Region-aware bridge modeling enables interpretable mesoscale representation of spatial transcriptomic tissue sections.

MOTIVATION: Spatial transcriptomics maps tissue architecture at high resolution, but spot-level maps are difficult to compare across sections, whereas whole-section averages obscure regional organization. We developed region-aware bridge modeling to aggregate interpretable biological program features into compact mesoscale summaries. RESULTS: Using public colorectal cancer and breast cancer 10x Genomics high-definition Visium sections, we constructed epithelial-like, fibroblast, smooth or myoepithelial, and extracellular matrix bridge features from cell-state indicators, curated gene-program

Fusion Strategy Toward Highly Crystalline Non-Fullerene Acceptor Enables Simultaneously Enhanced Current and Voltage in Organic Solar Cells.

Organic solar cells (OSCs) offer unique advantages, yet their photovoltaic performance is limited by the challenge of optimizing molecular packing in photoactive materials. While A-D-A'-D-A-type acceptors have improved efficiency by forming favorable 3D network packing, strategies to further enhance crystallinity without causing detrimental over-aggregation are scarce. Herein, we report a distinctive non-fullerene acceptor (NFA) congener, BrTh-2Cl, via a molecular fusion strategy. This design ingeniously integrates a highly crystalline brominated-thiophene unit with a chlorinated phenyl group

Exploring viscosity sensitivity of π-extended coumarin fluorogen-based PRPGs for precise photorelease of valproic acid: detection and defibrillation of TDP-43 aggregation.

Alteration of cellular microenvironment viscosity by protein aggregation plays a crucial role as a biophysical parameter that reflects abnormal cellular behaviour, leading to neurodegenerative diseases such as Parkinson's disease, Alzheimer's disease, amyotrophic lateral sclerosis (ALS), frontotemporal lobar degeneration (FTLD), etc. Herein, we report the design and development of a series of coumarin fluorogen-based photoremovable protecting groups (PRPGs, 5a-d) with variations in substitutions tethered with a π-extended linker that integrate viscosity sensing with visible-light-triggered rel

Size of Biomolecular Condensates Dictates Fate in Liquid-Solid Phase Transitions through Amorphous-Amyloid Competition.

Proteins function not only through intramolecular folding and intermolecular complex formation but also through phase transitions driven by intermolecular interactions. Such phases, including liquid-like condensates, amorphous aggregates (AAs), and amyloid fibrils, are linked to distinct biological functions and pathologies. Although the transition from liquid-like condensates to amyloids has been extensively studied, the kinetic relationships between amyloids and other metastable solid states under cell-sized confinement remain unclear, which may hinder the establishment of effective therapeu

Evaluation of Red Blood Cell Biomechanics in the Setting of Cancer-Associated Anemia and Chemotherapy.

INTRODUCTION: Red blood cells (RBCs) possess distinct biomechanical properties that enable their survival and efficient oxygen delivery. Cancer-associated anemia, frequently compounded by chemotherapy, is a major clinical challenge, yet little is known about how RBC biomechanics contribute to its pathophysiology. This study evaluates the biomechanical properties of RBCs in patients with cancer compared to controls and within patients before and after chemotherapy. METHODS: Biomechanical properties of RBCs were assessed in 110 women with breast, ovarian, or endometrial cancer, measured before

A microglia membrane biomimetic platinum-based MOF-loaded quercetin nanodrug delivery system for the treatment of Alzheimer's disease.

The aberrant deposition of β-amyloid (Aβ) is a central pathological hallmark of Alzheimer's disease (AD), triggering oxidative stress, metal ion dyshomeostasis, and excessive microglial activation in a self-perpetuating pathological cascade. To address these interconnected processes, a platinum-based metal-organic framework (Pt-MOF) with intrinsic antioxidant enzyme-mimetic activity was constructed and loaded with quercetin (Qu) to regulate microglial dysfunction. To enhance blood-brain barrier (BBB) penetration and inflammation-targeting capability, Pt-MOF/Qu was further camouflaged with micr

Taming Crystallization Kinetics via Side-Chain Steric Engineering Enables Over 21% Efficiency in Non-Halogenated-Solvent-Processed Organic Solar Cells.

