Reimagining VEGF Therapy: The Role of PLCγ2 in Pan-Organ Endothelial Networks.

OBJECTIVE: Ischemic tissue perfusion remains a major clinical challenge despite VEGF's central role in angiogenesis. Limited success of VEGF-based therapies underscores the need to understand endothelial heterogeneity, as distinct subsets exhibit differential responsiveness. This study explores conserved endothelial states that govern VEGF-driven angiogenesis across human organs and evaluates their functional significance. APPROACH: We performed single-cell and single-nucleus RNA-sequencing (scRNA-seq/snRNA-seq) analysis of endothelial cells (ECs) from eight human tissues using GTEx data, val

Integrative Multi-Omics and Machine Learning Reveal Senescent Fibroblast-Associated ADAMTS2 as a Critical Indicator of Intestinal Inflammation in IBD.

Although cellular senescence contributes to tissue dysfunction in inflammatory bowel disease (IBD), the heterogeneity of senescence at the single-cell resolution remains poorly defined. In this study, we systematically characterized the senescence landscape within the IBD microenvironment, with particular emphasis on fibroblast subpopulations and their systemic consequences. Single-cell RNA sequencing (scRNA-seq) dataset was applied to profile senescence-associated feature heterogeneity. Fibroblasts were classified into high-senescence (HS-Fibro) and low-senescence (LS-Fibro) groups according

Web-based gene expression analysis-paving the way to decode healthy and diseased ocular tissue.

BACKGROUND: Gene expression analysis using RNA sequencing has helped to improve the understanding of many diseases. Databases, such as the Gene Expression Omnibus database of the National Center for Biotechnology Information provide RNA sequencing raw data from various diseased tissue types but their analysis requires advanced bioinformatics skills. Therefore, specific ocular databases provide the transcriptional profiles of different ocular tissues and in addition enable intuitive web-based data analysis. OBJECTIVE: The aim of this narrative review is to provide an overview of ocular transcr

The novel human gene aprataxin is directly involved in DNA single-strand-break repair.

The cells of an ataxia-oculomotor apraxia type 1 (AOA1) patient, homozygous for a new aprataxin mutation (T739C), were treated with camptothecin, an inhibitor of DNA topoisomerase I which induces DNA single-strand breaks. DNA damage was evaluated by cytogenetic analysis of chromosomal aberrations. The results obtained showed marked and dose-related increases in induced chromosomal aberrations in the patient and her heterozygous mother compared to the intrafamilial wild-type control. The alkaline comet assay confirmed this pattern. Moreover, the AOA1 cells did not show hypersensitivity to ioniz

Integrated gene expression and mutation profiling of chemoresistance in canine mammary carcinoma cells.

Mammary gland tumors (MGTs) are common in female dogs, and are usually not treated with chemotherapy due to the lack of evidence of benefit for the patients. In clinical practice, when a female dog receives neoadjuvant or adjuvant chemotherapy, minimal improvement is achieved. Therefore, chemotherapy is not usually applied for these patients. We hypothesized that some canine MGTs do not respond to chemotherapy due to intrinsic chemoresistance. Chemoresistance can be a challenge and is often associated with overexpression of ABC transporters, such as MDR1, MRP1, MRP2, MRP3 and BCRP, which reduc

Exploring peripheral immune mechanisms and potential therapeutic drugs for Alzheimer's disease.

Alzheimer's disease (AD) is a neurodegenerative disorder characterised by cognitive decline. Peripheral immune dysregulation often precedes central immune abnormalities and may therefore provide valuable clues for the early diagnosis and stratification of AD. Current research on peripheral immunity in AD remains limited. This study provides new insights into the pathogenesis and therapeutic strategies for AD. Bioinformatics and single-cell RNA sequencing were used to analyse peripheral immune-related gene expression in AD. Key genes and immune cell populations linked to AD were identified. Que

Novel insights into the mechanism of formaldehyde-induced lung cancer: a network toxicology and molecular docking approach.

We explored the complex effects of formaldehyde (FA) on lung cancer through network toxicology and molecular docking techniques, focusing on understanding the molecular mechanisms by which FA affects lung cancer from a biological network perspective. With the information of formaldehyde and lung cancer targets provided by databases (ChEMBL, STITCH, GeneCards, OMIM), we identified key potential targets that are closely related to formaldehyde and lung cancer. Protein-protein interaction(PPI) networks for core targets were constructed using the STRING database, and ontology (GO) and Kyoto Encycl

Broad remodeling of the pulmonary immune landscape occurs during IAPA with specific functional deficits of neutrophil subsets.

