Modular Platform for Recombinant Production of Highly Toxic Bacterial Proteins.

Bacterial protein toxins constitute a vast, largely untapped reservoir of potent bioactive molecules for antimicrobial discovery and biotechnology. However, their intrinsic toxicity often prevents recombinant expression in microbial hosts, limiting biochemical and functional characterization. Here, we present a modular platform for the recombinant production of highly toxic bacterial proteins based on covalent fusion of polymorphic toxins (PTs) to their cognate immunity proteins (PIMs), enabling transient toxin neutralization during expression. This strategy enabled recombinant production and

Evaluating the effect of griseofulvin on the disintegration properties of tablets.

Understanding how active pharmaceutical ingredients (APIs) influence tablet disintegration remains essential for optimising oral drug product performance. In this work, we present an integrated, data-driven framework that combines deep-learning-based image segmentation, automated regime detection and mechanistic modelling to characterise liquid absorption and swelling behaviours from sessile drop experiments. Using this workflow, we investigated the effect of incorporating griseofulvin into directly compressed tablets containing a variety of excipient combinations. Across all formulations, the

Advancing Global Health Initiative Through Pediatric Vitamin A Supplementation.

Background: Malnutrition, particularly vitamin A deficiency, affects approximately 49% of children aged 6-59 months in sub-Saharan Africa, increasing the risks of morbidity, mortality, poor growth, and preventable blindness. Tanzania has followed the World Health Organization's (WHO) guidelines for biannual vitamin A supplementation (VAS) since 2004, but rural access remains limited. A preliminary assessment at a rural Tanzanian day care showed only 29% VAS coverage, indicating a substantial gap in coverage. Objective: To improve nutritional health among children aged 6-59 months through a com

Evaluation of radiation dose reduction effect of a foldable attached lead curtain in portable X-ray examinations in the neonatal intensive care unit: A Monte Carlo simulation study.

Mobile X-ray examinations in neonatal intensive care units (NICUs) are essential for the critically ill neonates. However, scattered radiation generated during imaging can lead to unnecessary radiation exposure for adjacent neonates and healthcare workers. Conventional mobile shielding devices are often challenging to implement effectively in NICU environments due to limited space and workflow constraints. Consequently, this study developed a foldable lead curtain that can be attached to a mobile X-ray unit and evaluated experimental measurements and Monte Carlo simulations. The lead curtain f

A hybrid framework for water-quality prediction using WOA-optimized EEMD and CNN-LSTM.

Accurate prediction of dissolved oxygen is important for water-quality management, yet monitoring series are usually noisy, nonlinear, and non-stationary, which limits conventional forecasting models. This study proposes a WOA-EEMD-CNN-LSTM framework in which EEMD is used to decompose the original series into intrinsic mode functions and a residual component for noise reduction and multi-scale representation, WOA is used to optimize the key EEMD parameters and improve decomposition quality, and CNN-LSTM is used to capture local features and long-term temporal dependencies for component-wise pr

Automatic recognition of Pd ions in high-resolution mass spectra of multicomponent samples without visible fine isotope structures.

Palladium plays a three-fold role in modern science and technology: it serves as a key active center in modern catalytic synthetic methods, occurs as unwanted trace contamination that influences new catalysts development and can lead to false-positive reports, and is an emerging environmental pollutant from automotive catalysts. High activity at ppm/ppb loadings and the necessity to analyze sub-micromolar concentrations of samples are common challenges in these applications, making reliable Pd identification essential. Mass spectrometry (MS) can detect Pd-containing ions in complex mixtures, b

"Precision planning for tissue expansion: A three-dimensional scanning-based predictive model to guide surgical timing".

Tissue expansion remains an essential reconstructive technique, yet the timing of expander removal is still largely determined by subjective clinical judgment. We developed a three-dimensional (3D) scanning-based quantitative workflow to objectively monitor expansion progress and provide a practical reference for surgical timing. In this study, serial 3D scans obtained throughout the expansion course were used to evaluate the relationship between the cumulative injected volume and incremental expansion area relative to baseline in a development cohort of 30 patients with congenital melanocytic

Leveraging high-throughput proteomics and AI-based protein folding to accelerate VAV1 molecular glue discovery.

