A comparison of two tests for the assessment of blood pressure responses to sodium.

In order to assess the congruity of two different methods for the characterization of blood pressure responsivity to alterations in sodium and extracellular fluid volume balance, we studied 40 normotensive and hypertensive humans. All subjects were initially studied with a protocol of rapid sodium and volume expansion induced by intravenous administration of 0.9% saline (2 L over 4 h) followed by a day of sodium and volume depletion achieved by a low (10 mmol) sodium diet and three 40 mg oral doses of furosemide. Subsequently the subjects underwent a dietary protocol consisting of 5 days of a

Regulation of Na channels of the rat cortical collecting tubule by aldosterone.

The activity of apical membrane Na channels in the rat cortical collecting tubule was studied during manipulation of the animals' mineralocorticoid status in vivo using a low-Na diet or the diuretic furosemide. Tubules were isolated and split open to expose the luminal membrane surface. Induction of Na channel activity was studied in cell-attached patches of the split tubules. No activity was observed with control animals on a normal diet. Channel activity could be induced by putting the animals on the low-Na diet for at least 48 h. The mean number of open channels per patch (NPo) was maximal

Amphotericin B nephrotoxicity.

The frequency of fungal infections is increasing. Amphotericin B remains the anti-fungal drug of choice for most systemic infections, but a limiting factor for its use is the development of nephrotoxicity. Amphotericin B-induced nephrotoxicity is manifested as azotaemia, renal tubular acidosis, impaired renal concentrating ability and electrolyte abnormalities like hypokalaemia and sodium and magnesium wasting. All these abnormalities occur to varying degrees in almost all patients receiving the drug. Upon withdrawal of therapy renal function gradually returns to baseline, although in some ins

Ion Homeostasis in Chloroplasts under Salinity and Mineral Deficiency : I. Solute Concentrations in Leaves and Chloroplasts from Spinach Plants under NaCl or NaNO(3) Salinity.

Spinach (Spinacia oleracea var "Yates") plants in hydroponic culture were exposed to stepwise increased concentrations of NaCl or NaNO(3) up to a final concentration of 300 millimoles per liter, at constant Ca(2+)-concentration. Leaf cell sap and extracts from aqueously isolated spinach chloroplasts were analyzed for mineral cations, anions, amino acids, sugars, and quarternary ammonium compounds. Total osmolality of leaf sap and photosynthetic capacity of leaves were also measured. For comparison, leaf sap from salt-treated pea plants was also analyzed. Spinach plants under NaCl or NaNO(3) sa

Vasopressin-central nervous system interactions in the development of DOCA hypertension.

DOCA-salt hypertension does not develop in rats with hereditary lack of vasopressin (DI rats) nor in rats with lesion of the anteroventral region of the third ventricle (AV3V), an area controlling vasopressin (VP) release. We examined, therefore, the effect of VP treatment on the development of DOCA salt hypertension in AV3V-lesioned (AV3V-L) normal Sprague-Dawley rats and in Brattleboro rats homozygous for diabetes insipidus (DI rats). We also examined changes in vascular reactivity in isolated, perfused kidneys in the experimental groups. Whereas sham-lesioned (SL) rats showed hypertension a

Intrinsic mineralocorticoid agonist activity of some nonsteroidal anti-inflammatory drugs. A postulated mechanism for sodium retention.

Because some nonsteroidal anti-inflammatory drugs (NSAID) induce salt and water retention and exhibit other steroid-like actions, studies were performed to ascertain whether these drugs possess intrinsic mineralocorticoid agonist activity. In vitro competitive binding assays utilizing tissue from adrenalectomized rats demonstrated that some NSAID can displace [3H]-aldosterone from renal cytoplasmic mineralocorticoid receptors. Displacement potency for these sites was in the sequence: aldosterone greater than spironolactone greater than phenylbutazone (PBZ) greater than aspirin (ASA) greater th

Effect of fatty acids on growth of Japanese encephalitis virus cultivated in BHK-21 cells and phospholipid metabolism of the infected cells.

Growth of Japanese encephalitis virus (JEV) in BHK-21 cells was stimulated in the presence of 20 to 40 mug of the sodium salt of oleic acid (cis-9-octadecenoic acid, 9-18:1) per ml supplemented in Waymouth medium. The stimulatory effect of the salt was highest when 9-18:1 was added after adsorption of the virus. Study of the effect of other fatty acids on growth of JEV showed the following results: the longer the chain length of the saturated fatty acid salt, the higher the stimulatory effect on viral growth. In contrast, polyunsaturated fatty acids had an inhibitory effect on viral growth. Th

THE EFFECT OF SERUM UPON THE GERMICIDAL ACTION OF SOAPS.

