Detection of Babesia bovis carrier cattle by using polymerase chain reaction amplification of parasite DNA.

Carrier cattle infected with Babesia bovis are difficult to detect because of the low numbers of parasites that occur in peripheral blood. However, diagnosis of low-level infections with the parasite is important for evaluating the efficacies of vaccines and in transmission and epidemiological studies. We used the polymerase chain reaction (PCR) to amplify a portion of the apocytochrome b gene from the parasite and tested the ability of this method to detect carrier cattle. The target sequence is associated with a 7.4-kb DNA element in undigested B. bovis genomic DNA (as shown previously), and

Antigenic specificity of the 72-kilodalton major surface glycoprotein of Leishmania braziliensis braziliensis.

We examined the expression and the antigenicity of the major surface polypeptides of Leishmania braziliensis braziliensis and Leishmania donovani chagasi, parasites which commonly coexist in the same endemic areas of Bolivia. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis protein profiles from surface-iodinated promastigotes showed the presence of a unique iodinatable polypeptide of 72 kDa on the L. b. braziliensis surface and of two major components of 65 and 50 kDa exposed at the surface of L. d. chagasi. Comparison of the peptide digestion profiles of the major iodinated polypept

Immunological consequences of infection and vaccination in South American trypanosomiasis.

Trypanosoma cruzi infection provokes a vigorous immune response that terminates the parasitaemia associated with the acute stage within two to three months of initial infection. Even so, a variable proportion of patients may develop severe Chagas' disease, often decades after initial infection. Recent experimental findings suggest that trypomastigotes of T. cruzi possess a surface bound neuraminidase and sugar binding protein by means of which they invade host cells--a mechanism very reminiscent of influenza virus. Studies of the antibody response to trypomastigotes in patients or murine model

Susceptibility to African trypanosomiasis of N'Dama and Zebu cattle in an area of Glossina morsitans submorsitans challenge.

The use of trypanotolerant livestock is considered to be an important strategy for the control of African animal trypanosomiasis. In order to define the extent of the differences in susceptibility and productivity, 10 Zebu cows (a breed considered trypanosusceptible) and 10 N'Dama cows (a breed recognised for trypanotolerance) were exposed to a natural field challenge from Glossina morsitans submorsitans Newstead. The animals were two-and-a-half to three years old and had not been previously exposed to trypanosomiasis. All Zebu died of trypanosomiasis within eight months of first exposure. In

Extraction-free LAMP-LFD assay for early diagnosis of congenital Chagas disease directly from neonatal dried blood spots.

BACKGROUND: Vertical transmission is a major driver of new Chagas disease cases. Early diagnosis in newborns is essential for effective treatment; however, conventional screening limitations and high patient attrition lead to severe under-registration. Simplified molecular tools can substantially improve early detection and treatment access. METHODOLOGY/PRINCIPAL FINDINGS: We developed a simplified loop-mediated isothermal amplification (LAMP)-based assay targeting the repetitive satellite DNA of Trypanosoma cruzi for qualitative detection in newborn dried blood spots (DBS). Tailored for reso

Ultrastructure of the wing-coupling apparatus in seven Triatominae species (Hemiptera: Reduviidae) from North America.

Triatomines are the primary vectors of Trypanosoma cruzi (Chagas), the causative organism of Chagas disease, one of the most important vector-borne diseases in Mexico. Triatomine dispersal encompasses a broad spectrum of flight capacities, ranging from short-distance, low-altitude flights to active, sustained dispersal in certain species. Although recent studies have examined the phenotypic and genetic variability of flight-related traits, the specific morphological constraints hindering their aerodynamic efficiency remain poorly understood. Therefore, this study aimed to provide a comparative

Mapping tsetse fly connectivity in Uganda with machine learning landscape genetics.

INTRODUCTION: Tsetse flies (genus Glossina ) are biting insects that transmit human and animal trypanosomiases across sub-Saharan Africa, and sustainable vector control depends on understanding dispersal barriers and reinvasion routes. Despite major progress toward elimination, Uganda remains at risk for both human forms of the disease ( Trypanosoma brucei gambiense and T. b. rhodesiense ) and planners still lack reliable maps of tsetse movement and reinvasion risk. METHODS AND RESULTS: We address this gap with machine-learning landscape genetics and species distribution models, integrating e

Left Atrial Thrombus and Cardioembolic Stroke in Chagas Cardiomyopathy Presenting with Atrial Flutter: A Case Report.

Background: Chagas disease, caused by Trypanosoma cruzi, remains endemic throughout Latin America but is increasingly reported in urban areas due to migration and vector adaptation. The cardiac form is the most severe manifestation, associated with arrhythmia, mural thrombus formation, and a high risk of cardioembolic events. Stroke secondary to Chagas cardiomyopathy is uncommon and poses diagnostic and therapeutic challenges. Case Presentation: A 58-year-old woman with serologic evidence of T. cruzi infection presented with sudden-onset dyspnea, oppressive chest pain, and left-sided weakness.

