First-in-human study and clinical case reports of the alveolar bone regeneration with the secretome from human mesenchymal stem cells.

BACKGROUND: Secreted growth factors and cytokines in the conditioned medium from bone marrow-derived mesenchymal stem cells (MSC-CM) have several effects on cell behavior. Our previous studies revealed that MSC-CM enhances bone regeneration by increasing cell mobilization, angiogenesis, and osteogenesis in vitro and in vivo. This clinical study was undertaken to evaluate the safety and use of MSC-CM for alveolar bone regeneration in eight patients who were diagnosed as needing bone augmentation prior to dental implant placement. METHODS: The protocol of this clinical study was approved by the

Oriented growth of rat Schwann cells on aligned electrospun poly(methyl methacrylate) nanofibers.

Transplanted Schwann cells have the potential to serve as a support for regenerating neurites after spinal cord injury. However, implanted Schwann cells die off rapidly once transplanted partly owing to the absence of a proper matrix support, with a glia scar and a cavity being present instead at the injury site. For this report, we evaluated aligned electrospun poly(methyl methacrylate) nanofibers as a Schwann cell-loading scaffold in vitro. By monitoring the fluorescence of green fluorescence protein-containing Schwann cells cultured on nanofibers, we found that aligned nanofibers provided b

Magnetic Resonance Imaging of Cardiac Strain Pattern Following Transplantation of Human Tissue Engineered Heart Muscles.

BACKGROUND: The use of tissue engineering approaches in combination with exogenously produced cardiomyocytes offers the potential to restore contractile function after myocardial injury. However, current techniques assessing changes in global cardiac performance after such treatments are plagued by relatively low detection ability. Since the treatment is locally performed, this detection could be improved by myocardial strain imaging that measures regional contractility. METHODS AND RESULTS: Tissue engineered heart muscles (EHMs) were generated by casting human embryonic stem cell-derived car

Ex vivo construction of a novel model of bioengineered bladder mucosa: A preliminary study.

OBJECTIVE: To generate and to evaluate ex vivo a novel model of bioengineered human bladder mucosa based on fibrin-agarose biomaterials. METHODS: We first established primary cultures of stromal and epithelial cells from small biopsies of the human bladder using enzymatic digestion and selective cell culture media. Then, a bioengineered substitute of the bladder lamina propria was generated using cultured stromal cells and fibrin-agarose scaffolds, and the epithelial cells were then subcultured on top to generate a complete bladder mucosa substitute. Evaluation of this substitute was carried

Bioactivity of novel carboxymethyl chitosan scaffold incorporating MTA in a tooth model.

AIM: To characterise the bioactivity of a novel carboxymethyl chitosan (CMCS) scaffold with and without incorporating mineral trioxide aggregate (MTA) in a tooth model. METHODOLOGY: Cross-linked CMCS scaffold (CaC) and MTA-coated CaC (CaMT) scaffold were prepared by freeze-drying. The bioactivity of the scaffolds was tested in vitro in four different mineralisation solutions (bulk system) and ex vivo in simulated body fluid (SBF) in the tooth model. After mineralisation, the mineral deposits on the scaffolds were analysed using scanning electron microscopy, energy dispersive X-ray, and induct

Haplotype-Phased Chromosome-Scale Genome Assemblies of Satsuma mandarin Uncover Structural and Transcriptional Divergence Between Parental Haplotypes and Structural Variations in Mutant Lineages.

Citrus, a widely cultivated Rutaceae crop, comprises numerous varieties, among which Satsuma mandarin (Citrus unshiu Marc.) is the most widely cultivated in Japan. In this study, a high-quality, chromosome-scale, haplotype-phased genome assembly of Satsuma mandarin ("Miyagawa wase") was generated using PacBio HiFi sequencing, Omni-C scaffolding, and a trio binning strategy incorporating its parental genomes (Kishu mandarin (C. kinokuni) and Kunenbo mandarin (C. nobilis)). This approach enabled the complete separation of parental haplotypes. To obtain accurate gene models, we performed long-rea

Contemporary Applications of Platelet-Rich Fibrin in Mandibular Surgery With Implications for Mandibular Fracture Management.

