Oral Mucosa Harbors a High Frequency of Endothelial Cells: A Novel Postnatal Cell Source for Angiogenic Regeneration.

Endothelial progenitor cells/endothelial cells (EPCs/ECs) have great potential to treat pathological conditions such as cardiac infarction, muscle ischemia, and bone fractures, but isolation of EPC/ECs from existing cell sources is challenging due to their low EC frequency. We have isolated endothelial progenitor (EP)-like cells from rat oral mucosa and characterized their yield, immunophenotype, growth, and in vivo angiogenic potential. The frequency of EP-like cells derived from oral mucosa is thousands of folds higher than EPCs derived from donor-match bone marrow samples. EP-like cells fro

High dose of TNF-α suppressed osteogenic differentiation of human dental pulp stem cells by activating the Wnt/β-catenin signaling.

Dental pulp stem cells (DPSCs) were a clonogenic, highly proliferative cells capable of self-renewal and multi-lineage differentiation including chondrocytes, adipocytes, neural cells and osteoblasts, which make it an attractive choice for bone regeneration and repair of craniofacial defects. Recent studies showed that tumor necrosis factor α (TNF-α) may affect osteoclastogenesis and bone formation. However, the effect and mechanism of TNF-α on DPSCs is not clear. In this study, we found that low dose TNF-α promoted mineralization and high dose TNF-α suppressed osteogenic differentiation of DP

Stem cell health and tissue regeneration in microgravity.

Exposure to microgravity causes significant mechanical unloading of mammalian tissues, resulting in rapid alterations of their physiology, which poses a significant risk for long-duration manned spaceflight. The immediate degenerative effects of spaceflight we understand best are those studied during short-term low-Earth-orbit experiments, and include rapid microgravity-adaptive bone and muscle loss, loss of cardiovascular capacity, defects in wound and bone fracture healing, and impaired immune function. Over the long-term, exposure to microgravity may cause severe deficits in mammalian stem

TNFR1-mediated signaling is important to induce the improvement of liver fibrosis by bone marrow cell infusion.

The importance of TNF-α signals mediated by tumor necrosis factor receptor type 1 (TNFR1) in inflammation and fibrosis induced by carbon tetrachloride (CCl(4)), and in post-injury liver regeneration including a GFP/CCl(4) model developed as a liver repair model by bone marrow cell (BMC) infusion, was investigated. In mice in which TNFR1 was suppressed by antagonist administration or by knockout, liver fibrosis induced by CCl(4) was significantly decreased. In these mice, intrahepatic macrophage infiltration and TGF-β1 expression were reduced and stellate cell activity was decreased; however, e

SEM analyses of bacterial contamination of e-PTFE membranes and GTR clinical results.

The aim of the present study was to determine whether bacterial contamination of expanded polytetrafluoroethylene (e-PTFE) membranes and the degree of membrane exposure could affect the clinical results of guided tissue regeneration (GTR). Seven patients (aged 42 to 58 years) presented with twenty intrabony defects that were treated by GTR + DFDBA (decalcified freeze-dried bone allograft). Gingival recession, probing depths, and clinical attachment levels were recorded before surgery and 10 months subsequently. Exposure of the membrane was recorded at the moment of removal. The membranes were

Clinical effectiveness of autologous platelet-rich plasma and Peptide-enhanced bone graft in the treatment of intrabony defects.

BACKGROUND: The goal of periodontal therapy has always been the regeneration of lost attachment apparatus. The use of platelet-rich plasma (PRP) plus anorganic bovine-derived matrix and peptide 15 (ABM/P-15) for periodontal regenerative therapy offers a potentially useful modality for treating periodontal osseous defects. However, it is unknown whether a combination of these materials enhances the outcome of periodontal regenerative therapy. Therefore, this study was carried out to compare the effectiveness of the two regenerative techniques (autologous PRP plus ABM/P-15 versus autologous PRP)

Ligand-Modulated Manganese Ferrite Nanozyme Hydrogel with Balanced Antioxidant Activity and Biocompatibility for Periodontitis Treatment.

Nanozymes hold significant promise for periodontitis treatment, owing to their high catalytic efficiency, multi-enzyme activity, robust stability, and low cost. However, the need for repeated administration and the complex oral environment present challenges in balancing biosafety and efficiency. Herein, we develop a ligand-modulated manganese ferrite nanozyme hydrogel (MFZ@PG) with balanced antioxidant activity and biocompatibility for periodontitis treatment through ZBP1/β-catenin signaling. MFZ@PG is synthesized by encapsulating zwitterionic dopamine sulfonate-modified manganese ferrite nan

From Bench to Bone: Translational Pathways of Nanomaterial-Based Therapies in Craniomaxillomandibulofacial and Cranial Vault Reconstruction.