This study reports a series of CRBN molecular glues that selectively induce proteasomal degradation of VAV1, a hematopoietic-specific signaling protein and key target in hematological malignancies and autoimmune diseases. Using unbiased global proteomics, we identify phenyl-glutarimide derivatives (e.g., NGT-201-12) as effective VAV1 degraders that engage its C-terminal SH3 domain (SH3-2). We elucidate a non-canonical RT-loop degron (RDxS motif, residues 796-799) in SH3-2, distinct from previously characterized G-loop degrons, supported by the physics- and AI-driven GluePlex workflow and site-

A Standardized In Vitro Protocol for Evaluating Human Dental Pulp Stem Cell Differentiation and Proliferation on Nanomaterial Scaffolds.

Reliable assessment of osteogenic and odontogenic differentiation, as well as stem cell proliferation, is fundamental to evaluating the biological performance of scaffold-based regenerative strategies. However, variability in experimental protocols, assay selection, and data interpretation often limits reproducibility and inter-study comparison. The protocol described in this article presents a standardized, reproducible workflow for the in vitro evaluation of osteogenic and odontogenic differentiation, as well as proliferative capacity, of stem cells cultured on biomaterial scaffolds. Controlled knowledge updating in memory-enabled AI agents: beyond model editing and recall.

Large language model agents that persist across sessions, tools, users, and changing environments do more than answer isolated prompts; they accumulate state. When new evidence arrives, the central question is where that change should live: transient context, external memory, tool or workflow definitions, activation states, or model parameters. Recent benchmarks compare update mechanisms and assess memory over time, while emerging architectures coordinate multiple memory types. In this Perspective, we examine how a decision layer should select among update substrates and evaluate whether its c

AI in Neuro-Oncology: An Overarching Review.

Artificial intelligence (AI) is increasingly being applied across neuro-oncology domains to analyze complex, high-dimensional, multimodal data. Despite major advances in the biological understanding and classification of CNS tumors, clinical outcomes for many of them remain poor, reflecting intra- and inter-tumor heterogeneity and limitations of conventional reductionist approaches. AI offers a paradigm shift to capture this heterogeneity and provide quantitative, reproducible diagnostic, predictive, and prognostic insights. This article describes a joint effort across the Response Assessment

In Silico to In Cerebro-Development of the Orexin Receptor Antagonist "Photorexant" for Photomodulation In Vitro and in Mouse Brains.

In this study, we report the first photoswitchable small molecule dual ligands for the orexin receptor subtypes 1 (OX1R) and 2 (OX2R) as molecular tools for optical control of orexin signaling. The orexin system is critical for several physiological processes and has been increasingly implicated in the pathogenesis of various psychiatric disorders. We implemented a prospective structure-based design approach, generating azobenzene-containing derivatives of the FDA-approved dual orexin receptor antagonist suvorexant with a computational workflow that featured a novel fragmentation and azobenzen

Reclassification of Noncanonical MET Splice-Site Variants Using RNA-Based Validation: A Practical Diagnostic Approach in Non-Small Cell Lung Cancer.

MET exon 14 (METex14) skipping alterations represent actionable oncogenic drivers in non-small cell lung cancer (NSCLC). However, noncanonical intronic splice-site variants are frequently classified as variants of uncertain significance (VUS) on DNA-based next-generation sequencing (NGS), limiting timely therapeutic decision making. We describe two patients with advanced NSCLC harboring novel intronic deletions affecting the MET exon 14 splice acceptor region, initially classified as VUS. Integrated transcript level analysis using RNA sequencing and orthogonal validation by reverse transcripti

Chemistry-informed Bayesian control and policy distillation for autonomous pH adjustment in diverse chemical systems.