Mandibular surgery encompasses a wide range of procedures, including the management of traumatic fractures and elective osteotomies, where optimal bone healing and soft-tissue regeneration are essential for successful clinical outcomes. Platelet-rich fibrin (PRF), a second-generation autologous platelet concentrate, has gained increasing attention because of its ability to provide a fibrin scaffold enriched with platelets, leukocytes, cytokines, and growth factors that promote angiogenesis, osteogenesis, and soft-tissue regeneration. This narrative review summarizes the current evidence regard

Repetitive Compressive Loading Downregulates the Expression of Autophagy-Related Factors, Autophagy Capacity and Cellular Activity in Human Osteoarthritic Chondrocytes.

Mechanical stress is thought to be involved in the pathogenesis and pathophysiology of osteoarthritis (OA). However, much remains to be elucidated regarding how chondrocytes sense and respond to mechanical stress (stress sensing and response factors). Additionally, it still remains unclear whether there are defensive responses and mechanisms to protect against pathological agents and mechanical stress in articular cartilage tissue. This study was designed to determine whether repetitive mechanical force, at physiologic levels, affects the expression of factors regulating autophagy such as the

Polyglactin 910 (Vicryl)-Wrapped Implants to Improve Implant Stability: A Preclinical Study.

INTRODUCTION: Use of textured breast implants and tissue expanders has markedly declined due to their association with BIA-ALCL. Alternatives for stabilization such as ADM are costly, and suture tabs for expander anchoring can cause pain and anchor point failure with resultant implant malposition. This study aims to explore an alternative approach using Polyglactin 910 mesh (Vicryl woven mesh) to wrap and stabilize smooth-surface implants. METHODS: Miniature 2-mL breast implants were manufactured using polydimethylsiloxane. Vicryl woven mesh was used to wrap smooth implants, with comparison g

Caveolin Scaffolding Domain (CSD) Peptide LTI-2355 Modulates the Phagocytic and Synthetic Activity of Lung-Derived Myeloid Cells in Idiopathic Pulmonary Fibrosis (IPF) and Post-Acute Sequelae of COVID Fibrosis (PASC-F).

Rationale: The role of the innate immune system in idiopathic pulmonary fibrosis (IPF) remains poorly understood. However, a functional myeloid compartment is required to remove dying cells and cellular debris, as well as to mediate innate immune responses against pathogens. Aberrant macrophage activity has been described in patients with post-acute sequelae of COVID fibrosis (PASC-F), and caveolin scaffolding domain (CSD) peptides have been found to attenuate inflammation and fibrosis in mouse lung injury models. Therefore, we examined, for the first time, the effects of CSD peptide LTI-2355

Interpenetrating polymeric network (IPNs) in ophthalmic drug delivery: Breaking the barriers.

To maintain the therapeutic drug concentration for a prolonged period of time in aqueous and vitreous humor is primary challenge for ophthalmic drug delivery. Majority of the locally administered drug into the eye is lost as to natural reflexes like blinking and lacrimation resulting in the short span of drug residence. Consequently, less than 5% of the applied drug penetrate through the cornea and reaches the intraocular tissues. The major targets for optimal ophthalmic drug delivery are increasing drug residence time in cul-de-sac of the eye, prolonging intraocular exposure, modulating drug

Disease-inducing potential of two leukemic cell lines in a xenografting model.

PURPOSE: Cryopreserved ovarian tissue transplant restores ovarian function in young cancer patients after gonadotoxic treatment. However, leukemia is associated with increased risk of malignant cell transmission. We aimed to assess the tumor-inducing potential of two different leukemic cell lines when xenografted to immunodeficient mice. METHODS: Fifty-four female immunodeficient mice were grafted with either 100, 200, 500, 1000, and 10,000 chronic myeloid leukemia in blast crisis (BV-173) cells or relapsed acute lymphoblastic leukemia (RCH-ACV) cells, embedded inside a fibrin scaffold along

Spleen Organoid Units Generate Functional Human and Mouse Tissue-Engineered Spleen in a Murine Model.

Introduction: Splenectomy is common after trauma or hematologic disease, and alters immune protection against pathogens, which may lead to fulminant infection with high mortality. Yet the spleen has demonstrable regenerative capacity and cells might be recovered and reimplanted at the time of injury or excision to avoid these risks. Methods: Tissue-engineered spleen (TESp) was generated from ActinGFP mice (mTESp) or human donor spleen (hTESp) through implantation of spleen organoid units (spleen OU), in NOD/SCID mice with concurrent splenectomy, on a biodegradable scaffold. Explants were evalu

Behavior and biocompatibility of rabbit bone marrow mesenchymal stem cells with bacterial cellulose membrane.

