By using retroviral mediated gene transfer technique, a primary schwannoma culture from a 56-year-old Neurofibromatosis type 2 (NF2) patient was immortalized with HPV E6-E7 genes. This cell line, HEI193, has a unique splice site mutation of the NF2 gene. Both immunocytochemistry and molecular biology techniques were used to demonstrate that this cell line is of Schwann cell origin. Comparison of the primary tumor with HEI193 revealed the same NF2 mutation and an identical pattern of allele loss at multiple loci, indicating that the established cell line had maintained many of the properties of

Most human diseases are substantially affected by genetic factors. It now seems clear that the pathogenesis of most diseases lies in complex interactions among the genotype, the environment, and the nature of the process that leads to cell, tissue, organ, or systemic injury. The information derived from the knowledge of the recent completion of the human genome, when combined with the sophisticated tools of molecular biology, will provide the framework for more rapid identification of the genes responsible for susceptibility to disease. Genetic approaches to complex disorders offer great poten

OBJECTIVE: To detail the biology and diagnostic usefulness of serum procalcitonin in critical illness.
DATA SOURCES: A review of articles published in peer reviewed journals from 1990 to 2001 and identified through a MEDLINE search on procalcitonin.
SUMMARY OF REVIEW: Procalcitonin (PCT) is a prohormone of calcitonin. Serum levels are elevated during sepsis and have been identified as a potential marker of infection in critical illness. However, its function and precise source of origin during sepsis remain unclear. The value of estimating serum PCT appears to be in the differentiation of in

The recent history of Pelizaeus-Merzbacher Disease (PMD) demonstrates paradigmatically the impact of basic biological research on clinical neurology and brain pathology: this rare and peculiar hereditary disease has become one of the best known disorders of its kind, through a cooperative research effort in neuropathology, human genetics, neurochemistry and molecular biology. PMD, a genetic dysmyelination restricted to the CNS, has been identified as a disease that involves the X chromosome-linked gene for myelin proteolipid protein (PLP), a major structural myelin component. Today more than 3

Leukoplakia has evolved as a clinico-pathologic concept over many years, with the current clinical designation being accepted worldwide. Reflective of the biology of leukoplakia is the concept of cellular atypia and epithelial dysplasia. Adding to a better understanding of leukoplakia in general has been the definition of relevant clinical subsets which, in some cases, includes etiology (snuff), while in other cases a verrucous clinical appearance will suggest a more aggressive anticipated behavior pattern. Tobacco usage, in many of its forms, remains the prime etiologic factor; however, other

Recent data indicate a previously unsuspected link between the complement system and adipocyte biology. Murine adipocytes produce key components of the alternative pathway of complement and are able to activate this pathway. This suggested to us an explanation for adipose tissue loss in partial lipodystrophy, a rare human condition usually associated with the immunoglobulin G(IgG) autoantibody nephritic factor (NeF) which leads to enhanced alternative pathway activation in vivo. We hypothesized that in the presence of NeF, there is dysregulated complement activation at the membrane of the adip

Recombinant DNA technology has made it possible to express foreign genes in viruses and bacteria. This has led to the idea of engineering live attenuated vaccines to express protective antigens from foreign pathogens. Vaccinia virus, the smallpox vaccine, has been used to ploneer this approach. Several hundred recombinants have been recorded in the literature and many of them have been shown to protect animals against challenge with the appropriate pathogen. Although mixed results have been obtained with experiments in humans and primates, and there is some concern over complications associate

Clinical characteristics of tumors metastatic to the orbit are related to primary tumor biology, and vary substantially among the various primary types. Common known primary sites include breast, lung, prostate, and melanoma. Tumor presentations can be classified into four generalized syndromes of mass, infiltrative, inflammatory, and functional effects. We found the infiltrative syndrome of presentation to be more common than for other types of orbital neoplasm. Accurate diagnosis often depends on recognition of the types of clinical syndromes and on the use of diagnostic modalities such as c

Periodic breathing (PB) has been related to both normal and pathologic respiratory system control in infants. However, comparison of the results of separate studies has been limited by the variability in procedures used by different investigators to quantify PB. In this study we scored 15 24-hr impedance pneumograms using the criteria of Parmelee et al. (Neuropediatrie 3:294-304, 1972), Christova-Gueorguieva (Biology of the Neonate 44:325-332, 1983), and Curzi-Dascalova, Kelly and Shannon (Pediatrics 63:355-360, 1979) and analyzed the resulting differences in several commonly used PB parameter

BACKGROUND: Meningiomas are the most common primary nervous system neoplasm in adults. There are few good cellular models, especially of high grade/malignant meningiomas, to use to identify new therapeutic agents and treatment regimens. The widely available, partially characterized, NF2- wildtype ( NF2 wt) Grade 3 malignant meningioma cell line IOMM-Lee can help meet this need.
METHODS: We generated new data to better characterize IOMM-Lee genomic and mtDNA variants, proliferation rate and colony-forming efficiency and sensitivity to ionizing radiation as a function of ATM kinase activity. A