1. Far more information about the effect of serum or other substances upon the germicidal action of soaps can be obtained by determining the germicidal titers over a wide range of pH than by determining the titer at a single pH. In this way a characteristic curve for each test substance is obtained. 2. The curve for a particular concentration of serum bears a definite relationship to the curve for salt solution (buffer) alone. Wherever the titer in salt solution is high, very small amounts of serum greatly diminish that titer. Wherever the titer in salt solution is low, small amounts of serum

Fast-Dissolving and Highly Soluble Dasatinib Multicomponent Crystals Efficiently Inhibit Leukemia Cell Viability.

Dasatinib (DAS) is a biopharmaceutical classification system (BCS) class II drug that is used orally to treat chronic myelogenous leukemia. However, its very low and pH-dependent aqueous solubility severely limits its oral bioavailability. To address this biopharmaceutical limitation, we designed more soluble multicomponent crystalline forms (MCCs) of DAS. Two novel salt solvate forms were successfully obtained via solvent evaporation through reactions with maleic and fumaric acids. A comprehensive solid-state evaluation of these salt solvate forms was conducted by using X-ray diffraction (SCX

Co-metabolism of Na+/K+ ion regulated physiological enhancement on selenium-accumulation in Saccharomyces yeasts.

BACKGROUND: Selenium is an important nutritional supplement that mainly exists naturally in soil as inorganic selenium. Saccharomyces cerevisiae cells are excellent medium for converting inorganic selenium in nature into organic selenium. RESULTS: Under the co-stimulation of sodium selenite (Na2SeO3) and potassium selenite (K2SeO3), the activity of selenophosphate synthetase (SPS) was improved up to about five folds more than conventional Na2SeO3 group with the total selenite salts content of 30 mg/L. Transcriptome analysis first revealed that due to the sharing pathway between sodium ion (Na

Salinity-mediated enhancement of quaternary ammonium compounds resistance and removal in endophytic bacteria LSE01.

The widespread usage of quaternary ammonium compounds (QACs) as disinfectants during the COVID-19 pandemic poses significant environmental risks, such as toxicity to organisms and the emergence of superbugs. In this study, different inorganic salts (NaCl, KCl, CaCl2, MgCl2) were used to induce endophytes LSE01 isolated from hyperaccumulating plants. After five generations of cultivation under 80 g/L NaCl, the minimum inhibitory concentration (MIC) of LSE01 to QACs increased by about 3-fold, while its degradation extent increased from 8% to 84% for C12BDMA-Cl and 5%-89% for C14BDMA-Cl. Exploring of multi-functional umami peptides from Stropharia rugosoannulata: Saltiness-enhancing effect and mechanism, antioxidant activity and potential target sites.

Umami peptides enhance flavor and offer potential health benefits. We analyzed the taste-value profiles of five novel umami peptides from Stropharia rugosoannulata using E-tongue, exhibiting significant saltiness characteristics. While the peptides PHEMQ and SEPSHF exhibited higher saltiness, their mixture with salt did not enhance saltiness compared to individual peptides. Surprisingly, SGCVNEL, which was initially weak in saltiness, showed remarkably enhanced saltiness when mixed with salt, possibly due to have strong binding with receptors. Molecular docking elucidated the salt-forming mech

Gitelman Syndrome and Hypertension: A Case Report.

In a patient with persistent hypokalemia, it is important to consider Gitelman syndrome, a rare, salt-wasting tubulopathy inherited in an autosomal recessive pattern. Gitelman syndrome leads to electrolyte abnormalities like hypokalemia, hypomagnesemia, and metabolic alkalosis. Typical clinical features include muscle cramps, fatigue, polydipsia, and salt cravings. Our case involves a female patient in her early 40s who visited the endocrinology clinic with symptoms of polyuria, constipation, muscle weakness, and fatigue. Electrolyte abnormalities included hypokalemia, hypomagnesemia, hypochlo

Development of New Innovative Synthetic Organic Chemistry Using Lone Pairs of Oxygen Atoms.

