In the peripheral nervous system, neurons derived from specific rostrocaudal levels of the neuraxis selectively synapse on targets that arise from corresponding body positions. To identify molecules involved in such position-dependent connectivity, we used subtractive hybridization to isolate genes selectively expressed in rostral or caudal skeletal muscle. One mRNA that was more abundant in neck than in hindlimb muscles encoded the mouse ortholog of human AL-1 and chick RAGS, membrane-associated ligands of Eph tyrosine kinases that have recently been implicated in cortical axon fasciculation

Animal studies have shown the importance of the nucleus tractus solitarii, a collection of neurons in the brain stem, in the acute regulation of blood pressure. Impulses arising from the carotid and aortic baroreceptors converge in this center, where the first synapse of the baroreflex is located. Stimulation of the nucleus tractus solitarii provides an inhibitory signal to other brain stem structures, particularly the rostral ventrolateral medulla, resulting in a reduction in sympathetic outflow and a decrease in blood pressure. Conversely, experimental lesions of the nucleus tractus solitari

Presentation of an acoustic signal to one ear can suppress sound-evoked activity recorded at the opposite ear. The suppression appears to be mediated by medial olivocochlear (MOC) efferent neurons synapsing with outer hair cells (OHCs) and acting through the MOC neurotransmitter, acetylcholine (ACh). The purpose of the present investigation was to study the suppression of distortion product otoacoustic emissions (DPOAEs) by contralateral sound and to examine whether the suppression could be blocked by known antagonists of olivocochlear (OC) efferent activity. Urethane-anesthetized guinea pigs

1. There is not yet a consensus as to which of the neuronal membranes, which molecular component of any particular membrane or what specific function of the membrane is critical for general anaesthesia. 2. However, when considering anaesthetic effects on different synapses, with neurotransmitter receptors, ion channels etc., the unifying central concept is action at a membrane level. 3. This paper will review the general evidence for this unifying hypothesis, and consider the apparent exceptions and limitations. 4. The membrane hypothesis is usually stated in the form of the Meyer and Overton

The effects of prenatal protein malnutrition on kindling-induced changes in inhibitory modulation of dentate granule cell activity were examined by analysis of extracellular field potentials recorded from the granule cell layer of the dentate gyrus in response to paired-pulse stimulation of the perforant pathway in freely-moving rats. Since we have shown that kindling results in enhanced synaptic transmission at the level of the perforant path/granule cell synapse (see preceding paper), we sought to determine if the kindling process might induce changes in inhibitory modulation of granule cell

We have investigated the actin content and ultrastructure of two kinds of presumed sensory projections on the lip epidermis of beroid ctenophores. Transmission electron microscopy showed that conical pegs contain a large bundle of densely packed, parallel microfilaments. Rhodamine-phalloidin brightly stained the pegs, confirming that they contain filamentous actin. Epidermal cells with actin pegs also bear a single long cilium with an onion-root structure, previously described as arising from a different type of cell. The actin peg and onion-root cilium project side-by-side, defining a polariz

The pinealocytes--the main cellular elements of the pineal organ--are polarized, displaying a (photo)receptor and an axonic effector cell pole. The receptor endings are of two main types: they bear rod-type or cone-type outer segments characterized by the presence of immunoreactive opsin-, S-antigen- and vitamin A-binding sites. The effector pole may form ribbon-containing synapses on the secondary pineal neurons, and/or neurohormonal terminals on the basal lamina of the pineal nervous tissue. Applying potassium pyroantimonate (PPA) to electron-microscopic histochemistry, we found in the frog

In unanaesthetized cats the biochemical mechanisms and the functional characteristics of the emetic action of injection of noradrenaline and McN-A-343, a ganglionic muscarinic stimulant into the cerebral ventricle (i.c.v.) through chronically implanted cannulae were investigated. Both produced dose-dependent and shortlasting emetic response. The emesis evoked by noradrenaline was abolished, whereas the emesis induced by McN-A-343 was not completely blocked after ablation of the area postrema. Further, the emetic response to noradrenaline as well as to McN-A-343 was attenuated or blocked in cat

Alpha-adrenergic activity mediated by the autonomic nervous system, consisting of the release of norepinephrine and its engagement by the alpha receptors, is a recognized pathway for arteriolar vasoconstriction. The pathway is well traveled in acute reactions to emotion and stress, but this complicity in chronic hypertension other than pheochromocytoma has been more difficult to show. Among the reasons for the difficulty is the failure to develop an animal model of hypertension mediated through the nervous system. Another reason is the uncertainty as to whether or not measurements of plasma no

Stimulation of the trigeminal nerve or ganglion in the cat caused a frequency-dependent reduction in carotid vascular resistance. Systemic arterial blood pressure (SABP) decreased at low frequencies (0.2 to 5 sec-1) and increased at higher frequencies, thus increasing carotid blood flow at the higher frequencies. The effect on resistance was predominantly ipsilateral and was unaltered by cervical sympathectomy, but was abolished or substantially reduced by section of the trigeminal root proximal to the ganglion. Diminution of carotid vascular resistance was replicated by stimulation of the gre