Craniomaxillofacial and cranial vault reconstruction remain limited by donor-site morbidity, infection, inadequate vascularization, incomplete osseointegration, and poor adaptation of conventional grafts to complex patient-specific anatomy. Nanomaterial-based therapies offer a translationally attractive strategy. Nanoscale hydroxyapatite, calcium phosphates, mesoporous silica, metallic nanoparticles, bioactive glasses, polymeric carriers, injectable nanocomposite hydrogels, extracellular vesicle-inspired systems, and nanostructured coatings can act on several targets at once, including osteoge

Stabilization of Bone Morphogenetic Protein-2 at Physiological pH: Contrasting Roles of CHAPS and Arginine in Aggregation Inhibition.

Bone morphogenetic protein-2 (BMP-2) is a key osteoinductive growth factor employed clinically in spinal fusion and fracture repair where bone regeneration is insufficient. However, its therapeutic efficacy is limited by low solubility and aggregation at physiological pH. This study investigates BMP-2 aggregation and identifies additives that stabilize its native, biologically active dimer form under physiological conditions. We demonstrate that 3-[(3-cholamidopropyl) dimethylammonio]-1-propanesulfonate (CHAPS) efficiently refolded monomeric BMP-2 from inclusion bodies into dimeric form but fa

Millimeter-Scale Dual-Opposing RNA-Gradient Hydrogel for Interfacial Gene Silencing.

Tissue interface restoration poses significant challenges in tissue engineering, particularly in areas requiring gradients of cellularity, biochemical composition, and mechanical properties essential for tissue-specific functions. Recent advancements in microfluidic technology have enabled the creation of hydrogels with spatially defined gradients of biological molecules for engineering gradient tissues that mimic the natural heterogeneity of the native extracellular matrix. However, these gradients are typically outside relevant millimeter length scales for biological interfaces. To address t

Synergistic Antibacterial and Osteogenic Activities of Cu/Sr-Empowered 13-93 Bioglass in PMMA Composites for Sequential Promotion of Infectious Osseointegration.

Poly(methyl methacrylate) (PMMA) is among the most prevalent antibiotic delivery systems for the prevention and treatment of bone infections. Nevertheless, its restricted antibiotic release efficacy generally does not eradicate pathogens at localized infection sites, potentially resulting in bacterial resistance, and it has inadequate capacity to facilitate osseointegration. This study involved the synthesis of a copper (Cu)/strontium (Sr)-doped 13-93 bioactive glass, which was subsequently combined with a PMMA matrix to produce a 13-93 Cu/Sr-doped cement (13-93CuSr/PC). The Cu and Sr in 13-93

Bridging skeletal stem cell diversity and human skeletal modeling.

Skeletal stem cells (SSCs) underlie skeletal development, homeostasis, regeneration, and aging, yet their identities and functions are highly heterogeneous across anatomical sites and life stages. Mouse genetic studies have identified multiple SSC populations-each residing in distinct niches such as the growth plate, periosteum, and bone marrow-and revealed their dynamic regulation across developmental, homeostatic, regenerative, and aging contexts. However, translating these insights to humans remains challenging due to species differences and limited access to physiological human skeletal ti

Rebooting blood vessel repair: implications of the SEMA-VR CardioLink-15 trial.

PURPOSE OF REVIEW: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) reduce major adverse cardiovascular events (MACE), heart failure, and mortality through undetermined mechanisms independent of glycemic control. This review examines emerging evidence that GLP-1RAs overcome vascular regenerative cell exhaustion (VRCE), a pathological depletion of bone marrow-derived progenitor cells that mediate vessel repair. RECENT FINDINGS: VRCE, the progressive loss of circulating progenitor cells that mediate vessel regeneration, has recently emerged as an underappreciated driver of MACE risk in indi

Selection of a novel MSC-targeted aptamer Pt-1 and its functionalized hydrogel for tissue regeneration.