Adjusting pH of an arbitrary solution to a specified target remains difficult to automate when buffering behavior is unknown, nonlinear, and composition-dependent. We formulate pH adjustment as a sequential inference-and-control problem and develop a chemistry-informed Particle Filtering (PF) Bayesian controller that updates an explicit dose-pH response model. Across five independently generated benchmark sets of 3000 simulation tasks, the PF controller achieved 95.4 ± 0.6% success, requiring 4.84 ± 0.09 additions among successful tasks. PF state-action trajectories were subsequently distilled

Alginate-Methacrylate-RGD MicroSpheres (Alg-MR-MS) for cell expansion and delivery in tissue engineering.

Clinically relevant cell numbers are essential for many tissue engineering applications, requiring scalable expansion platforms. Microcarrier-based bioreactor systems address this demand; however, conventional non-dissolvable carriers require harsh enzymatic or mechanical detachment, reducing cell viability and integrity. Although dissolvable microcarriers improve recovery, most are designed only for harvesting and lack functionality for downstream tissue engineering. Here, we present a sequential dual-crosslinkable alginatemethacrylate-RGD microsphere system (Alg-MR-MS) that integrates scalab

Precision Injection-Compression Molding and Functional Integration of Hollow Microneedle Microfluidic Chips for Minimally Invasive Continuous Health Monitoring.

Real-time biomarker monitoring through interstitial fluid (ISF) sampling using hollow microneedle (HMN) arrays is crucial for personalized precision medicine and chronic disease management. However, clinical translation faces a critical bottleneck: the absence of stable, cost-effective, scalable high-precision manufacturing technologies. This work systematically establishes a comprehensive end-to-end solution encompassing precise molding and functional integration. We employed injection-compression molding with multi-objective optimization algorithms to elucidate relationships between critical

A reproducible entropy-weighted Gower-CLARA-TOPSIS procedure for mixed-type MSME segmentation and prioritization.

In this paper a reproducible approach for segmentation of mixed-type data and the within-cluster prioritizing is presented. The procedure keeps raw categorical values for Gower dissimilarity, binary or count representations only for entropy weighting, applies CLARA to repeated samples with PAM medoids, and evaluates weighted and Top-N feature configurations with the Silhouette Coefficient, medoid-based Davies-Bouldin Index, and Calinski-Harabasz Index. The chosen configuration is then utilized to rank within clusters, instead of between clusters using TOPSIS to evaluate the economic-operationa

A multitask framework for automated multi-frame right upper quadrant ultrasound interpretation and clinical decision support.

Ultrasound is a cornerstone of emergency and hepatobiliary imaging, yet its interpretation remains highly operator-dependent and time-sensitive. Here, we present a multitask vision-language agent (VLM) developed to assist with right upper quadrant (RUQ) ultrasound interpretation across the diagnostic workflow. The system was trained on data from a single tertiary academic medical center (9189 cases, 594,099 images) and evaluated on datasets from two independent institutions (1704 cases, 90,616 images; and 108 cases, 3240 images). Femur-First Cut Kinematically Aligned Unicompartmental Knee Arthroplasty: A Seamless Surgical Technique from UKA to TKA.

Kinematic alignment (KA) is based on a resurfacing philosophy that restores the patient's native joint line orientation and preserves physiological soft-tissue balance. In calipered KA total knee arthroplasty, reconstruction of the distal femoral articular surface is the first and most critical step because it establishes the cylindrical axis that governs knee kinematics. Although unicompartmental knee arthroplasty (UKA) shares the same resurfacing philosophy, most currently available techniques begin with tibial resection, making femoral component positioning dependent on tibial alignment, ga

Injection-free, spatially controlled delivery of glioblastoma cells to human brain slice cultures using soft cryogel biomaterials.

Organotypic brain slice culture has emerged as a valuable ex vivo system for studying brain development, function, and disease in a multicellular tissue context. More recently, this has extended to the use of human tissue derived from fetal, post-mortem, or discarded surgical specimens. In glioblastoma research, focal delivery of exogenous cells to brain slices is often technically challenging, particularly when spatial control is required. Here, we present a simple and experimentally accessible method for localized cell delivery to human brain slice cultures using soft donut-shaped cryogel bi