The development of high-performance, non-halogenated organic solar cells (OSCs) is essential for sustainable commercialization. However, the high boiling points of halogen-free solvents often trigger excessive pre-aggregation of non-fullerene acceptors (NFAs) and unfavorable phase separation, severely limiting device performance. Herein, a side-chain steric engineering strategy is employed to develop a weakly crystalline NFA, TPD-Y, featuring a thieno[3,4-c]pyrrole-4,6-dione unit. The reduced crystallinity of TPD-Y effectively suppresses aggregation-caused quenching (ACQ), yielding a high phot

Reshaping the immune landscape: next-generation microglia-targeted therapies for Alzheimer's disease.

BACKGROUND: Alzheimer's disease (AD) is a significant global health challenge characterized as a multifactorial neurodegenerative disorder, involving amyloid-β (Aβ) and Tau aggregation, neuroinflammation and progressive neuronal injury. While Amyloid-targeted therapies have achieved a breakthrough in prevention of Aβ aggregation, the strategies face notable limitations in achieving curative outcomes and management of amyloid-independent central nervous system (CNS) dysfunction. Consequently, targeting microglia, the central immune cells of the brain, has emerged as a promising strategy to enha

A Comprehensive Review on Compounds and Herbal Approaches against Amyloid Beta Aggregation in Alzheimer's Disease.

Alzheimer's disease is a neurodegenerative disorder that affects brain cells over time, progressively reducing cognitive function, interfering with memory retention, and making daily tasks more difficult. The development of amyloid-beta (Aβ) plaques plays an important role in the progression of the disease, contributing to neurological swelling and cognitive impairment. Aβ induces oxidative damage and produces anti-inflammatory cytokines while reducing the association with vascular RAGE and acetylcholine. This article aims to present the latest and relevant research on treatment approaches tar

Symmetry-to-Asymmetry-Engineered Aggregation-Induced Second Near-Infrared Emission Nanoplatform for Nitric Oxide-Enhanced Phototheranostics of Fungal Biofilms.

Fungal biofilm infections pose a serious and growing threat to human health. To address this challenge, we developed a multifunctional nanoplatform (ASE@B NPs) that integrates a second near-infrared (NIR-II) photothermal agent, biofilm-targeting anti-microbial function, and photothermally triggered nitric oxide (NO) release. This system enables precise targeting of the infected microenvironment through a carrier containing acid-responsive segments, thereby achieving combined anti-fungal and anti-inflammatory effects against fungal biofilms. A key to this advance was the enhancement of photothe

Steric hindrance engineering on Iridium(III) photosensitizers: unlocking aggregation-induced emission for enhanced photodynamic therapy.

Ir(III) complexes have gained significant attention as photosensitizers (PSs) in fluorescence imaging-guided photodynamic therapy (PDT) due to their tunable structures and rich photophysical properties. However, the development of Ir(III)-based PSs that possess both strong emission and high reactive oxygen species (ROS) generation capabilities in the biological microenvironment still remains challenges. Herein, we propose a rational molecular design strategy to concurrently boost both photoluminescence and ROS generation in Ir(III)-based PSs through steric hindrance engineering. The introducti

Coronin1A Regulates the Trafficking of Alpha Synuclein in Microglia.

Microglia respond to cytotoxic protein aggregates associated with the progression of neurodegenerative disease. Pathological protein aggregates activate the microglial NLRP3 inflammasome resulting in proinflammatory signaling, secretion, and potentially pyroptotic cell death. We characterized mixed sex primary mouse microglia exposed to microbial stressors and alpha synuclein preformed fibrils (αsyn PFFs) to identify cellular mechanisms related to Parkinson's disease. Microglia package and release the endosome fate regulator Coronin1A (Coro1A) in EVs in an Nlrp3-dependent manner in widely used

Isomer-driven supramolecular polymorphism of 4-hydroxytamoxifen modulating nanostructure-dependent cellular uptake and cytotoxicity.

Geometric isomerism in small-molecule drugs has traditionally been studied in the context of receptor binding or metabolic stability. However, its influence on supramolecular behavior and downstream biological function remains underexplored. Here, we investigate how the (Z)- and (E)-isomers of 4-hydroxytamoxifen (4-OHT) self-assemble in aqueous environments and how this supramolecular divergence impacts anticancer activity. The (Z)-isomer forms small, amorphous nanoflake assemblies that display enhanced colloidal dispersibility and efficient cellular uptake, while the (E)-isomer assembles into

Investigations of 3-Hydroxy Chromone Derivatives as Multipotent Therapeutics for the Treatment of Alzheimer's Disease: In Silico and In Vitro Interventions and Fluorescence Studies.

