Prenatal diagnosis in 3,000 women for chromosome, X-linked, and metabolic disorders.

In 3,000 women referred for prenatal diagnosis, 110(3.7%) abnormal fetuses were detected and 85 therapeutic terminations were performed. These were five main reasons for referral. Among the 2,227 women referred because of maternal age 35 years and older, there were 51 (2.3%) who had aneuploid fetuses. In the 297 women referred because of a previous child with Down syndrome, 3 aneuploid fetuses (1.0%) were detected. Of the 55 couples where one spouse was a carrier of a balanced chromosome rearrangement, 6 chromosomally abnormal fetuses were found (10.9%) (all the offspring of maternal carriers)

Rapid, reliable, comprehensive and sensitive detection of MDCC in AML by an optimized panel of 6 FISH probes.

CPX-351 is approved for myelodysplasia related changes (MRC)-type AML which can be classified by MDS-defining cytogenetic changes (MDCC). According to WHO 2022 this entity is renamed AML-MR (myelodysplasia-related) and molecular features are considered too. In a prospective study we aimed to prove that a rapid identification of AML-MRC/MR cases with MDCC is practicable and reliable using a streamlined 6-probes FISH-panel. We assessed feasibility, success rate, sensitivity to detect cytogenetic aberrations, especially MDCC, turnaround times (TAT), and reliability of peripheral blood (pb) vs. bo

Decoding Genetic Risk Factors in Recurrent Pregnancy Loss: An Integrative Chromosomal and Bioinformatics Approach.

Recurrent pregnancy loss (RPL) is characterized by two or more consecutive pregnancy losses, often associated with genetic, immunological, endocrine, and anatomical abnormalities. Among these, chromosomal abnormalities, including aneuploidies and submicroscopic copy number variations (CNVs), play a critical role in adverse pregnancy outcomes. A total of 125 fetal specimens were collected, of which 118 were included after applying predefined exclusion criteria. DNA isolated from products of conception and fetal tissues was subjected to quantitative fluorescent PCR (QF-PCR) for rapid aneuploidy

Tongue strength and endurance in relation to oral cavity morphology among children with Down syndrome in the permanent dentition period.

CONTEXT: Down syndrome is a genetic condition caused by trisomy 21, often accompanied by orofacial hypotonia and anomalies in stomatognathic development. A hypotonic tongue may alter oral cavity morphology, particularly palatal depth, anterior dental relationships, and lip competence, thereby affecting mastication, swallowing, and speech. AIMS: To evaluate the association between tongue strength and endurance with oral cavity morphology in children with Down syndrome during the permanent dentition period. SETTINGS AND DESIGN: A cross-sectional analytic study was conducted among children with

Chronic lymphocytic leukemia with IGH::BCL3-translocation is characterized by a homogeneous and distinct genetic and epigenetic landscape.

Approximately 1% of chronic lymphocytic leukemia (CLL) cases harbor a translocation juxtaposing the immunoglobulin heavy chain (IGH) and B-cell lymphoma 3 (BCL3) loci. Aiming at comprehensive molecular characterization of IGH::BCL3-positive B-cell neoplasms, we here investigated samples from 84 patients using fluorescence in situ hybridization (FISH), whole-genome and targeted sequencing, and DNA methylation analyses. Junctional sequences obtained in 27 patients showed breakpoints upstream of BCL3 in all CLL cases. IGH breaks were presumably driven by aberrant class-switch recombination in 26/

Integrated multiomic profiling reveals 2 distinct splenic marginal zone lymphoma subgroups with prognostic relevance.

Splenic marginal zone lymphoma (SMZL) is a rare B-cell malignancy with notable genetic, epigenetic, and clinical heterogeneity. In this study, we used coding and noncoding sequencing (n = 74), including whole-genome sequencing (WGS) of 24 paired tumor-normal samples, targeted sequencing (n = 55), and DNA methylation in 126 patients to characterize the disease. From WGS, we identified recurrent, predominantly clonal coding mutations in KLF2 (50%), KMT2D (25%), and NOTCH2 (25%), alongside rare mutations in FLNC (8%), novel mutations in FAM135B (17%), and noncoding mutational hot spots in BCL6, P

A novel clustering of down syndrome and coeliac disease complicated by enteritis and small intestinal strictures: a case series.

Down syndrome (DS) increases risk for autoimmune conditions, including coeliac disease (CeD). Here, we report five adults with DS and biopsy-proven CeD who developed enteritis and symptomatic small bowel stricturing, a clustering not previously described in this population. All five patients had confirmed DS and CeD, and despite apparent adherence to a gluten-free diet, all developed small bowel strictures requiring endoscopic intervention, most commonly at the duodenal D1/D2 junction. Stricture histology demonstrated active CeD with varying degrees of inflammation and fibrosis. Three patients

A Nomogram of Fetal Nasal Bone Length at 16-24 Weeks of Gestation in North Indian Pregnant Women: A Prospective Study.

Introduction Fetal nasal bone length (NBL) is a crucial sonographic marker in prenatal screening for chromosomal abnormalities, particularly trisomy 21. However, its diagnostic accuracy is influenced by ethnic variations, and normative data for North Indian populations remain scarce. Objective This study aimed to establish a gestational age-specific nomogram for fetal NBL between 16+1 and 24 weeks of gestation in North Indian pregnant females, and to evaluate its correlation with standard fetal biometric parameters. Methods A prospective observational study was conducted on 438 structurally an

Clinical and Molecular Presentation of a Patient with Paternal Uniparental Isodisomy of Chromosome 16.

Uniparental disomies (UPDs) are among the causes of imprinting disorders. Specific phenotypes of most causative UPDs have been described. Here, we describe the case of a 2-year-old female patient who presented a syndromic phenotype. Chromosomal microarray analysis revealed UPD of the whole chromosome 16. Microsatellite analysis demonstrated paternal origin of the UPD and its isodisomic pattern (UPiD (16) pat). Mosaic trisomy 16 was not detected using the FISH method. Whole-exome sequencing revealed no pathogenetic genetic variants sufficient to explain the syndromic phenotype nor unmasked path
