Molecular and cellular aspects of age-related cognitive decline and Alzheimer's disease.

As the population of people aged 60 or older continues to rise, it has become increasingly important to understand the molecular basis underlying age-related cognitive decline. In fact, a better understanding of aging biology will help us identify ways to maintain high levels of cognitive functioning throughout the aging process. Many cellular and molecular aspects of brain aging are shared with other organ systems; however, certain age-related changes are unique to the nervous system due to its structural, cellular and molecular complexity. Importantly, the brain appears to show differential

Macroscopic Neural Oscillation during Skilled Reaching Movements in Humans.

The neural mechanism of skilled movements, such as reaching, has been considered to differ from that of rhythmic movement such as locomotion. It is generally thought that skilled movements are consciously controlled by the brain, while rhythmic movements are usually controlled autonomously by the spinal cord and brain stem. However, several studies in recent decades have suggested that neural networks in the spinal cord may also be involved in the generation of skilled movements. Moreover, a recent study revealed that neural activities in the motor cortex exhibit rhythmic oscillations correspo

Immature teratoma of the ovary with distant metastases: favorable prognosis and insights into chemotherapeutic retroconversion.

We present a case of a young woman with an immature teratoma of the right ovary that showed systemic metastases. The patient initially experienced abdominal distention at the age of 15 years. Radiographic assessment revealed a right ovarian tumor; thus, right salpingo-oophorectomy was performed, and the resected ovarian tumor showed a multilocular cystic lesion with partially solid areas. Pathologic diagnosis was an immature teratoma, grade 2. As brain, lung, and liver metastases were discovered within 2 years after the operation, sequential resections of the metastatic foci were performed bef

Delayed transplantation of human neural precursor cells improves outcome from focal cerebral ischemia in aged rats.

Neural precursor cell (NPC) transplantation may have a role in restoring brain function after stroke, but how aging might affect the brain's receptivity to such transplants is unknown. We reported previously that transplantation of human embryonic stem cell (hESC)-derived NPCs together with biomaterial (Matrigel) scaffolding into the brains of young adult Sprague-Dawley rats 3 weeks after distal middle cerebral artery occlusion (MCAO) reduced infarct volume and improved neurobehavioral performance. In this study, we compared the effect of NPC and Matrigel transplants in young adult (3-month-ol

Widespread decrease of type 1 cannabinoid receptor availability in Huntington disease in vivo.

UNLABELLED: The type 1 cannabinoid receptor (CB1) is a crucial modulator of synaptic transmission in the brain. Animal and postmortem human data suggest that mutant huntingtin represses CB1 transcription. Our aim was to measure CB1 levels in the brains of Huntington disease (HD) patients in vivo. METHODS: Twenty symptomatic HD patients and 14 healthy controls underwent PET with the novel CB1 ligand N-[2-(3-cyano-phenyl)-3-(4-(2-(18)F-fluorethoxy)phenyl)-1-methylpropyl]-2-(5-methyl-2-pyridyloxy)-2-methylproponamide. RESULTS: We observed a profound decrease of CB1 availability throughout the g

[Neurogenesis in the pathologies of the nervous system].

Neural stem cells (NSCs) are the self-renewing, multipotent cells that generate neurons, astrocytes, and oligodendrocytes in the nervous system. Contrary to the long-held dogma, neurogenesis occurs in discrete areas of the adult brain, the hippocampus and the subventricular zone, and NSCs reside in the adult central nervous system. Recent studies have shown that neurogenesis is increased in the diseased brains, after strokes and traumatic brain injuries, and that new neuronal cells are generated at the sites of injury, where they replace some of the degenerated nerve cells. Thus, the central n

[Complications of the early postoperative period in craniocerebral injuries and ways of preventing them].

The author analysed the postoperative complications and the errors in their recognition as well as neglects and omissions of the postoperative management of 49 of 755 patients subjected to operation for a craniocerebral injury. In 6.5% of cases, the postoperative period was characterized by a complicated course. Inspection of the wound in 40 patients and brain autopsy in 9 patients who had died without undergoing a repeated intervention revealed large hemorrhage in 29 (57%) and edema of the brain in 20 (43%) cases. In 7 patients, the condition deteriorated due to dislocation of the median-stem

Astrocyte Enrichment of 3D Cortical Constructs Enhances Brain Repair.

Regenerative medicine offers a promising approach to treat brain injuries, yet challenges persist in promoting neuronal survival and integration. Recent studies demonstrate that human cells implanted into rodent brains can exhibit plasticity, integrate into neural circuits and alleviate functional deficits. However, integration is often poor, with inadequate vascularization, and insufficient support cells such as astrocytes. Astrocytes play a crucial role in neuronal development and recovery by releasing growth factors, facilitating synaptogenesis, and promoting blood vessel formation. This st

Human brainstem activation underpinning offset analgesia changes in perceived pain intensity. An ultra-high field functional magnetic resonance imaging investigation.