BACKGROUND: Tissue engineering has been shown to exhibit great potential for the creation of biomaterials capable of developing into functional tissues. Cellular expansion and integration depends on the quality and surface-determinant factors of the scaffold, which are required for successful biological implants. The objective of this research was to characterize and evaluate the in vitro characteristics of rabbit bone marrow mesenchymal stem cells (BM-MSCs) associated with a bacterial cellulose membrane (BCM). We assessed the adhesion, expansion, and integration of the biomaterial as well as

Caveolin-1 protects B6129 mice against Helicobacter pylori gastritis.

Caveolin-1 (Cav1) is a scaffold protein and pathogen receptor in the mucosa of the gastrointestinal tract. Chronic infection of gastric epithelial cells by Helicobacter pylori (H. pylori) is a major risk factor for human gastric cancer (GC) where Cav1 is frequently down-regulated. However, the function of Cav1 in H. pylori infection and pathogenesis of GC remained unknown. We show here that Cav1-deficient mice, infected for 11 months with the CagA-delivery deficient H. pylori strain SS1, developed more severe gastritis and tissue damage, including loss of parietal cells and foveolar hyperplasi

Immune characterization of mesenchymal stem cells in human umbilical cord Wharton's jelly and derived cartilage cells.

Mesenchymal stem cells derived from human umbilical cord Wharton's jelly (hWJMSCs) became prospective seed cell candidate for tissue engineering and cell-based therapy because of its variety source, easy procurement, robust proliferation, and high purity compared with bone marrow- and adipose-derived MSCs. Such neonatal stem cells can be isolated from a variety of extraembryonic tissues and appear to be more primitive and have greater multi-potentiality than their adult counterparts. In this study, we investigated the immune characters of hWJMSCs and its derived cartilage cells (hWJMSC-Cs) by

HPMA-RGD hydrogels seeded with mesenchymal stem cells improve functional outcome in chronic spinal cord injury.

Chronic spinal cord injury (SCI) is characterized by tissue loss and a stable functional deficit. While several experimental therapies have proven to be partly successful for the treatment of acute SCI, treatment of chronic SCI is still challenging. We studied whether we can bridge a chronic spinal cord lesion by implantation of our newly developed hydrogel based on 2-hydroxypropyl methacrylamide, either alone or seeded with mesenchymal stem cells (MSCs), and whether this treatment leads to functional improvement. A balloon-induced compression lesion was performed in adult 2-month-old male Wis

Gene disruption of tissue transglutaminase.

Transglutaminase 2 (TGase 2), or tissue transglutaminase, catalyzes either epsilon-(gamma-glutamyl)lysine or N(1), N(8)-(gamma-glutamyl)spermidine isopeptide bonds. TGase 2 expression has been associated with apoptosis, and it has been proposed that its activation should lead to the irreversible assembly of a cross-linked protein scaffold in dead cells. Thus, TGase 2-catalyzed protein polymerization contributes to the ultrastructural changes typical of dying apoptotic cells; it stabilizes the integrity of the apoptotic cells, preventing the release of harmful intracellular components into the

Dentin-Pulp Complex Tissue Regeneration via Three-Dimensional Cell Sheet Layering.

The dentin-pulp complex is a unique structure in teeth that contains both hard and soft tissues. Generally, deep caries and trauma cause damage to the dentin-pulp complex, and if left untreated, this damage will progress to irreversible pulpitis. The aim of this study was to fabricate a layered cell sheet composed of rat dental pulp (DP) cells and odontogenic differentiation of pulp (OD) cells and to investigate the ability to regenerate the dentin-pulp complex in a scaffold tooth. We fabricated two single cell sheets composed of DP cells (DP cell sheet) or OD cells (OD cell sheet) and a layer

MSC Origin and Biomechanical Conditioning Determine ECM Maturation in Tissue-Engineered Matrix.

Background: The extracellular matrix (ECM) plays a central role in the mechanical strength and functional integration of tissue-engineered matrix (TEM), particularly in cardiovascular and load-bearing applications. Mesenchymal stromal cells (MSCs) from different sources may vary in their ECM-forming potential. Methods: In this study, adipose-derived (hADMSC), bone marrow-derived (hBMSC), and umbilical cord-derived MSCs (hUCMSC) were compared with human dermal fibroblasts (HDFBs) as a reference. Cells were seeded onto polyglycolic acid (PGA)/poly-4-hydroxybutyrate (P4HB) scaffolds and cultured
