbit, dog, rat and man. As well as a qualitative microscopic study, a differential cell count was carried out to determine the percentage of the light and dark variants of chief cells and of sustentacular cells present. The investigation showed that the carotid bodies enlarge in cattle, guinea-pigs and rabbits living in the hypobaric hypoxia of high altitude to which they have to acclimatize. The carotid bodies are

Control of heme and cytochrome P-450 metabolism by inorganic metals, organometals and synthetic metalloporphyrins.

The heme-cytochrome P-450 complexes represent sensitive metabolic systems for examining the biological impact of metals on important cellular functions. Many metals, both in the inorganic form and bound to organic moieties, potently induce heme oxygenase, the rate limiting enzyme of heme degradation. The resulting increase in the rate of heme breakdown is reflected in a marked depression of cellular cytochrome P-450 content and impairment of the oxidative metabolism of natural and foreign chemicals dependent on this hemeprotein. Organometal complexes do not mimic in all their aspects the actio

Transposable element splicing reveals a tumor-enriched class of gene-independent transcripts.

Transposable elements (TEs) are broadly reactivated in tumors, but their repetitive nature has limited transcript-level characterization. Here, we systematically investigate the TE-derived splicing landscape across pan-cancer samples and define a splice-resolved subset of TE-derived transcripts, termed TEtrans, with tumor enrichment and gene independence. TEtrans initiate within TEs, spliced within or between TE loci, and are composed predominantly of TE sequences. Their expression was associated with stemness and inflammation regulators in a context-dependent manner. Functional analyses ident

From "Passive Response" to "Controllable Design": The Evolution of Crystal Structure and Morphology of Hollow FePt Nanochains by Annealing Treatment.

Atomic migration behavior in reactive atmospheres, especially the competitive mechanism between short- and long-range transport, is vital to determine the stability and performance of alloy nanomaterials. In this work, taking hollow Fe3Pt7 nanochains as a model system, in situ scanning electron microscopy and molecular dynamics simulations were combined to reveal the effects of annealing temperature and reaction atmosphere on the structure evolution. Vacuum annealing from room temperature to 900 °C results in size compression, pore formation, collapse of the hollow structure, and phase transit

Optical Responses From Zero Optical-Path-Length Object.

Optical path length (OPL) is a key concept that determines elementary and advanced optical effects from classical optical components to cutting-edge nanophotonic devices. In this paper, we treat objects with their physical and OPL thickness simultaneously vanishing and show that such objects in principle can produce remarkably strong optical effects under certain conditions associated with dipole-preserving compression of an ordinary three-dimensional medium. Such two-dimensional (2D) sheets essentially produce elementary optical responses of reflection, diffraction, and localization in the sa

Colorimetric sensor array based on trimetallic MOF-derived Zn-Co-Cu nanozymes for discrimination of seven antioxidants.

Maintaining redox homeostasis relies heavily on antioxidants, and deviations in their normal levels are intimately linked to the onset of multiple diseases. Herein, a series of Zn-Co-Cu trimetallic MOF-derived nitrogen-doped carbon nanozymes (ZnCoCu-NCx) were synthesized via a core-shell MOF pyrolysis strategy. By tuning the Co/Cu ratio, a series of compositionally tunable nanozymes with distinct oxidase-like activities were obtained. Owing to the synergistic interaction among the three metallic constituents and the construction of bamboo-shaped N-doped carbon nanotube architectures, the as-pr

Present tense: a phenomenology of grief in neurosurgery.

This paper applies phenomenological insights to the neurosurgeon's lived experience of grief, approaching it not as a discrete psychological reaction, but as an enduring aspect of professional life shaped by repeated encounters with loss, complication, and responsibility. For neurosurgeons, grief is not limited to moments of patient loss but emerges gradually through the cumulative weight of adverse outcomes, moral uncertainty, and the recognition of medicine's limits. Despite its pervasiveness, grief is often treated as something to be managed, resolved, or privately endured. Drawing on pheno

Magnetic Properties of Ge-Doped Fe3GaTe2 van der Waals Ferromagnets.

Recently, van der Waals magnetic materials have garnered extensive research attention. Among them, Fe3GaTe2 possesses strong magnetic anisotropy and a Curie temperature (TC) above room temperature, exhibiting great application potential in spintronics. Furthermore, Fe3GaTe2 shows good responsiveness to elemental doping. However, doping at Fe atomic sites with magnetic metal atoms, while modifying TC of Fe3GaTe2, disrupts its intrinsic ferromagnetic coupling interactions. This leads the doped Fe3GaTe2 to exhibit properties such as antiferromagnetism, which is unfavorable for the exploitation of

The Sponge Loop: How an Ancient Animal-Microbe Engine Fuels Modern Ocean Ecosystems.

Sponges can be considered biological engines that fuel the ecology of shallow-water to deep-sea ecosystems through the sponge loop. Here, we discuss new insights on the role of the sponge holobiont in the uptake, transformation, and transfer of resources that fuel the many food webs in which sponge engines operate. The tightly coupled animal-microbe partnership consists of many intrinsic traits (e.g., growth form, tissue architecture, and number and diversity of microbial symbionts) that depend on a huge variety of environmental factors (e.g., temperature, light, and both organic and inorganic

Molecular mimicry in the agroecosystem: A new paradigm for understanding how pesticide residues drive the emergence of antimicrobial resistance.

Antimicrobial resistance (AMR) is a mounting global crisis, with environmental dissemination of antibiotic resistance genes (ARGs) emerging as a critical driver. Agroecosystems, chronically exposed to complex mixtures of bioactive chemicals, including pesticides, represent an underrecognized hotspot for AMR evolution. This review synthesizes established mechanisms by which pesticides select for resistance and introduces a novel hypothesis: molecular mimicry as a hidden driver. Evidence highlights three key pathways: cross-resistance via multidrug efflux pumps; coselection on mobile genetic ele

A generalized multilayer reaction-diffusion model for tumor dynamics under hyperthermic therapy with spatial heterogeneity.

Local hyperthermia is an effective treatment for advanced malignant tumors, whose multilayer structure and physiological heterogeneity strongly influence their response to therapy. In this work, a generalized multilayer reaction-diffusion model is developed to study tumor-volume dynamics under localized hyperthermic treatment. The tumor is represented as a set of interacting biological layers, allowing spatial variation in key parameters such as diffusion and proliferation. The model is formulated with appropriate initial, interface and boundary conditions and solved using the finite element m

Multifractal evolution of shale fracture and pore structures under uniaxial compression.

The evolution of fracture–pore structures critically controls the mechanical stability and permeability of shale reservoirs. To clarify their coupled behavior during stress-induced failure, we investigate the multifractal evolution of fractures and pores in shale under uniaxial compression using digital rock reconstruction and discrete element modeling. The proposed fracture-damage framework reproduces fracture nucleation, propagation, and coalescence, and reveals a strong correspondence between energy conversion and damage development. Multifractal analyses show that pore heterogeneity primar

A novel variable stiffness actuator with a rotary magnetorheological damper.

A novel rotary variable stiffness actuator (VSA) is presented to meet the requirements of the variable stiffness drive for a robot joint. Considering the characteristics of a rotary magnetorheological damper (MRD) that has controllable torque and fast response, a servo motor and a rotary MRD are combined to form a VSA. The principle of variable stiffness drive is analyzed according to the frame of the VSA. The rotary MRD for the VSA is designed, and the electromagnetic field distribution in the damper is analyzed with the finite element method. Then, the torque and magnetomotive force value of