Rotavirus is a segmented double-stranded RNA (dsRNA) virus that causes severe gastroenteritis in young children. We have established an efficient simplified rotavirus reverse genetics (RG) system that uses 11 T7 plasmids, each expressing a unique simian SA11 (+)RNA, and a cytomegalovirus support plasmid for the African swine fever virus NP868R capping enzyme. With the NP868R-based system, we generated recombinant rotavirus (rSA11/NSP3-FL-UnaG) with a genetically modified 1.5-kb segment 7 dsRNA encoding full-length nonstructural protein 3 (NSP3) fused to UnaG, a 139-amino-acid green fluorescent

Immune checkpoint blockade targeting PD-1/PD-L1 has reshaped cancer therapy across multiple tumor types. However, primary and acquired resistance remain major barriers to durable clinical benefit. Increasing evidence indicates that immunotherapy resistance is not driven by isolated molecular alterations, but by coordinated changes involving tumor-intrinsic immune escape, immunosuppressive tumor microenvironments, systemic inflammatory remodeling, and adaptive biomarker evolution under therapeutic pressure. This review focuses on pan-cancer biomarkers of immune therapy resistance from the persp

Myeloid differentiation primary response 88 (MyD88), a central adaptor protein governing Toll-like receptor (TLR) and interleukin-1 receptor (IL-1R) signaling cascades, is increasingly recognized as a pivotal mediator of neuroimmune interactions and neuromodulation. Beyond its canonical immune functions, emerging evidence reveals widespread MyD88 expression throughout the nervous system, where it plays functional roles in both glial populations and neuronal networks. While previous reviews have largely focused on glial mechanisms, recent studies highlight a complex, often overlooked aspect: th

Mitochondrial transfer and transplantation (mitoTT) has emerged as an experimental and potentially transformative therapeutic strategy, drawing attention for its ability to modulate cellular bioenergetics, stress responses, and tissue repair. Endogenous mitochondrial transfer can occur through tunneling nanotubes, extracellular vesicles, cell fusion, gap junction-associated communication, and phagocytosis-like internalization. These routes differ substantially in distance, directionality, cargo integrity, regulatory control, and physiological relevance. Preclinical studies indicate that exogen

Leptomeningeal disease (LMD) is one of the most devastating complications of advanced non-small cell lung cancer (NSCLC), historically associated with profound neurologic morbidity and poor survival. Advances in cerebrospinal fluid (CSF)-based diagnostics, improved molecular characterization, and the development of central nervous system (CNS)-penetrant targeted therapies have begun to reshape both disease assessment and treatment expectations, particularly in oncogene-driven NSCLC. Available data suggest that outcomes in LMD are highly heterogeneous and are influenced by molecular subtype, pe

The rapid dissemination of phylogenetically diverse porcine reproductive and respiratory syndrome virus type 1 (PRRSV-1) strains in China necessitates advanced research tools tailored to contemporary isolates. To address the scarcity of such resources, we developed a reverse-genetics platform for the prevalent Chinese PRRSV-1 strain GXFS20220129. A full-length infectious cDNA clone was constructed and used to rescue replication-competent virus (rGXFS/F3/ic), Its growth kinetics were showed slightly attenuated compared to the parental strain but maintained a similar overall growth pattern. Proteolysis-Targeting Chimeras (PROTACs) represent a novel and promising cancer treatment strategy considered a direct alternative to conventional small-molecule inhibitors. PROTACs selectively degrade disease-causing proteins (including previously 'undruggable' targets such as transcription factors and scaffolding proteins) by harnessing the cellular ubiquitin proteasome system. In this review, we look at the most recent developments in PROTAC technology and their oncology applications. Versatility, while maintaining substrate selectivity and degradation efficiency, has also been enhanced by

The emerging field of cancer neuroscience has reshaped our understanding of tumor biology, showing that the nervous system is an active participant in the tumor microenvironment. It modulates tumor progression through synaptic communication with cancer cells, paracrine signaling via neurotransmitters and neuropeptides, and indirect regulation of the tumor immune microenvironment. Recent discoveries have shown that neurons form synapses with brain tumors and peripheral cancers, driving tumor cell proliferation and invasion. Neurotransmitter and neuropeptide signaling activates pathways in cance

PURPOSE OF REVIEW: Anaphylaxis is a life-threatening systemic hypersensitivity reaction with highly variable clinical expression and unpredictable severity. Increasing evidence suggests that host-microbiota interactions may contribute to interindividual variability in allergic sensitization and effector responses. This review summarizes current mechanistic, preclinical, and emerging human data supporting a role for the microbiota in modulating anaphylaxis risk and severity.

Osteoimmunology explores the interplay between the skeletal system and the immune system. It focuses on the identification of shared signaling pathways and the mechanisms of mutual regulation. Bone homeostasis depends on the balance between the activity of bone-forming osteoblasts and bone-resorbing osteoclasts. Interestingly, the crosstalk between osteoclasts, osteoblasts, osteocytes, and immune cells not only shapes bone remodeling but also influences immune responses. A central pathway in osteoclast differentiation is the receptor activator of nuclear factor-κB ligand (RANKL) signaling, whi