Five-hydroxytryptamine (5-HT) causes a hyperpolarization and increased conductance of the leech body wall muscle cell membrane. If 5-HT is applied in the absence of the Cl minus ion, the response appears as a depolarization, whereas if 5-HT is applied in the absence of the K+ ion, the response is a hyperpolarization. In both cases, the conductance of the muscle cell membrane is increased. Stimulation of the peripheral nerve to the body wall muscle produces a complex junctional potential in muscle cells. Exposing the muscle to d-tubocurarine (d-TC) eliminates the excitatory component (EJP) of t

Antigen-specific T cells constitute a low percentage of peripheral blood mononuclear cells (PBMCs) and require antigen presentation by autologous antigen-presenting cells for activation and cytokine secretion. Conventional interferon-γ release assay (IGRA) averages rare activation events beyond cellular resolution, while single-cell compartmentalization disrupts the intracellular synapses for T cell immunity. Here, we developed mSTAR (matrix-based Specific T-cell Activation Reporter), an active-matrix digital microfluidic platform that partitions microliters of PBMCs into a nanoliter-scale mat

Neuroimmune mechanisms are central to Alzheimer's disease (AD), but a single continuum of "more inflammation" does not explain why similar amyloid-β (Aβ) and tau burdens are associated with different clinical trajectories, tissue responses, and treatment risks. We present a testable neuroimmune-state framework that integrates, rather than replaces, the AT(N) system, cellular-phase models, and transcriptional descriptions of disease-associated microglia and reactive astrocytes. The framework treats neuroimmune states as multidimensional, regionally heterogeneous configurations defined by pathol

α-Synuclein is a presynaptic protein that has been extensively studied for its role in Parkinson's disease. Multiplication and missense variants in the gene encoding α-synuclein are causes of familial PD, and aggregated α-synuclein is a major component of Lewy bodies and Lewy neurites, the histopathological hallmark of the disease. However, the native function of α-synuclein remains unclear, though several lines of evidence suggest it modulates presynaptic function. α-Synuclein binds the vesicular SNARE protein synaptobrevin-2/VAMP2, and increased levels of α-synuclein potentiate SNARE complex

Leber congenital amaurosis (LCA) and Early-onset severe retinal dystrophy (EOSRD) manifest within the first months and the first years of life, respectively. They are the leading cause of severe vision impairment in childhood. Using next generation sequencing, we identified eight families of patients with LCA/EOSRD carrying biallelic combination of six germline variants in DDX41 , encoding a DEAD-box ATPase RNA helicase involved in RNA splicing, innate immunity and hematopoiesis. In fibroblasts from a patient carrying the homozygous missense variant c.1187T>C (p. Ile396Thr) and in the retina o

Information about head motion and gravity is conveyed to the brain by vestibular nerve afferents which are subdivided by their spontaneous firing properties into regular and irregular subtypes, thought to be differentially responsible for vestibulo-ocular versus vestibulo-spinal reflexes. In the vestibular periphery, afferents make glutamatergic synapses with type II hair cells (HCs) in all vertebrates. During the evolutionary transition to land, however, amniotes (reptiles, birds, and mammals) additionally developed type I vestibular HCs in which unique calyceal afferent terminals cover the b

Microglia are critical modulators of neuronal activity, regulating brain function through removal of dying neurons, pruning non-functional synapses, and producing ligands that support neuronal survival. Microglial morphologies change with aging, shifting from a homeostatic, surveillant state (ramified morphology) to a more reactive state (intermediate, amoeboid, and dystrophic morphology). Common marmosets (Callithrix jacchus) are a valuable translational nonhuman primate model for aging, and studies of neuroinflammation and neurodegenerative disease. These small, genetically diverse platyrrhi

The eye, long viewed as immune-privileged, is now recognized as a dynamic immunological environment where innate and adaptive mechanisms intersect to maintain retinal homeostasis. In aging and age-related retinal diseases such as age-related macular degeneration (AMD), this balance deteriorates as chronic, low-grade inflammation replaces immune quiescence. Adaptive immune involvement in AMD is reflected by increased antiretinal autoantibodies and systemic immune changes, including accelerated aging of CD8+ T-cells. Complement activation further amplifies adaptive responses; C5a stimulates T-ce

OBJECTIVE: To investigate the therapeutic effects of gomisin E on insomnia in aged rats and to elucidate the underlying mechanisms.
METHODS: An aging-related insomnia rat model was established by intraperitoneal injection of D-galactose combined with p-chlorophenylalanine (PCPA). Rats were treated with gomisin E at different doses (5, 15, and 45 mg/kg). Sleep quality, depressive-like behaviors, and cognitive function were evaluated using the pentobarbital sodium-induced sleep test, sucrose preference test, and Morris water maze test, respectively. Potential targets and signaling pathways were

Proprotein convertase subtilisin-kexin type 9 (PCSK9) has recently emerged as a significant mediator that links metabolic dysfunction to neurodegeneration related to Alzheimer's disease (AD). It is a well-known and crucial component involved in cholesterol homeostasis. However, its function in the central nervous system (CNS) is still in its early stages. Normally, it is engaged with the breakdown of cholesterol in the body, but within the brain, PCSK9 has been seen to disrupt the homeostasis of cholesterol and its uptake. Receptors such as LDL receptor-related protein-1 (LRP-1) and low-densit
