Genetic and metabolomic pathways linking frailty, chronic kidney disease, and kidney failure: a Mendelian randomization and multi-omics analysis.

Global population aging highlights frailty as a critical health concern, yet its genetic basis remains poorly understood. We employed Mendelian randomization (MR) and National Health and Nutrition Examination Survey (NHANES) to examine causal relationships between frailty index (FI) and 36 urological diseases, investigating shared genetic mechanisms. The FI was calculated using genome-wide association data, and bidirectional MR analysis with multiple statistical methods was applied to investigate causal links between FI and 36 urological diseases. Complementary observational analyses using NHA

The role of inflammatory cytokines and blood metabolites in osteoporosis: A Mendelian randomization study on pathways and therapeutic targets.

The precise mechanisms underlying the development of osteoporosis (OP) remain unclear, but evidence suggests that inflammatory cytokines and blood metabolites play a significant role. We aim to investigate, as a strictly genomic study based on genome-wide association studies (GWAS) summary statistics, the causal relationships between inflammatory cytokines, blood metabolites, and OP by using Mendelian randomization (MR) analysis and uncover potential therapeutic targets. We utilized inflammatory cytokines from a GWAS summary containing 8293 healthy participants, 1400 blood metabolites from GWA

No genetic association between iron deficiency anemia and ischemic stroke and its subtypes: a bidirectional two-sample Mendelian randomization study.

BACKGROUND: Observational researches have suggested a connection between iron deficiency anemia (IDA) and an increased likelihood of ischemic stroke (IS), yet establishing causality is challenging owing to the inherent limitations of such studies, including their vulnerability to confounding factors and the potential for reverse causation. This study employs a bidirectional two-sample Mendelian randomization (MR) approach to assess the causal linkage between IDA and IS and its subtypes. METHODS: Identifiable single nucleotide polymorphisms (SNPs) with significant links to either IDA or IS and

Association between blood mitochondrial DNA copy number and mental disorders: A bidirectional two-sample mendelian randomization study.

BACKGROUND: Mitochondria is essential for cellular energy production, oxidative stress, and apoptosis. Mitochondrial DNA (mtDNA) encodes essential proteins for mitochondrial function. Although several studies have explored the association between changes in mtDNA copy number (mtDNA-CN) and risk of mental disorders, the results remain debated. This study used a bidirectional two-sample Mendelian randomization (MR) analysis to examine the genetic causality between mtDNA-CN and mental disorders. METHODS: Genome-wide association study (GWAS) data for mtDNA-CN were sourced from UK biobank, involvi