Oxygen atoms have a lone pair of electrons, so they have high chelation ability, high nucleophilic ability, stabilizing ability of adjacent cations, and take a chelate or oxocarbenium ion structure with Lewis acids and metals. I took advantage of these properties to develop three new reactions, 1) asymmetric synthesis of chiral quaternary carbon centers, 2) asymmetric synthesis using acetal functions, and 3) organic chemistry using acetal-type reactive salt chemical species, and applied them to biologically active natural products synthesis. New reactions described here are all innovative and

Application of Multiple Regression and Design of Experiments for Modelling the Effect of Monoethylene Glycol in the Calcium Carbonate Scaling Process.

To avoid gas hydrate formation during oil and gas production, companies usually employ thermodynamic inhibitors consisting of hydroxyl compounds, such as monoethylene glycol (MEG). However, these inhibitors may cause other types of fouling during production such as inorganic salt deposits (scale). Calcium carbonate is one of the main scaling salts and is a great concern, especially for the new pre-salt wells being explored in Brazil. Hence, it is important to understand how using inhibitors to control gas hydrate formation may be interacting with the scale formation process. Multiple regressio

Role of the kidneys in resistant hypertension.

Resistant hypertension is a failure to achieve goal BP (<140/90 mm Hg for the overall population and <130/80 mm Hg for those with diabetes mellitus or chronic kidney disease) in a patient who adheres to maximum tolerated doses of 3 antihypertensive drugs including a diuretic. The kidneys play a critical role in long-term regulation of blood pressure. Blunted pressure natriuresis, with resultant increase in extracellular fluid volume, is an important cause of resistant hypertension. Activation of the renin-angiotensin-aldosterone system, increased renal sympathetic nervous system activity and i

Hypothesis: Cystic fibrosis carrier geography reflects interactions of tuberculosis and hypertension with vitamin D deficiency, altitude and temperature. Vitamin D deficiency effects and CF carrier advantage.

Interactions between selective factors (hypertension and tuberculosis) and environmental effects (vitamin D deficiency [VDD], temperature, and altitude) largely explain cystic fibrosis (CF) carrier geography. For VDD sequelae such as hypertension and tuberculosis vulnerability, clinical evidence of carrier protection is supported by indications that decreased CF arylsulfatase B activity suppresses tuberculosis, and that excess CF salt loss decreases blood pressure. A need for salt retention in the tropics selected against CF carriers despite possible advantages against cholera, typhoid, and ot

Effect of pressure on [2]pseudorotaxane formation and decomplexation and their corresponding activation volumes.

In this study, we investigated the effect of pressure on the formation and decomplexation of [2]pseudorotaxanes. High pressure accelerated the formation of [2]pseudorotaxanes in an aprotic nonpolar solvent (CDCl(3)/CD(3)CN) via the slipping approach when using two crown ether/secondary ammonium salt systems: dibenzo[24]crown-8/bis(cyclohexylmethyl)ammonium salt (1a/2a) and tetrabenzo[24]crown-8/dibenzylammonium salt (1b/2b). The influence of pressure on the rate constants for the formation of the [2]pseudorotaxanes 3a and 3b revealed activation volumes (DeltaV(double dagger)) of -2.5 and -4.6

Iodine-deficiency disorders.

2 billion individuals worldwide have insufficient iodine intake, with those in south Asia and sub-Saharan Africa particularly affected. Iodine deficiency has many adverse effects on growth and development. These effects are due to inadequate production of thyroid hormone and are termed iodine-deficiency disorders. Iodine deficiency is the most common cause of preventable mental impairment worldwide. Assessment methods include urinary iodine concentration, goitre, newborn thyroid-stimulating hormone, and blood thyroglobulin. In nearly all countries, the best strategy to control iodine deficienc

Role of macula densa neuronal nitric oxide synthase in renal diseases.

Macula densa cells have an important role in the regulation of glomerular blood flow and glomerular filtration by its regulation of afferent arteriolar vascular tone. Nitric oxide derived from neuronal nitric oxide synthase (nNOS) in macula densa can dilate afferent arterioles. Macula densa nNOS is important for renin secretion, and its expression is regulated by dietary salt, renal angiotensin II, intracellular pH, and other factors. In salt-sensitive hypertension, nNOS is suppressed, whereas in SHR or in the early phase of diabetes, nNOS is increased in macula densa along with NADPH oxidase,