Glossina from the Republic of the Congo: species identification by MALDI-TOF MS and research of associated micro-organisms.

Human African trypanosomiasis (HAT) and Animal African trypanosomiasis (AAT), transmitted by Glossina species, remain major health and economic burdens in Africa. Accurate vector identification is essential for effective control strategies. However, current identification methods of Glossina species based on morphological and/or molecular techniques have several limitations that often hinder reliable species-level classification. This study assessed matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) as an alternative or complementary approach to morphol

Evaluation of Quality of Care in Chagas Disease Cardiomyopathy.

BACKGROUND: Chagas disease (ChD), a neglected tropical disease caused by Trypanosoma cruzi, affects around 7.5 to 10 million people globally, primarily in Latin America. Chronic Chagas cardiomyopathy (CCM) is the most severe clinical form, leading to substantial cardiovascular morbidity and mortality. Despite existing guidelines, fragmented health systems, low provider awareness, and limited access to care hinder effective disease management. OBJECTIVES: We aimed to define the key components of the CCM quality of care (structure, process, and outcomes) for main clinical activities at the thre

Germline-targeted baboon apolipoprotein L-1 protects mice against African trypanosomes.

Some primates are immune to infection by most African trypanosome parasites due to apolipoprotein L-1 (APOL1), a primate-specific ion channel-forming protein. Our long-term objective has been to reduce African trypanosomiasis in livestock by genetic bioengineering of cattle with primate APOL1. To select which primate APOL1, we analyzed Papio ssp. APOL1 proteins and found that Papio hamadryas APOL1 was a strong candidate for transgenic animal production based on its trypanosome-killing capacity, ion channel properties, and stability. We generated seven transgenic murine lines based on the P. ha

Towards new care strategies in a suburban healthcare center in La Matanza, Argentina: performance evaluation of rapid diagnostic tests for Chagas disease in a non-endemic area.

INTRODUCTION: Chagas disease, caused by the protozoan parasite Trypanosoma cruzi, remains a major public health problem, affecting approximately seven million people worldwide. Although historically restricted to endemic regions, migration has led to its global distribution, increasing the relevance of congenital transmission in non-endemic areas. If not timely diagnosed and treated, infection can progress to chronic Chagas cardiomyopathy in nearly 30 % of patients, generating substantial social and healthcare burdens. Diagnosis requires concordant results from two or three serological assays

First whole-genome sequence of Triatoma sanguisuga (Le Conte, 1855), vector of Chagas disease.

Triatoma sanguisuga is the most widespread triatomine bug species in the United States. The species vectors the human parasite Trypanosoma cruzi, which causes Chagas disease. Vector-borne Chagas disease is rarely diagnosed in the United States, but T. sanguisuga has been implicated in a handful of cases. Despite its public health importance, little is known about the genomics or population genetics of T. sanguisuga. Here, we used long-read sequencing to assemble the first whole-genome sequence for T. sanguisuga using DNA extracted from 1 adult specimen from Delaware. The final size of the geno

Mass balance, pharmacokinetics, metabolism, and excretion of radiolabeled acoziborole, a potential novel treatment for human African trypanosomiasis, following single microtracer oral dose to humans.

Acoziborole is an oxaborole 6-carboxamide active against Trypanosoma brucei gambiense and rhodesiense, the parasites responsible for human African trypanosomiasis. This open-label, phase I study in six healthy male participants evaluated the mass balance, pharmacokinetics, metabolism pathways, and excretion of a single oral 960 mg dose of [14C]-acoziborole. Blood, plasma, and feces were collected for 120 days and urine for 16 days. The excretion balance and systemic exposure of total circulating radioactivity were determined using accelerator mass spectrometry. Metabolism profiling was perform

Development of chimeric multivalent proteins for serological diagnosis of African animal trypanosomosis.

The control of African animal trypanosomosis (AAT) relies on accurate diagnostic tools. Serological diagnosis using ELISA is well-suited for surveillance due to its high-throughput capacity, low cost, and adaptability to rapid formats. However, the WOAH-recommended antibody ELISA for AAT, based on trypanosome lysates purified from rodent blood, can lack specificity and presents standardization challenges as well as ethical concerns. Recombinant proteins offer a solution to standardization, often improving specificity, though potentially at the expense of sensitivity. Combining multiple recombi

Severity-dependent IgG epitope profiling in COVID-19 reveals differential recognition of pathogen-derived antigens.

BACKGROUND: The contribution of antibody-mediated responses to COVID - 19 outcomes remains unclear, particularly regarding cross-reactivity with unrelated pathogens. While co-infections are known to influence disease progression, the broader landscape of IgG reactivity during SARS-CoV-2 infection has not been systematically explored. METHODS: We employed a high-density peptide microarray containing 4,344 linear epitopes from 37 viruses, 27 bacteria, 17 parasites, and 8 fungi to characterize serum IgG repertoires from individuals with moderate (n = 39) or severe (n = 40) COVID - 19.
