Creation of two tomographic voxel models of paediatric patients in the first year of life.

Tomographic computational models, based on regional segmentation of CT or MRI medical images, have increasingly been proposed as replacements for current stylized mathematical models of human anatomy used in radiation dosimetry studies. While much effort has been devoted towards the creation of adult models, few research studies have been initiated to address the need for models supporting paediatric radiology dosimetry. In this study, two tomographic models were created using a combination of automatic and manual segmentation via a program created in-house using IDL version 5.5. The first mod

Fate map of the chicken neural plate at stage 4.

A detailed fate map was obtained for the early chick neural plate (stages 3d/4). Numerous overlapping plug grafts were performed upon New-cultured chick embryos, using fixable carboxyfluorescein diacetate succinimidyl ester to label donor chick tissue. The specimens were harvested 24 hours after grafting and reached in most cases stages 9-11 (early neural tube). The label was detected immunocytochemically in wholemounts, and cross-sections were later obtained. The positions of the graft-derived cells were classified first into sets of purely neural, purely non-neural and mixed grafts. Comparis

Medical evaluation of overweight children and adolescents: reports from pediatricians, pediatric nurse practitioners, and registered dietitians.

OBJECTIVE: The primary aim of this study was to determine how pediatric health care providers identify overweight in children and adolescents and how they evaluate obesity-related medical complications. This information can guide development of programs to help providers improve their evaluation practices. A secondary objective was to examine the association of certain provider characteristics with recommended evaluation practices. METHODS: A random sample of pediatricians, pediatric nurse practitioners (PNPs), and registered dietitians received a questionnaire about their evaluation of overw

Peptides from the PKD repeats of polycystin, the PKD1 gene product, modulate pattern formation in the developing kidney.

Mutations in the PKD1 gene cause the majority of cases of autosomal dominant polycystic kidney disease. The PKD1 gene codes for a protein of unknown function, polycystin-1, that is predicted to be a receptor. Its large extracellular domain contains 16 copies of novel motif, the PKD repeat, that is likely to be a ligand binding domain based on its similarity to immunoglobulin domains. These observations suggested that soluble fragments of the extracellular domain of polycystin-1 could be used as competitive inhibitors of polycystin function in a suitable model system. Polycystin-1 is highly exp

Torso signalling regulates terminal patterning in Drosophila by antagonising Groucho-mediated repression.

Patterning of the non-segmental termini of the Drosophila embryo depends on signalling via the Torso receptor tyrosine kinase (RTK). Activation of Torso at the poles of the embryo triggers restricted expression of the zygotic gap genes tailless (tll) and huckebein (hkb). In this paper, we show that the Groucho (Gro) corepressor acts in this process to confine terminal gap gene expression to the embryonic termini. Embryos lacking maternal gro activity display ectopic tll and hkb transcription; the former leads, in turn, to lack of abdominal expression of the Krüppel and knirps gap genes. We sho

Microbiome varies by body site in the yellow mud turtle, Kinosternon flavescens, from the Chihuahuan Desert.

Microbiomes are important to the ecology and evolution of their associated animal hosts. These important functions range broadly from individual animal health to population-level adaptations. Turtle-associated microbiomes are under increasing investigation, given turtles' conservation status and the unique natural history of the shell as an evolutionary-developmental novelty. Many components of the turtle-microbiome interaction remain understudied, including how microbial communities assemble based on host and environmental factors. Here, we hypothesized that age, habitats, and body sites woul

Mandibular biomechanics rehabilitated with different prosthetic restorations under normal and impact loading scenarios.

BACKGROUND: Restorative treatment options for edentulous patients range from traditional dentures to fixed restorations. The proper selection of materials greatly influences the longevity and stability of fixed restorations. Most prosthetic parts are frequently fabricated from titanium. Ceramics (e.g. zirconia) and polymers (e.g. PEEK and BIOHPP) have recently been included in these fabrications. The mandibular movement produces complex patterns of stress and strain. Mandibular fractures may result from these stresses and strains exceeding the critical limits because of the impact force from f

An interaction between fetal sex and placental weight and efficiency predicts intrauterine growth in response to maternal protein insufficiency and gestational exposure window in a mouse model of FASD.

BACKGROUND: Individuals exposed to gestational stressors such as alcohol exhibit a spectrum of growth patterns, suggesting individualized responses to the stressors. We hypothesized that intrauterine growth responses to gestational alcohol are modified not only by the stressor's severity but by fetal sex and the placenta's adaptive capacity. METHODS: Pregnant C57BL/6J mice were assigned to one of three groups. Group 1 consumed a normal protein diet (18% protein by weight) and received 4.5 g alcohol/kg body weight (NP-Alc-8) or isocaloric maltodextrin (NP-MD-8) daily from embryonic day (E) 8.5

Combination of oxymatrine and diammonium glycyrrhizinate significantly mitigates mice allergic contact dermatitis induced by dinitrofluorobenzene.