Chromone-based small organic molecules are designed and synthesized as putative multipotent ligands to intervene in several interlinked pathological pathways of Alzheimer's disease. The synthesized compounds were evaluated as acetylcholinesterase, monoamine oxidase, and amyloid β aggregation inhibitors using biochemical assays. Most of the compounds were found to inhibit the enzymes in a lower micromolar concentration range. In the series, two compounds, i.e., NSS-16 and NSS-18, displayed a balanced activity profile with the IC50 values of 1.77 and 1.53 μM against AChE and 2.06 and 1.51 μM aga

Isomerism Effect of 3D Dimeric Acceptors for Non-Halogenated Solvent-Processed Organic Solar Cells with 20 % Efficiency.

Organic photovoltaic materials that can be processed via non-halogenated solvents are crucial for the large-area manufacturing of organic solar cells (OSCs). However, the limited available of electron acceptors with adequate solubility and favorable molecular packing presents a challenge in achieving efficient non-halogenated solvent-processed OSCs. Herein, inspired by the three-dimensional dimeric acceptor CH8-4, we employed a molecular isomerization strategy to synthesize its isomers, CH8-4A and CH8-4B, by tuning the position of fluorine (F) atom in the central unit. The differing intramolec

Platelet aggregation response in children with infantile haemangioma under propranolol treatment.

Infantile haemangioma is characterized by the proliferation of immature endothelial cells. A small portion requires treatment. Beta blockers are a good treatment option; however, it has been shown that beta blockers cause a decrease in the aggregation response of platelets. Twenty healthy children and 20 children with a diagnosis of infantile haemangioma who were started on propranolol treatment were included in the study. All patients were given propranolol 1 mg/kg/day in two doses. Haemangioma size, appearance features, blood count parameters and percentage of the maximal amplitude of platel

Preparation of highly-dispersed iron pyrophosphate nanoparticles via ultrasound-assistant carboxymethyl cellulose modification to enhance their liposome encapsulation and cell absorption.

Ferric pyrophosphate (FePP) is commonly used in fortified foods because of its subtle metallic odor/color. However, the insoluble nature of FePP restricts its intestinal absorption, thereby diminishing its efficacy. Liposomes can enhance the transmembrane absorption of encapsulated ingredients but encounter difficulties when encapsulating insoluble compounds. In this study, it was initially attempted to boost FePP solubility by reducing its particle size. Nevertheless, nanoscale FePP (N-FePP) exhibited a strong tendency to aggregate. To address this, an ultrasound-assisted carboxymethyl cellul

C9orf72 Dipeptide Repeat Proteinopathy Is Linked to Increased Histone H3 Phosphorylation on Serine 10.

Amyotrophic lateral sclerosis (ALS) and frontotemporal dementia (FTD) are fatal illnesses forming a neurodegenerative disease continuum. While most ALS/FTD cases are sporadic, a small proportion of cases are linked to mutations in many genes. Among these, hexanucleotide repeat expansions in the C9orf72 gene are the most common and lead to the formation of dipeptide repeat proteins (DPRs), including a proline-arginine dipeptide (PR), which aggregate in the cytoplasm of decaying neurons. As genetics alone fails to explain the etiology of ALS/FTD, it is possible that epigenetic mechanisms - such

Dual Targeting and Cascaded Aggregation of Luminogen for Washing-Free and Enhanced Fluorescence Imaging of Intracellular Bacteria.

The invasion and intracellular survival of Staphylococcus aureus (S. aureus) within phagocytic cells is a major cause of serious antibiotic resistance and recurrent infections in clinical settings, making the detection of intracellular S. aureus highly desirable prior to initiating effective treatment. In this study, we proposed a dual-targeting and cascaded aggregation-induced emission luminogen (AIEgen) probe for washing-free and enhanced fluorescence imaging of intracellular S. aureus. The AIEgen probes consist of an AIE molecule (TPE), cleavable peptide (YVADC(StBu)GKK), targeting peptide

Both single mutation and molecular conjugation on the mutant prevent calcitonin from forming amyloid fibrils.

Human calcitonin (hCT) is a peptide hormone made up of 32 amino acids, produced by the parafollicular cells of the thyroid gland. Its main role is to decrease blood calcium levels by inhibiting osteoclast activity and reducing calcium reabsorption in the kidneys and intestines. This characteristic positions hCT as a potential treatment for bone-related conditions, such as osteoporosis and Paget's disease. However, hCT has a strong tendency to form amyloid fibrils, which can lead to a loss of its biological function. This research aims to stabilize hCT's conformation through molecular conjugati