Offset analgesia is a pain modulatory phenomenon whereby a slight decrease in the intensity of a painful stimulus results in a large, disproportionate reduction in perceived pain. While human brain imaging studies have begun to map the brain circuitry underpinning offset analgesia, the brainstem circuits responsible for this phenomenon remain unknown. Furthermore, it is now recognised that previously described analgesic paradigms can evoke pain increases in some individuals, but whether the same brainstem circuitry is responsible for inhibitory and paradoxical facilitatory responses remains to

Atypical Molecular Features of Pediatric Tectal Glioma: A Single Institutional Series.

We present 4 children (diagnosed between 1 and 8 y, 3 females and 1 male) with molecularly distinct tectal gliomas (2 KRAS mutant, 1 EGFR mutant, 1 SRGAP3-RAF-1 fusion) that contributes to the growing literature of this uncommonly biopsied tumor. The patient with EGFR R222C mutation had a more severe course, earlier diagnosis, subsequent leptomeningeal metastatic disease, required more aggressive therapies, and died 9 years after diagnosis. Patients with KRAS mutations and SRGAP3-RAF-1 fusion had a more indolent course. Our series expands the molecular phenotype of tectal glioma with the poten

"Focused Ultrasound-mediated Drug Delivery in Humans - a Path Towards Translation in Neurodegenerative Diseases".

The blood-brain barrier (BBB) has a major protective function in preventing the entry of harmful molecules into the brain, but is simultaneously limiting the delivery of drugs, restricting their potential clinical application in neurodegenerative diseases. Recent preclinical evidence demonstrates that following application of focused ultrasound with microbubbles (FUS+MB), the BBB becomes reversibly accessible to compounds that normally are brain-impermeable, suggesting FUS+MB as a promising new platform for delivery of therapeutic agents into the central nervous system. As a step towards trans

Glycosaminoglycan scaffolding and neural progenitor cell transplantation promotes regenerative immunomodulation in the mouse ischemic brain.

Ischemic stroke is a leading cause of morbidity and mortality, with limited treatments that can facilitate brain regeneration. Neural progenitor cells (NPCs) hold promise for replacing tissue lost to stroke, and biomaterial approaches may improve their efficacy to overcome hurdles in clinical translation. The immune response and its role in stroke pathogenesis and regeneration may interplay with critical mechanisms of stem cell and biomaterial therapies. Cellular therapy can modulate the immune response to reduce toxic neuroinflammation early after ischemia. However, few studies have attempted

Imaging Patterns Characterizing Mitochondrial Leukodystrophies.

BACKGROUND AND PURPOSE: Achieving a specific diagnosis in leukodystrophies is often difficult due to clinical and genetic heterogeneity. Mitochondrial defects cause 5%-10% of leukodystrophies. Our objective was to define MR imaging features commonly shared by mitochondrial leukodystrophies and to distinguish MR imaging patterns related to specific genetic defects. MATERIALS AND METHODS: One hundred thirty-two patients with a mitochondrial leukodystrophy with known genetic defects were identified in the data base of the Amsterdam Leukodystrophy Center. Numerous anatomic structures were systema

The Paramedian Supracerebellar Approach: A Less Disruptive and More Flexible Operative Corridor to the Pineal and Posterior Upper Brainstem Regions.

The paramedian supracerebellar craniotomy is an underrecognized route to the midline and paramedian regions of the upper posterior brainstem. As compared with its midline supracerebellar counterpart, this less disruptive approach preserves the majority of the midline bridging veins, requires less cerebellar retraction, and is significantly more efficient. In this offering, I will emphasize the realities of this flexible route and its remarkable advantages in reaching deep-seated lesions.

NADH-linked mitochondrial respiration in the developing mouse brain is sex-, age- and tissue-dependent.

Mitochondria play a major role in the brain. Apart from energy production, mitochondria regulate key factors in the activation of cell signaling pathways such as survival, proliferation, and differentiation. While all these processes occur during the physiological development of the brain, it is surprising that the mitochondrial functions and functioning in the brain during the postnatal development remain poorly explored. In this work, we collected samples of brainstem and cortex of mice at postnatal ages 3 (P3), 21 (P21), and at adulthood (3 months old) and evaluated the mitochondrial oxygen

Neural mechanisms of single corrective steps evoked in the standing rabbit.

Single steps in different directions are often used for postural corrections. However, our knowledge about the neural mechanisms underlying their generation is scarce. This study was aimed to characterize the corrective steps generated in response to disturbances of the basic body configuration caused by forward, backward or outward displacement of the hindlimb, as well as to reveal location in the CNS of the corrective step generating mechanisms. Video recording of the motor response to translation of the supporting surface under the hindlimb along with contact forces and activity of back and

Patient 1: A 35-year-old woman became deep coma because of intracranial hemorrhage after pulmonary surgery. Patient 2: A 39-year-old woman became deep coma because of cerebellar hemorrhage after hepatic surgery. Scalp-recorded digital electroencephalography (EEG) showed electrocerebral inactivity in both cases. In addition, both EEG showed repetitive discharges at bilateral frontopolar electrodes in response to photic stimuli. The amplitude and latency of the discharges was 17 μV and 24 msec in case 1, and 9 μV and 27 msec in case 2 respectively. The activity at left frontopolar electrode disa