Mesenchymal stem cell (MSC)-based therapies present substantial opportunities for regenerative medicine, yet the absence of lineage-specific tools for precise cell recruitment constitutes a primary limitation. In this study, we developed a novel ssDNA aptamer, designated as Pt-1, specifically evolved against human dental pulp stem cells (hDPSCs) using Cell-SELEX technology. Pt-1 exhibited high target specificity, strong binding affinity, and favorable physiological stability. Notably, Pt-1 identified a distinct subpopulation within heterogeneous DPSCs; the Pt-1high subset displayed significant

Localized Delivery of Mesenchymal Stem Cell Spheroids via an Injectable Hydrogel for Rheumatoid Arthritis Therapy.

Rheumatoid arthritis (RA) is an autoimmune disorder marked by chronic joint inflammation and tissue damage. While current systemic therapies mainly reduce inflammation, they often lack regenerative capacity and cause off-target effects. To address this, we developed an injectable collagen-hyaluronic acid (Col-HA-NHS) hydrogel for intra-articular delivery of umbilical mesenchymal stem cell (UMSC) spheroids. The hydrogel demonstrated excellent shear-thinning and regelable properties, biocompatibility, and controlled degradation. Encapsulated UMSC spheroids (S-UMSCs) exhibited enhanced paracrine

Multifunctional injectable hydrogel with Zn-metformin coordination for synergistic anti-infection, antioxidant, and osteogenic therapy in periodontitis.

Periodontitis, a chronic inflammatory disease induced by pathogenic microorganisms. Effective management requires the elimination of plaque biofilms, the suppression of oxidative stress, and the regeneration of periodontal bone. Nevertheless, current treatments are often constrained by inadequate debridement of complex periodontal pockets, potential adverse effects, and emerging antibiotic resistance. To address these deficiencies, this work develops an injectable protocatechuic acid-grafted carboxymethyl chitosan hydrogel incorporating Zn-Metformin coordination (PgC@ZM). This hydrogel targets

Research progress and prospects of exosomes from diverse cellular sources in the treatment of knee osteoarthritis: a narrative review.

Knee osteoarthritis (KOA) is a highly prevalent degenerative joint disease characterized by osteophyte formation at joint margins, subchondral bone sclerosis, and progressive degeneration of the articular cartilage. In advanced stages, it can result in severe functional impairment of the knee joint, imposing a substantial burden on patients' quality of life and on healthcare systems. Conventional intra-articular treatments, such as hyaluronic acid and corticosteroid injections, provide temporary pain relief but fail to achieve true tissue repair or regeneration. In recent years, several novel

Targeting bone marrow mesenchymal stem cells with traditional Chinese medicine active ingredients: Restoring hematopoietic microenvironment homeostasis to prevent and treat myelosuppression.

Myelosuppression is a common and dose-limiting severe complication in tumor radiotherapy and chemotherapy. The disruption of hematopoietic microenvironment homeostasis mediated by bone marrow mesenchymal stem cells (BMSCs) may be one of the important pathological factors for its persistent non-healing. Traditional Chinese medicine (TCM) can regulate BMSCs function through multi-target and multi-pathway mechanisms, reshape the hematopoietic microenvironment, and support hematopoietic regeneration. This article systematically explains the network mechanisms by which BMSCs regulate hematopoiesis

Integrin Signaling Imbalance in Periodontitis: A Stage-Dependent Link Between Inflammation, Bone Resorption and Regenerative Failure.

Periodontitis is a chronic inflammatory disease driven largely by dysregulated host responses that lead to destruction of periodontal tissues. Integrins are heterodimeric transmembrane receptors that regulate cell adhesion and bidirectional signaling in epithelial cells, immune cells, periodontal ligament fibroblasts, and osteoclasts. During disease progression, integrin-related responses may shift across overlapping molecular phases. Epithelial integrins such as α3β1 and α6β4 support barrier integrity, whereas α5β1 may facilitate microbial interaction and inflammatory signaling. β2 integrins

Low DYNLL1 Resists Porphyromonas gingivalis-Induced Epithelial Jamming-Like State.

Incomplete regeneration of the junctional epithelium (JE) following periodontal therapy is associated with persistent alveolar bone resorption and disease recurrence. While pathogen clearance is essential, the host-intrinsic mechanisms dictating epithelial recovery remain obscure. Here, we reveal that Porphyromonas gingivalis challenge induces an aberrant jamming-like phase transition in the epithelium during the early recovery window, mechanically restricting the epithelial remodeling required for proper JE reconstruction. Through a genome-wide CRISPR-Cas9 screen, we identified Dynein Light C