UNLABELLED: This study investigated the safety and effect of oxymatrine (OMT) and/or diammonium glycyrrhizinate (DG) on allergic contact dermatitis (ACD) induced by 1-fluoro-2,4-dinitrofluorobenzene (DNFB) in ICR mice. Mice were topically smeared with vehicle (control) or DNFB on their ear and skin to induce ACD. The mice were randomized and injected with saline as the model, treated intraperitoneally with dexamethasone (DEX), 45 or 90 mg·kg−1 OMT and/or DG daily beginning one day post the first smearing for two weeks. The body weights, the severity of ear and skin inflammation, the levels of

Dementia with Lewy bodies and Parkinson's disease-dementia: current concepts and controversies.

Dementia with Lewy bodies (DLB) and Parkinson's disease-dementia (PDD), although sharing many clinical, neurochemical and morphological features, according to DSM-5, are two entities of major neurocognitive disorders with Lewy bodies of unknown etiology. Despite considerable clinical overlap, their diagnosis is based on an arbitrary distinction between the time of onset of motor and cognitive symptoms: dementia often preceding parkinsonism in DLB and onset of cognitive impairment after onset of motor symptoms in PDD. Both are characterized morphologically by widespread cortical and subcortical

Propagation of alpha-synuclein pathology: hypotheses, discoveries, and yet unresolved questions from experimental and human brain studies.

Progressive aggregation of alpha-synuclein (αS) through formation of amorphous pale bodies to mature Lewy bodies or in neuronal processes as Lewy neurites may be the consequence of conformational protein changes and accumulations, which structurally represents "molecular template". Focal initiation and subsequent spread along anatomically connected structures embody "structural template". To investigate the hypothesis that both processes might be closely associated and involved in the progression of αS pathology, which can be observed in human brains, αS amyloidogenic precursors termed "seeds"

Folinic acid protects against suppression of growth by methotrexate in mice.

The objective of this study was to investigate whether folinic acid supplementation would protect young mice against suppression of growth by methotrexate (MTX). Four equal groups of Balb/c young male mice (5 animals in each group; mean+/-SD body weight 9.64+/-0.85 g, in their rapid growth phase) were subjected to the following drug treatment: One group was given MTX (3.5 mg/kg body weight) intraperitoneally on every 2nd day, another received folinic acid (7.0 mg/kg body weight) intraperitoneally every 2nd day. The third group was given both of these drugs (MTX on every 2nd day and folinic aci

Distribution of IGF-I and -II, IGF binding proteins (IGFBPs) and IGFBP mRNA in ocular fluids and tissues: potential sites of synthesis of IGFBPs in aqueous and vitreous.

Levels of insulin-like growth factor-I and II (IGF-I and IGF-II) in bovine aqueous humor are twice those found in the vitreous (aqueal IGF-I = 0.62 nM, vitreal IGF-I = 0.30 nM; aqueal IGF-II = 0.028 nM, vitreal IGF-II = 0.017 nM). IGF-I and II binding assays and IGF-II Western ligand blots indicate that aqueous and vitreous humor have equal overall levels of binding (binding assays, mean +/- S.E.M. bound/free per 50 microliters of fluid: vitreal IGF-II = 7.28 +/- 1.6, IGF-I = 0.3 +/- 0.078; aqueal IGF-II = 7.21 +/- 0.072; IGF-I = 0.3 +/- 0.078). Body fat, menarche, fitness and fertility.

Many well-trained athletes, ballet dancers and women who diet excessively have secondary or primary amenorrhoea. Less extensive training or weight loss may result in anovulatory menstrual cycles, or a shortened luteal phase. These disruptions of reproductive ability are due to hypothalamic dysfunction, which is correlated with weight loss or excessive leanness. It is proposed that these associations are causal and that the high percentage of body fat (26-28%) in the mature human female may influence reproduction directly. Four mechanisms are known: (i) adipose tissue converts androgens to oest

Multidimensional body image and self-esteem: a latent profile analysis differentiating orthorexia nervosa and exercise addiction from disordered eating.

BACKGROUND: Orthorexia Nervosa (ON), the obsessive preoccupation with healthy eating, and Exercise Addiction (EA), the behavioral addiction to exercise, are controversially discussed as potential mental disorders. The unclear distinction between these conditions and disordered eating, especially regarding restrictive eating and instrumental exercise behaviors, contributes to this debate. The objective of the study was to ascertain whether latent profiles can be identified that support the independence of ON and EA from disordered eating. It was hypothesized that ON profiles would be characteri

Short- and Long-Term Impact of Exogenous Hormones on Thermoregulation in Women.

OBJECTIVES: How the body changes its methods of thermoregulation as part of the female ovarian cycle is a crucial aspect of human adaptive strategies to negotiate variable environments and tasks. The use of exogenous hormones to manage the ovarian cycle is a regular part of modern life but its long-term impact on thermoregulation, particularly while moving, continues to be understudied. METHODS: Here, we specifically track adults (N = 35) including men, women of known ovarian phases, and women on an estrogen-progesterone combination pill, doing the same walking and resting protocol while thei