BACKGROUND: Influenza-associated pulmonary aspergillosis (IAPA) is a severe complication of influenza infection associated with prolonged intensive care unit stay and increased mortality. Although impaired antifungal immunity has been implicated in IAPA pathogenesis, the cell type-specific immune mechanisms driving susceptibility remain incompletely understood. We aimed to characterize the pulmonary immune landscape during IAPA using single-cell transcriptomics and functional neutrophil assays. METHODS: Male C57BL/6 mice were assigned to naïve control, influenza A/PR/8/34 (H1N1) infection, A.

Periaortic lymphatic vessels protect against thoracic aortic dissection through mobilizing immune response.

AIMS: Inflammation and the immune response are crucial factors in the progression of thoracic aortic dissection (TAD), while lymphatic vessels (LVs) play a key role in regulating the inflammatory response by facilitating the drainage of inflammatory mediators, and immune cell trafficking. Here, we investigated the role of LVs in the pathogenesis of TAD. METHODS AND RESULTS: Aortas collected from TAD patients and mice both showed a reduction in the number of periaortic LVs during TAD progression. Genetic linage tracing, tissue clearing technology, and fluorochrome tracers were used to explore

Glycoprotein 130 Antagonism Counteracts Metabolic and Inflammatory Alterations to Enhance Right Ventricle Function in Pulmonary Artery Banded Pigs.

BACKGROUND: Right ventricular dysfunction (RVD) is a risk factor for death in multiple cardiovascular diseases, but RV-enhancing therapies are lacking. Inhibition of glycoprotein-130 (GP130) signaling with the small molecule SC144 improves RV function in rodent RVD via anti-inflammatory and metabolic mechanisms. However, SC144's efficacy and molecular effects in a translational large animal model of RVD are unknown. METHODS: 4-week-old castrated male pigs underwent pulmonary artery banding (PAB). After 3 weeks, PAB pigs were randomized into 2 groups (daily injections of SC144 [2.2 mg/kg, PAB-

Airway epithelial stem cell renewal and differentiation: overcoming challenging steps towards clinical-grade tissue engineering.

BACKGROUND: Despite their life-saving potential, tissue engineering approaches for the treatment of extensive tracheal and bronchial defects still face significant limitations. A major challenge is the inability to regenerate a functional airway epithelium containing the appropriate amount of stem cells required for long-term tissue renewal following transplantation of the bioengineered graft. In this scenario, extensive cell culture characterization, validation assays and quality controls are needed to guide each step of the regeneration process. METHODS: Stem cell depletion is often due to

Integrative Multi-Omics and Machine Learning Reveal Shared Biomarkers in Type 2 Diabetes and Atherosclerosis.

Atherosclerosis (AS) is a leading cause of death and disability in type 2 diabetes mellitus (T2DM). However, the shared molecular mechanisms linking T2DM and atherosclerosis have not been fully elucidated. We analyzed AS- and T2DM-related gene expression profiles from the Gene Expression Omnibus (GEO) database to identify overlapping differentially expressed genes and co-expression signatures. Functional enrichment (Gene Ontology (GO)/Kyoto Encyclopedia of Genes and Genomes (KEGG)) and protein-protein interaction (PPI) network analyses were then used to describe the pathways and interaction mo

Single cell RNA sequencing of human FAPs reveals different functional stages in Duchenne muscular dystrophy.

Background: Duchenne muscular dystrophy is a genetic disease produced by mutations in the dystrophin gene characterized by early onset muscle weakness leading to severe and irreversible disability. Muscle degeneration involves a complex interplay between multiple cell lineages spatially located within areas of damage, termed the degenerative niche, including inflammatory cells, satellite cells (SCs) and fibro-adipogenic precursor cells (FAPs). FAPs are mesenchymal stem cell which have a pivotal role in muscle homeostasis as they can either promote muscle regeneration or contribute to muscle de

SLC39A1 promotes pancreatic cancer metastasis by elevating intracellular zinc and activating the Src/FAK signaling pathway.

Pancreatic cancer (PC) is a highly aggressive malignancy, and zinc ion homeostasis is closely linked to its metastatic potential, though the underlying regulatory mechanisms remain unclear. To investigate this, single-cell RNA sequencing data from primary and metastatic PC were downloaded from the GEO database to analyze differences in zinc ion levels and identify key zinc-regulating proteins. A PC mouse model was further established, and experiments including immunofluorescence, immunohistochemistry, and zinc ion level detection were conducted to comprehensively evaluate the impact of zinc io
